    Learning by Playing: A Curriculum Utilizing Improvisational Theater to Develop Prosocial Behavior Skills and Self-efficacy in Young Adolescents

    Prosocial behavior requires competency in social-emotional skills including interpersonal skills, conflict resolution, and emotional regulation. During the developmental stage of early adolescence, youth are learning these skills and are more likely to engage in prosocial behavior if they feel efficacious in them. However, most social-emotional learning programs focus on conceptual knowledge in these areas without providing sufficient opportunities for practice, which does not allow students to develop sufficient procedural knowledge and self-efficacy. Improvisational theater offers a solution to this problem; used in concert with social-emotional lessons, it provides practice scenarios and procedural learning opportunities. This project consists of a curriculum built on these principles intended for use in a Montessori middle school environment. Its structure aligns with both Montessori secondary best practices and the fluid nature of interpersonal skills, allowing the teacher to adapt the curriculum to the needs of the class. It contains notes on implementation, lesson plans, and supplemental documents. In the discussion, students describe the perceived benefits of the program, including improved self-efficacy and a sense of community. Future directions include peer review, curriculum expansion, and research on efficacy

    Monitoring boreal forest biomass and carbon storage change by integrating airborne laser scanning, biometry and eddy covariance data

    AbstractThis study presents a comparison and integration of three methods commonly used to estimate the amount of forest ecosystem carbon (C) available for storage. In particular, we examine the representation of living above- and below-ground biomass change (net accumulation) using plot-level biometry and repeat airborne laser scanning (ALS) of three dimensional forest plot structure. These are compared with cumulative net CO2 fluxes (net ecosystem production, NEP) from eddy covariance (EC) over a six-year period within a jack pine chronosequence of four stands (~94, 30, 14 and 3years since establishment from 2005) located in central Saskatchewan, Canada. Combining the results of the two methods yield valuable observations on the partitioning of C within ecosystems. Subtracting total living biomass C accumulation from NEP results in a residual that represents change in soil and litter C storage. When plotted against time for the stands investigated, the curve produced is analogous to the soil C dynamics described in Covington (1981). Here, ALS biomass accumulation exceeds EC-based NEP measured in young stands, with the residual declining with age as stands regenerate and litter decomposition stabilizes. During the 50–70year age-period, NEP and live biomass accumulation come into balance, with the soil and litter pools of stands 70–100years post-disturbance becoming a net store of C. Biomass accumulation was greater in 2008–2011 compared to 2005–2008, with the smallest increase in the 94-year-old “old jack pine” stand and greatest in the 14-year-old “harvested jack pine 1994” stand, with values of 1.4 (±3.2) tCha−1 and 12.0 (±1.6) tCha−1, respectively. The efficiency with which CO2 was stored in accumulated biomass was lowest in the youngest and oldest stands, but peaked during rapid regeneration following harvest (14-year-old stand). The analysis highlights that the primary source of uncertainty in the data integration workflow is in the calculation of biomass expansion factors, and this aspect of the workflow needs to be implemented with caution to avoid large error propagations. We suggest that the adoption of integrated ALS, in situ and atmospheric flux monitoring frameworks is needed to improve spatio-temporal partitioning of C balance components at sub-decadal scale within rapidly changing forest ecosystems and for use in national carbon accounting programs

    Hypothesis-driven genome-wide association studies provide novel insights into genetics of reading disabilities

    Reading Disability (RD) is often characterized by difficulties in the phonology of the language. While the molecular mechanisms underlying it are largely undetermined, loci are being revealed by genome-wide association studies (GWAS). In a previous GWAS for word reading (Price, 2020), we observed that top single-nucleotide polymorphisms (SNPs) were located near to or in genes involved in neuronal migration/axon guidance (NM/AG) or loci implicated in autism spectrum disorder (ASD). A prominent theory of RD etiology posits that it involves disturbed neuronal migration, while potential links between RD-ASD have not been extensively investigated. To improve power to identify associated loci, we up-weighted variants involved in NM/AG or ASD, separately, and performed a new Hypothesis-Driven (HD)–GWAS. The approach was applied to a Toronto RD sample and a meta-analysis of the GenLang Consortium. For the Toronto sample (n = 624), no SNPs reached significance; however, by gene-set analysis, the joint contribution of ASD-related genes passed the threshold (p~1.45 × 10–2, threshold = 2.5 × 10–2). For the GenLang Cohort (n = 26,558), SNPs in DOCK7 and CDH4 showed significant association for the NM/AG hypothesis (sFDR q = 1.02 × 10–2). To make the GenLang dataset more similar to Toronto, we repeated the analysis restricting to samples selected for reading/language deficits (n = 4152). In this GenLang selected subset, we found significant association for a locus intergenic between BTG3-C21orf91 for both hypotheses (sFDR q < 9.00 × 10–4). This study contributes candidate loci to the genetics of word reading. Data also suggest that, although different variants may be involved, alleles implicated in ASD risk may be found in the same genes as those implicated in word reading. This finding is limited to the Toronto sample suggesting that ascertainment influences genetic associations

    Treatment of cyclic vomiting syndrome with co-enzyme Q10 and amitriptyline, a retrospective study

    <p>Abstract</p> <p>Background</p> <p>Cyclic vomiting syndrome (CVS), which is defined by recurrent stereotypical episodes of nausea and vomiting, is a relatively-common disabling condition that is associated with migraine headache and mitochondrial dysfunction. Co-enzyme Q10 (Co-Q) is a nutritional supplement that has demonstrated efficacy in pediatric and adult migraine. It is increasingly used in CVS despite the complete lack of studies to demonstrate its value in treatment</p> <p>Methods</p> <p>Using an Internet-based survey filled out by subjects with CVS or their parents, the efficacy, tolerability and subject satisfaction in CVS prophylaxis were queried. Subjects taking Co-Q (22 subjects) were compared against those taking amitriptyline (162 subjects), which is the general standard-of-care.</p> <p>Results</p> <p>Subjects/parents reported similar levels of efficacy for a variety of episode parameters (frequency, duration, number of emesis, nausea severity). There was a 50% reduction in at least one of those four parameters in 72% of subjects treated with amitriptyline and 68% of subjects treated Co-Q. However, while no side effects were reported on Co-Q, 50% of subjects on amitriptyline reported side effects (P = 5 × 10<sup>-7</sup>), resulting in 21% discontinuing treatment (P = 0.007). Subjects/parents considered the benefits to outweigh the risks of treatment in 47% of cases on amitriptyline and 77% of cases on Co-Q (P = 0.008).</p> <p>Conclusion</p> <p>Our data suggest that the natural food supplement Co-Q is potentially efficacious and tolerable in the treatment of CVS, and should be considered as an option in CVS prophylaxis. Our data would likely be helpful in the design of a double-blind clinical trial.</p

    Language and reading impairments are associated with increased prevalence of non-right handedness

No clear pattern has emerged, possibly because of small samples, publication bias, and heterogeneous criteria across studies. Non-right-handedness (NRH) frequency was assessed in N = 2503 cases with reading and/or language impairment and N = 4316 sex-matched controls identified from 10 distinct cohorts (age range 6–19 years old; European ethnicity) using a priori set criteria. A meta-analysis (Ncases = 1994) showed elevated NRH % in individuals with language/reading impairment compared with controls (OR = 1.21, CI = 1.06–1.39, p = .01). The association between reading/language impairments and NRH could result from shared pathways underlying brain lateralization, handedness, and cognitive functions.Publisher PDFPeer reviewe

    Expression and Function of Serotonin 2A and 2B Receptors in the Mammalian Respiratory Network

    Neurons of the respiratory network in the lower brainstem express a variety of serotonin receptors (5-HTRs) that act primarily through adenylyl cyclase. However, there is one receptor family including 5-HT2A, 5-HT2B, and 5-HT2C receptors that are directed towards protein kinase C (PKC). In contrast to 5-HT2ARs, expression and function of 5-HT2BRs within the respiratory network are still unclear. 5-HT2BR utilizes a Gq-mediated signaling cascade involving calcium and leading to activation of phospholipase C and IP3/DAG pathways. Based on previous studies, this signal pathway appears to mediate excitatory actions on respiration. In the present study, we analyzed receptor expression in pontine and medullary regions of the respiratory network both at the transcriptional and translational level using quantitative RT-PCR and self-made as well as commercially available antibodies, respectively. In addition we measured effects of selective agonists and antagonists for 5-HT2ARs and 5-HT2BRs given intra-arterially on phrenic nerve discharges in juvenile rats using the perfused brainstem preparation. The drugs caused significant changes in discharge activity. Co-administration of both agonists revealed a dominance of the 5-HT2BR. Given the nature of the signaling pathways, we investigated whether intracellular calcium may explain effects observed in the respiratory network. Taken together, the results of this study suggest a significant role of both receptors in respiratory network modulation

    Genome-wide analyses of individual differences in quantitatively assessed reading- and language-related skills in up to 34,000 people

    The use of spoken and written language is a fundamental human capacity. Individual differences in reading- and language-related skills are influenced by genetic variation, with twin-based heritability estimates of 30 to 80% depending on the trait. The genetic architecture is complex, heterogeneous, and multifactorial, but investigations of contributions of single-nucleotide polymorphisms (SNPs) were thus far underpowered. We present a multicohort genome-wide association study (GWAS) of five traits assessed individually using psychometric measures (word reading, nonword reading, spelling, phoneme awareness, and nonword repetition) in samples of 13,633 to 33,959 participants aged 5 to 26 y. We identified genome-wide significant association with word reading (rs11208009, P = 1.098 × 10−8) at a locus that has not been associated with intelligence or educational attainment. All five reading-/language-related traits showed robust SNP heritability, accounting for 13 to 26% of trait variability. Genomic structural equation modeling revealed a shared genetic factor explaining most of the variation in word/nonword reading, spelling, and phoneme awareness, which only partially overlapped with genetic variation contributing to nonword repetition, intelligence, and educational attainment. A multivariate GWAS of word/nonword reading, spelling, and phoneme awareness maximized power for follow-up investigation. Genetic correlation analysis with neuroimaging traits identified an association with the surface area of the banks of the left superior temporal sulcus, a brain region linked to the processing of spoken and written language. Heritability was enriched for genomic elements regulating gene expression in the fetal brain and in chromosomal regions that are depleted of Neanderthal variants. Together, these results provide avenues for deciphering the biological underpinnings of uniquely human traits
