    Predictive model for the emergence of Xylotrechus arvicola (Coleoptera: Cerambycidae) in La Rioja vineyards (Spain)

    Xylotrechus arvicola (Coleoptera, Cerambycidae) (Olivier) is a polyphagous borer which attacks different woody species in natural habitats in Spain. This coleopteran is also becoming a real impacting pest in several Spanish wine producing territories. Infested vines show adult exit holes, rachitic shoots, a higher incidence of wood fungal diseases and fragility. At the beginning of the 1990s, a heavy increase of infestation was observed in La Rioja vineyards, the only Qualified Guarantee of Origin Region (A.O.C.) in Spain. Due to the endophytic development of the larvae of X. arvicola, adults are the designated target for the integrated management. Therefore, the aim of the present paper was to study the flight period of the pest and to calculate the value of degree-days for the emergence of both sexes from data collected in the field during 2003-2008 in a Tempranillo cultivar in Tirgo (La Rioja). Adult emergences occurred from the end of May to mid-August and a certain degree of protandry was detected. The predictive models obtained for each year fit a sinusoidal curve in all cases, except for males in 2004, when the adjustment was polynomial. The obtained correlation coefficients were considerably high, as the value 0.99 was exceeded in 72 % of the cases. These results obtained in La Rioja vineyards suggest that control strategies aimed at the reduction of the adult population of X. arvicola in the view of the integrated management, should be performed between mid-June and the end of July.

    A Possible Third Body in the X-Ray System GRS 1747-312 and Models with Higher-Order Multiplicity

    GRS 1747-312 is a bright Low-Mass X-ray Binary in the globular cluster Terzan 6, located at a distance of 9.5 kpc from the Earth. It exhibits regular outbursts approximately every 4.5 months, during which periodic eclipses are known to occur. These eclipses have only been observed in the outburst phase, and are not clearly seen when the source is quiescent. Recent Chandra observations of the source were performed in June 2019 and April, June, and August of 2021. Two of these observations captured the source during its outburst, and showed clear flux decreases at the expected time of eclipse. The other two observations occurred when the source was quiescent. We present the discovery of a dip that occurred during the quiescent state. The dip is of longer duration and its time of occurrence does not fit the ephemeris of the shorter eclipses. We study the physical characteristics of the dip and determine that it has all the properties of an eclipse by an object with a well defined surface. We find that there are several possibilities for the nature of the object causing the 5.3 ks eclipse. First, GRS 1747-312 may be an X-ray triple, with an LMXB orbited by an outer third object, which could be an M-dwarf, brown dwarf, or planet. Second, there could be two LMXBs in close proximity to each other, likely bound together. Whatever the true nature of the eclipser, its presence suggests that the GRS 1747-312 system is unique.Comment: 38 pages, 30 figures, submitted to MNRA

    A cohort study of post-weaning multisystemic wasting syndrome and PCV2 in 178 pigs from birth to 14 weeks on a single farm in England

    Our hypothesis was that pigs that develop post-weaning multisystemic wasting syndrome (PMWS) are detectable from an early age with signs of weight loss and other clinical and serological abnormalities. Therefore, the objective of this study was to investigate the temporally varying and fixed events linked with the clinical incidence of PMWS by comparing affected and unaffected pigs in a cohort of 178 male piglets. Piglets were enrolled at birth and examined each week. Samples of blood were collected at regular intervals. The exposures measured were porcine circovirus type 2 (PCV2) antibody titres in all 178 and PCV2 antigen in a subset of 75 piglets. We also observed piglet health and measured their weight, and a post-mortem examination was performed by an external laboratory on all pigs between 6 and 14 weeks of age that died. From the cohort, 14 (8%) pigs died from PMWS and 4% from other causes. A further 37 pigs between 6 and 14 weeks of age died from PMWS (30) and ileitis and other causes (7). PMWS was only apparent in pigs from 1 to 2 weeks before death when they wasted rapidly. There were no other characteristic clinical signs and no obvious gross clinical lesions post-mortem. There was no strong link with PCV2 antibody throughout life but PCV2 antigen level was higher from 4 to 6 weeks of age in pigs that died from PMWS compared with pigs that died from other causes

    Shrinkage, liquid formation and mass losses of WC-19 vol% FeNiCoCr alloys during sintering have been inves- tigated in compositions either with coarse or submicron WC powders. Mass losses detected by thermogravimetry are compatible with carbothermal reduction of the different oxides present in the powder mixtures. Hardness and fracture toughness of materials based on submicron WC powders are within tolerances of those reported for WC- Co materials with similar microstructures.     EPAC-lung: pooled analysis of circulating tumour cells in advanced non-small cell lung cancer

    Introduction: We assessed the clinical validity of circulating tumour cell (CTC) quantification for prognostication of patients with advanced non-small cell lung cancer (NSCLC) by undertaking a pooled analysis of individual patient data. Methods: Nine European NSCLC CTC centres were asked to provide reported/unreported pseudo-anonymised data for patients with advanced NSCLC who participated in CellSearch CTC studies from January 2003 to March 2017. We used Cox regression models, stratified by centres, to establish the association between CTC count and survival. We assessed the added value of CTCs to prognostic clinicopathological models using likelihood ratio (LR) statistics and c-indices. Results: Seven out of nine eligible centres provided data for 550 patients with prognostic information for overall survival. CTC counts of ≥2 and ≥ 5 per 7·5 mL were associated with reduced progression-free survival (≥2 CTCs: hazard ratio [HR] = 1.72, p < 0·001; ≥5 CTCs: HR = 2.21, p < 0·001) and overall survival (≥2 CTCs: HR = 2·18, p < 0·001; ≥5 CTCs: HR = 2·75, p < 0·001), respectively. Survival prediction was significantly improved by addition of baseline CTC count to LR clinicopathological models (log-transformed CTCs p < 0·001; ≥2 CTCs p < 0·001; ≥5 CTCs p ≤ 0·001 for both survival end-points), whereas moderate improvements were observed with the use of c-index models. There was some evidence of between-centre heterogeneity, especially when examining continuous counts of CTCs. Conclusions: These data confirm CTCs as an independent prognostic indicator of progression-free survival and overall survival in advanced NSCLC and also reveal some evidence of between-centre heterogeneity. CTC count improves prognostication when added to full clinicopathological predictive models

    El presente trabajo se enmarca dentro de un proyecto de extensión que se desarrolla durante el 2012, acreditado y financiado por la UNLP, denominado “Identidad, territorio y salud. Construcción de identidades colectivas y transferencia institucional en torno al primer nivel de atención en el Barrio Federal de Ensenada”. Este barrio cuenta con una población aproximada de 480 habitantes y está ubicado en la zona de Gran La Plata, Pcia. de Buenos Aires. El objetivo del trabajo consiste en fortalecer la apropiación y promoción de la salud colectiva, entendida ésta como un proceso construido históricamente por cada sociedad, que se crea y recrea en todos los aspectos de la vida cotidiana de los sujetos. La propuesta metodológica se sustenta en un proceso dialógico entre los sujetos involucrados: vecinos de barrio Federal y los integrantes del equipo del trabajo extensionista. Desde este marco (pedagógico horizontal) desarrollamos actividades grupales de carácter lúdico-recreativo, que a primera vista no aparentan tener una relación con el proceso salud-enfermedad, propio del imaginario social, pero implícitamente, estarían abarcando y generando cambios en el significado asignado del mismo. Partimos de la idea de fortalecer el sentirse bien, no en el estricto sentido biológico, sino como aquella capacidad para crear momentos y actividades con pares en un mismo territorio y generando lazos entre vecinos y entre éstos y la institución de salud del barrio. De este modo, las actividades se desarrollan principalmente en la sala de espera de la unidad sanitaria y en la plaza circundante. Hemos definido tres líneas de trabajo: artístico, narrativo-plástico y de producción audiovisual y medioambiente. En los encuentros se trabaja con diferentes recursos, propios de disciplinas como música, plástica, teatro, danza, escultura, circo, artes audiovisuales, literatura, entre otros. El carácter interdisciplinario de este proyecto, conformado por estudiantes de las carreras de Trabajo Social y Antropología, un artista escultor y dirigido y coordinado por docentes y graduados de las Facultades de Ciencias Medicas, Humanidades y Ciencias de la Educación y Trabajo Social, enriquece las perspectivas de abordaje y fortalece la visión social, integral y colectiva de la salud.     Study protocol for the multicentre cohorts of Zika virus infection in pregnant women, infants, and acute clinical cases in Latin America and the Caribbean: the ZIKAlliance consortium.

    BACKGROUND: The European Commission (EC) Horizon 2020 (H2020)-funded ZIKAlliance Consortium designed a multicentre study including pregnant women (PW), children (CH) and natural history (NH) cohorts. Clinical sites were selected over a wide geographic range within Latin America and the Caribbean, taking into account the dynamic course of the ZIKV epidemic. METHODS: Recruitment to the PW cohort will take place in antenatal care clinics. PW will be enrolled regardless of symptoms and followed over the course of pregnancy, approximately every 4 weeks. PW will be revisited at delivery (or after miscarriage/abortion) to assess birth outcomes, including microcephaly and other congenital abnormalities according to the evolving definition of congenital Zika syndrome (CZS). After birth, children will be followed for 2 years in the CH cohort. Follow-up visits are scheduled at ages 1-3, 4-6, 12, and 24 months to assess neurocognitive and developmental milestones. In addition, a NH cohort for the characterization of symptomatic rash/fever illness was designed, including follow-up to capture persisting health problems. Blood, urine, and other biological materials will be collected, and tested for ZIKV and other relevant arboviral diseases (dengue, chikungunya, yellow fever) using RT-PCR or serological methods. A virtual, decentralized biobank will be created. Reciprocal clinical monitoring has been established between partner sites. Substudies of ZIKV seroprevalence, transmission clustering, disabilities and health economics, viral kinetics, the potential role of antibody enhancement, and co-infections will be linked to the cohort studies. DISCUSSION: Results of these large cohort studies will provide better risk estimates for birth defects and other developmental abnormalities associated with ZIKV infection including possible co-factors for the variability of risk estimates between other countries and regions. Additional outcomes include incidence and transmission estimates of ZIKV during and after pregnancy, characterization of short and long-term clinical course following infection and viral kinetics of ZIKV.     Characterization of Italian honeys (Marche Region) on the basis of their mineral content and some typical quality parameters

    <p>Abstract</p> <p>Background</p> <p>The characterization of three types of Marche (Italy) honeys (Acacia, Multifloral, Honeydew) was carried out on the basis of the their quality parameters (pH, sugar content, humidity) and mineral content (Na, K, Ca, Mg, Cu, Fe, and Mn). Pattern recognition methods such as principal components analysis (PCA) and linear discriminant analysis (LDA) were performed in order to classify honey samples whose botanical origins were different, and identify the most discriminant parameters. Lastly, using ANOVA and correlations for all parameters, significant differences between diverse types of honey were examined.</p> <p>Results</p> <p>Most of the samples' water content showed good maturity (98%) whilst pH values were in the range 3.50 – 4.21 confirming the good quality of the honeys analysed. Potassium was quantitatively the most relevant mineral (mean = 643 ppm), accounting for 79% of the total mineral content. The Ca, Na and Mg contents account for 14, 3 and 3% of the total mineral content respectively, while other minerals (Cu, Mn, Fe) were present at very low levels. PCA explained 75% or more of the variance with the first two PC variables. The variables with higher discrimination power according to the multivariate statistical procedure were Mg and pH. On the other hand, all samples of acacia and honeydew, and more than 90% of samples of multifloral type have been correctly classified using the LDA. ANOVA shows significant differences between diverse floral origins for all variables except sugar, moisture and Fe.</p> <p>Conclusion</p> <p>In general, the analytical results obtained for the Marche honeys indicate the products' high quality. The determination of physicochemical parameters and mineral content in combination with modern statistical techniques can be a useful tool for honey classification.</p

    Inadequate control of thyroid hormones sensitizes to hepatocarcinogenesis and unhealthy aging

    An inverse correlation between thyroid hormone levels and longevity has been reported in several species and reduced thyroid hormone levels have been proposed as a biomarker for healthy aging and metabolic fitness. However, hypothyroidism is a medical condition associated with compromised health and reduced life expectancy. Herein, we show, using wild-type and the Pax8 ablated model of hypothyroidism in mice, that hyperthyroidism and severe hypothyroidism are associated with an overall unhealthy status and shorter lifespan. Mild hypothyroid Pax8 +/- mice were heavier and displayed insulin resistance, hepatic steatosis and increased prevalence of liver cancer yet had normal lifespan. These pathophysiological conditions were precipitated by hepatic mitochondrial dysfunction and oxidative damage accumulation. These findings indicate that individuals carrying mutations on PAX8 may be susceptible to develop liver cancer and/or diabetes and raise concerns regarding the development of interventions aiming to modulate thyroid hormones to promote healthy aging or lifespan in mammals

    Final results from the PERUSE study of first-line pertuzumab plus trastuzumab plus a taxane for HER2-positive locally recurrent or metastatic breast cancer, with a multivariable approach to guide prognostication

    Background: The phase III CLinical Evaluation Of Pertuzumab And TRAstuzumab (CLEOPATRA) trial established the combination of pertuzumab, trastuzumab and docetaxel as standard first-line therapy for human epidermal growth factor receptor 2 (HER2)-positive locally recurrent/metastatic breast cancer (LR/mBC). The multicentre single-arm PERtUzumab global SafEty (PERUSE) study assessed the safety and efficacy of pertuzumab and trastuzumab combined with investigator-selected taxane in this setting. Patients and methods: Eligible patients with inoperable HER2-positive LR/mBC and no prior systemic therapy for LR/mBC (except endocrine therapy) received docetaxel, paclitaxel or nab-paclitaxel with trastuzumab and pertuzumab until disease progression or unacceptable toxicity. The primary endpoint was safety. Secondary endpoints included progression-free survival (PFS) and overall survival (OS). Prespecified subgroup analyses included subgroups according to taxane, hormone receptor (HR) status and prior trastuzumab. Exploratory univariable analyses identified potential prognostic factors; those that remained significant in multivariable analysis were used to analyse PFS and OS in subgroups with all, some or none of these factors. Results: Of 1436 treated patients, 588 (41%) initially received paclitaxel and 918 (64%) had HR-positive disease. The most common grade 653 adverse events were neutropenia (10%, mainly with docetaxel) and diarrhoea (8%). At the final analysis (median follow-up: 5.7 years), median PFS was 20.7 [95% confidence interval (CI) 18.9-23.1] months overall and was similar irrespective of HR status or taxane. Median OS was 65.3 (95% CI 60.9-70.9) months overall. OS was similar regardless of taxane backbone but was more favourable in patients with HR-positive than HR-negative LR/mBC. In exploratory analyses, trastuzumab-pretreated patients with visceral disease had the shortest median PFS (13.1 months) and OS (46.3 months). Conclusions: Mature results from PERUSE show a safety and efficacy profile consistent with results from CLEOPATRA and median OS exceeding 5 years. Results suggest that paclitaxel is a valid alternative to docetaxel as backbone chemotherapy. Exploratory analyses suggest risk factors that could guide future trial design
