    Control of embryonic Xenopus morphogenesis by a Ral-GDS/Xral branch of the Ras signalling pathway.

    Ras proteins mediate biological responses through various effectors and play a key role in relaying the Fibroblast Growth Factor (FGF) mesoderm induction signal during embryogenesis of the frog, Xenopus laevis. One Ras effector pathway involves the activation of the small G protein Ral. In the present study, we have investigated the role of key components in the Ral branch of FGF and Ras signalling during early Xenopus development. Treatment of animal caps with bFGF, which converts prospective ectoderm to mesoderm, activates Xral. The Ras mutant 12V37G, which can bind to Ral-GDS but not Raf, also activates Xral as well as causing developmental defects and cortical F-actin disassembly. A similar phenotype is induced by Ral-GDS itself. FGF-induced expression of several signature mesodermal genes, by contrast, is independent of Xral signalling. This and other data suggest that the RalB branch of Ras and FGF signalling regulates the actin cytoskeleton and morphogenesis in a transcriptionally independent manner. We also find Xral to be specifically activated in the marginal zone of Xenopus embryos, and find that disruption of the Ral pathway in this region prevents closure of the blastopore during gastrulation. We conclude that Ral signalling is autonomously required by mesodermal cells to effect essential morphogenetic changes during Xenopus gastrulation

    Size Dependent Transport of Floating Plastics Modeled in the Global Ocean

    Plastic has been detected in the ocean in most locations where scientists have looked for it. While ubiquitous in the environment, plastic pollution is heterogeneous, and plastics of varying composition, shape, and size accumulate differently in the global ocean. Many physical and biological processes influence the transport of plastics in the marine environment. Here we focus on physical processes and how they can naturally sort floating plastics at the ocean surface and within its interior. We introduce a new open-source GPU-accelerated numerical model, ADVECT, which simulates the three-dimensional dispersal of large arrays of modelled ocean plastics with varying size, shape, and density. We use this model to run a global simulation and find that buoyant particles are sorted in the ocean according to their size, both at the surface due to wind-driven drift and in the water column due to their rising velocity. Finally, we compare our findings with recent literature reporting the size distribution of plastics in the ocean and discuss which observations can and cannot be explained by the physical processes encoded in our model

    RLIP mediates downstream signalling from RalB to the actin cytoskeleton during Xenopus early development.

    The Ras protein activates at least three different pathways during early development. Two of them regulate mesodermal gene expression and the third is thought to participate in the control of actin cytoskeleton dynamics via the Ral protein. From a yeast two-hybrid screen of a Xenopus maternal cDNA library, we identified the Xenopus orthologue of the Ral interacting protein (RLIP, RIP1 or RalBP1), a putative effector of small G protein Ral. Previously we observed that a constitutively activated form of Ral GTPase (XralB G23V) induced bleaching of the animal hemisphere and disruption of the cortical actin cytoskeleton. To demonstrate that RLIP is the effector of RalB in early development, we show that the artificial targeting of RLIP to the membrane induces a similar phenotype to that of activated RalB. We show that overexpression of the Ral binding domain (RalBD) of XRLIP, which binds to the effector site of Ral, acts in competition with the endogenous effector of Ral and protects against the destructive effect of XralB G23V on the actin cytoskeleton. In contrast, the XRLIP has a synergistic effect on the activated form of XralB, which is dependent on the RalBD of RLIP. We provide evidence for the involvement of RLIP by way of its RalBD on the dynamics of the actin cytoskeleton and propose that signalling from Ral to RLIP is required for gastrulation

    Mimetic desire in autism spectrum disorder

    International audienceMimetic desire (MD), the spontaneous propensity to pursue goals that others pursue, is a case of social influence that is believed to shape preferences. Autism spectrum disorder (ASD) is defined by both atypical interests and altered social interaction. We investigated whether MD is lower in adults with ASD compared to typically developed adults and whether MD correlates with social anhedonia and social judgment, two aspects of atypical social functioning in autism. Contrary to our hypotheses, MD was similarly present in both ASD and control groups. Anhedonia and social judgment differed between the ASD and control groups but did not correlate with MD. These results extend previous findings by suggesting that basic mechanisms of social influence are preserved in autism. The finding of intact MD in ASD stands against the intuitive idea that atypical interests stem from reduced social influence and indirectly favors the possibility that special interests might be selected for their intrinsic properties

    Similar Comparative Low and High Doses of Deltamethrin and Acetamiprid Differently Impair the Retrieval of the Proboscis Extension Reflex in the Forager Honey Bee (Apis mellifera)

    In the present study, the effects of low (10 ng/bee) and high (100 ng/bee) doses of acetamiprid and deltamethrin insecticides on multi-trial learning and retrieval were evaluated in the honey bee Apis mellifera. After oral application, acetamiprid and deltamethrin at the concentrations used were not able to impair learning sessions. When the retention tests were performed 1 h, 6 h, and 24 h after learning, we found a significant difference between bees after learning sessions when drugs were applied 24 h before learning. Deltamethrin-treated bees were found to be more sensitive at 10 ng/bee and 100 ng/bee doses compared to acetamiprid-treated bees, only with amounts of 100 ng/bee and at 6 h and 24 h delays. When insecticides were applied during learning sessions, none of the tested insecticides was able to impair learning performance at 10 ng/bee or 100 ng/bee but retention performance was altered 24 h after learning sessions. Acetamiprid was the only one to impair retrieval at 10 ng/bee, whereas at 100 ng/bee an impairment of retrieval was found with both insecticides. The present results therefore suggest that acetamiprid and deltamethrin are able to impair retrieval performance in the honey bee Apis mellifera

    Imidacloprid and thiacloprid neonicotinoids bind more favourably to cockroach than to honeybee alpha6 nicotinic acetylcholine receptor: insights from computational studies

    The binding interactions of two neonicotinoids, imidacloprid (IMI) and thiacloprid (THI) with the extracellular domains of cockroach and honeybee alpha6 nicotinic acetylcholine receptor (nAChR) subunits in an homomeric receptor have been studied through docking and molecular dynamics (MD) simulations. The binding mode predicted for the two neonicotinoids is validated through the good agreement observed between the theoretical results with the crystal structures of the corresponding complexes with Ac-AChBP, the recognized structural surrogate for insects nAChR extracellular ligand binding domain. The binding site of the two insect alpha6 receptors differs by only one residue of loop D, a serine residue (Ser83) in cockroach being replaced by a lysine residue (Lys108) in honeybee. The docking results show very close interactions for the two neonicotinoids with both alpha6 nAChR models, in correspondence to the trends observed in the experimental neonicotinoid-Ac-AChBP complexes. However, the docking parameters (scores and energies) are not significantly different between the two insect alpha6 nAChRs to draw clear conclusions. The MD results bring distinct trends. The analysis of the average interaction energies in the two insects alpha6 nAChRs shows indeed better affinity of neonicotinoids bound to alpha6 cockroach compared to honeybee nAChR. This preference is explained by tighter contacts with aromatic residues (Trp and Tyr) of the binding pocket. Interestingly, the non-conserved residue Lys108 of loop D of alpha6 honeybee nAChR interacts through van der Waals contacts with neonicotinoids, which appear more favourable than the direct or water mediated hydrogen-bond interaction between the OH group of Ser83 of alpha6 cockroach nAChR and the electronegative terminal group of the two neonicotinoids (nitro in IMI and cyano in THI). Finally, in both insects nAChRs, THI is consistently found to bind more favourably than IMI

    En contexte archéologique, l’analyse pollinique de spéléothèmes offre l’opportunité de caractériser la composition de l’environnement végétal de l’Homme préhistorique. L’évaluation des processus taphonomiques affectant la pluie pollinique lors du dépôt dans la calcite souligne les limites et les potentiels de ce type de support. Les résultats obtenus sur des sites préhistoriques du pourtour nord‑méditerranéen sont confrontés et corrélés à l’ensemble de l’information paléoécologique livrée par les données multidisciplinaires. Ces études palynologiques tentent aussi d’estimer la distribution spatiale des écosystèmes végétaux, la composante locale étant souvent la plus significative. Quand la formation des spéléothèmes coïncide avec des niveaux archéologiques, l’environnement local de l’Homme préhistorique peut être précisé.Pollen analysis on speleothems carry out detailed vegetal environment data in archaeological context. Pollen rain recorded in calcite is subject to a variety of taphonomic processes, pointing out limits and potentials of pollen analyses on speleothems. These records are evaluated in the light of multidisciplinary palaeoecological information from Northern Mediterranean prehistoric settlements. A main contribution of local vegetation with regard to spatial distribution of the different vegetal ecosystems is observed. When speleothem growths and archaeological layer deposits are synchronous, local environment of the prehistoric populations can be accurately documented

    Isochrone ages for field dwarfs: method and application to the age-metallicity relation

    A new method is presented to compute age estimates from theoretical isochrones using temperature, luminosity and metallicity data for individual stars. Based on Bayesian probability theory, this method avoids the systematic biases affecting simpler strategies, and provides reliable estimates of the age probability distribution function for late-type dwarfs. This method is especially relevant for G dwarfs in the 3-15 Gyr range of ages, crucial to the study of the chemical and dynamical history of the Galaxy. We apply our method to the classic sample of Edvardsson et al. (1993), who derived the age-metallicity relation (AMR) of a sample of 189 field dwarfs with precisely determined abundances. Using new parallax, temperature and metallicity data, our age determination for the Edvardsson et al. sample indicates that the intrinsic dispersion in the AMR is at most 0.15 dex and probably lower. In particular, we show that old, metal-rich objects ([Fe/H]\sim 0.0 dex, age > 5 Gyr) and young, metal-poor objects ([Fe/H]<-0.5 dex, age < 6 Gyr) in many observed AMR plots are artifacts caused by a too simple treatment of the age determination, and that the Galactic AMR is monotonically increasing and rather well-defined. Incidentally, our results tend to restore confidence in the method of age determination from chromospheric activity for field dwarfs.Comment: 20 pages, 11 figures, to be published in MNRA

    A Methodology to Characterize Riverine Macroplastic Emission Into the Ocean

    Land-based macroplastic is considered one of the major sources of marine plastic debris. However, estimations of plastic emission from rivers into the oceans remain scarce and uncertain, mainly due to a severe lack of standardized observations. To properly assess global plastic fluxes, detailed information on spatiotemporal variation in river plastic quantities and composition are urgently needed. In this paper, we present a new methodology to characterize riverine macroplastic dynamics. The proposed methodology was applied to estimate the plastic emission from the Saigon River, Vietnam. During a 2-week period, hourly cross-sectional profiles of plastic transport were made across the river width. Simultaneously, sub-hourly samples were taken to determine the weight, size and composition of riverine macroplastics (&gt;5 cm). Finally, extrapolation of the observations based on available hydrological data yielded new estimates of daily, monthly and annual macroplastic emission into the ocean. Our results suggest that plastic emissions from the Saigon River are up to four times higher than previously estimated. Importantly, our flexible methodology can be adapted to local hydrological circumstances and data availability, thus enabling a consistent characterization of macroplastic dynamics in rivers worldwide. Such data will provide crucial knowledge for the optimization of future mediation and recycling efforts
