    L'adoption d'une nouvelle pratique est un processus dynamique et complexe influencé par de nombreux facteurs. Lorsque de nouvelles normes sont établies concernant l'utilisation des marqueurs biochimiques du syndrome coronarien aigu (MBSCA), les professionnels en biochimie clinique doivent considérer leur intégration à la pratique existante. La vitesse de l'adoption et la nature du changement effectué peuvent varier d'un hÎpital à un autre, causant des retards dans l'utilisation optimale des nouveaux MBSCA. L'objectif général de cette étude est de comprendre le rÎle de la critique par les pairs des données probantes dans l'adoption des nouveaux MBSCA. Les participants sont des professionnels en biochimie clinique membres de l'Association des médecins biochimistes du Québec (AMBQ) et du Canadian Society of Clinical Chemistry (CSCC).     A comparative study of GRACE with continental evapotranspiration estimates in Australian semi-arid and arid basins: sensitivity to climate variability and extremes

    This study examines the dynamics and robustness of large-scale evapotranspiration products in water-limited environments. Four types of ET products are tested against rainfall in two large semi-arid to arid Australian basins from 2003 to 2010: two energy balance ET methods which are forced by optical satellite retrievals from MODIS; a newly developed land surface model (AWRA); and one approach based on observations from the Gravity Recovery and Climate Experiment (GRACE) and rainfall data. The two basins are quasi (Murray-Darling Basin: 1.06 million km(2)) and completely (Lake Eyre Basin: 1.14 million km(2)) endorheic. During the study period, two extreme climatic events-the Millennium drought and the strongest La Nina event-were recorded in the basins and are used in our assessment. The two remotely-sensed ET products constrained by the energy balance tended to overestimate ET flux over water-stressed regions. They had low sensitivity to climatic extremes and poor capability to close the water balance. However, these two remotely-sensed and energy balance products demonstrated their superiority in capturing spatial features including over small-scale and complicated landscapes. AWRA and GRACE formulated in the water balance framework were more sensitive to rainfall variability and yielded more realistic ET estimates during climate extremes.     Three-Dimensional Folding and Functional Organization Principles of the Drosophila Genome

    SummaryChromosomes are the physical realization of genetic information and thus form the basis for its readout and propagation. Here we present a high-resolution chromosomal contact map derived from a modified genome-wide chromosome conformation capture approach applied to Drosophila embryonic nuclei. The data show that the entire genome is linearly partitioned into well-demarcated physical domains that overlap extensively with active and repressive epigenetic marks. Chromosomal contacts are hierarchically organized between domains. Global modeling of contact density and clustering of domains show that inactive domains are condensed and confined to their chromosomal territories, whereas active domains reach out of the territory to form remote intra- and interchromosomal contacts. Moreover, we systematically identify specific long-range intrachromosomal contacts between Polycomb-repressed domains. Together, these observations allow for quantitative prediction of the Drosophila chromosomal contact map, laying the foundation for detailed studies of chromosome structure and function in a genetically tractable system

    Rapid and concomitant gut microbiota and endocannabinoidome response to diet-induced obesity in mice

    The intestinal microbiota and the expanded endocannabinoid (eCB) system, or endocannabinoidome (eCBome), have both been implicated in diet-induced obesity and dysmetabolism. These systems were recently suggested to interact during the development of obesity. We aimed at identifying the potential interactions between gut microbiota composition and the eCBome during the establishment of diet-induced obesity and metabolic complications. Male mice were fed a high-fat, high-sucrose (HFHS) diet for 56 days to assess jejunum, ileum, and cecum microbiomes by 16S rRNA gene metataxonomics as well as ileum and plasma eCBome by targeted liquid chromatography-tandem mass spectrometry (LC-MS/MS). The HFHS diet induced early (3 days) and persistent glucose intolerance followed by weight gain and hyperinsulinemia. Concomitantly, it induced the elevation of the two eCBs, anandamide, in both ileum and plasma, and 2-arachidonoyl-glycerol, in plasma, as well as alterations in several other N-acylethanolamines and 2-acylglycerols. It also promoted segment-specific changes in the relative abundance of several genera in intestinal microbiota, some of which were observed as early as 3 days following HFHS diet. Weight-independent correlations were found between the relative abundances of, among others, Barnesiella, Eubacterium, Adlercreutzia, Parasutterella, Propionibacterium, Enterococcus, and Methylobacterium and the concentrations of anandamide and the anti-inflammatory eCBome mediator N-docosahexaenoyl-ethanolamine. This study highlights for the first time the existence of potential interactions between the eCBome, an endogenous system of multifunctional signaling lipids, and several intestinal genera during early and late HFHS-induced dysmetabolic events, with potential impact on the host capability of adapting to increased intake of fat and sucrose

    Chemometric perspectives on plankton community responses to natural iron fertilisation over and downstream of the Kerguelen Plateau in the Southern Ocean

    International audienceWe examined phytoplankton community responses to natural iron fertilisation at 32 sites over and downstream from the Kerguelen Plateau in the Southern Ocean during the austral spring bloom in October–November 2011. The community structure was estimated from chemical and isotopic measurements (particulate organic carbon – POC; 13C-POC; particulate nitrogen – PN; 15N-PN; and biogenic silica – BSi) on size-fractionated samples from surface waters (300, 210, 50, 20, 5, and 1 ÎŒm fractions). Higher values of 13C-POC (vs. co-located 13C values for dissolved inorganic carbon – DIC) were taken as indicative of faster growth rates and higher values of 15N-PN (vs. co-located 15N-NO3 source values) as indicative of greater nitrate use (rather than ammonium use, i.e. higher f ratios).Community responses varied in relation to both regional circulation and the advance of the bloom. Iron-fertilised waters over the plateau developed dominance by very large diatoms (50–210 ÎŒm) with high BSi / POC ratios, high growth rates, and significant ammonium recycling (lower f ratios) as biomass built up. In contrast, downstream polar frontal waters with a similar or higher iron supply were dominated by smaller diatoms (20–50 ÎŒm) and exhibited greater ammonium recycling. Stations in a deep-water bathymetrically trapped recirculation south of the polar front with lower iron levels showed the large-cell dominance observed on the plateau but much less biomass. Comparison of these communities to surface water nitrate (and silicate) depletions as a proxy for export shows that the low-biomass recirculation feature had exported similar amounts of nitrogen to the high-biomass blooms over the plateau and north of the polar front. This suggests that early spring trophodynamic and export responses differed between regions with persistent low levels vs. intermittent high levels of iron fertilisation

    Implication Statement During the pandemic, the MapIt app was integrated into an occupational therapy program to support remote learning of built environment adaptation. MapIt maps rooms in a home, then generates a 3D model for viewing and taking virtual measurements. The students express that the use of MapIt during their training leads to embodying the roles expected of an occupational therapist. To inspire other good pedagogical ideas, this article presents how MapIt can support learning in authentic situations, a key element of a professionalization path, approaching the realities experienced by patients, clients, or caregivers.ÉnoncĂ© des implications de la recherche Durant la pandĂ©mie, l’application MapIt a Ă©tĂ© intĂ©grĂ©e dans un programme d’ergothĂ©rapie pour soutenir l’apprentissage Ă  distance de l’adaptation de l’environnement bĂąti.     Interleukin-18 produced by bone marrow- derived stromal cells supports T-cell acute leukaemia progression

    International audienceDevelopment of novel therapies is critical for T-cell acute leukae-mia (T-ALL). Here, we investigated the effect of inhibiting the MAPK/MEK/ERK pathway on T-ALL cell growth. Unexpectedly, MEK inhibitors (MEKi) enhanced growth of 70% of human T-ALL cell samples cultured on stromal cells independently of NOTCH activa-tion and maintained their ability to propagate in vivo. Similar results were obtained when T-ALL cells were cultured with ERK1/ 2-knockdown stromal cells or with conditioned medium from MEKi-treated stromal cells. Microarray analysis identified interleu-kin 18 (IL-18) as transcriptionally up-regulated in MEKi-treated MS5 cells. Recombinant IL-18 promoted T-ALL growth in vitro, whereas the loss of function of IL-18 receptor in T-ALL blast cells decreased blast proliferation in vitro and in NSG mice. The NFKB pathway that is downstream to IL-18R was activated by IL-18 in blast cells. IL-18 circulating levels were increased in T-ALL-xeno-grafted mice and also in T-ALL patients in comparison with controls. This study uncovers a novel role of the pro-inflammatory cytokine IL-18 and outlines the microenvironment involvement in human T-ALL development
