    Expressão e análise antigênica da proteína RTP36 recombinante da amostra São Paulo de Ehrlichia canis para testes sorológicos

    Ehrlichia canis is the main etiological agent of canine monocytic ehrlichiosis (CME), a globally canine infectious disease. In Brazil, CME is considered to be endemic, and its prevalence can reach 65% in some states. The diagnosis of ehrlichiosis is important for treatment and epidemiological purposes. The E. canis TRP36 (Tandem Repeat Protein) protein elicits the earliest acute-phase antibody response observed during the course of the disease. This study aimed to generate the recombinant TRP36 protein from E. canis São Paulo strain and to evaluate its potential as a tool for the serologic diagnosis of CME. The E. canis São Paulo isolate was cultivated in DH82 lineage cells, and its genomic DNA was obtained. The bacterial DNA fragment encoding the entire ORF of TRP36 was cloned into the pBAD/Thio-TOPO vector and transformed into Escherichia coli DH10B competent cells with the trp36-bearing plasmid for protein expression. To evaluate the protein antigenicity, 16 canine serum samples were previously tested (by PCR and the commercial SNAP4Dx serological test). The results were in accordance with the SNAP4Dx test. Experiments using this recombinant protein as an antigen, targeting the development of a serologic test based on ELISA methodology, are the next step to produce a reliable, affordable and useful diagnostic tool for CME in Brazil.Ehrlichia canis é o principal agente etiológico da erliquiose monocítica canina (EMC), uma doença infecciosa canina globalmente dispersa. No Brasil, a EMC é considerada endêmica, e a infecção pode atingir 65% em cães em alguns estados. O diagnóstico de erliquiose é importante para fins de tratamento e epidemiológicos. A proteína TRP36 de E. canis leva a uma resposta humoral com produção de anticorpos em fase aguda, encontrada durante o curso da doença. O objetivo deste estudo foi obter a proteína TRP36 recombinante da amostra São Paulo de E. canis e avaliar seu potencial como ferramenta para o diagnóstico sorológico da CME. O isolado de E. canis São Paulo foi cultivado em células da linhagem DH82 e o DNA genômico foi obtido. O fragmento de DNA bacteriano que codifica toda a ORF de TRP36 foi clonado no vetor pBAD / Thio-TOPO e transformado em células competentes Escherichia coli DH10B, com o plasmídeo portador de trp36 para expressão de proteínas. Para avaliar a antigenicidade da proteína, 16 amostras de soro canino foram previamente analisadas (por PCR e teste sorológico comercial SNAP4Dx). Os resultados estavam de acordo com o teste SNAP4Dx. Os experimentos que utilizam essa proteína recombinante como antígeno, visando ao desenvolvimento de um teste sorológico baseado no ELISA, são o próximo passo para produzir um teste de diagnóstico confiável, acessível e útil para o diagnóstico da EMC no Brasil

    ESTRATÉGIAS DA GESTÃO PARA IMPLANTAÇÃO DO MODELO DA REDE CEGONHA EM UMA MATERNIDADE PÚBLICA DE CURITIBA

    The objective was to analyze the strategies adopted for the implementation of the guidelines of the Stork Network from managers’ perspective. An exploratory research with a quantitative and qualitative approach was undertaken. The data were collected in 2015 through interviews with three strategic managers from a common-risk maternity hospital in a capital in the South of Brazil. The analysis method of the Collective Subject Discourse was used with the software Qualiquantisoft®. The following categories emerged as results, among others: Strategies for the implementation of the Care Model, and Strategies for the structural reorganization of the management model, which lead to the implementation of the care model recommended by the Stork Network. In the light of this research, it can be concluded that the management’s proposal to implement a humanized care model based on scientific evidence is under construction, with valuable advances, the teams’ effort and the valuation of the professional who drives the change actions, putting in practice the continuous improvement of care.La finalidad fue analizar estrategias adoptadas para la implementación de las directivas de la Red Cigüeña, bajo la perspectiva de gestores. Se trata de una investigación exploratoria, con aproximación cuanti-cualitativa. Los datos fueron recolectados en 2015, mediante entrevistas con tres gestores del nivel estratégico de una maternidad de riesgo habitual en una capital del sur de Brasil. Fue utilizado el método de análisis del Discurso del Sujeto Colectivo con apoyo delsoftware Qualiquantisoft®. Como resultados, emergieron las categorías, entre otros: Estrategias para implantación del Modelo Asistencial, y Estrategias para reorganización estructural del modelo gerencial, que llevaron a la implantación del modelo asistencial preconizado por la RedCigüeña. Se pudo concluir, a la luz de esta investigación, que la propuesta de la gestión en implantar un modelo humanizado, pautado en evidencias científicas, está en proceso de construcción, con avances valiosos, empeño de los equipos y valuación del profesional, impulsando las acciones de cambio, concretizando la práctica del cuidado en permanente perfeccionamiento.Objetivou-se analisar estratégias adotadas para a implantação das diretrizes da Rede Cegonha, sob a perspectiva de gestores. Trata-se de uma pesquisa exploratória, de abordagem quanti-qualitativa, cuja coleta de dados ocorreu em 2015, mediante entrevistas a três gestores do nível estratégico de uma maternidade de risco habitual em uma capital do sul do Brasil. Utilizou-se o método de análise do Discurso do Sujeito Coletivo com apoio do software Qualiquantisoft®. Como resultados, emergiram as categorias, entre outras: Estratégias para implantação do Modelo Assistencial, e Estratégias para reorganização estrutural do modelo gerencial, que foram condizentes à implantação do modelo assistencial preconizado pela Rede Cegonha. Pode-se concluir, sob a luz desta pesquisa, que a proposta da gestão em implantar um modelo humanizado, pautado em evidências científicas, está em processo de construção, com avanços valiosos, empenho das equipes e valorização do profissional impulsionando as ações de mudança, concretizando a efetivação do cuidado em permanente aprimoramento

    The Ministry of Health, through the Secretariat of Primary Health Care and in partnership with the Secretariat of Health Surveillance, built and implemented strategies of Primary Health Care (PHC) within the scope of support to local managers and in partnership with the Council National Secretary of State (CONASS) and Municipal Health (CONASEMS) to combat COVID-19. These actions have PHC as the main responsible for several areas and physical, human and financial resources, as well as allowing to boost national progress towards the use of information and communication technologies and new partnerships for conducting research.El Ministerio de Salud, a través de la Secretaría de Atención Primaria de Salud y en asociación con la Secretaría de Vigilancia Sanitaria, elaboró ​​e implementó estrategias de Atención Primaria de Salud (APS) dentro del alcance del apoyo a los gerentes locales y en asociación con la Secretaría Consejo Nacional de Salud Estado (CONASS) y Salud Municipal (CONASEMS) para combatir COVID-19.     This work shows the results of a Proficiency Testing performed by a partnership between INMETRO and ANP. The performance of 49 Brazilian laboratories (using the z-score statistical test) in determining 10 quality parameters of ethanol fuel and biodiesel was evaluated. The certified reference values were provided by INMETRO, allowing a more rigorous assessment of the laboratories. For hydrous ethanol, the acidity parameter showed the lowest number of laboratories with satisfactory results (48%), while 85% of the laboratories presented satisfactory results for ethanol content. For biodiesel, the percentage of laboratories with satisfactory results ranged from 46% (kinematic viscosity) to 92% (acid number)

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     ATLANTIC EPIPHYTES: a data set of vascular and non-vascular epiphyte plants and lichens from the Atlantic Forest

    Epiphytes are hyper-diverse and one of the frequently undervalued life forms in plant surveys and biodiversity inventories. Epiphytes of the Atlantic Forest, one of the most endangered ecosystems in the world, have high endemism and radiated recently in the Pliocene. We aimed to (1) compile an extensive Atlantic Forest data set on vascular, non-vascular plants (including hemiepiphytes), and lichen epiphyte species occurrence and abundance; (2) describe the epiphyte distribution in the Atlantic Forest, in order to indicate future sampling efforts. Our work presents the first epiphyte data set with information on abundance and occurrence of epiphyte phorophyte species. All data compiled here come from three main sources provided by the authors: published sources (comprising peer-reviewed articles, books, and theses), unpublished data, and herbarium data. We compiled a data set composed of 2,095 species, from 89,270 holo/hemiepiphyte records, in the Atlantic Forest of Brazil, Argentina, Paraguay, and Uruguay, recorded from 1824 to early 2018. Most of the records were from qualitative data (occurrence only, 88%), well distributed throughout the Atlantic Forest. For quantitative records, the most common sampling method was individual trees (71%), followed by plot sampling (19%), and transect sampling (10%). Angiosperms (81%) were the most frequently registered group, and Bromeliaceae and Orchidaceae were the families with the greatest number of records (27,272 and 21,945, respectively). Ferns and Lycophytes presented fewer records than Angiosperms, and Polypodiaceae were the most recorded family, and more concentrated in the Southern and Southeastern regions. Data on non-vascular plants and lichens were scarce, with a few disjunct records concentrated in the Northeastern region of the Atlantic Forest. For all non-vascular plant records, Lejeuneaceae, a family of liverworts, was the most recorded family. We hope that our effort to organize scattered epiphyte data help advance the knowledge of epiphyte ecology, as well as our understanding of macroecological and biogeographical patterns in the Atlantic Forest.     A REESTRUTURAÇÃO PRODUTIVA NA ÁREA DA SAÚDE E DA ENFERMAGEM OBSTÉTRICA

    Esta discusión pretende presentar dimensiones de reestructuración productiva en el sector de salud en Brasil, discutiendo la relación con el trabajo de la enfermería obstétrica. Su construcción se basó en artículos sobre reestructuración productiva y salud, disponibles en la Biblioteca Virtual de Salud, reflejando el desarrollo del proceso de cuidar de esta especialidad. Verificamos que reestructuración productiva en salud se puede vislumbrar en la gerencia y la organizacíon del trabajo en salud. A pesar de los impactos negativos en el ámbito del trabajo, en obstetricia hubo apreciación del trabajo vivo en un contexto favorable a cambios en la asistencia al parto, a través de la inserción de la enfermería obstétrica e implementación de tecnologías de cuidado. Concluimos que, además de centrarse en el trabajo vivo y tecnologías livianas, estas concepciones constituyen avance tecnológico, pues modificaron y resignificaron el cuidar, iniciando el proceso de inversión del núcleo tecnológico en obstetricia
