CASP-12 as a measure of quality of life at older ages: comparing near and older elderly

    Get PDF
It is well known that European governments face to an ageing society as result of the increasing life expectancy and the decreasing birth rates in recent decades. A growing part of the European population is living their “Third age”, and this new paradigm has become increasingly important and has become a major concern in public policy. It stands to reason that better conditions at these ages (health, economic status, happiness, family, etc.) surely influence this approach. The aim of this paper is to determinate the key factors that affect the Quality of Life (QoL) of the elderly in the European countries. Additionally, we try to identify if significant differences are resulted in QoL of the elderly in regions across Europe (southern, northern and eastern). We have used data from the most recent wave released in SHARE (Survey of Health, Ageing and Retirement in Europe, wave 7) for all European countries participants in the survey. Then, we used the SHARE version of the CASP-12 scale to determinate if there are significant discrepancies between European countries. Despcritive results shown significant differences raised in QoL of the elderly by regions across Europe (northern and eastern) if life satisfaction is self-reported. However, using CASP-12 as key indicator of quality of life and life satisfaction the results are more heterogenous.Universidad de Málaga. Campus de Excelencia Internacional Andalucía Tech

How genomics has informed our understanding of the pathogenesis of osteoporosis

    Get PDF
Osteoporosis is a skeletal disorder characterized by compromised bone strength that predisposes a person to an increased risk of fracture. Osteoporosis is a complex trait that involves multiple genes, environmental factors, and gene-gene and gene-environment interactions. Twin and family studies have indicated that between 25% and 85% of the variation in bone mass and other skeletal phenotypes is heritable, but our knowledge of the underlying genes is limited. Bone mineral density is the most common assessment for diagnosing osteoporosis and is the most often used quantitative value in the design of genetic studies. In recent years, our understanding of the pathophysiology of osteoporosis has been greatly facilitated by advances brought about by the Human Genome Project. Genetic approaches ranging from family studies of monogenic traits to association studies with candidate genes, to whole-genome scans in both humans and animals have identified a small number of genes that contribute to the heritability of bone mass. Studies with transgenic and knockout mouse models have revealed major new insights into the biology of many of these identified genes, but much more needs to be learned. Ultimately, we hope that by revealing the underlying genetics and biology driving the pathophysiology of osteoporosis, new and effective treatment can be developed to combat and possibly cure this devastating disease. Here we review the rapidly evolving field of the genomics of osteoporosis with a focus on important gene discoveries, new biological/physiological paradigms that are emerging, and many of the unanswered questions and hurdles yet to be overcome in the field

    Get PDF

Transcriptional profiling reveals functional links between RasGrf1 and Pttg1 in pancreatic beta cells

Background: Our prior characterization of RasGrf1 deficient mice uncovered significant defects in pancreatic islet count and size as well as beta cell development and signaling function, raising question about the mechanisms linking RasGrf1 to the generation of those"pancreatic" phenotypes. Results: Here, we compared the transcriptional profile of highly purified pancreatic islets from RasGrf1 KO mice to that of WT control animals using commercial oligonucleotide microarrays. RasGrf1 elimination resulted in differential gene expression of numerous components of MAPK- and Calcium-signaling pathways, suggesting a relevant contribution of this GEF to modulation of cellular signaling in the cell lineages integrating the pancreatic islets. Whereas the overall transcriptional profile of pancreatic islets was highly specific in comparison to other organs of the same KO mice, a significant specific repression of Pttg1 was a common transcriptional alteration shared with other tissues of neuroectodermal origin. This observation, together with the remarkable pancreatic phenotypic similarities between RasGrf1 KO and Pttg1 KO mice suggested the possibility of proximal functional regulatory links between RasGrf1 and Pttg1 in pancreatic cell lineages expressing these proteins. Analysis of the mPttg1 promoter region identified specific recognition sites for numerous transcription factors which were also found to be differentially expressed in RasGrf1 KO pancreatic islets and are known to be relevant for Ras-ERK signaling as well as beta cell function. Reporter luciferase assays in BT3 insulinoma cells demonstrated the ability of RasGrf1 to modulate mPttg1 promoter activity through ERK-mediated signals. Analysis of the phenotypic interplay between RasGrf1 and Pttg1 in double knockout RasGrf1/Pttg1 mice showed that combined elimination of the two loci resulted in dramatically reduced values of islet and beta cell count and glucose homeostasis function which neared those measured in single Pttg1 KO mice and were significantly lower than those observed in individual RasGrf1 KO mice. Conclusions: The specific transcriptional profile and signaling behavior of RasgGrf1 KO pancreatic islets, together with the dominance of Pttg1 over RasGrf1 with regards to the generation of these phenotypes in mouse pancreas, suggest that RasGrf1 is an important upstream component of signal transduction pathways regulating Pttg1 expression and controlling beta cell development and physiological response

Rye (Secale cereale L.) and squarrose clover (Trifolium squarrosum L.) cover crops can increase their allelopathic potential for weed control when used mixed as dead mulch

Cover crops are essential tools in agro-ecosystems for reduc- ing the reliance on synthetic inputs and associated environmental risks. Alongside their benefits to soil fertility, cover crops can con- trol weeds by their competitive and allelopathic attributes. Laboratory and field experiments were conducted to assess the allelopathic potential of two cover crop species, rye (Secale cereale L.) and squarrose clover (Trifolium squarrosum L.), alone or in a mixture, on seed germination and growth of arable weeds. Aqueous extracts of the two cover crops and their mixture were tested in a bioassay on Conyza canadensis (L). Cronq., Amaranthus retroflexus L. and Digitaria sanguinalis (L.) Scop. In vitro effects of aqueous extracts varied in a dose-dependent manner, with cover crops and weed species. All three extracts were able to reduce the germination of A. retroflexus (–87%) consider- ably. Inhibitory effects by rye and mixture extracts on radicle growth of all weed species ranged between 51 and 82%. Rye extract was the best at reducing shoot length of C. canadensis and D. sanguinalis (–39 to 44%), while squarrose clover was more effective on A. retroflexus (–79%). Plant extracts also delayed the germination time of weed species with a substantial effect of the mixture on C. canadensis seeds. In the field experiment, no signif- icant weed suppression was provided by cover crop residues incorporated as green manure compared to control plots, despite tillage being more effective in reducing weed density than no-till. Still, the mulch of the mixture controlled weed emergence signif- icantly better than single cover crop mulches. The chemical char- acterization of cover crop residues, both shoots and roots, revealed a notable richness of allelopathic phenolic acids and flavonoids, which may constitute potential natural herbicides through slow decomposition. From the analysis of the aqueous extracts, other non-analysed and/or unidentified water-soluble allelopathic com- pounds should underlie the phytotoxicity observed in vitro, at least for rye. For cover crop mixture, positive interactions among plant materials leading to a better release of allelochemicals and weed- ing effectiveness are discussed according to chemical profiles and field data. Our study demonstrated the allelopathic activity of the cover crops and their potential to be included in weed manage- ment strategies according to cropping system needs. Additional trials are needed to confirm the performance of cover crop residues under field conditions
