    Microsatellite Analysis of Trophy Largemouth Bass from Arkansas Reservoirs

    The Arkansas Game and Fish Commission (AGFC) has introduced Florida largemouth bass (FLMB; Micropterus salmoides floridanus) to water bodies historically containing the northern largemouth bass (NLMB; Micropterus salmoides salmoides) subspecies since the late 1970s in an attempt to produce a trophy LMB fishery. Since 2006, the AGFC has been biannually sampling reservoirs stocked with FLMB to determine levels of admixture. Here, total sampling efforts between 2006 and 2011 have been combined, and LMB heavier than 2,268 g (5 lb) were analyzed in an effort to investigate distribution of bass by their genetic composition designated as trophy LMB by the AGFC. Of the 148 trophy LMB sampled, 123 possessed FLMB alleles (83.1%). Thirty-two of the heaviest 50 (64.0%) LMB sampled, including a potential state record that was nullified, were genetically confirmed to be FLMB.     PDE8 controls CD4(+) T cell motility through the PDE8A-Raf-1 kinase signaling complex

    The levels of cAMP are regulated by phosphodiesterase enzymes (PDEs), which are targets for the treatment of inflammatory disorders. We have previously shown that PDE8 regulates T cell motility. Here, for the first time, we report that PDE8A exerts part of its control of T cell function through the V-raf-1 murine leukemia viral oncogene homolog 1 (Raf-1) kinase signaling pathway. To examine T cell motility under physiologic conditions, we analyzed T cell interactions with endothelial cells and ligands in flow assays. The highly PDE8-selective enzymatic inhibitor PF-04957325 suppresses adhesion of in vivo myelin oligodendrocyte glycoprotein (MOG) activated inflammatory CD4(+) T effector (Teff) cells to brain endothelial cells under shear stress. Recently, PDE8A was shown to associate with Raf-1 creating a compartment of low cAMP levels around Raf-1 thereby protecting it from protein kinase A (PKA) mediated inhibitory phosphorylation. To test the function of this complex in Teff cells, we used a cell permeable peptide that selectively disrupts the PDE8A-Raf-1 interaction. The disruptor peptide inhibits the Teff-endothelial cell interaction more potently than the enzymatic inhibitor. Furthermore, the LFA-1/ICAM-1 interaction was identified as a target of disruptor peptide mediated reduction of adhesion, spreading and locomotion of Teff cells under flow. Mechanistically, we observed that disruption of the PDE8A-Raf-1 complex profoundly alters Raf-1 signaling in Teff cells. Collectively, our studies demonstrate that PDE8A inhibition by enzymatic inhibitors or PDE8A-Raf-1 kinase complex disruptors decreases Teff cell adhesion and migration under flow, and represents a novel approach to target T cells in inflammation

    The distribution and movement patterns of four woodland caribou herds in Quebec and Labrador

    Recent studies of woodland caribou (Rangifer tarandus caribou) in northern Quebec and central Labrador have demonstrated similar patterns of seasonal movements and distribution among four herds. Aerial surveys and radio-telemetry indicated that animals occupied forest-wetland habitat at densities of 0.03 caribou km2, or lower, for most of the year. Although females were widely dispersed at calving individuals demonstrated fidelity toward specific calving locations, in successive years. Caribou did not form large post-calving aggregations. Movement was greatest in the spring, prior to calving, and in the fall, during or immediately after rutting. Caribou were generally sedentary during summer and winter, although some moved relatively long distances to late-winter range. Although the herds occupy continuous range across Quebec and Labrador, our data indicate that the herds are largely discreete and should be managed individually

    T cell receptors for the HIV KK10 epitope from patients with differential immunologic control are functionally indistinguishable

    HIV controllers (HCs) are individuals who can naturally control HIV infection, partially due to potent HIV-specific CD8+ T cell responses. Here, we examined the hypothesis that superior function of CD8+ T cells from HCs is encoded by their T cell receptors (TCRs). We compared the functional properties of immunodominant HIV-specific TCRs obtained from HLA-B*2705 HCs and chronic progressors (CPs) following expression in primary T cells. T cells transduced with TCRs from HCs and CPs showed equivalent induction of epitope-specific cytotoxicity, cytokine secretion, and antigen-binding properties. Transduced T cells comparably, albeit modestly, also suppressed HIV infection in vitro and in humanized mice. We also performed extensive molecular dynamics simulations that provided a structural basis for similarities in cytotoxicity and epitope cross-reactivity. These results demonstrate that the differential abilities of HIV-specific CD8+ T cells from HCs and CPs are not genetically encoded in the TCRs alone and must depend on additional factors

    CADMIUM GEOCHEMISTRY OF SOILS AND WILLOW IN A METAMORPHIC BEDROCK TERRAIN AND ITS POSSIBLE RELATION TO MOOSE HEALTH, SEWARD PENINSULA, ALASKA

    The regional geochemistry of soil and willow over Paleozoic metamorphic rocks in the Seward Peninsula, Alaska is potentially high in cadmium (Cd), and willow, a preferred browse of moose, bioaccumulates Cd. Local moose show clinical signs of tooth wear and breakage and have been declining in population for unknown reasons. Willow leaves (all variants of Salix pulchra) and A-, B-, and C-horizon soils were sampled near 2 mining prospects suspected to be high in Cd. Although Al, Cd, Co, Cu, Fe, Mo, Ni, Pb, and Zn were examined, our focus in this exploratory study was on the level of Cd in the 3 soil horizons and willow between and within the 2 prospects and their vicinity. We used an unbalanced, one-way, hierarchical analysis of variance (ANOVA) to investigate the geochemistry of soils and willow at various distance scales across the 2 prospect areas that were separated by ~80 km; sites within a location were approximately 0.5 km apart and replicate samples were separated by ~0.05 km. Cd concentration was significantly different in willow between and within sites, and within sites for all soil horizons. Specifically, this exploratory study identified highly elevated levels of Cd in willow growing over Paleozoic bedrock in the Seward Peninsula at both prospects and over the Paleozoic geologic unit in general.     Earthquake lubrication and healing explained by amorphous nanosilica

    During earthquake propagation, geologic faults lose their strength, then strengthen as slip slows and stops. Many slip-weakening mechanisms are active in the upper-mid crust, but healing is not always well-explained. Here we show that the distinct structure and rate-dependent properties of amorphous nanopowder (not silica gel) formed by grinding of quartz can cause extreme strength loss at high slip rates. We propose a weakening and related strengthening mechanism that may act throughout the quartz-bearing continental crust. The action of two slip rate-dependent mechanisms offers a plausible explanation for the observed weakening: thermally-enhanced plasticity, and particulate flow aided by hydrodynamic lubrication. Rapid cooling of the particles causes rapid strengthening, and inter-particle bonds form at longer timescales. The timescales of these two processes correspond to the timescales of post-seismic healing observed in earthquakes. In natural faults, this nanopowder crystallizes to quartz over 10s–100s years, leaving veins which may be indistinguishable from common quartz veins
