    A review of independent living adaptations

    This study will explore and test potential changes to the structural and delivery mechanisms for independent living adaptations. This is in order to move towards the Welsh Government goal of ensuring that adaptations can be delivered more speedily to those who need them. In doing so, it will help reduce inherent inequalities in the current system by ensuring joined up solutions, prevention and early intervention activity. This Review was commissioned in the light of some £35 million being spent annually by local authorities in Wales on Disabled Facilities Grants (DFGs) and the Welsh Government’s own spending of approximately £8 million on Physical Adaptations Grants (PAG) for social tenants. The Review was carried out between May and November 2014 and addressed three aims: mapping the current system performance Indicators testing the feasibility of options for change

    Increased male reproductive success in Ts65Dn “Down syndrome” mice

    The Ts65Dn mouse is trisomic for orthologs of about half the genes on Hsa21. A number of phenotypes in these trisomic mice parallel those in humans with trisomy 21 (Down syndrome), including cognitive deficits due to hippocampal malfunction that are sufficiently similar to human that “therapies” developed in Ts65Dn mice are making their way to human clinical trials. However, the impact of the model is limited by availability. Ts65Dn cannot be completely inbred and males are generally considered to be sterile. Females have few, small litters and they exhibit poor care of offspring, frequently abandoning entire litters. Here we report identification and selective breeding of rare fertile males from two working colonies of Ts65Dn mice. Trisomic offspring can be propagated by natural matings or by in vitro fertilization (IVF) to produce large cohorts of closely related siblings. The use of a robust euploid strain as recipients of fertilized embryos in IVF or as the female in natural matings greatly improves husbandry. Extra zygotes cultured to the blastocyst stage were used to create trisomic and euploid embryonic stem (ES) cells from littermates. We developed parameters for cryopreserving sperm from Ts65Dn males and used it to produce trisomic offspring by IVF. Use of cryopreserved sperm provides additional flexibility in the choice of oocyte donors from different genetic backgrounds, facilitating rapid production of complex crosses. This approach greatly increases the power of this important trisomic model to interrogate modifying effects of trisomic or disomic genes that contribute to trisomic phenotypes

    Spatial and Seasonal Distribution of American Whaling and Whales in the Age of Sail

    American whalemen sailed out of ports on the east coast of the United States and in California from the 18th to early 20th centuries, searching for whales throughout the world’s oceans. From an initial focus on sperm whales (Physeter macrocephalus) and right whales (Eubalaena spp.), the array of targeted whales expanded to include bowhead whales (Balaena mysticetus), humpback whales (Megaptera novaeangliae), and gray whales (Eschrichtius robustus). Extensive records of American whaling in the form of daily entries in whaling voyage logbooks contain a great deal of information about where and when the whalemen found whales. We plotted daily locations where the several species of whales were observed, both those caught and those sighted but not caught, on world maps to illustrate the spatial and temporal distribution of both American whaling activity and the whales. The patterns shown on the maps provide the basis for various inferences concerning the historical distribution of the target whales prior to and during this episode of global whaling

    Cadmium accumulation and interactions with zinc, copper, and manganese, analysed by ICP-MS in a long-term Caco-2 TC7 cell model

    The influence of long-term exposure to cadmium (Cd) on essential minerals was investigated using a Caco-2 TC7 cells and a multi-analytical tool: microwave digestion and inductively coupled plasma mass spectrometry. Intracellular levels, effects on cadmium accumulation, distribution, and reference concentration ranges of the following elements were determined: Na, Mg, Ca, Cr, Fe, Mn, Co, Ni, Cu, Zn, Mo, and Cd. Results showed that Caco-2 TC7 cells incubated long-term with cadmium concentrations ranging from 0 to 10 lmol Cd/l for 5 weeks exhibited a significant increase in cadmium accumulation. Furthermore, this accumulation was more marked in cells exposed long-term to cadmium compared with controls, and that this exposure resulted in a significant accumulation of copper and zinc but not of the other elements measured. Interactions of Cd with three elements: zinc, copper, and manganese were particularly studied. Exposed to 30 lmol/l of the element, manganese showed the highest inhibition and copper the lowest on cadmium intracellular accumulation but Zn, Cu, and Mn behave differently in terms of their mutual competition with Cd. Indeed, increasing cadmium in the culture medium resulted in a gradual and significant increase in the accumulation of zinc. There was a significant decrease in manganese from 5 lmol Cd/l exposure, and no variation was observed with copper.     Thermally Triggered Hydrogel Injection Into Bovine Intervertebral Disc Tissue Explants Induces Differentiation Of Mesenchymal Stem Cells And Restores Mechanical Function.

    We previously reported a synthetic Laponite® crosslinked pNIPAM-co-DMAc (L-pNIPAM-co-DMAc) hydrogel which promotes differentiation of mesenchymal stem cells (MSCs) to nucleus pulposus (NP) cells without additional growth factors. The clinical success of this hydrogel is dependent on: integration with surrounding tissue; the capacity to restore mechanical function; as well as supporting the viability and differentiation of delivered MSCs. Bovine NP tissue explants were injected with media (control), human MSCs (hMSCs) alone, acellular L-pNIPAM-co-DMAc hydrogel or hMSCs incorporated within the L-pNIPAM-co-DMAc hydrogel and maintained at 5% O2 for 6 weeks. Viability of native NP cells and delivered MSCs was maintained. Furthermore hMSCs delivered via the L-pNIPAM-co-DMAc hydrogel differentiated and produced NP matrix components: aggrecan, collagen type II and chondroitin sulphate, with integration of the hydrogel with native NP tissue. In addition L-pNIPAM-co-DMAc hydrogel injected into collagenase digested bovine discs filled micro and macro fissures, were maintained within the disc during loading and restored IVD stiffness. The mechanical support of the L-pNIPAM-co-DMAc hydrogel, to restore disc height, could provide immediate symptomatic pain relief, whilst the delivery of MSCs over time regenerates the NP extracellular matrix; thus the L-pNIPAM-co-DMAc hydrogel could provide a combined cellular and mechanical repair approach

    A Runaway Black Hole in COSMOS: Gravitational Wave or Slingshot Recoil?

    We present a detailed study of a peculiar source in the COSMOS survey at z=0.359. Source CXOCJ100043.1+020637 (CID-42) presents two compact optical sources embedded in the same galaxy. The distance between the 2, measured in the HST/ACS image, is 0.495" that, at the redshift of the source, corresponds to a projected separation of 2.46 kpc. A large (~1200 km/s) velocity offset between the narrow and broad components of Hbeta has been measured in three different optical spectra from the VLT/VIMOS and Magellan/IMACS instruments. CID-42 is also the only X-ray source having in its X-ray spectra a strong redshifted broad absorption iron line, and an iron emission line, drawing an inverted P-Cygni profile. The Chandra and XMM data show that the absorption line is variable in energy by 500 eV over 4 years and that the absorber has to be highly ionized, in order not to leave a signature in the soft X-ray spectrum. That these features occur in the same source is unlikely to be a coincidence. We envisage two possible explanations: (1) a gravitational wave recoiling black hole (BH), caught 1-10 Myr after merging, (2) a Type 1/ Type 2 system in the same galaxy where the Type 1 is recoiling due to slingshot effect produced by a triple BH system. The first possibility gives us a candidate gravitational waves recoiling BH with both spectroscopic and imaging signatures. In the second case, the X-ray absorption line can be explained as a BAL-like outflow from the foreground nucleus (a Type 2 AGN) at the rearer one (a Type 1 AGN), which illuminates the otherwise undetectable wind, giving us the first opportunity to show that fast winds are present in obscured AGN.Comment: 13 figures; submitted to ApJ.
