    Cosmological Evolution of Supermassive Black Holes. II. Evidence for Downsizing of Spin Evolution

    The spin is an important but poorly constrained parameter for describing supermassive black holes (SMBHs). Using the continuity equation of SMBH number density, we explicitly obtain the mass-dependent cosmological evolution of the radiative efficiency for accretion, which serves as a proxy for SMBH spin. Our calculations make use of the SMBH mass function of active and inactive galaxies (derived in the first paper of this series), the bolometric luminosity function of active galactic nuclei (AGNs), corrected for the contribution from Compton-thick sources, and the observed Eddington ratio distribution. We find that the radiative efficiency generally increases with increasing black hole mass at high redshifts (z>~1), roughly as \eta \propto M_bh^0.5, while the trend reverses at lower redshifts, such that the highest efficiencies are attained by the lowest mass black holes. Black holes with M_bh>~10^8.5M_sun maintain radiative efficiencies as high as \eta~0.3-0.4 at high redshifts, near the maximum for rapidly spinning systems, but their efficiencies drop dramatically (by an order of magnitude) by z~0. The pattern for lower mass holes is somewhat more complicated but qualitatively similar. Assuming that the standard accretion disk model applies, we suggest that the accretion history of SMBHs and their accompanying spins evolve in two distinct regimes: an early phase of prolonged accretion, plausibly driven by major mergers, during which the black hole spins up, then switching to a period of random, episodic accretion, governed by minor mergers and internal secular processes, during which the hole spins down. The transition epoch depends on mass, mirroring other evidence for "cosmic downsizing" in the AGN population; it occurs at z~2 for high-mass black holes, and somewhat later, at z~1, for lower-mass systems.Comment: To appear in the ApJ, 11 pages and 9 figure

    Disparities in Rate, Triggers, and Management in Pediatric and Adult Cases of Suspected Drug-Induced Anaphylaxis in Canada

    INTRODUCTION: Data is sparse on drug-induced anaphylaxis (DIA) and there have not been studies assessing the differences in clinical characteristics and management of DIA between adults and children. OBJECTIVE: We assessed the percentage, diagnosis, and management of DIA among all anaphylaxis visits in three pediatric and one adult emergency departments (ED) across Canada. METHODS: Children presenting to the Montreal Children\u27s Hospital (MCH), British Columbia Children\u27s Hospital (BCCH), and Children\u27s Hospital at London Health Sciences Center and adults presenting to Hôpital du Sacré-Coeur with anaphylaxis were recruited as part of the Cross-Canada Anaphylaxis Registry. A standardized data form documenting the reaction and management was completed and patients were followed annually to determine assessment by allergist and use of confirmatory tests. RESULTS: From June 2012 to May 2016, 51 children were recruited from the pediatric centers and 64 adults from the adult center with drug-induced anaphyalxis. More than half the cases were prospectively recruited. The percentage of DIA among all cases of anaphylaxis was similar in all three pediatric centers but higher in the adult center in Montreal. Most reactions in children were triggered by non-antibiotic drugs, and in adults, by antibiotics. The majority of adults and a third of children did not see an allergist after the initial reaction. In those that did see an allergist, diagnosis was established by either a skin test or an oral challenge in less than 20% of cases. CONCLUSIONS: Our results reveal disparities in rate, culprit, and management of DIA in children versus adults. Further, most cases of suspected drug allergy are not appropriately diagnosed. Guidelines to improve assessment and diagnosis of DIA are required

    Risk stratification by pre-operative cardiopulmonary exercise testing improves outcomes following elective abdominal aortic aneurysm surgery : a cohort study

    Background: In 2009, the NHS evidence adoption center and National Institute for Health and Care Excellence (NICE) published a review of the use of endovascular aneurysm repair (EVAR) of abdominal aortic aneurysms (AAAs). They recommended the development of a risk-assessment tool to help identify AAA patients with greater or lesser risk of operative mortality and to contribute to mortality prediction. A low anaerobic threshold (AT), which is a reliable, objective measure of pre-operative cardiorespiratory fitness, as determined by pre-operative cardiopulmonary exercise testing (CPET) is associated with poor surgical outcomes for major abdominal surgery. We aimed to assess the impact of a CPET-based risk-stratification strategy upon perioperative mortality, length of stay and non-operative costs for elective (open and endovascular) infra-renal AAA patients. Methods: A retrospective cohort study was undertaken. Pre-operative CPET-based selection for elective surgical intervention was introduced in 2007. An anonymized cohort of 230 consecutive infra-renal AAA patients (2007 to 2011) was studied. A historical control group of 128 consecutive infra-renal AAA patients (2003 to 2007) was identified for comparison. Comparative analysis of demographic and outcome data for CPET-pass (AT ≥ 11 ml/kg/min), CPET-fail (AT < 11 ml/kg/min) and CPET-submaximal (no AT generated) subgroups with control subjects was performed. Primary outcomes included 30-day mortality, survival and length of stay (LOS); secondary outcomes were non-operative inpatient costs. Results: Of 230 subjects, 188 underwent CPET: CPET-pass n = 131, CPET-fail n = 35 and CPET-submaximal n = 22. When compared to the controls, CPET-pass patients exhibited reduced median total LOS (10 vs 13 days for open surgery, n = 74, P < 0.01 and 4 vs 6 days for EVAR, n = 29, P < 0.05), intensive therapy unit requirement (3 vs 4 days for open repair only, P < 0.001), non-operative costs (£5,387 vs £9,634 for open repair, P < 0.001) and perioperative mortality (2.7% vs 12.6% (odds ratio: 0.19) for open repair only, P < 0.05). CPET-stratified (open/endovascular) patients exhibited a mid-term survival benefit (P < 0.05). Conclusion: In this retrospective cohort study, a pre-operative AT > 11 ml/kg/min was associated with reduced perioperative mortality (open cases only), LOS, survival and inpatient costs (open and endovascular repair) for elective infra-renal AAA surgery

    On the radiative efficiencies, Eddington ratios, and duty cycles of luminous high-redshift quasars

    We investigate the characteristic radiative efficiency \epsilon, Eddington ratio \lambda, and duty cycle P_0 of high-redshift active galactic nuclei (AGN), drawing on measurements of the AGN luminosity function at z=3-6 and, especially, on recent measurements of quasar clustering at z=3-4.5 from the Sloan Digital Sky Survey. The free parameters of our models are \epsilon, \lambda, and the normalization, scatter, and redshift evolution of the relation between black hole mass \mbh and halo virial velocity V_vir. We compute the luminosity function from the implied growth of the black hole mass function and the quasar correlation length from the bias of the host halos. We test our adopted formulae for the halo mass function and halo bias against measurements from the large N-body simulation developed by the MICE collaboration. The strong clustering of AGNs observed at z=3 and, especially, at z=4 implies that massive black holes reside in rare, massive dark matter halos. Reproducing the observed luminosity function then requires high efficiency \epsilon and/or low Eddington ratio \lambda, with a lower limit (based on 2\sigma agreement with the measured z=4 correlation length) \epsilon> 0.7\lambda/(1+0.7\lambda), implying \epsilon > 0.17 for \lambda > 0.25. Successful models predict high duty cycles, P_0~0.2, 0.5, and 0.9 at z=3.1, 4.5 and 6, respectively, and they require that the fraction of halo baryons locked in the central black hole is much larger than the locally observed value. The rapid drop in the abundance of the massive and rare host halos at z>7 implies a proportionally rapid decline in the number density of luminous quasars, much stronger than simple extrapolations of the z=3-6 luminosity function would predict.     MicroRNAs in cardiac arrhythmia: DNA sequence variation of MiR-1 and MiR-133A in long QT syndrome.

    Long QT syndrome (LQTS) is a genetic cardiac condition associated with prolonged ventricular repolarization, primarily a result of perturbations in cardiac ion channels, which predisposes individuals to life-threatening arrhythmias. Using DNA screening and sequencing methods, over 700 different LQTS-causing mutations have been identified in 13 genes worldwide. Despite this, the genetic cause of 30-50% of LQTS is presently unknown. MicroRNAs (miRNAs) are small (∼ 22 nucleotides) noncoding RNAs which post-transcriptionally regulate gene expression by binding complementary sequences within messenger RNAs (mRNAs). The human genome encodes over 1800 miRNAs, which target about 60% of human genes. Consequently, miRNAs are likely to regulate many complex processes in the body, indeed aberrant expression of various miRNA species has been implicated in numerous disease states, including cardiovascular diseases. MiR-1 and MiR-133A are the most abundant miRNAs in the heart and have both been reported to regulate cardiac ion channels. We hypothesized that, as a consequence of their role in regulating cardiac ion channels, genetic variation in the genes which encode MiR-1 and MiR-133A might explain some cases of LQTS. Four miRNA genes (miR-1-1, miR-1-2, miR-133a-1 and miR-133a-2), which encode MiR-1 and MiR-133A, were sequenced in 125 LQTS probands. No genetic variants were identified in miR-1-1 or miR-133a-1; but in miR-1-2 we identified a single substitution (n.100A> G) and in miR-133a-2 we identified two substitutions (n.-19G> A and n.98C> T). None of the variants affect the mature miRNA products. Our findings indicate that sequence variants of miR-1-1, miR-1-2, miR-133a-1 and miR-133a-2 are not a cause of LQTS in this cohort

    Cell_motility: a cross-platform, open source application for the study of cell motion paths

    BACKGROUND: Migration is an important aspect of cellular behaviour and is therefore widely studied in cell biology. Numerous components are known to participate in this process in a highly dynamic manner. In order to obtain a better insight in cell migration, mutants or drugs are used and their motive phenotype is then linked with the disturbing factors. One of the typical approaches to study motion paths of individual cells relies on fitting mean square displacements to a persistent random walk function. Since the numerous calculations involved often rely on diverse commercial software packages, the analysis can be expensive, labour-intensive and error-prone work. Additionally, due to the nature of algorithms employed the calculations involved are not readily reproducible without access to the exact software package(s) used. RESULTS: We here present the cell_motility software, an open source Java application under the GNU-GPL license that provides a clear and concise analysis workbench for large amounts of cell motion data. Apart from performing the necessary calculations, the software also visualizes the original motion paths as well as the results of the calculations to help the user interpret the data. The application features an intuitive graphical user interface as well as full user and developer documentation and both source and binary files can be freely downloaded from the project website at . CONCLUSION: In providing a free, open source software solution for the automated processing of cell motion data, we aim to achieve two important goals: labs can greatly simplify their data analysis pipeline as switching between different computational software packages becomes obsolete (thus reducing the chances for human error during data manipulation and transfer) and secondly, to provide scientists in the field with a freely available common platform to perform their analyses, enabling more efficient data quality control through peer reviewing

    Broad Line Region Physical Conditions along the Quasar Eigenvector 1 Sequence

    [Abridged] We compare broad emission line profiles and estimate line ratios for all major emission lines between Ly-alpha and H-beta in a sample of six quasars. The sources were chosen with two criteria in mind: the existence of high quality optical and UV spectra as well as the possibility to sample the spectroscopic diversity in the 4D Eigenvector 1 context . In the latter sense each source occupies a region (bin) in the FWHM(H-beta) vs. optical FeII strength plane that is significantly different from the others. High S/N H-beta emission line profiles are used as templates for modeling the other lines (Ly-alpha, CIV 1549, HeII 1640, Al III 1860, Si III] 1892, and Mg II 2800). We can adequately model all broad lines assuming the existence of three components distinguished by blueshifted, unshifted and redshifted centroids (indicated as blue, broad and very broad component respectively). The broad component (high electron density, low ionization parameter; high column density) is present in almost all type-1 quasars and therefore corresponds most closely to the classical broad line emitting region (the reverberating component). The blue component emission (lower electron density; high ionization; low column density) arises in less optically thick gas; it is often thought to arise in an accretion disk wind. The least understood component involves the very broad component (high ionization and large column density). It is perhaps the most distinguishing characteristic of quasars with FWHM H-beta > 4000 km/s that belong to the so-called Population B of our 4DE1 space. Population A quasars (FWHM H-beta < 4000 km/s) are dominated by broad component emission in H-beta and blue component emission in CIV 1549 and other high ionization lines. 4DE1 appears to be the most useful current context for revealing and unifying spectral diversity in type-1 quasars.Comment: 7 Tables, 5 Figures; accepted for publication in Monthly Notices of the Royal Astronomical Societ
