    Sex differences and correlates of poor glycaemic control in type 2 diabetes: a cross-sectional study in Brazil and Venezuela.

    OBJECTIVE: Examine whether glycaemic control varies according to sex and whether the latter plays a role in modifying factors associated with inadequate glycaemic control in patients with type 2 diabetes (T2D) in Brazil and Venezuela. DESIGN, SETTING AND PARTICIPANTS: This was a cross-sectional, nationwide survey conducted in Brazil and Venezuela from February 2006 to June 2007 to obtain information about glycaemic control and its determinants in patients with diabetes mellitus attending outpatient clinics. MAIN OUTCOME MEASURES: Haemoglobin A1c (HbA1c) level was measured by liquid chromatography, and patients with HbA1c ≥7.0% (53 mmol/mol) were considered to have inadequate glycaemic control. The association of selected variables with glycaemic control was analysed by multivariate linear regression, using HbA1c as the dependent variable. RESULTS: A total of 9418 patients with T2D were enrolled in Brazil (n=5692) and in Venezuela (n=3726). They included 6214 (66%) women and 3204 (34%) men. On average, HbA1c levels in women were 0.13 (95% CI 0.03 to 0.24; p=0.015) higher than in men, after adjusting for age, marital status, education, race, country, body mass index, duration of disease, complications, type of healthcare, adherence to diet, adherence to treatment and previous measurement of HbA1c. Sex modified the effect of some factors associated with glycaemic control in patients with T2D in our study, but had no noteworthy effect in others. CONCLUSIONS: Women with T2D had worse glycaemic control than men. Possible causes for poorer glycaemic control in women compared with men include differences in glucose homeostasis, treatment response and psychological factors. In addition, sex modified factors associated with glycaemic control, suggesting the need to develop specific treatment guidelines for men and women

    Gesture Facilitates Children’s Creative Thinking

    Gestures help people think and can help offer new ideas to problem solvers. We conducted two experiments exploring the self-oriented function of gesture in a novel domain; creative thinking. In Experiment 1 we explored the relationship between children’s spontaneous gesture production and their ability to generate novel uses for everyday items (Alternative Uses Task). There was a significant correlation between children’s creative fluency and their gesture production, with the majority of children’s gestures depicting an action upon the target object. Restricting children from gesturing did not significantly reduce their fluency. In Experiment 2 we encouraged children to gesture and this significantly boosted their creative idea generation. These findings demonstrate that gestures serve an important self-oriented function and can assist creative thinking

    Bond Behavior between Recycled Concrete Containing CDW and Different Types of Steel Bars

    The operation of reinforced concrete structures is directly associated with the adhesion between the steel bar and the concrete, which allows the internal forces to be transferred to the reinforcement during the process of loading the structural elements. The modification of the concrete composition, with the introduction of recycled aggregate from construction and demolition waste (CDW), affects the steel-concrete interface and can modify the bonding stress, which is also influenced by the type and diameter of the bar used. In this work, the influence of the recycled fine aggregate (RFA) and types of steel bar on the steel-concrete bond was experimentally evaluated using the pullout test. Conventional concrete and recycled concrete, with RFA replacement level of 25%, were produced. Two types of steel rebars (i.e.,plain and deformed) with diameters of 10.0 and 16.0 mm were considered in this paper. The results indicate a reduction in the adhesion stress with the introduction of recycled aggregate, but this trend is influenced by the diameter of the bar used. The use of ribbed bars modifies the stress bonslip behavior, with an increase in the average bond strength, which is also observed with the reduction of the diameter of the bar

    A Broad Requirement for TLS Polymerases η and κ, and Interacting Sumoylation and Nuclear Pore Proteins, in Lesion Bypass during C. elegans Embryogenesis

    Translesion synthesis (TLS) polymerases are specialized DNA polymerases capable of inserting nucleotides opposite DNA lesions that escape removal by dedicated DNA repair pathways. TLS polymerases allow cells to complete DNA replication in the presence of damage, thereby preventing checkpoint activation, genome instability, and cell death. Here, we characterize functional knockouts for polh-1 and polk-1, encoding the Caenorhabditis elegans homologs of the Y-family TLS polymerases η and κ. POLH-1 acts at many different DNA lesions as it protects cells against a wide range of DNA damaging agents, including UV, γ-irradiation, cisplatin, and methyl methane sulphonate (MMS). POLK-1 acts specifically but redundantly with POLH-1 in protection against methylation damage. Importantly, both polymerases play a prominent role early in embryonic development to allow fast replication of damaged genomes. Contrary to observations in mammalian cells, we show that neither POLH-1 nor POLK-1 is required for homologous recombination (HR) repair of DNA double-strand breaks. A genome-wide RNAi screen for genes that protect the C. elegans genome against MMS–induced DNA damage identified novel components in DNA damage bypass in the early embryo. Our data suggest SUMO-mediated regulation of both POLH-1 and POLK-1, and point towards a previously unrecognized role of the nuclear pore in regulating TLS

    This project, though officially designated by the English Department as a creative thesis, is really a hybrid work that combines creative writing with literary criticism. The work is structured as a dream vision, a literary genre popular in the Middle Ages in which a narrator receives some form of instruction or wisdom through an allegorical dream. Examples include The Pearl, The Romance of the Rose, and Chaucer\u27s House of Fame. In this thesis, the allegorical space of the dream vision provides a platform for a series of essays structured as dialogues. These dialogues explore the aesthetics and politics of modern fantasy and supernatural literature, focusing particularly on the opposition of this literature, which often draws on ancient and medieval source material, to modern capitalist society. The discourses themselves are not strictly critical, but incorporate subjectivity, metaphor, and symbolism in their investigation of cultural texts

    Identifying nonalcoholic fatty liver disease patients with active fibrosis by measuring extracellular matrix remodeling rates in tissue and blood.

    Excess collagen synthesis (fibrogenesis) in the liver plays a causal role in the progression of nonalcoholic fatty liver disease (NAFLD). Methods are needed to identify patients with more rapidly progressing disease and to demonstrate early response to treatment. We describe here a novel method to quantify hepatic fibrogenesis flux rates both directly in liver tissue and noninvasively in blood. Twenty-one patients with suspected NAFLD ingested heavy water (2 H2 O, 50-mL aliquots) two to three times daily for 3-5 weeks prior to a clinically indicated liver biopsy. Liver collagen fractional synthesis rate (FSR) and plasma lumican FSR were measured based on 2 H labeling using tandem mass spectrometry. Patients were classified by histology for fibrosis stage (F0-F4) and as having nonalcoholic fatty liver or nonalcoholic steatohepatitis (NASH). Magnetic resonance elastography measurements of liver stiffness were also performed. Hepatic collagen FSR in NAFLD increased with advancing disease stage (e.g., higher in NASH than nonalcoholic fatty liver, positive correlation with fibrosis score and liver stiffness) and correlated with hemoglobin A1C. In addition, plasma lumican FSR demonstrated a significant correlation with hepatic collagen FSR.ConclusionUsing a well-characterized cohort of patients with biopsy-proven NAFLD, this study demonstrates that hepatic scar in NASH is actively remodeled even in advanced fibrosis, a disease that is generally regarded as static and slowly progressive. Moreover, hepatic collagen FSR correlates with established risks for fibrotic disease progression in NASH, and plasma lumican FSR correlates with hepatic collagen FSR, suggesting applications as direct or surrogate markers, respectively, of hepatic fibrogenesis in humans.     Theoretical study of the hydrated Gd3+ ion: Structure, dynamics, and charge transfer

    The dynamical processes taking place in the first coordination shells of the gadolinium (III) ion are important for improving the contrast agent efficiency in magnetic-resonance imaging. An extensive study of the gadolinium (III) ion solvated by a water cluster is reported, based on molecular dynamics simulations. The AMOEBA force field [P. Y. Ren and J. W. Ponder, J. Phys. Chem. B 107, 5933 (2003)] that includes many-body polarization effects is used to describe the interactions among water molecules, and is extended here to treat the interactions between them and the gadolinium ion. In this purpose accurate ab initio calculations have been performed on Gd3+-H2O for extracting the relevant parameters. Structural data of the first two coordination shells and some dynamical properties such as the water exchange rate between the first and second coordination shells are compared to available experimental results. We also investigate the charge transfer processes between the ion and its solvent, using a fluctuating charges model fitted to reproduce electronic structure calculations on [Gd(H2O)n]3+ complexes, with n ranging from 1 to 8. Charge transfer is seen to be significant (about one electron) and correlated with the instantaneous coordination of the ion

    Genetic structure of black rat populations in a rural plague focus in Madagascar

    Correspondance: [email protected] genetic structure of reservoir populations is a key characteristic in understanding the persistence of infectious diseases in natural systems. In the Highlands of Madagascar, where plague has persisted since 1920, the black rat, Rattus rattus (L., 1758), is the sole species acting as a reservoir of the disease. Ecological surveys have shown a clear correlation between the locations of the plague-persistence area in Madagascar (above 800 m elevation) and the distribution area of one endemic plague vector, the flea Synopsyllus fonquerniei, which is found exclusively on rats living outdoors. This clear habitat segregation has led to the suggestion that R. rattus populations in the central highlands are divided into indoor- and outdoor-dwelling populations. Using eight microsatellite markers, we analysed the genetic structure of R. rattus populations living within a human plague focus in relation to habitat and geographic distance. We found that habitat by itself was not a structuring factor, unlike geographic distance. Nevertheless, the significant genotypic differentiation of R. rattus populations that was found at a fine spatial scale might relate to differences in population dynamics between rats in indoor and outdoor habitats
