Effects of working memory training in young and old adults
Many cognitive abilities, including working memory and reasoning ability, decline with progressing age. In this study, we investigated whether four weeks of intensive working memory training would enhance working memory and reasoning performance in an age-comparative setting. Groups of 34 young (19-36years) and 27 older (62-77years) adults practiced tasks representing the three functional categories in the facet model of working memory capacity: storage and processing, relational integration, and supervision. The data were compared to those of a young and an old active control group who practiced tasks with low working memory demands. A cognitive test battery measuring near and far transfer was administered before and after training. Both age groups showed increased working memory performance in the trained tasks and in one structurally similar, but nontrained, task. Young adults also improved in a task measuring word-position binding in working memory. However, we found no far transfer to reasoning in either age group. The results provide evidence that working memory performance can be improved throughout the life span. However, in contrast to a previous study in which each facet of working memory capacity was trained separately, the present study showed that training multiple functional categories simultaneously induces less transfe
New Perspective in the Formulation and Characterization of Didodecyldimethylammonium Bromide (DMAB) Stabilized Poly(Lactic-co-Glycolic Acid) (PLGA) Nanoparticles
Over the last few decades the establishment of nanoparticles as suitable drug carriers with the transport of drugs across biological barriers such as the gastrointestinal barrier moved into the focus of many research groups. Besides drug transport such carrier systems are well suited for the protection of drugs against enzymatic and chemical degradation. The preparation of biocompatible and biodegradable nanoparticles based on poly(lactic-co-glycolic acid) (PLGA) is intensively described in literature, while especially nanoparticles with cationic properties show a promising increased cellular uptake. This is due to the electrostatic interaction between the cationic surface and the negatively charged lipid membrane of the cells. Even though several studies achieved the successful preparation of nanoparticles stabilized with the cationic surfactants such as didodecyldimethylammonium bromide (DMAB), in most cases insufficient attention was paid to a precise analytical characterization of the nanoparticle system. The aim of the present work was to overcome this deficit by presenting a new perspective in the formulation and characterization of DMAB-stabilized PLGA nanoparticles. Therefore these nanoparticles were carefully examined with regard to particle diameter, zeta potential, the effect of variation in stabilizer concentration, residual DMAB content, and electrolyte stability. Without any steric stabilization, the DMAB-modified nanoparticles were sensitive to typical electrolyte concentrations of biological environments due to compression of the electrical double layer in conjunction with a decrease in zeta potential. To handle this problem, the present study proposed two modifications to enable electrolyte stability. Both polyvinyl alcohol (PVA) and polyethylene glycol (PEG) modified DMAB-PLGA-nanoparticles were stable during electrolyte addition. Furthermore, in contrast to unmodified DMAB-PLGA-nanoparticles and free DMAB, such modifications led to a lower cytotoxic activity against Caco-2 cells. In conclusion this study offers a closer and critical point of view on preparation, in vitro and analytical evaluation of DMAB-stabilized PLGA nanoparticles for the physiological use

Loop corrections to the sphaleron transition rate in the minimal standard model
The baryon number dissipation rate due to sphaleron transitions at high
temperatures in the minimal standard model is evaluated. We find that this rate
can be considerably suppressed by one loop contributions of bosonic and
fermionic fluctuations which are particularly important for a small mass of the
Higgs boson and a large top quark mass. Fixing the latter to its recently
stated value of 174 GeV the complete erasure of the baryon asymmetry is
prevented within the framework of the minimal standard model if the Higgs mass
is less than about 66 GeV.Comment: 11 pages (LaTex) plus 2 figures (uuencoded postscript files);

Incorporation of doxorubicin in different polymer nanoparticles and their anticancer activity
Background: Nanoparticles are under investigation as carrier systems for anticancer drugs. The expression of efflux transporters such as the ATP-binding cassette (ABC) transporter ABCB1 is an important resistance mechanism in therapy-refractory cancer cells. Drug encapsulation into nanoparticles has been shown to bypass efflux-mediated drug resistance, but there are also conflicting results. To investigate whether easy-to-prepare nanoparticles made of well-tolerated polymers may circumvent transporter-mediated drug efflux, we prepared poly(lactic-co-glycolic acid) (PLGA), polylactic acid (PLA), and PEGylated PLGA (PLGA-PEG) nanoparticles loaded with the ABCB1 substrate doxorubicin by solvent displacement and emulsion diffusion approaches and assessed their anticancer efficiency in neuroblastoma cells, including ABCB1-expressing cell lines, in comparison to doxorubicin solution.
Results: The resulting nanoparticles covered a size range between 73 and 246 nm. PLGA-PEG nanoparticle preparation by solvent displacement led to the smallest nanoparticles. In PLGA nanoparticles, the drug load could be optimised using solvent displacement at pH 7 reaching 53 µg doxorubicin/mg nanoparticle. These PLGA nanoparticles displayed sustained doxorubicin release kinetics compared to the more burst-like kinetics of the other preparations. In neuroblastoma cells, doxorubicin-loaded PLGA-PEG nanoparticles (presumably due to their small size) and PLGA nanoparticles prepared by solvent displacement at pH 7 (presumably due to their high drug load and superior drug release kinetics) exerted the strongest anticancer effects. However, nanoparticle-encapsulated doxorubicin did not display increased efficacy in ABCB1-expressing cells relative to doxorubicin solution.
Conclusion: Doxorubicin-loaded nanoparticles made by different methods from different materials displayed substantial discrepancies in their anticancer activity at the cellular level. Optimised preparation methods resulted in PLGA nanoparticles characterised by increased drug load, controlled drug release, and high anticancer efficacy. The design of drug-loaded nanoparticles with optimised anticancer activity at the cellular level is an important step in the development of improved nanoparticle preparations for anticancer therapy. Further research is required to understand under which circumstances nanoparticles can be used to overcome efflux-mediated resistance in cancer cells

First Measurement of the Ru(p,)Rh Cross Section for the p-Process with a Storage Ring
This work presents a direct measurement of the Ru()Rh cross section via a novel technique using a storage ring,
which opens opportunities for reaction measurements on unstable nuclei. A
proof-of-principle experiment was performed at the storage ring ESR at GSI in
Darmstadt, where circulating Ru ions interacted repeatedly with a
hydrogen target. The Ru()Rh cross section between 9
and 11 MeV has been determined using two independent normalization methods. As
key ingredients in Hauser-Feshbach calculations, the -ray strength
function as well as the level density model can be pinned down with the
measured () cross section. Furthermore, the proton optical potential
can be optimized after the uncertainties from the -ray strength
function and the level density have been removed. As a result, a constrained
Ru()Rh reaction rate over a wide temperature range is
recommended for -process network calculations.Comment: 10 pages, 7 figs, Accepted for publication at PR
