    STABILITY OF MEAN VALUES: APPLICATION OF THE BATES METHOD TO FIELD HOCKEY

    A limitation within biomechanics research is an insufficient number of trials are analysed. Conjecture exists over the minimum number of trials required to achieve a stable mean. This study was conducted to identify the minimum number of trials necessary for mean values of selected kinematic variables to stabilise, using the progressive standard deviation method adapted from Bates et al. (1983). Three-dimensional kinematic data of the field hockey hit was collected using a 10-camera Vicon motion analysis system and analysed using Visual 3D (C-Motion). Mean values for selected parameters were calculated and the progressive change in standard deviation assessed. Results showed the number of trials needed for mean stabilisation is both parameter and task specific. The last threshold approach should be used to account for variability within parameters

    The influence of 3-D interfacial structure and morphology on the mechanical behavior of nanocomposites

    2-dimensional (2D) sharp interfaces with distinct boundaries demarcating an abrupt discontinuity in material properties in nanolayered composites have been studied for almost twenty years and are responsible for enhanced behaviors such as strength, radiation damage tolerance, and deformability. However, 2-D interfaces have their limitations with respect to deformability and toughness. 3D interfaces are defined as heterophase interfaces that extend out of plane into the two crystals on either side and are chemically, crystallographically, and/or topologically divergent, in three dimensions, from both crystals they join. Here, we present the mechanical behavior of two different classes of nanocomposites: 1.) nanolayered Cu/Nb containing interfaces with 3D character and 2.) Tungsten-based 3D ordered mesoporous composites consisting of a porous W scaffolding with silicon carbide or silicon nitride infill. Micropillar compression results show that the strength of Cu/Nb nanocomposites containing 3D interfaces is significantly greater than those containing 2D interfaces. Shear banding in 3D Cu/Nb is observed during pillar compression with retention of continuous layers across the shear band. We will present our recent results on deformation of such 3-D interfaces and structures, and describe this evolution mechanistically through the use of atomistic simulations.     Microbial diversity and activity in the Nematostella vectensis holobiont: insights from 16S rRNA gene sequencing, isolate genomes, and a pilot-scale survey of gene expression

    We have characterized the molecular and genomic diversity of the microbiota of the starlet sea anemone Nematostella vectensis, a cnidarian model for comparative developmental and functional biology and a year-round inhabitant of temperate salt marshes. Molecular phylogenetic analysis of 16S rRNA gene clone libraries revealed four ribotypes associated with N. vectensis at multiple locations and times. These associates include two novel ribotypes within the ε-Proteobacterial order Campylobacterales and the Spirochetes, respectively, each sharing 99% 16S rRNA identity with Endozoicomonas elysicola and Pseudomonas oleovorans, respectively. Species-specific PCR revealed that these populations persisted in N. vectensis asexually propagated under laboratory conditions. cDNA indicated expression of the Campylobacterales and Endozoicomonas 16S rRNA in anemones from Sippewissett Marsh, MA. A collection of bacteria from laboratory raised N. vectensis was dominated by isolates from P. oleovorans and Rhizobium radiobacter. Isolates from field-collected anemones revealed an association with Limnobacter and Stappia isolates. Genomic DNA sequencing was carried out on 10 cultured bacterial isolates representing field- and laboratory-associates, i.e., Limnobacter spp., Stappia spp., P. oleovorans and R. radiobacter. Genomes contained multiple genes identified as virulence (host-association) factors while S. stellulata and L. thiooxidans genomes revealed pathways for mixotrophic sulfur oxidation. A pilot metatranscriptome of laboratory-raised N. vectensis was compared to the isolate genomes and indicated expression of ORFs from L. thiooxidans with predicted functions of motility, nutrient scavenging (Fe and P), polyhydroxyalkanoate synthesis for carbon storage, and selective permeability (porins). We hypothesize that such activities may mediate acclimation and persistence of bacteria in a N. vectensis holobiont defined by both internal and external gradients of chemicals and nutrients in a dynamic coastal habitat.Massachusetts Institute of Technology.     Direct Observation of Broadband Coating Thermal Noise in a Suspended Interferometer

    We have directly observed broadband thermal noise in silica/tantala coatings in a high-sensitivity Fabry-Perot interferometer. Our result agrees well with the prediction based on indirect, ring-down measurements of coating mechanical loss, validating that method as a tool for the development of advanced interferometric gravitational-wave detectors.Comment: Final version synchronized with publication in Phys.     Developmental changes in word recognition threshold from two to five years of age in children with different middle ear status

    The aims were to: (1) provide word recognition thresholds (WRTs) at 31, 43, and 61 months of age; (2) investigate developmental changes over time; (3) investigate the relationship between OME and WRT, and (4) investigate the relationship between WRT and hearing thresholds. Around 1000 children were tested longitudinally as part of the ALSPAC study, using an adaptive measure of word recognition in quiet. Mean WRTs were 28, 23, and 23 dB (A) at 31, 43, and 61 months, respectively. Normal auditory development is associated with a mean improvement in WRT of 5 dB between age 31 and 61 months. There was a mean increase in WRT of 5 dB and 15 dB when OME was present in one and two ears, respectively. Thus, both unilateral and bilateral OME results in a detrimental effect on hearing ability for speech. Additionally, early and ‘persistent’ OME is associated with greater disability. However by 61 months, previous OME status was not significant. To our knowledge, this is the largest longitudinal study reporting WRT in preschool children with different middle ear status

    High-fat diet exacerbates SIV pathogenesis and accelerates disease progression

American Society for Clinical Investigation.Consuming a high-fat diet (HFD) is a risk factor for obesity and diabetes; both of these diseases are also associated with systemic inflammation, similar to HIV infection. A HFD induces intestinal dysbiosis and impairs liver function and coagulation, with a potential negative impact on HIV/SIV pathogenesis. We administered a HFD rich in saturated fats and cholesterol to nonpathogenic (African green monkeys) and pathogenic (pigtailed macaques) SIV hosts. The HFD had a negative impact on SIV disease progression in both species. Thus, increased cell-associated SIV DNA and RNA occurred in the HFD-receiving nonhuman primates, indicating a potential reservoir expansion. The HFD induced prominent immune cell infiltration in the adipose tissue, an important SIV reservoir, and heightened systemic immune activation and inflammation, altering the intestinal immune environment and triggering gut damage and microbial translocation. Furthermore, HFD altered lipid metabolism and HDL oxidation and also induced liver steatosis and fibrosis. These metabolic disturbances triggered incipient atherosclerosis and heightened cardiovascular risk in the SIV-infected HFD-receiving nonhuman primates.     Characterisation of aggregates of cyclodextrin-drug complexes using Taylor Dispersion Analysis

    There is a need to understand the nature of aggregation of cyclodextrins (CDs) with guest molecules in increasingly complex formulation systems. To this end an innovative application of Taylor dispersion analysis (TDA) and comparison with dynamic light scattering (DLS) have been carried out to probe the nature of ICT01-2588 (ICT-2588), a novel tumor-targeted vascular disrupting agent, in solvents including a potential buffered formulation containing 10% hydroxypropyl-β-cyclodextrin. The two hydrodynamic sizing techniques give measurement responses are that fundamentally different for aggregated solutions containing the target molecule, and the benefits of using TDA in conjunction with DLS are that systems are characterised through measurement of both mass- and z-average hydrodynamic radii. Whereas DLS measurements primarily resolve the large aggregates of ICT01-2588 in its formulation medium, methodology for TDA is described to determine the size and notably to quantify the proportion of monomers in the presence of large aggregates, and at the same time measure the formulation viscosity. Interestingly TDA and DLS have also distinguished between aggregate profiles formed using HP-β-CD samples from different suppliers. The approach is expected to be widely applicable to this important class of drug formulations where drug solubility is enhanced by cyclodextrin and other excipients
