    The melanoma-associated antigen 1 (MAGEA1) protein stimulates the E3 ubiquitin-ligase activity of TRIM31 within a TRIM31-MAGEA1-NSE4 complex

    The MAGE (Melanoma-associated antigen) protein family members are structurally related to each other by a MAGEhomology domain comprised of 2 winged helix motifs WH/A and WH/B. This family specifically evolved in placental mammals although single homologs designated NSE3 (non-SMC element) exist in most eukaryotes. NSE3, together with its partner proteins NSE1 and NSE4 form a tight subcomplex of the structural maintenance of chromosomes SMC5–6 complex. Previously, we showed that interactions of the WH/B motif of the MAGE proteins with their NSE4/EID partners are evolutionarily conserved (including the MAGEA1-NSE4 interaction). In contrast, the interaction of the WH/A motif of NSE3 with NSE1 diverged in the MAGE paralogs. We hypothesized that the MAGE paralogs acquired new RING-finger containing partners through their evolution and form MAGE complexes reminiscent of NSE1-NSE3-NSE4 trimers. In this work, we employed the yeast 2-hybrid system to screen a human RING-finger protein library against several MAGE baits. We identified a number of potential MAGE-RING interactions and confirmed several of them (MDM4, PCGF6, RNF166, TRAF6, TRIM8, TRIM31, TRIM41) in co-immunoprecipitation experiments. Among these MAGE-RING pairs, we chose to examine MAGEA1-TRIM31 in detail and showed that both WH/A and WH/B motifs of MAGEA1 bind to the coiled-coil domain of TRIM31 and that MAGEA1 interaction stimulates TRIM31 ubiquitin-ligase activity. In addition, TRIM31 directly binds to NSE4, suggesting the existence of a TRIM31-MAGEA1-NSE4 complex reminiscent of the NSE1-NSE3-NSE4 trimer. These results suggest that MAGEA1 functions as a co-factor of TRIM31 ubiquitin-ligase and that the TRIM31-MAGEA1-NSE4 complex may have evolved from an ancestral NSE1-NSE3-NSE4 complex

The research involved 56 testers who provided detailed feedback. To improve the communication skills of the participants, it is necessary to repeat the simulations and analyze the individual aspects of the communication in depth

    Sunflower Plants as Bioindicators of Environmental Pollution with Lead (II) Ions

    In this study, the influence of lead (II) ions on sunflower growth and biochemistry was investigated from various points of view. Sunflower plants were treated with 0, 10, 50, 100 and/or 500 ÎĽM Pb-EDTA for eight days. We observed alterations in growth in all experimental groups compared with non-treated control plants. Further we determined total content of proteins by a Bradford protein assay. By the eighth day of the experiment, total protein contents in all treated plants were much lower compared to control. Particularly noticeable was the loss of approx. 8 ÎĽg/mL or 15 ÎĽg/mL in shoots or roots of plants treated with 100 mM Pb-EDTA. We also focused our attention on the activity of alanine transaminase (ALT), aspartate transaminase (AST) and urease. Activity of the enzymes increased with increasing length of the treatment and applied concentration of lead (II) ions. This increase corresponds well with a higher metabolic activity of treated plants. Contents of cysteine, reduced glutathione (GSH), oxidized glutathione (GSSG) and phytochelatin 2 (PC2) were determined by high performance liquid chromatography with electrochemical detection. Cysteine content declined in roots of plants with the increasing time of treatment of plants with Pb-EDTA and the concentration of toxic substance. Moreover, we observed ten times higher content of cysteine in roots in comparison with shoots. The observed reduction of cysteine content probably relates with its utilization for biosynthesis of GSH and phytochelatins, because the content of GSH and PC2 was similar in roots and shoots and increased with increased treatment time and concentration of Pb-EDTA. Moreover, we observed oxidative stress caused by Pb-EDTA in roots where the GSSG/GSH ratio was about 0.66. In shoots, the oxidative stress was less distinctive, with a GSSG/GSH ratio 0.14. We also estimated the rate of phytochelatin biosynthesis from the slope of linear equations plotted with data measured in the particular experimental group. The highest rate was detected in roots treated with 100 ÎĽM of Pb-EDTA. To determine heavy metal ions many analytical instruments can be used, however, most of them are only able to quantify total content of the metals. This problem can be overcome using laser induced breakdown spectroscopy, because it is able to provide a high spatial-distribution of metal ions in different types of materials, including plant tissues.     This bachelor thesis focuses on the mobile devices market and the possibilities of their use for presentation of key business data using reporting applications. The goal of the thesis is to provide a comprehensive view of the mobile devices market as well as available reporting applications of commercial suppliers who play a crucial role in this segment. Then compare these applications using a multi-criteria evaluation based on a list of criteria and determine the most suitable application that has received the highest rating in this comparison. The final output is a table of compared applications with the ratings assigned to the individual criteria. The thesis serves as a basic orientation in the mobile reporting applications market. Application comparison results can be a useful source of information for companies planning to integrate them into their enterprise information system

    Diplomová práce se zaměřuje na analýzu využití agilních metodik řízení projektů ve zkoumané bankovní společnosti. Cílem práce je na základě dotazníkového šetření identifikovat hlavní přínosy a omezení agilně řízených projektů ve vybrané bankovní společnosti procházející agilní transformací. Dalším cílem je z výsledků šetření vytvořit sadu doporučení a návrhů na možná zlepšení, která poté může být využita i pro jiné společnosti, procházející agilní transformací. Dílčím cílem je poté popis samotného procesu agilní transformace uvedené bankovní společnosti.Teoretická část se věnuje vysvětlení klíčových pojmů týkajících se agilních metodik, dále popisu procesu agilní transformace bankovní společnosti, způsobu rozdělení týmů a jejich fungování, spolu s používanými praktikami. Praktická část zahrnuje popis použité metody kvantitativního šetření a sestavení otázek pro jeho realizaci, spolu s výběrem vhodných respondentů. Na základě provedeného šetření jsou vyhodnoceny jeho výsledky, zjištěny přínosy a omezení vyplývající z agilní transformace společnosti, a poté vytvořena sada návrhů a doporučení pro zlepšení fungování agilních týmů.This diploma thesis focuses on the analysis of the use of agile project management methodologies in the studied banking company. The main goal of this thesis is to identify the main benefits and limitations of agile managed projects in a selected banking company undergoing agile transformation, on the grounds of a questionnaire survey. Another goal is to create a set of recommendations and suggestions for possible improvements from the results of the survey, which can then be used for other companies undergoing agile transformation. A partial goal is then to describe the very process of agile transformation of the banking company.The theoretical part is devoted to the explanation of key concepts related to agile methodologies, as well as a description of the process of agile transformation of a banking company, the way teams are divided and their functioning, together with the practices used. The practical part includes a description of the used method of quantitative research, the compilation of questions for this research, along with the selection of suitable respondents. Based on the survey, its results are evaluated, the benefits and limitations resulting from the agile transformation of the company are identified, and then a set of proposals and recommendations for improving the functioning of agile teams is created
