    An investigation of factors leading to the reluctance of SaaS ERP adoption in Namibian SMEs

    This paper aims to contribute to the growing study of Cloud ERP in small and medium enterprises (SMEs). A qualitative study was done in Namibian SMEs to investigate factors that lead to the reluctance of Cloud ERP in SMEs in developing countries. Data was collected from fourteen different SMEs. A Diffusion of Innovation (DoI) framework was chosen to understand how this relatively new technology is being adopted. The study has found attitude towards change, satisfaction with the existing system, lack of knowledge, compatibility issues, unreliable internet connectivity and data security concerns as the main factors that lead to this reluctance. Furthermore, it has also found that perceived low cost has a positive effect on the intention to adopt cloud ERP. The study concludes that the presence of Cloud ERP providers will encourage Cloud ERP adoption, as there seems to be lack of knowledge among prospective adopters. In addition, with unreliable internet connectivity concerns, these providers can consider partnering up with internet service providers to provide affordable reliable internet services

    Molecular profile of the rat peri-infarct region four days after stroke: Study with MANF

    The peri-infarct region after ischemic stroke is the anatomical location for many of the endogenous recovery processes, and the molecular events in the peri-infarct region remain poorly characterized. In this study, we examine the molecular profile of the peri-infarct region on post-stroke day four, time when reparative processes are ongoing. We used a multiomics approach, involving RNA sequencing, and mass spectrometry-based proteomics and metabolomics to characterize molecular changes in the peri-infarct region. We also took advantage of our previously developed method to express transgenes in the peri-infarct region where self-complementary adeno-associated virus (AAV) vectors were injected into the brain parenchyma on post-stroke day 2. We have previously used this method to show that mesencephalic astrocyte-derived neurotrophic factor (MANF) enhances functional recovery from stroke and recruits phagocytic cells to the peri-infarct region. Here, we first analyzed the effects of stroke to the peri-infarct region on post-stroke day 4 in comparison to sham-operated animals, finding that stroke induced changes in 3345 transcripts, 341 proteins, and 88 metabolites. We found that after stroke genes related to inflammation, proliferation, apoptosis, and regeneration were upregulated, whereas genes encoding neuroactive ligand receptors and calcium-binding proteins were downregulated. In proteomics, we detected upregulation of proteins related to protein synthesis and downregulation of neuronal proteins. Metabolomic studies indicated that in after stroke tissue there is increase in saccharides, sugar phosphates, ceramides and free fatty acids and decrease of adenine, hypoxantine, adenosine and guanosine. We then compared the effects of post-stroke delivery AAV1-MANF delivery to AAV1-eGFP (enhanced green fluorescent protein). MANF administration increased the expression of 77 genes, most of which were related to immune response. In proteomics, MANF administration reduced S100A8 and S100A9 protein levels. In metabolomics, no significant differences between MANF and eGFP treatment were detected, but relative to sham surgery group, most of the changes in lipids were significant in the AAV-eGFP group only. This work describes the molecular profile of the peri-infarct region during recovery from ischemic stroke, and establishes a resource for further stroke studies. These results provide further support for parenchymal MANF as a modulator of phagocytic function.Peer reviewe

    De novo assembly of genomes from long sequence reads reveals uncharted territories of Propionibacterium freudenreichii

    Background: Propionibacterium freudenreichii is an industrially important bacterium granted the Generally Recognized as Safe (the GRAS) status, due to its long safe use in food bioprocesses. Despite the recognized role in the food industry and in the production of vitamin B12, as well as its documented health-promoting potential, P. freudenreichii remained poorly characterised at the genomic level. At present, only three complete genome sequences are available for the species. Results: We used the PacBio RS II sequencing platform to generate complete genomes of 20 P. freudenreichii strains and compared them in detail. Comparative analyses revealed both sequence conservation and genome organisational diversity among the strains. Assembly from long reads resulted in the discovery of additional circular elements: two putative conjugative plasmids and three active, lysogenic bacteriophages. It also permitted characterisation of the CRISPR-Cas systems. The use of the PacBio sequencing platform allowed identification of DNA modifications, which in turn allowed characterisation of the restriction-modification systems together with their recognition motifs. The observed genomic differences suggested strain variation in surface piliation and specific mucus binding, which were validated by experimental studies. The phenotypic characterisation displayed large diversity between the strains in ability to utilise a range of carbohydrates, to grow at unfavourable conditions and to form a biofilm. Conclusion: The complete genome sequencing allowed detailed characterisation of the industrially important species, P. freudenreichii by facilitating the discovery of previously unknown features. The results presented here lay a solid foundation for future genetic and functional genomic investigations of this actinobacterial species.Peer reviewe

    Large methane releases lead to strong aerosol forcing and reduced cloudiness

    The release of vast quantities of methane into the atmosphere as a result of clathrate destabilization is a potential mechanism for rapid amplification of global warming. Previous studies have calculated the enhanced warming based mainly on the radiative effect of the methane itself, with smaller contributions from the associated carbon dioxide or ozone increases. Here, we study the effect of strongly elevated methane (CH4) levels on oxidant and aerosol particle concentrations using a combination of chemistry-transport and general circulation models. A 10-fold increase in methane concentrations is predicted to significantly decrease hydroxyl radical (OH) concentrations, while moderately increasing ozone (O3). These changes lead to a 70 % increase in the atmospheric lifetime of methane, and an 18 % decrease in global mean cloud droplet number concentrations (CDNC). The CDNC change causes a radiative forcing that is comparable in magnitude to the longwave radiative forcing ("enhanced greenhouse effect") of the added methane. Together, the indirect CH4-O3 and CH4-OH-aerosol forcings could more than double the warming effect of large methane increases. Our findings may help explain the anomalously large temperature changes associated with historic methane releases

    Cellular, extracellular and extracellular vesicular miRNA profiles of pre-ovulatory follicles indicate signaling disturbances in polycystic ovaries

    Cell-free RNAs have the potential to act as a means of gene expression regulation between cells and are therefore used as diagnostic markers describing the state of tissue environment. The origin and functions of such RNAs in human ovarian follicle, the environment of oocyte maturation, are unclear. The current study investigates the difference in the microRNA profiles of fertile women and polycystic ovary syndrome (PCOS) patients in three compartments from the same preovulatory follicle: mural granulosa cells (MGC), cell-free follicular fluid (FF), and extracellular vesicles (EV) of the FF by small RNA sequencing. In silico analysis was used for the prediction and over-representation of targeted pathways for the detected microRNAs. PCOS follicles were distinguished from normal tissue by the differential expression of 30 microRNAs in MGC and 10 microRNAs in FF (FDR < 0.1) that commonly regulate cytokine signaling pathways. The concentration of EV-s was higher in the FF of PCOS patients (p = 0.04) containing eight differentially expressed microRNAs (p < 0.05). In addition, we present the microRNA profiles of MGC, FF, and EV in the fertile follicle and demonstrate that microRNAs loaded into EVs target mRNAs of distinct signaling pathways in comparison to microRNAs in FF. To conclude, the three follicular compartments play distinct roles in the signaling disturbances associated with PCOS

Embora esforços considerĂĄveis tenham sido dedicados com o intuito de definir "qualidade do solo", ainda nĂŁo hĂĄ um conceito amplamente aceito pela comunidade cientifica. A seleção de Ă­ndices qualitativos para definir qualidade do solo Ă© uma tarefa extremamente difĂ­cil, e diversas propriedades quĂ­micas, fĂ­sicas e biolĂłgicas tem sido sugeridas como potenciais indicadores. A matĂ©ria orgĂąnica do solo estĂĄ associada com processos quĂ­micos, fĂ­sicos e biolĂłgicos no solo, e, portanto, Ă© considerada um dos melhores indicadores de qualidade do solo. O manejo do solo pode influenciar significativamente a dinĂąmica do carbono orgĂąnico e o ciclo de N, P, e S. Entretanto, mudanças na concentração total da matĂ©ria organica em resposta ao manejo pode ser dificil de ser detectada devido Ă  variabilidade natural do solo. Quando comparada com a matĂ©ria orgĂąnica total do solo, a fração mais prontamente disponĂ­vel, como o carbono orgĂąnico dissolvido (COD), Ă© mais sensĂ­vel Ă s mudanças no manejo do solo a curto e mĂ©dio prazo e, portanto, pode ser utilizada como indicador fundamental de qualidade do solo ou das alteraçÔes das condiçÔes naturais. Considerable attempts have been made to define soil quality, but a general concept has not yet been accepted by the scientific community. The selection of quantitative indices for soil quality is extremely difficult, and a considerable number of chemical, physical, and biochemical properties have been suggested as potential indicators of soil quality. Because soil organic matter (SOM) can be associated with different soil chemical, physical and biological processes, it has been widely considered as one of the best soil quality indicator. Land use can significantly influence dynamics of organic carbon and N, P, and S cycle. However, changes in total soil organic carbon (SOC) contents in response to land use may be difficult to detect because of the natural soil variability. In the short to medium term, biological properties and readily decomposable fractions of SOC, such as dissolved organic carbon (DOC), are much more sensitive to soil management than is SOM as a whole, and can be used as a key indicator of soil natural functions. Despite the fact that labile C accounts for a small portion of the total organic matter in the soils, DOC is the most mobile and important C-source for microorganisms, and can easily reflect the effects of land use on soil quality. Although several methods are used to characterize DOC, the factors influencing mineralization and bioavailability of elements associated with organic matter (N, P, and S) remains unclear. Future research should focus on the processes that govern DOC and nutrient dynamics and how they affect soil quality

    Physical activity levels as a quantifier in police officers and cadets

    Objectives: The aim of the present study was to determine the physical activity levels of active duty police officers and police academy cadets in different life domains and intensities. These parameters were treated as potential quantifiers that could be used when assessing individuals preparing for work as future police officers. Material and Methods: The study recruited 153 active police officers and 176 cadets attending a police academy and administered a diagnostic survey, the long-form version of the International Physical Activity Questionnaire, while in the statistical analysis the Student's t-test for independent groups was applied. Results: It was determined that police officers present high physical activity levels within the work domain, which are developed from initial training at a police academy and then throughout their police career. Conclusions: Such data are important in the light of the role police officers play in public safety as well as the prominence of physical activity within a particular profession and how it can be targeted and tailored to their needs
