Diversidad genética de papas nativas (Solanum spp.) del distrito de Vilcashuamán, Ayacucho- Perú, mediante AFLP
En este presente trabajo, la diversidad genética de 30 morfotipos de papas nativas de Vilcashuamán (Ayacucho) fue evaluada mediante la técnica de polimorfismo de longitud de fragmentos amplificados (AFLP). La extracción de ADN se realizó con el método de CTAB modificado, usando hojas frescas de plantas de cuatro semanas de cultivo en invernadero. Partiendo de 200 mg de tejido vegetal se logró obtener entre 300 a 500 ng/μL de ADN de buena calidad. La digestión enzimática del ADN se realizó utilizando EcoRI y MseI, y se emplearon 12 combinaciones de primers, de las cuales se eligieron las dos combinaciones más polimórficas (E13 – M49 y E38 – M49). El análisis estadístico se realizó con el programa NTSYs 2.10 usando el coeficiente de Simple Matching logrando obtener valores de PIC (índice de contenido polimórfico) de 0.45 y 0.40 para las combinaciones E38 – M49 y E13 – M49, respectivamente. En total se lograron identificar 68 bandas claramente diferenciables, de las cuales el 55.8% fueron bandas polimórficas. El análisis de agrupamiento según el algoritmo UPGMA originó un dendograma con un índice de correlación cofenética de r= 0.7; a un coeficiente de similitud de 0.6; se establecieron ocho grupos genéticos y a un coeficiente de 1 no se encontraron morfotipos duplicados. Los resultados obtenidos demuestran el alto poder informativo del AFLP y la alta variabilidad de las papas nativas estudiadas

The Iron abundance in Galactic Planetary Nebulae
We constrain the iron abundance in a sample of 33 low-ionization Galactic
planetary nebulae (PNe) using [Fe III] lines and correcting for the
contribution of higher ionization states with ionization correction factors
(ICFs) that take into account uncertainties in the atomic data. We find very
low iron abundances in all the objects, suggesting that more than 90% of their
iron atoms are condensed onto dust grains. This number is based on the solar
iron abundance and implies a lower limit on the dust-to-gas mass ratio, due
solely to iron, of M_dust/M_gas>1.3x10^{-3} for our sample. The depletion
factors of different PNe cover about two orders of magnitude, probably
reflecting differences in the formation, growth, or destruction of their dust
grains. However, we do not find any systematic difference between the gaseous
iron abundances calculated for C-rich and O-rich PNe, suggesting similar iron
depletion efficiencies in both environments. The iron abundances of our sample
PNe are similar to those derived following the same procedure for a group of 10
Galactic H II regions. These high depletion factors argue for high depletion
efficiencies of refractory elements onto dust grains both in molecular clouds
and AGB stars, and low dust destruction efficiencies both in interstellar and

"Counterpart" method for abundance determinations in HII regions
We suggest a new way of the determining abundances and electron temperatures
in HII regions from strong emission lines. Our approach is based on the
standard assumption that HII regions with similar intensities of strong
emission lines have similar physical properties and abundances. A "counterpart"
for a studied HII region may be chosen among HII regions with well-measured
abundances (reference HII regions) by comparison of carefully chosen
combinations of strong line intensities. Then the abundances in the
investigated HII region can be assumed to be the same as that in its
counterpart. In other words, we suggest to determine the abundances in HII
regions "by precedent". To get more reliable abundances for the considered HII
region, a number of reference HII regions is selected and then the abundances
in the target HII region is estimated through extra-/interpolation. We will
refer to this method of abundance determination as the counterpart method or,
for brevity, the C method. We define a sample of reference HII regions and
verify the validity of the C method. We find that this method produces reliable
abundances. Finally, the C method is used to obtain the radial abundance
distributions in the extended discs of the spiral galaxies M83, NGC4625 and NGC

The VVV Templates Project. Towards an Automated Classification of VVV Light-Curves. I. Building a database of stellar variability in the near-infrared
Context. The Vista Variables in the V\'ia L\'actea (VVV) ESO Public Survey is
a variability survey of the Milky Way bulge and an adjacent section of the disk
carried out from 2010 on ESO Visible and Infrared Survey Telescope for
Astronomy (VISTA). VVV will eventually deliver a deep near-IR atlas with
photometry and positions in five passbands (ZYJHK_S) and a catalogue of 1-10
million variable point sources - mostly unknown - which require
classifications. Aims. The main goal of the VVV Templates Project, that we
introduce in this work, is to develop and test the machine-learning algorithms
for the automated classification of the VVV light-curves. As VVV is the first
massive, multi-epoch survey of stellar variability in the near-infrared, the
template light-curves that are required for training the classification
algorithms are not available. In the first paper of the series we describe the
construction of this comprehensive database of infrared stellar variability.
Methods. First we performed a systematic search in the literature and public
data archives, second, we coordinated a worldwide observational campaign, and
third we exploited the VVV variability database itself on (optically)
well-known stars to gather high-quality infrared light-curves of several
hundreds of variable stars. Results. We have now collected a significant (and
still increasing) number of infrared template light-curves. This database will
be used as a training-set for the machine-learning algorithms that will
automatically classify the light-curves produced by VVV. The results of such an

An evaluation of potential reference genes for stability of expression in two salmonid cell lines after infection with either Piscirickettsia salmonis or IPNV
<p>Abstract</p> <p>Background</p> <p>Due to the limited number of species specific antibodies against fish proteins, differential gene expression analyses are vital for the study of host immune responses. Quantitative real-time reverse transcription PCR (qRT-PCR) is one of the most powerful tools for this purpose. Nevertheless, the accuracy of the method will depend on the careful selection of genes whose expression are stable and can be used as internal controls for a particular experimental setting.</p> <p>Findings</p> <p>The expression stability of five commonly used housekeeping genes [beta-actin (<it>ACTB</it>), elongation factor 1-alpha (<it>EF1A</it>), ubiquitin (<it>UBQ</it>), glyceraldehyd-3-phosphate dehydrogenase (<it>GAPDH</it>) and tubulin alpha (<it>TUBA</it>)] were monitored in salmonid cell lines CHSE-214 and RTS11 after infection with two of the most fastidious fish pathogens, the facultative bacterium <it>Piscirickettsia salmonis </it>and the aquabirnavirus IPNV (Infectious Pancreatic Necrosis Virus). After geNorm analysis, <it>UBQ </it>and <it>EF1A </it>appeared as the most stable, although <it>EF1A </it>was slightly upregulated at late stages of <it>P. salmonis </it>infection in RTS11. <it>ACTB </it>instead, showed a good performance in each case, being always considered within the three most stable genes of the panel. In contrast, infection-dependent differential regulation of <it>GAPDH </it>and <it>TUBA </it>was also demonstrated.</p> <p>Conclusion</p> <p>Based on the data presented here with the cell culture models CHSE-214 and RTS11, we suggest the initial choice of <it>UBQ</it>, <it>ACTB </it>and <it>EF1A </it>as reference genes in qRT-PCR assays for studying the effect of <it>P. salmonis </it>and IPNV on the host immune response.</p
Biodiversity of Fusarium species in Mexico associated with ear rot in maize, and their identification using a phylogenetic approach
Fusariumproliferatum, F. subglutinans, and F. verticillioides are known causes of ear and kernel rot in maize worldwide. In Mexico, only F. verticillioides and F. subglutinans, have been reported previously as causal agents of this disease. However, Fusarium isolates with different morphological characteristics to the species that are known to cause this disease were obtained in the Highland-Valley region of this country from symptomatic and symptomless ears of native and commercial maize genotypes. Moreover, while the morphological studies were not sufficient to identify the correct taxonomic position at the species level, analyses based in the Internal Transcribed Spacer region and the Nuclear Large Subunit Ribosomal partial sequences allowed for the identification of F. subglutinans, F. solani, and F. verticillioides, as well as four species (F. chlamydosporum, F. napiforme, F. poae, and F. pseudonygamai) that had not previously been reported to be associated with ear rot. In addition, F. napiforme and F. solani were absent from symptomless kernels. Phylogenetic analysis showed genetic changes in F. napiforme, and F. pseudonygamai isolates because they were not true clones, and probably constitute separate sibling species. The results of this study suggest that the biodiversity of Fusarium species involved in ear rot in Mexico is greater than that reported previously in other places in the world. This new knowledge will permit a better understanding of the relationship between all the species involved in ear rot disease and their relationship with maize
Anatomical and Physiological Plasticity in Leymus chinensis (Poaceae) along Large-Scale Longitudinal Gradient in Northeast China
Although it has been widely accepted that global changes will pose the most important constrains to plant survival and distribution, our knowledge of the adaptive mechanism for plant with large-scale environmental changes (e.g. drought and high temperature) remains limited.An experiment was conducted to examine anatomical and physiological plasticity in Leymus chinensis along a large-scale geographical gradient from 115° to 124°E in northeast China. Ten sites selected for plant sampling at the gradient have approximately theoretical radiation, but differ in precipitation and elevation. The significantly increasing in leaf thickness, leaf mass per area, vessel and vascular diameters, and decreasing in stoma density and stoma index exhibited more obvious xerophil-liked traits for the species from the moist meadow grassland sites in contrast to that from the dry steppe and desert sites. Significant increase in proline and soluble sugar accumulation, K(+)/Na(+) for the species with the increasing of stresses along the gradient showed that osmotic adjustment was enhanced.Obvious xerophytic anatomical traits and stronger osmotic adjustment in stress conditions suggested that the plants have much more anatomical and physiological flexibilities than those in non-stress habitats along the large-scale gradient
Calbindin-D32k Is Localized to a Subpopulation of Neurons in the Nervous System of the Sea Cucumber Holothuria glaberrima (Echinodermata)
Members of the calbindin subfamily serve as markers of subpopulations of neurons within the vertebrate nervous system. Although markers of these proteins are widely available and used, their application to invertebrate nervous systems has been very limited. In this study we investigated the presence and distribution of members of the calbindin subfamily in the sea cucumber Holothuria glaberrima (Selenka, 1867). Immunohistological experiments with antibodies made against rat calbindin 1, parvalbumin, and calbindin 2, showed that these antibodies labeled cells and fibers within the nervous system of H. glaberrima. Most of the cells and fibers were co-labeled with the neural-specific marker RN1, showing their neural specificity. These were distributed throughout all of the nervous structures, including the connective tissue plexi of the body wall and podia. Bioinformatics analyses of the possible antigen recognized by these markers showed that a calbindin 2-like protein present in the sea urchin Strongylocentrotus purpuratus, corresponded to the calbindin-D32k previously identified in other invertebrates. Western blots with anti-calbindin 1 and anti-parvalbumin showed that these markers recognized an antigen of approximately 32 kDa in homogenates of radial nerve cords of H. glaberrima and Lytechinus variegatus. Furthermore, immunoreactivity with anti-calbindin 1 and anti-parvalbumin was obtained to a fragment of calbindin-D32k of H. glaberrima. Our findings suggest that calbindin-D32k is present in invertebrates and its sequence is more similar to the vertebrate calbindin 2 than to calbindin 1. Thus, characterization of calbindin-D32k in echinoderms provides an important view of the evolution of this protein family and represents a valuable marker to study the nervous system of invertebrates
