    Two Lane Traffic Simulations using Cellular Automata

    We examine a simple two lane cellular automaton based upon the single lane CA introduced by Nagel and Schreckenberg. We point out important parameters defining the shape of the fundamental diagram.     A multicenter, open-label, long-term safety and tolerability study of DFN-02, an intranasal spray of sumatriptan 10 mg plus permeation enhancer DDM, for the acute treatment of episodic migraine.

    BackgroundDFN-02 is a novel intranasal spray formulation composed of sumatriptan 10 mg and a permeation-enhancing excipient comprised of 0.2% 1-O-n-Dodecyl-β-D-Maltopyranoside (DDM). This composition of DFN-02 allows sumatriptan to be rapidly absorbed into the systemic circulation and exhibit pharmacokinetics comparable to subcutaneously administered sumatriptan. Rapid rate of absorption is suggested to be important for optimal efficacy. The objective of this study was to evaluate the safety and tolerability of DFN-02 (10 mg) in the acute treatment of episodic migraine with and without aura over a 6-month period based on the incidence of treatment-emergent adverse events and the evaluation of results of clinical laboratory tests, vital signs, physical examination, and electrocardiograms.MethodsThis was a multi-center, open-label, repeat-dose safety study in adults with episodic migraine with and without aura. Subjects diagnosed with migraine with or without aura according to the criteria set forth in the International Classification of Headache Disorders, 2nd edition, who experienced 2 to 6 attacks per month with fewer than 15 headache days per month and at least 48 headache-free hours between attacks, used DFN-02 to treat their migraine attacks acutely over the course of 6 months.ResultsA total of 173 subjects was enrolled, 167 (96.5%) subjects used at least 1 dose of study medication and were evaluable for safety, and 134 (77.5%) subjects completed the 6-month study. A total of 2211 migraine attacks was reported, and 3292 doses of DFN-02 were administered; mean per subject monthly use of DFN-02 was 3.6 doses. Adverse events were those expected for triptans, as well as for nasally administered compounds. No new safety signals emerged. Dysgeusia and application site pain were the most commonly reported treatment-emergent adverse events over 6 months (21% and 30.5%, respectively). Most of the treatment-emergent adverse events were mild. There were 5 serious adverse events, all considered unrelated to the study medication; the early discontinuation rate was 22.5% over the 6-month treatment period.ConclusionDFN-02 was shown to be well tolerated when used over 6 months to treat episodic migraine acutely

    Gene Expression Profiling and Network Analysis Reveals Lipid and Steroid Metabolism to Be the Most Favored by TNFα in HepG2 Cells

    BACKGROUND: The proinflammatory cytokine, TNFalpha, is a crucial mediator of the pathogenesis of several diseases, more so in cases involving the liver wherein it is critical in maintaining liver homeostasis since it is a major determiner of hepatocyte life and death. Gene expression profiling serves as an appropriate strategy to unravel the underlying signatures to envisage such varied responses and considering this, gene transcription profiling was examined in control and TNFalpha treated HepG2 cells. METHODS AND FINDINGS: Microarray experiments between control and TNFalpha treated HepG2 cells indicated that TNFalpha could significantly alter the expression profiling of 140 genes; among those up-regulated, several GO (Gene Ontology) terms related to lipid and fat metabolism were significantly (p<0.01) overrepresented indicating a global preference of fat metabolism within the hepatocyte and those within the down-regulated dataset included genes involved in several aspects of the immune response like immunoglobulin receptor activity and IgE binding thereby indicating a compromise in the immune defense mechanism(s). Conserved transcription factor binding sites were identified in identically clustered genes within a common GO term and SREBP-1 and FOXJ2 depicted increased occupation of their respective binding elements in the presence of TNFalpha. The interacting network of "lipid metabolism, small molecule biochemistry" was derived to be significantly overrepresented that correlated well with the top canonical pathway of "biosynthesis of steroids". CONCLUSIONS: TNFalpha alters the transcriptome profiling within HepG2 cells with an interesting catalog of genes being affected and those involved in lipid and steroid metabolism to be the most favored. This study represents a composite analysis of the effects of TNFalpha in HepG2 cells that encompasses the altered transcriptome profiling, the functional analysis of the up- and down- regulated genes and the identification of conserved transcription factor binding sites. These could possibly determine TNFalpha mediated alterations mainly the phenotypes of hepatic steatosis and fatty liver associated with several hepatic pathological states

    Assessment of medical students by OSPE method in pathology

    ABSTRACT: Objective structured practical examination (OSPE) is a practical exam system wherein there is a series of stations at which students work through tasks designed to test various skills and are tested using agreed check lists with observers sitting at some of the stations. It has been found to be objective, valid and reliable. It also eliminates examiner bias. The purpose of this study is to introduce OSPE both as an evaluation and a teaching tool and to draw attention to its advantages and disadvantages. We designed an OSPE that tested all the above objectives satisfactorily. The OSPE exam was conducted in the department of pathology during the second internal assessment of the students. This method was the compared with the conventional method of practical exam conducted during the first internal assessment of the same batch of students. The results showed that OSPE tests different desired components of competence better. It is an objective, valid and reliable method and gets rid of variation due to examiner bias. It has a better discrimination index on merit. Interpretative exercises are the most relevant part and deserve the lion&rsquo;s share of total marks. Students feel more comfortable and less stressed to perform the exam.     Cell membrane stability- an important criterion for selection of heat tolerant genotypes in wheat (Triticum aestivum L.)

    Cell membrane stability, grain filling rate, grain filling duration, canopy temperature and grain yield were used to evaluate performance of 100 diverse bread wheat (Triticum aestivum L.) genotypes under timely sown and late sown heat stress conditions for two cropping season. The genotypes differed significantly for all the traits show-ing considerable variation for improvement of characters. The genotypes WH1165 had significant high grain yield (14.6* g and 11.4g) and (11.3* g and 11.4* g) followed by cell membrane stability under timely sown and heat stress conditions, respectively indicating potential tolerance against heat stress. Correlation coefficients revealed that cell membrane stability (0.451**) and (0.639**) in timely sown and in late sown conditions, respectively were the most important trait followed by grain filling rate (0.882** and 0.744**) under timely sown and late sown conditions respec-tively. Results revealed that bread wheat genotypes which had high value of cell membrane stability had high grain yield showed potential photorespiration and high grain filling rate under heat stress condition. Twenty two genotypes WH1021, WH1155, VL803, WH787, NW1014, Raj3765, HD1869, 2042, WH1124, HD2285, WH1133, HUW234, 4066, Sonak, UP2425, UP2473, PBW503, PBW373, PBW533, SGP13, HD2643 and WH789 were identified as heat tolerant genotypes based on their relative performance in yield components, grain yield and heat susceptibility indi-ces. These genotypes were found to be ideal candidates to be used in developing heat tolerant wheat varieties. Canopy temperature, membrane thermostability and grain filling rate have also shown strong correlation with grain yield. Because of this association, these traits constitute the best available ‘tool’ for genetic improvement of wheat suitable for cultivation under heat stressed environments. Thus, these could be used as indirect selection criteria for developing heat tolerant wheat genotypes that would provide sufficient yields to meet the ever increasing wheat demand

    A comparative study of the use of tiger-specific and heterologous microsatellite markers for population genetic studies of the Bengal tiger (Panthera tigris tigris)

    Comparison of genetic diversity indices of heterologous and species-specific microsatellite loci within a species may provide a panel of appropriate markers for genetic studies, but few studies have carried out such comparisons. We examined and compared the genetic characteristics of tiger-specific and heterologous loci in eight captive Bengal tigers. The mean polymorphic information content (PIC) value of the tiger-specific microsatellite loci (n = 15) was 0.447, and the number of alleles was from 2 to 4 per locus. In comparison, the heterologous microsatellite loci (n = 15) had a mean PIC value of 0.539, and the number of alleles per locus was three to five. Our findings indicate that the heterologous markers have a higher frequency (n = 11) of polymorphic microsatellite loci and number of alleles per locus compared with tiger-specific loci. We pooled the highly polymorphic (PIC &gt; 0.5) tiger-specific loci (n = 5) and heterologous microsatellite loci (n = 11) except one and noted a higher mean observed heterozygosity and PIC values of 0.668 and 0.575, respectively, compared with the heterologous and tiger-specific loci taken alone. Using a locus selection criterion of PIC &gt; 0.5, we recommend a combined panel of 16 highly polymorphic loci for genetic studies of the wild population of the Bengal tigers and suggest that either a combination of tiger-specific and heterologous microsatellite primers or heterologous primers be used in genetic studies related to the ecology, biology, socio-biology and behavior of Bengal tigers as &gt;13 loci are needed in such studies.Keywords: Bengal tiger, highly polymorphic, tiger-specific, heterologous, microsatellite loci

    Speech and Language Characteristics in Kabuki Syndrome

    The aim of this study was to delineate the language difficulties in Kabuki Syndrome. At presentation, the child with Kabuki Syndrome had waddling gait, dolicocephaly, and frontal bossing. On administering an Indian standardized test (Linguistic Profile Test), different aspects of spontaneous language production were analysed. Receptive language skills were within normal limits, but expressive language skills showed deficits in semantic and syntactic areas. Articulation errors appeared mainly due to poor oral-motor coordination and hypotonia. A dull flat pattern of speech was characteristic to the child

    Role of methotrexate in ectopic pregnancy

    Background: Ectopic pregnancy is an acute emergency in obstetric if not timely diagnosed and timely treated. Ectopic pregnancy is leading cause of death in first trimester. Ectopic pregnancy can be managed surgically or medically. Medical management with Methotrexate administration avoids anesthesia in surgery, is cost effective and also offers success rate comparable to surgical management. Aim and objectives were to study the role of methotrexate in ectopic pregnancyMethods: This will be a retrospective observational study conducted in Obstetrics and Gynecology department of AMC MET medical college. Study group constitutes of 30 females with ectopic pregnancy. Preliminary blood investigations, ultrasonography and beta-human chorionic gonadotropin (b-hcg) level will be tested. Patients will be treated with single dose of methotrexate 50 mg/M2. Follow up b-hcg level will be done after 48 hours. Response and tolerance to methotrexate will be monitored.Results: The success rate of methotrexate therapy in our study was 83.33% (n=25) and 16.66% (n=5) required surgical intervention with tubal ruptured and abdominal pain.Conclusions: Methotrexate treatment of ectopic pregnancies is safe and effective with no major side effects. It has the advantage of tubal conservation and saves patients from surgical intervention

    An analytical study of abnormal uterine bleeding in women of child bearing age group

    Background: AUB (abnormal uterine bleeding) is defined as any variation from the normal menstrual cycle including alteration in its frequency, regularity of menses, duration of flow and amount of blood loss. In India, the reported prevalence of AUB is 17.9%. It can occur any time between menarche to menopause. A good clinician tries to recognize and identify the causative factors responsible for the disease, reverse the abnormality and induce or restore the cyclic predictable menses which should have normal volume and duration.Methods: 200 cases of AUB fitting the selection criteria were taken from OPD and gynecology ward between September 2020 to June 2021.Results: AUB was more common in the age group 41-45 years, that is, perimenopausal age group, more in multiparous women and women with previous surgeries on uterus and adnexa. Heavy menstrual bleeding was the most common complaint. Endometrial hyperplasia was the most common finding on ultrasound examination. Medical therapy was beneficial in some patients, rest had to undergo surgical intervention later.Conclusions: Transvaginal sonography is very accurate in assessing the endometrium as well as uterus and adnexa and diagnosing their abnormalities. Medical therapy is the first line of management in most cases. Dilatation and curettage should be used along with hysteroscopy for better results. LNG-IUS gives very good result in suitable cases. Hysterectomy is the final measure if everything else fails. Vaginal hysterectomy is preferred wherever possible
