    Highly Sensitive and Selective VOC Sensor Systems Based on Semiconductor Gas Sensors: How to?

    Monitoring of volatile organic compounds (VOCs) is of increasing importance in many application fields such as environmental monitoring, indoor air quality, industrial safety, fire detection, and health applications. The challenges in all of these applications are the wide variety and low concentrations of target molecules combined with the complex matrix containing many inorganic and organic interferents. This paper will give an overview over the application fields and address the requirements, pitfalls, and possible solutions for using low-cost sensor systems for VOC monitoring. The focus lies on highly sensitive metal oxide semiconductor gas sensors, which show very high sensitivity, but normally lack selectivity required for targeting relevant VOC monitoring applications. In addition to providing an overview of methods to increase the selectivity, especially virtual multisensors achieved with dynamic operation, and boost the sensitivity further via novel pro-concentrator concepts, we will also address the requirement for high-performance gas test systems, advanced solutions for operating and read-out electronic, and, finally, a cost-efficient factory and on-site calibration. The various methods will be primarily discussed in the context of requirements for monitoring of indoor air quality, but can equally be applied for environmental monitoring and other fields

    Geobacter, Anaeromyxobacter and Anaerolineae populations are enriched on anodes of root exudate-driven microbial fuel cells in rice field soil

    Plant-based sediment microbial fuel cells (PMFCs) couple the oxidation of root exudates in living rice plants to current production. We analysed the composition of the microbial community on anodes from PMFC with natural rice field soil as substratum for rice by analysing 16S rRNA as an indicator of microbial activity and diversity. Terminal restriction fragment length polymorphism (TRFLP) analysis indicated that the active bacterial community on anodes from PMFCs differed strongly compared with controls. Moreover, clones related to Deltaproteobacteria and Chloroflexi were highly abundant (49% and 21%, respectively) on PMFCs anodes. Geobacter (19%), Anaeromyxobacter (15%) and Anaerolineae (17%) populations were predominant on anodes with natural rice field soil and differed strongly from those previously detected with potting soil. In open circuit (OC) control PMFCs, not allowing electron transfer, Deltaproteobacteria (33%), Betaproteobacteria (20%), Chloroflexi (12%), Alphaproteobacteria (10%) and Firmicutes (10%) were detected. The presence of an electron accepting anode also had a strong influence on methanogenic archaea. Hydrogenotrophic methanogens were more active on PMFC (21%) than on OC controls (10%), whereas acetoclastic Methanosaetaceae were more active on OC controls (31%) compared with PMFCs (9%).     Cell sorting in a Petri dish controlled by computer vision.

    Fluorescence-activated cell sorting (FACS) applying flow cytometry to separate cells on a molecular basis is a widespread method. We demonstrate that both fluorescent and unlabeled live cells in a Petri dish observed with a microscope can be automatically recognized by computer vision and picked up by a computer-controlled micropipette. This method can be routinely applied as a FACS down to the single cell level with a very high selectivity. Sorting resolution, i.e., the minimum distance between two cells from which one could be selectively removed was 50-70 micrometers. Survival rate with a low number of 3T3 mouse fibroblasts and NE-4C neuroectodermal mouse stem cells was 66 +/- 12% and 88 +/- 16%, respectively. Purity of sorted cultures and rate of survival using NE-4C/NE-GFP-4C co-cultures were 95 +/- 2% and 62 +/- 7%, respectively. Hydrodynamic simulations confirmed the experimental sorting efficiency and a cell damage risk similar to that of normal FACS

    Multispecies Purification of Testicular Germ Cells

    Advanced methods of cellular purification are required to apply genome technology to the study of spermatogenesis. One approach, based on flow cytometry of murine testicular cells stained with Hoechst-33342 (Ho-FACS), has been extensively optimized and currently allows the isolation of 9 germ cell types. This staining technique is straightforward to implement, highly effective at purifying specific germ cell types and yields sufficient cell numbers for high throughput studies. Ho-FACS is a technique that does not require species-specific markers, but whose applicability to other species is largely unexplored. We hypothesized that, due to the similar cell physiology of spermatogenesis across mammals, Ho-FACS could be used to produce highly purified subpopulations of germ cells in mammals other than mouse. To test this hypothesis, we applied Ho-FACS to 4 mammalian species that are widely used in testis research - Rattus norvegicus, Cavia porcellus, Canis familiaris and Sus scrofa domesticus We successfully isolated 4 germ cell populations from these species with average purity of 79% for spermatocytes, and 90% for spermatids and 66% for spermatogonia. Additionally, we compare the performance of mechanical and chemical dissociation for each species, and propose an optimized gating strategy to better discriminate round and elongating spermatids in the mouse, which can potentially be applied to other species.     Global mantle flow and the development of seismic anisotropy : differences between the oceanic and continental upper mantle

The definitive version was published in Journal of Geophysical Research 112 (2007): B07317, doi:10.1029/2006JB004608.Viscous shear in the asthenosphere accommodates relative motion between Earth's surface plates and underlying mantle, generating lattice-preferred orientation (LPO) in olivine aggregates and a seismically anisotropic fabric. Because this fabric develops with the evolving mantle flow field, observations of seismic anisotropy can constrain asthenospheric flow patterns if the contribution of fossil lithospheric anisotropy is small. We use global viscous mantle flow models to characterize the relationship between asthenospheric deformation and LPO and compare the predicted pattern of anisotropy to a global compilation of observed shear wave splitting measurements. For asthenosphere >500 km from plate boundaries, simple shear rotates the LPO toward the infinite strain axis (ISA, the LPO after infinite deformation) faster than the ISA changes along flow lines. Thus we expect the ISA to approximate LPO throughout most of the asthenosphere, greatly simplifying LPO predictions because strain integration along flow lines is unnecessary. Approximating LPO with the ISA and assuming A-type fabric (olivine a axis parallel to ISA), we find that mantle flow driven by both plate motions and mantle density heterogeneity successfully predicts oceanic anisotropy (average misfit 13°). Continental anisotropy is less well fit (average misfit 41°), but lateral variations in lithospheric thickness improve the fit in some continental areas. This suggests that asthenospheric anisotropy contributes to shear wave splitting for both continents and oceans but is overlain by a stronger layer of lithospheric anisotropy for continents.     Serious Complication of Cement Augmentation for Damaged Pilot Hole

    Polymethl methacrylate (PMMA) screw reinforcement is frequently used in osteoporotic bone as well as in damaged pilot holes. However, PMMA use can be dangerous, since the amount of applied cement is uncontrolled. A 47-year-old male with traumatic cervical spondylolisthesis at C6-7 underwent anterior cervical plate fixation. During repeated drilling and tapping for false trajectory correction, a pilot hole was damaged. Although it was an unconventional method, PMMA augmentation was tried. However, PMMA was accidentally injected to the cervical spinal cord owing to lack of fluoroscopic guidance. The PMMA was surgically removed after corpectomy and durotomy. The patient had left side hemiparesis (Grade 2/5) immediately post operation. The patient improved spontaneously (Grade 4/5) except for 4th and 5th digit extension. Here, we report a rare complication of PMMA extrusion in the spinal cord during a damaged pilot hole injection, which has not previously been described
