    We developed and tested a single multiplex polymerase chain reaction (PCR) that detects enterotoxigenic, enteropathogenic, enteroinvasive, and Shiga-toxin–producing Escherichia coli. This PCR is specific, sensitive, and rapid in detecting target isolates in stool and food. Because of its simplicity, economy, and efficiency, this protocol warrants further evaluation in large, prospective studies of polymicrobial substances

    Nanoencapsulated Clove Oil Applied as an Anesthetic at Slaughtering Decreases Stress, Extends the Freshness, and Lengthens Shelf Life of Cultured Fish

    In the aquaculture industry, fish are stunned using a wide range of methods, but all of them trigger stress responses and affect the fish flesh quality. Chilled water is considered one of the most efficient methods, but even this is not a stress‐free experience for the fish. Anesthetics included in the ice slurry or in water could decrease this stress and delay the loss of flesh quality. In this work, we analyze the effect of clove oil (CO) nanoencapsulated in β‐cyclodextrins (β‐CD) (CO + β‐CD), incorporated in the stunning bath, on the stress response and the organoleptic attributes of fresh marine and freshwater fish from four economically important fish species: Atlantic salmon, European seabass, Nile tilapia, and Rainbow trout. CO + β‐CD reduces the time required to induce anesthesia, independently of water salinity, habitat or water temperature. The plasmatic glucose and cortisol levels decreased in all four species, although the concentrations of CO varied between species. Moreover, plasmatic lactate level differed between the marine and freshwater fish. The use of CO + β‐CD extended the shelf life of fish from all the species studied (by 3–7 days). In conclusion, using CO encapsulated in β‐CD for anesthetizing fish can be regarded as an improved fish‐stunning technique that reduces the anesthesia‐induction time, decreases the stress response, and extends the shelf life of fresh fish.Versión del edito

    Stark effect in a wedge-shaped quantum box

    The effect of an external applied electric field on the electronic ground state energy of a quantum box with a geometry defined by a wedge is studied by carrying out a variational calculation. This geometry could be used as an approximation for a tip of a cantilever of an atomic force microscope. We study theoretically the Stark effect as function of the parameters of the wedge: its diameter, angular aperture and thickness; as well as function of the intensity of the external electric field applied along the axis of the wedge in both directions; pushing the carrier towards the wider or the narrower parts. A confining electronic effect, which is sharper as the wedge dimensions are smaller, is clearly observed for the first case. Besides, the sign of the Stark shift changes when the angular aperture is changed from small angles to angles theta>pi. For the opposite field, the electronic confinement for large diameters is very small and it is also observed that the Stark shift is almost independent with respect to the angular aperture.Comment: 23 pages, 9 figures, 1 tabl

    Vaccination of Gilthead Seabream After Continuous Xenoestrogen Oral Exposure Enhances the Gut Endobolome and Immune Status via GPER1

    In fish culture settings, the exogenous input of steroids is a matter of concern. Recently, we unveiled that in the gilthead seabream (Sparus aurata), the G protein-coupled estrogen receptor agonist G-1 (G1) and the endocrine disruptor 17α-ethinylestradiol (EE2) are potent modulators in polyreactive antibody production. However, the integral role of the microbiota upon immunity and antibody processing in response to the effect of EE2 remains largely unexplored. Here, juvenile seabreams continuously exposed for 84 days to oral G1 or EE2 mixed in the fish food were intraperitoneally (i.p.) immune primed on day 42 with the model antigen keyhole limpet hemocyanin (KLH). A critical panel of systemic and mucosal immune markers, serum VTG, and humoral, enzymatic, and bacteriolytic activities were recorded and correlated with gut bacterial metagenomic analysis 1 day post-priming (dpp). Besides, at 15 dpp, animals received a boost to investigate the possible generation of specific anti-KLH antibodies at the systemic and mucosal interphases by the end of the trial. On day 43, EE2 but not G1 induced a significant shift in the serum VTG level of naive fish. Simultaneously, significant changes in some immune enzymatic activities in the serum and gut mucus of the EE2-treated group were recorded. In comparison, the vaccine priming immunization resulted in an attenuated profile of most enzymatic activities in the same group. The gut genes qPCR analysis exhibited a related pattern, only emphasized by a significant shift in the EE2 group’s il1b expression. The gut bacterial microbiome status underwent 16S rRNA dynamic changes in alpha diversity indices, only with the exposure to oral G1, supporting functional alterations on cellular processes, signaling, and lipid metabolism in the microbiota. By the same token, the immunization elevated the relative abundance of Fusobacteria only in the control group, while this phylum was depleted in both the treated groups. Remarkably, the immunization also promoted changes in the bacterial class Betaproteobacteria and the estrogen-associated genus Novosphingobium. Furthermore, systemic and mucosal KLH-specific immunoglobulin (Ig)M and IgT levels in the fully vaccinated fish showed only slight changes 84 days post-estrogenic oral administration. In summary, our results highlight the intrinsic relationship among estrogens, their associated receptors, and immunization in the ubiquitous fish immune regulation and the subtle but significant crosstalk with the gut endobolome.Versión del edito

    Association of myostatin, a cytokine released by muscle, with inflammation in rheumatoid arthritis

Herein we explore the association between myostatin levels and inflammatory parameters in rheumatoid arthritis (RA). One hundred twenty-seven women without rheumatic diseases and 84 women with a diagnosis of RA were assessed in a cross-sectional study. Outcomes reflecting the activity of the arthritis including Disease Activity Score (DAS28-ESR) and impairment in functioning by the Health Assessment Questionnaire-Disability Index were assessed in RA. We obtained Skeletal muscle mass index (SMI), fat-free mass index (FFMI), and fat mass index using dual-energy x-ray absorptiometry. Serum myostatin was determined by enzyme-linked immunosorbent assay. Myostatin levels were correlated with disease activity and parameters of muscle mass. The SMI was lower and concentration of myostatin was higher in RA patients than in controls (P = .008 and P < .001, respectively). Myostatin significantly positively correlated with C-reactive protein (rho = 0.48, P < .001), erythrocyte sedimentation rate (rho = 0.28, P = .009), and DAS28-ESR (rho = 0.22, P = .04), and negatively correlated with SMI (rho = −0.29, P = .008), (FFMI) (rho = −0.24, P = .027). In the multivariate logistic regression analysis, levels of myostatin remained associated with disease activity in RA (P = .027). In our study, myostatin was associated with disease activity in RA patients, suggesting a mechanistic link between myostatin, muscle wasting and inflammation in RA

    Breast cancer is recognized worldwide as a major health problem among women due to its high incidence and high mortality and morbidity rates. Breast reconstruction is an approach of great value for those patients who underwent mastectomy, impacting their quality of life and psychological stress. The deep inferior epigastric artery perforator (DIEP) flap was described as the preferred graft for breast reconstruction with an autologous flap by surgeons because it represented a decrease in complications for the time and obtained better results. DIEP flap reconstruction requires microsurgical skills as well as continuous monitoring of the patient to identify and resolve possible associated complications.

    Productive Performance of Sheep in an Agropastoral System on the Coast of Oaxaca, Mexico

    Objective: To determine the prolificacy and weight of lambs at birth (BW) in twogenotypes of hair sheep within an agropastoral system on the coast of Oaxaca, Mexico,and to evaluate the effect of two sources of selenium on the performance of lambs fromsaid system during the fattening phase.Design/Methodology/Approach: The study was carried out in an agropastoral system(coconut palm and Cynodon dactylon grass) in Río Grande, Oaxaca. Two experimentswere conducted; the first studied 58 ewes distributed in a completely randomized design(CRD) with factorial arrangement of 2x2x3, and focused on the following fixed effects:genotype, number of births, and body condition (BC). The measured variables wereprolificacy and BW. The second experiment studied 23 lambs of 4 months in age thatwere offspring of the females in experiment 1. They were distributed in a CRD withthree treatments: T1, without selenium provided; T2, with barium selenate provided; T3,with selenomethionine provided. The dependent variables were daily weight gain(DWG), dry material consumption (DMC), and feed conversion (FC). Results: The Pelibuey ewes with one and two births and the Black Belly ewes with onebirth demonstrated the highest averages in prolificacy (P?0.05). The lowest averages(P?0.05) were detected in ewes with BC 4 irrespective of number of births. The BW wasaffected (P?0.05) by genotype; Black Belly lambs weighed 680 g (P?0.05) more thatPelibuey lambs. The DWG, DMC and FC behaved similarly (P?0.05) betweentreatments and their general averages were 0.114 kg day -1 , 0.679 kg day -1 , and 6.18,respectively.Study Limitations/Implications: In the agropastoral system, the BC of ewes limitstheir reproductive performance. The weight of the lambs at the start of fatteningdetermines their performance during fattening.Findings/Conclusions: In the agropastoral system, Pelibuey and Black Belly femalespresent adequate prolificacy averages and BW; however, it is very important that theyremain within BC 3. Neither of the two sources of selenium improves DWG, DMC, andFC of lambs fattened in pens in the agropastoral system

    Long-Term Habitat Degradation Drives Neotropical Macrophyte Species Loss While Assisting the Spread of Invasive Plant Species

    The spread of invasive macrophyte species is a pressing threat to neotropical shallow lakes. Yet there are few studies addressing the full extent of biotic and abiotic changes that may occur in response to invasive species. Less is known of how other human-induced stressors such as eutrophication and lake draining may interact over time with invasive macrophytes to influence biodiversity. We combined limnological observations with paleoecological data from Fúquene Lake, Colombia, a eutrophic neotropical shallow lake, to provide information on the current and long-term (decades-centuries) dynamics of the spread of two well-established invasive plants Eichhornia crassipes and Egeria densa. We found a unique in macrophyte species composition in areas currently dominated by Egeria and Eichhornia. Eichhornia-dominated areas had 14 macrophyte species, turbid (secchi=19 ± 6 cm) and poorly oxygenated (3.94 ± 2.61 ppm) waters whereas Egeria-rich areas supported 5 species and had clearer (secchi=51 ± 12 cm) and better-oxygenated (6.06 ± 2.4 ppm) waters. Historical macrophyte community shifts were linked to eutrophication and lake level variation and characterized by the loss of charophytes and bryophytes before 1500 CE and subsequent reductions in Nymphaea sp., Potamogeton illinoensis and Najas guadalupensis in the early 1900s (lake draining). Eichhornia crassipes (since 1500 CE) and E. densa (early 1900s) occurred well before proposed dates of introduction (1950s and 1990 respectively). Both species have rapidly expanded since the 1990s along with Azolla filiculoides in response to an inflow water diversion scheme and heavy nutrient loads. Our results suggest that the spread of Eichhornia and Egeria was not responsible for native macrophyte species loss, but that their current dominance is exerting synergistic and antagonistic secondary effects on plant assemblages through habitat modification, competitive exclusion and promotion of habitat heterogeneity across the lake. It could therefore be misleading to suggest that invasive plants causes macrophyte species loss in degraded lakes. We suggest that aggressive species like Eichhornia, Azolla, and Egeria require hydrologically stable and eutrophic environments to spread; thus, management actions should focus on controlling these two factors. Our study demonstrates the need to use a long-term approach to fully-understand the effects of invasive macrophytes

    Morphological Analysis of Major Segments of Coronary Artery Occlusion: Importance in Myocardial Revascularization Surgery

    Revascularization surgery should ensure morphological similarity between the coronary artery and the graft. This is an important factor for its duration and permeability. The aim of this study was to analyze the morphological characteristics and morphometrics of the coronary artery segments with greater occlusion. This was an observational, cross-sectional descriptive study that consisted of two phases. A macroscopic phase in which 11 cadaveric hearts were extracted and coronary dominance and length of the anterior interventricular artery (AIA), the right coronary artery (RCA) and the circumflex artery (CXA) were determined. In the microscopic phase a total of 77 segments of these arteries were obtained and the luminal diameter, wall thickness, and amount of elastic fibers and the presence and size of the atheroma were determined. Right coronary dominance was the most frequent. Total vessel length was 15.65±1.17 cm for the AIA, 12.67±2.02 cm for the RCA and 8.79±2.5 cm for the CXA. Diameters ranged from 2.3 mm in the proximal segments and between 1.1 mm to 1.8 mm in the distal segments. Wall thickness in the proximal segments was between 354 µm and 396 µm and in the distal segments it ranged from 120 µm to 305 µm. The amount of elastic fibers showed that they were muscular arteries. Atheromas were present in 35% in the CXA, and in 32.5% in the AIA and the RCA. The largest ones were found in the proximal segments. This study examined the morphology and morphometry of the segments of the coronary arteries that are more frequently occluded. It provides information on the most significant parameters to be considered for election of the vascular graft in myocardial revascularization surgery
