    A presença de compostos bioativos na amora-preta confere propriedades funcionais que estimulam a elaboração de produtos derivados. Entretanto, fatores como o processamento e o armazenamento são essenciais para a estabilidade desses produtos e para a manutenção da funcionalidade. Assim, o objetivo deste trabalho foi avaliar a influência do congelamento sobre as características físico-químicas e o potencial antioxidante de néctar de amora-preta durante 90 dias de armazenamento. O produto foi avaliado a cada 15 dias quanto aos teores totais de compostos fenólicos, antocianinas, ácido ascórbico, atividade antioxidante, sólidos solúveis totais, pH, acidez total e viscosidade aparente. Ao final do estudo verificou-se que o congelamento, como método de conservação, mantém a estabilidade físico-química e o potencial antioxidante do néctar de amora-preta

    Retinal nerve fiber layer thickness analysis in normal and glaucomatous dogs using laser polarimetry diagnosis

    Once the glaucomatous damage is progressive and irreversible, studies on the glaucoma onset as well as its development have been discussed. It is known that the early diagnostisis is extremely important to the stabilization and treatment. The retinal nerve fibers layer thickness analysis ''in vivo'' was proposed in human ophthalmology in order to establish the thickness changing, due to glaucoma, and have shown that such findings can be even detected six years priors to the first clinical signs. However, in Veterinary Medicine, such data need to be investigated and discussed. This study used two groups of dogs, a glaucomatous group and a normal group, that had been submitted to the retinal nerve fibers layer analysis through the GDx Nerve Fiber Analyzer. Statistical data showed that the nerve fiber layer of the glaucomatous group was thinner (p < 0.05), sustaining ganglion cells axons loss in glaucomatous eyes, compared to normal eyes.As lesões glaucomatosas são progressivas e irreversíveis. Estudos sobre o aparecimento e comportamento desta enfermidade vêm sendo discutidos na oftalmologia em geral. O diagnóstico precoce é de extrema importância para o tratamento e estabilização da mesma. A análise da espessura da camada de fibras nervosas retinianas ''in vivo'' foi proposta na oftalmologia humana, para estabelecer as alterações produzidas pelo glaucoma, e tem demonstrado que tais alterações podem ser detectadas até seis anos antes de alterações de campo visual e aumento de pressão intraocular. Entretanto, na Medicina Veterinária, estes dados carecem de estudo e discussão. Neste experimento foram utilizados dois grupos de cães, com olhos normais e olhos glaucomatosos, que foram submetidos à análise das fibras nervosas retinianas através do Analisador de Fibras Nervosas GDx. Os resultados estatísticos demonstraram que a camada de fibras nervosas dos olhos dos cães glaucomatosos estava mais delgada (p < 0,05) quando comparados aos olhos de cães normais, confirmando a perda de axônios das células ganglionares em olhos de cães com glaucoma

    Increased prevalence of unstable HLA-C variants in HIV-1 rapid-progressor patients

    HIV-1 infection in the absence of treatment results in progression toward AIDS. Host genetic factors play a role in HIV-1 pathogenesis, but complete knowledge is not yet available. Since less-expressed HLA-C variants are associated with poor HIV-1 control and unstable HLA-C variants are associated with higher HIV-1 infectivity, we investigated whether there was a correlation between the different stages of HIV-1 progression and the presence of specific HLA-C allotypes. HLA-C genotyping was performed using allele-specific PCR by analyzing a treatment-naïve cohort of 96 HIV-1-infected patients from multicentric cohorts in the USA, Canada, and Brazil. HIV-1-positive subjects were classified according to their different disease progression status as progressors (Ps, n = 48), long-term non-progressors (LTNPs, n = 37), and elite controllers (ECs, n = 11). HLA-C variants were classified as stable or unstable according to their binding stability to β2-microglobulin/peptide complex. Our results showed a significant correlation between rapid progression to AIDS and the presence of two or one unstable HLA-C variants (p-value: 0.0078, p-value: 0.0143, respectively). These findings strongly suggest a link between unstable HLA-C variants both at genotype and at allele levels and rapid progression to AIDS. This work provides further insights into the impact of host genetic factors on AIDS progression

    Anthropogenic geomorphic change as a potential generator of renewable geologic resources in the humid Pampa of Argentina

    The nature and amount of sediments in various lakes, intermittent swampy areas and river sectors in the humid Pampa (Buenos Aires Province, Argentina) were analysed. The aim was to determine whether recent sediments in such environments could serve as an alternative resource to the brick industry, for minimising the current, high environmental impact of soil mining. Sediment sequences were obtained, and the thicknesses of the upper sediment layers, corresponding to the suballuvial (approx. 1400-700 years BP) and alluvial (approx. 250 years BP to present) were determined. Sediment samples were collected and analysed for grain size and Atterberg limits. Suitable sediments were then selected to determine the optimal brick materials and their technical properties. Similarly, control bricks were prepared with ceramic pastes of local industries. The results show that the quality of the former is similar or superior to that of the latter. The initial estimates of the available resource indicate a long-term supply for the industry. Estimates of the current sedimentation rates indicate that resource renewal might occur at a rate comparable to current consumption. The sedimentation rates have increased significantly in the past two centuries - more so in the past few decades (the Anthropocene?) - with increasing human modification of geomorphic processes. If the results presented here are confirmed, a highly sustainable model can be implemented in the brick industry.Facultad de Ciencias Naturales y Muse

    Leishmania (Viannia) naiffi : rare enough to be neglected?

    In the Brazilian Amazon, American tegumentary leishmaniasis (ATL) is endemic and presents a wide spectrum of clinical manifestations due, in part, to the circulation of at least seven Leishmaniaspecies. Few reports of Leishmania (Viannia) naiffiinfection suggest that its occurrence is uncommon and the reported cases present a benign clinical course and a good response to treatment. This study aimed to strengthen the clinical and epidemiological importance of L. (V.) naiffiin the Amazon Region (Manaus, state of Amazonas) and to report therapeutic failure in patients infected with this species. Thirty Leishmania spp samples isolated from cutaneous lesions were characterised by multilocus enzyme electrophoresis. As expected, the most common species was Leishmania (V.) guyanensis (20 cases). However, a relevant number ofL. (V.) naiffi patients (8 cases) was observed, thus demonstrating that this species is not uncommon in the region. No patient infected withL. (V.) naiffievolved to spontaneous cure until the start of treatment, which indicated that this species may not have a self-limiting nature. In addition, two of the patients experienced a poor response to antimonial or pentamidine therapy. Thus, either ATL cases due to L. (V.) naifficannot be as uncommon as previously thought or this species is currently expanding in this region

    Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Nirmatrelvir/ritonavir in COVID-19 patients with haematological malignancies:a report from the EPICOVIDEHA registry

    Background: Nirmatrelvir/ritonavir treatment decreases the hospitalisation rate in immunocompetent patients with COVID-19, but data on efficacy in patients with haematological malignancy are scarce. Here, we describe the outcome of nirmatrelvir/ritonavir treatment in a large cohort of the latter patients. Methods: This is a retrospective cohort study from the multicentre EPICOVIDEHA registry (NCT04733729) on patients with haematological malignancy, who were diagnosed with COVID-19 between January and September 2022. Patients receiving nirmatrelvir/ritonavir were compared to those who did not. A logistic regression was run to determine factors associated with nirmatrelvir/ritonavir administration in our sample. Mortality between treatment groups was assessed with Kaplan–Meier survival plots after matching all the patients with a propensity score. Additionally, a Cox regression was modelled to detect factors associated with mortality in patients receiving nirmatrelvir/ritonavir. Findings: A total of 1859 patients were analysed, 117 (6%) were treated with nirmatrelvir/ritonavir, 1742 (94%) were treated otherwise. Of 117 patients receiving nirmatrelvir/ritonavir, 80% had received ≥1 anti-SARS-CoV-2 vaccine dose before COVID-19 onset, 13% of which received a 2nd vaccine booster. 5% were admitted to ICU. Nirmatrelvir/ritonavir treatment was associated with the presence of extrapulmonary symptoms at COVID-19 onset, for example anosmia, fever, rhinitis, or sinusitis (aOR 2.509, 95%CI 1.448–4.347) and 2nd vaccine booster (aOR 3.624, 95%CI 1.619–8.109). Chronic pulmonary disease (aOR 0.261, 95%CI 0.093–0.732) and obesity (aOR 0.105, 95%CI 0.014–0.776) were not associated with nirmatrelvir/ritonavir use. After propensity score matching, day-30 mortality rate in patients treated with nirmatrelvir/ritonavir was 2%, significantly lower than in patients with SARS-CoV-2 directed treatment other than nirmatrelvir/ritonavir (11%, p = 0.036). No factor was observed explaining the mortality difference in patients after nirmatrelvir/ritonavir administration. Interpretation: Haematological malignancy patients were more likely to receive nirmatrelvir/ritonavir when reporting extrapulmonary symptoms or 2nd vaccine booster at COVID-19 onset, as opposed to chronic pulmonary disease and obesity. The mortality rate in patients treated with nirmatrelvir/ritonavir was lower than in patients with targeted drugs other than nirmatrelvir/ritonavir.
