    Media coverage attacking the character and trustworthiness of a scientist can diminish public faith in the research findings of that scientist. Ralph M. Barnes, Heather M. Johnston, Noah MacKenzie, Stephanie J. Tobin and Chelsea M. Taglang have investigated the degree to which such attacks do undermine trust in that scientist's research, and the relative impact of various types of ad hominem attacks. Perhaps surprisingly, purely ad hominem attacks, such as accusations of a financial conflict of interest, for example, prove just as effective in undermining public faith in research findings as direct criticism of the empirical foundations of a science claim

    Comparative Population Genomics and Biophysical Modeling of Shrimp Migration in the Gulf of Mexico Reveals Current-Mediated Connectivity

    The Gulf of Mexico experiences frequent perturbations, both natural and anthropogenic. To better understand the impacts of these events, we must inventory natural variability within the ecosystem, communities, species, and populations, and contextualize these findings in relation to physical features. Here, we present an integrated study of comparative population genomics and biophysical oceanography. Targeting three species of mesopelagic shrimp common to the Gulf of Mexico midwater (Acanthephyra purpurea, Systellaspis debilis, and Robustosergia robusta), we analyzed genetic diversity and population connectivity as proxies for species health and resilience, respectively. We also simulated a range of vertical migratory behaviors for the shrimp to infer the relationship between diel vertical migration and horizontal transmission between the Gulf of Mexico and the greater Atlantic Ocean. This study aims to establish biological baselines and characterize these values in terms of the prevailing oceanographic feature of the midwater: the Gulf Loop Current. Generally, the oplophorid species (A. purpurea and S. debilis) exhibit lower genetic diversity and higher interpopulation homogeneity compared to the sergestid (R. robusta). Biophysical simulations suggest the differences in vertical migratory regimes between these two groups have important implications for horizontal transport out of the Gulf of Mexico. Because of the difference in vertical migration patterns, access to the Gulf Loop Current varies across taxa and impacts inter-basin migration. Our findings suggest a negative correlation between surface abundance and genetic diversity in these three shrimp species. We hypothesize that this correlation may be due to the relationships between surface abundance and access to the fastest moving waters of the Gulf Loop Current

    The Need for Reporting Metabolic Sampling Interval in Publication: An Example Using Maximal VO2 Values and Running Economy

    Background: Knowledge of metabolic outcomes, such as maximal oxygen consumption (VO2) or running economy, has wide-ranging application. Metabolic outcomes are widely reported in literature yet the metabolic sampling interval (example: breath-by-breath, 30-sec average) utilized for collection is rarely ever stated. Purpose: The purposes of the present investigation were to probe the potential discrepancies created when analyzing running economy and VO2max raw metabolic data with four different metabolic sampling intervals. Methods: Five recreationally-active and endurance-trained subjects were included in the present analysis and four metabolic sampling intervals were analyzed: 30-sec average, 20-sec average, 8-breath, and 4-breath. Subjects engaged in 4-min running economy phases at 55 and 65% of their VO2max before entering into a maximal protocol purposed to elicit VO2max in 8-12 minutes. Utilizing the steady state and maximal VO2 data, metabolic sampling intervals were analyzed for their effect on reported VO2 values. Results: For running economy at 55%, there was no differences found (f = 0.207; df = 1.862; p = 0.799) between sampling frequencies when analyzed by repeated measures analysis of variance and corrected with Greenhouse-Geisser for a violation of sphericity. For running economy at 65%, there were also no differences ­found (f = 1.456; df = 3; p = 0.799) between sampling frequencies. For inspection, the relative VO2 values were: 27.2 (±3.1), 27.9 (±4.1), 28.4 (±3.6), and 28.8 (±5.1) for the 30-sec, 20-sec, 8-breath, and 4-breath average, respectively. Maximal VO2 values of 53.0 (±6.6), 55.1 (±7.2), 55.1 (±7.2), and 59.6 (±9.4) for the 30-sec, 20-sec, 8-breath, and 4-breath average, respectively, were found to be significantly different (f = 21.062; df = 1.278; p \u3c 0.001) after adjusting for a violation of sphericity (p \u3c 0.001). Bonferroni analysis indicated differences between the 30-sec average and all other averages and also the 20-sec and 8-breath averages when compared against the 4-breath average. The 4-breath average yielded the highest VO2max value. Coincidentally, the 20-sec and 8-breath averages were identical. Conclusion: In the present investigation of raw metabolic data, sampling interval was found to impact the maximal oxygen consumption (VO2max) values but not running economy values when investigating a small sample of data with four select sampling intervals. The report of maximal VO2 is rather common in the literature and knowing sample interval is vital for between-study comparison, determination of regression-related activities, or for pre-post comparison of data from the same or different labs

    Dystrophin Gene Mutation Location and the Risk of Cognitive Impairment in Duchenne Muscular Dystrophy

    Contains fulltext : 88828.pdf (publisher's version ) (Open Access)BACKGROUND: A significant component of the variation in cognitive disability that is observed in Duchenne muscular dystrophy (DMD) is known to be under genetic regulation. In this study we report correlations between standardised measures of intelligence and mutational class, mutation size, mutation location and the involvement of dystrophin isoforms. METHODS AND RESULTS: Sixty two male subjects were recruited as part of a study of the cognitive spectrum in boys with DMD conducted at the Sydney Children's Hospital (SCH). All 62 children received neuropsychological testing from a single clinical psychologist and had a defined dystrophin gene (DMD) mutation; including DMD gene deletions, duplications and DNA point mutations. Full Scale Intelligence Quotients (FSIQ) in unrelated subjects with the same mutation were found to be highly correlated (r = 0.83, p = 0.0008), in contrast to results in previous publications. In 58 cases (94%) it was possible to definitively assign a mutation as affecting one or more dystrophin isoforms. A strong association between the risk of cognitive disability and the involvement of groups of DMD isoforms was found. In particular, improvements in the correlation of FSIQ with mutation location were identified when a new classification system for mutations affecting the Dp140 isoform was implemented. SIGNIFICANCE: These data represent one of the largest studies of FSIQ and mutational data in DMD patients and is among the first to report on a DMD cohort which has had both comprehensive mutational analysis and FSIQ testing through a single referral centre. The correlation between FSIQ results with the location of the dystrophin gene mutation suggests that the risk of cognitive deficit is a result of the cumulative loss of central nervous system (CNS) expressed dystrophin isoforms, and that correct classification of isoform involvement results in improved estimates of risk

    The title compound, C8H8N2O4, was prepared via the nitration of p-xylene. The mol­ecules are stacked along the c axis in an antiparallel manner. The two nitro groups are rotated relative to the benzene ring with dihedral angles of 44.50 (7) and 31.67 (8)°. The tilt of the nitro groups allows the formation of C—H⋯O inter­actions between the ring C—H and nitro groups of adjacent mol­ecules creating puckered sheets perpendicular to the c axis. The H atoms of the methyl group in the 5-position are disordered (60° rotation) with an occupancy of 0.616 (19) for the major component. The crystal was found to be a non-merohedral twin with a twin law [−1 −0.002 0.005, 0.00031 −1 0.002, 0.118 −0.007 1] corresponding to a rotation of 180° about the reciprocal axis (001) and refined to give a minor component fraction of 0.320 (2)

    Cluster randomised controlled trial of a theory-based multiple behaviour change intervention aimed at healthcare professionals to improve their management of type 2 diabetes in primary care

    Background: National diabetes audits in the UK show room for improvement in the quality of care delivered to people with type 2 diabetes in primary care. Systematic reviews of quality improvement interventions show that such approaches can be effective but there is wide variability between trials and little understanding concerning what explains this variability. A national cohort study of primary care across 99 UK practices identified modifiable predictors of healthcare professionals' prescribing, advising and foot examination. Our objective was to evaluate the effectiveness of an implementation intervention to improve six guideline-recommended health professional behaviours in managing type 2 diabetes in primary care: prescribing for blood pressure and glycaemic control, providing physical activity and nutrition advice and providing updated diabetes education and foot examination. Methods: Two-armed cluster randomised trial involving 44 general practices. Primary outcomes (at 12 months follow-up): from electronic medical records, the proportion of patients receiving additional prescriptions for blood pressure and insulin initiation for glycaemic control and having a foot examination; and from a patient survey of a random sample of 100 patients per practice, reported receipt of updated diabetes education and physical activity and nutrition advice. Results: The implementation intervention did not lead to statistically significant improvement on any of the six clinical behaviours. 1,138,105 prescriptions were assessed. Intervention (29% to 37% patients) and control arms (31% to 35%) increased insulin initiation relative to baseline but were not statistically significantly different at follow-up (IRR 1.18, 95% CI 0.95-1.48). Intervention (45% to 53%) and control practices (45% to 50%) increased blood pressure prescription from baseline to follow-up but were not statistically significantly different at follow-up (IRR 1.05, 95% CI 0.96 to 1.16). Intervention (75 to 78%) and control practices (74 to 79%) increased foot examination relative to baseline; control practices increased statistically significantly more (OR 0.84, 95% CI 0.75-0.94). Fewer patients in intervention (33%) than control practices (40%) reported receiving updated diabetes education (OR = 0.74, 95% CI 0.57-0.97). No statistically significant differences were observed in patient reports of having had a discussion about nutrition (intervention = 73%; control = 72%; OR = 0.98, 95% CI 0.59-1.64) or physical activity (intervention = 57%; control = 62%; OR = 0.79, 95% CI 0. 56-1.11). Development and delivery of the intervention cost 1191 pound per practice. Conclusions: There was no measurable benefit to practices' participation in this intervention. Despite widespread use of outreach interventions worldwide, there is a need to better understand which techniques at which intensity are optimally suited to address the multiple clinical behaviours involved in improving care for type 2 diabetes.Peer reviewe
