Liposomes isolation and characterization by microencapsulation
Liposomes are microscopic vesicles composed of concentric lipid bilayers entrapping aqueous compartments. Proteins and lipids are among biocompatible and biodegradable materials used for microencapsulation, being albumin an interesting protein due to its role as a drugs carrier. The aim of the study was to encapsulate ciprofloxacin loaded liposomes into Albusomes® a novel pharmaceutical vehicle based on albumin coated liposomes. The vesicle preparation was carried out using the lipid hydration method and sonication method using a mixture of egg phosphatidylcholine (EPC), diethyldodeccyiamonium (DDA) and cholesterol (Ch). The liposomes were encapsulated with bovine serum albumin by induced flocculation. The pellet, containing the albumin coated liposomes with ciprofloxacin trapped into water vesicle core and albumin coating layers, was isolated by centrifugation. Concentration of ciprofloxacin was quantified in the supernatant and the absence of lipid vesicles was confirmed. The proposed vehicle (Albusomes®) showed to be able to encapsulate liposomes with a high drug entrapment efficacy (86.0 ± 5.0% and 93.52±3.94%). for lipid hydration method and sonication method, respectively.Los liposomas son vesículas microscópicas compuestas de bicapas lipídicas concéntricas que alternan con compartimentos acuosos. Las proteínas y los lípidos son materiales biocompatibles y biodegradables utilizados para la microencapsulación de fármacos, siendo la albúmina una de las proteína de interés debido a su importante papel como un transportador. El objetivo del estudio fue encapsular liposomas cargados con ciprofloxacino en Albusomas®, un novedoso vehículo farmacéutico basado en liposomas recubiertos de albúmina. La preparación de las vesículas se llevó a cabo mediante el método de hidratación de lípidos y el método de sonicación, utilizando una mezcla de fosfatidilcolina de huevo (EPC), dietildodecilamonio (DDA) y colesterol (Ch). Los liposomas se encapsularon con albúmina bovina por floculación inducida. El sedimento, aislado por centrifugación contiene las microesferas formadas por los liposomas recubiertos de albúmina con ciprofloxacino en el núcleo acuoso, y en las capas de revestimiento de albúmina. Se determinó la concentración de ciprofloxacino en el sobrenadante y se comprobó la ausencia de liposomas en el mismo. El vehículo propuesto (Albusomas®) demostró ser capaz de encapsular los liposomas con una alta eficacia de encapsulación de ciprofloxacino (86,0 ± 5,0% y 93,52±3,94%) para el método de hidratación de lípidos y el método de sonicación, respectivamente
Drug delivery from liposomes in different biorelevant media
Liposomes provide ideal vesicular systems for controlled release and vectorization of drugs through pulmonary administration, due to their similarity to cell membranes and their great versatility./nSildenafil citrate, represents an optimal model of drug to be incorporated into these lipid vesicles for the treatment of pulmonary hypertension./nThe objective of the work was to characterize the behavior of liposomes loaded with sildenafil and evaluate the transfer of the drug in different bio-resistant media./nLiposomes of two sizes (0.45 and 0.20 microns) were developed by sonication. Subsequently, the drug release test was carried out in different biorelevant media (simulated plasma fluid SBF, simulated pulmonary fluid SPF and PBS)./nAccording to the results obtained, a more sustained and consistent rate of release of the drug was observed over time in the case of liposomes, regardless of the medium. Posteriormente se llevó a cabo el ensayo de liberación del fármaco en diferentes medios biorrelevantes (fluido plasmático simulado SBF, fluido pulmonar simulado SPF y PBS)./nSegún los resultados obtenidos se observó una velocidad de liberación del fármaco más sostenida y constante a lo largo del tiempo en el caso de los liposomas, independientemente del medio. En los medios PBS y SPF, se liberó una mayor cantidad de fármaco desde los liposomas con mayor tamaño (0,45 µm), sin embargo, en el SBF las cinéticas de cesión de ambos tamaños de liposomas (0,45 y 0,20 µm) son muy similares, no encontrándose diferencias significativas en el tiempo o cantidad de fármaco liberada
Requisitos de almacenamiento de información e identificación de actores para una biblioteca digital de bienes muebles
Desarrollar una biblioteca digital no es una tarea sencilla. En ella
intervienen técnicos y expertos que deben tener una fluida comunicación que
permita que el sistema resultante se adecue a las necesidades del usuario. Este
trabajo presenta las necesidades de almacenamiento que requiere una biblioteca
digital para la gestión del patrimonio mueble andaluz, así como la identificación
de los actores que van a interactuar con el sistema. Al final del trabajo se
presenta una aproximación metodológica para el desarrollo de sistemas para el
tratamiento de bibliotecas digitales que se está elaborando en el contexto del
proyecto de investigación MADEIRA(Metodologías y Arquitecturas para la

Aplicación Integrada de la Biblioteca Digital del Patrimonio Histórico Andaluz
En este trabajo se describen las necesidades del Instituto Andaluz de
Patrimonio Histórico para desarrollar el Sistema de Información Integrado de
Patrimonio Histórico Andaluz. El objetivo es crear un sistema de acceso y
gestión uniforme a la información dispersa en diversas bases de datos que han
sido creadas por grupos de trabajo independientes. Para ello, se propone un
modelo de arquitectura, basado en XML, que permite el acceso uniforme a
bases de datos heterogéneas y distribuidas
INTEREVO-TR: Interactive Evolutionary Test Generation with Readability Assessment
Automated test case generation has proven to be useful to reduce the usually high expenses of software testing. However, several studies have also noted the skepticism of testers regarding the comprehension of generated test suites when compared to manually designed ones. This fact suggests that involving testers in the test generation process could be helpful to increase their acceptance of automatically-produced test suites. In this paper, we propose incorporating interactive readability assessments made by a tester into EvoSuite, a widely-known evolutionary test generation tool. Our approach, InterEvo-TR, interacts with the tester at different moments during the search and shows different test cases covering the same coverage target for their subjective evaluation. The design of such an interactive approach involves a schedule of interaction, a method to diversify the selected targets, a plan to save and handle the readability values, and some mechanisms to customize the level of engagement in the revision, among other aspects. To analyze the potential and practicability of our proposal, we conduct a controlled experiment in which 39 participants, including academics, professional developers, and student collaborators, interact with InterEvo-TR. Our results show that the strategy to select and present intermediate results is effective for the purpose of readability assessment. Furthermore, the participants' actions and responses to a questionnaire allowed us to analyze the aspects influencing test code readability and the benefits and limitations of an interactive approach in the context of test case generation, paving the way for future developments based on interactivity

Design of a liposomal formulation aimed to vaccines administration
Antigen is not the single component involved in the pharmacological response for vaccines but additional factors such as formulation and adjuvant compounds play a relevant role. Several strategies based on the use of liposomes are currently assayed in this field. According to this, the aim of the present work was the design, preparation and characterization of a pharmaceutical vehicle able to produce a rapid delivery of adjuvants and a slow release of antigen. From the know-how a theoretical vehicle was design and proposed as optimal formulation and this was then prepared and characterized. The “proposed formulation” is based on albumin coated liposomes made of egg phosphatidylcholine, cholesterol and diethyldodeccyiamonium. The applied procedure carried out in absence of organic solvents allowed for obtaining liposomes with a 20-70 nm aerodynamic diameter range and a zeta potential mean value of 61.9 ±2.08 mV and also for its coating with albumin. Coated particles showed a spherical shape and a size range of 2-10 µm which does not fulfil parenteral formulations requirements but are optimal for nasal or dermatological administration routes.Los efectos de una vacuna dependen no solo del antígeno,
sino también de factores relacionados con la formulación; la elección del sistema de liberación junto con el empleo de potenciadores inmunológicos son aspectos de gran relevancia. Actualmente existen numerosas estrategias en desarrollo en este campo basados en la utilización de liposomas. El objetivo de este trabajo ha sido el diseño y la preparación de un vehículo para la administración de vacunas, capaz de liberar inmediatamente el coadyuvante y más lentamente el antígeno. Analizando los conocimientos previos se realizó, en una primera fase, el diseño teórico del vehículo y seguidamente la preparación y caracterización del mismo. La “formulación propuesta” se basa en la microencapsulación de liposomas, constituidos por fosfatidilcolina, colesterol y dimetildioctadecil amonio, con albumina bovina. El procedimiento experimental aplicado, que transcurre en ausencia de disolventes orgánicos, permite obtener liposomas con un potencial zeta de 61,9 ±2,08 mV y tamaño entre 20 y 70 nm así como su inclusión en partículas esféricas de albúmina, cuyo rango de tamaño resultó ser de 2-10 µm. Con estas características el vehículo no podría administrarse por vía parenteral pero si nasal o transdérmica
Diseño del Modelo Navegacional para Sistemas de Tratamiento de Bibliotecas Digitales
Los sistemas de tratamiento de bibliotecas digitales nacen como
sistemas complejos en los que se mezclan importantes requisitos de
almacenamiento de información, debido a los múltiples medios que se usan en
ellos, con grandes necesidades funcionales. Pero además en el entorno de las
bibliotecas digitales se añaden unas necesidades, que son propias de las
aplicaciones multimedia y páginas web, como son el uso de los enlaces y el
ofrecer una atractiva interfaz de usuario. Estas nuevas necesidades se hacen
patentes principalmente en la fase de diseño de la aplicación, puesto que es en
ella donde se le añadirán a la aplicación. En este trabajo, se propone cómo se
puede incorporar el diseño de los aspectos de navegación a una aplicación
Xanthan Gum–Konjac Glucomannan Blend Hydrogel for Wound Healing
Hydrogels are considered to be the most ideal materials for the production of wound dressings since they display a three-dimensional structure that mimics the native extracellular matrix of skin as well as a high-water content, which confers a moist environment at the wound site. Until now, different polymers have been used, alone or blended, for the production of hydrogels aimed for this biomedical application. From the best of our knowledge, the application of a xanthan gum-konjac glucomannan blend has not been used for the production of wound dressings. Herein, a thermo-reversible hydrogel composed of xanthan gum-konjac glucomannan (at different concentrations (1% and 2% w/v) and ratios (50/50 and 60/40)) was produced and characterized. The obtained data emphasize the excellent physicochemical and biological properties of the produced hydrogels, which are suitable for their future application as wound dressings.info:eu-repo/semantics/publishedVersio
The role of the surface evapotranspiration in regional climate modelling: Evaluation and near-term future changes
Surface evapotranspiration (SFCEVP) plays an essential role in the climate, being the link between hydrological and energy cycles. Therefore, how it is approximated and its implication in the regional climate are important aspects to understand the effects of climate change, especially in transitional zones such as the Iberian Peninsula (IP). This study aims to investigate the spatiotemporal patterns of the SFCEVP using a regional climate model (RCM), the Weather Research and Forecasting (WRF) model. To this purpose, a set of WRF simulations were completed using different driving data. On the first hand, a recent present (1980–2017) simulation driven by the ERA-Interim reanalysis was carried out to evaluate the suitability of the RCM performance. On the other hand, two global climate models (GCMs) from the CMIP5 initiative, the CESM1 and the MPI-ESM-LR, were used as driving data to evaluate the GCM-RCM coupling, which is essential to climate change applications. Finally, projected changes were also investigated for a near-term future (2021–2050) paradigm. In general, the results pointed out the WRF model as a valuable tool to study the spatiotemporal patterns of the SFCEVP in the IP, showing an overall and acceptable ability at different spatial and temporal scales. Concerning projections, the results indicate that the IP is likely to undergo significant changes in the SFCEVP in the near future. These changes will be more apparent over the southernmost, and particularly during spring and summer, being in the latter season the SFCEVP fundamentally reduced. These results agree with projected changes in soil moisture, which is probably associated with changes in precipitation patterns. Additionally, the results reveal the major role of SFCEVP in modulating the climate over this region, which is involved in the complex land-atmosphere processes.Departamento de Física Aplicada, Facultad de Ciencias, Universidad de Granad
