    Zinc oxide nanoparticles selectively induce apoptosis in human cancer cells through reactive oxygen species

It has been reported that ZnO NPs induce selective killing of cancer cells. However, the underlying molecular mechanisms behind the anticancer response of ZnO NPs remain unclear.Methods and results: We investigated the cytotoxicity of ZnO NPs against three types of cancer cells (human hepatocellular carcinoma HepG2, human lung adenocarcinoma A549, and human bronchial epithelial BEAS-2B) and two primary rat cells (astrocytes and hepatocytes). Results showed that ZnO NPs exert distinct effects on mammalian cell viability via killing of all three types of cancer cells while posing no impact on normal rat astrocytes and hepatocytes. The toxicity mechanisms of ZnO NPs were further investigated using human liver cancer HepG2 cells. Both the mRNA and protein levels of tumor suppressor gene p53 and apoptotic gene bax were upregulated while the antiapoptotic gene bcl-2 was downregulated in ZnO NP-treated HepG2 cells. ZnO NPs were also found to induce activity of caspase-3 enzyme, DNA fragmentation, reactive oxygen species generation, and oxidative stress in HepG2 cells.Conclusion: Overall, our data demonstrated that ZnO NPs selectively induce apoptosis in cancer cells, which is likely to be mediated by reactive oxygen species via p53 pathway, through which most of the anticancer drugs trigger apoptosis.     Factors influencing farmers' participation in water management: a community development perspective

    Participation is believed to a systematic involvement of the significant number of people, in diverse activities to manage their own problems, carrying the concept of togetherness, which enhance the advantages. It always oscillates and depends on the background in which it occurs. Farmers' participation is critical to improve on-farm water management and crop productivity. The study was conducted to identify the factors involved in the fluctuation of participation. This paper reports the results, in which, farmer participation through planning, implementation, monitoring and evaluation of water-user associations were measured, and it was linked to various socioeconomic, demographic, and farm and irrigation characteristics. Multiple linear regression analysis reveals that the participation was positively influenced by age, education, residential locality, house type, lack of on-farm facilities, underground water use for irrigation purpose and location of watercourses on the canal network. However, residential locality and on-farm facilities have significantly negative relationships with the dependent variable in Sindh province of Pakistan

    Stigmergy co-ordinates multicellular collective behaviours during Myxococcus xanthus surface migration

    Surface translocation by the soil bacterium Myxococcus xanthus is a complex multicellular phenomenon that entails two motility systems. However, the mechanisms by which the activities of individual cells are coordinated to manifest this collective behaviour are currently unclear. Here we have developed a novel assay that enables detailed microscopic examination of M. xanthus motility at the interstitial interface between solidified nutrient medium and a glass coverslip. Under these conditions, M. xanthus motility is characterised by extensive micro-morphological patterning that is considerably more elaborate than occurs at an air-surface interface. We have found that during motility on solidified nutrient medium, M. xanthus forges an interconnected furrow network that is lined with an extracellular matrix comprised of exopolysaccharides, extracellular lipids, membrane vesicles and an unidentified slime. Our observations have revealed that M. xanthus motility on solidified nutrient medium is a stigmergic phenomenon in which multi-cellular collective behaviours are co-ordinated through trail-following that is guided by physical furrows and extracellular matrix materials

    Quantifying techno-economic indicators\u27 impact on isolated renewable energy systems

    Addressing climate change with the rising global energy usage necessitates electricity sector decarbonization by rapidly moving toward flexible and efficient off-grid renewable energy systems (RESs). This paper analyzes the wind and solar micro-grids, with batteries and pumped hydro storage for a robust off-grid RES techno-economic operation, while considering diverse multi-objective optimization cases. This research has considered the RES variable operational losses in the developed methodology and relations between different indicators are evaluated, revealing a basic understanding between them. The results reveal that the reliability index is inversely related to the oversupply index, while directly related to the system self-sufficiency index. The cost of energy is more sensitive to technical indicators rather than the storage cost and so can be used as a primary monetary index. Energy and cost balance analysis showed that 16%-20% of the used energy was drained in RES operational losses, which were usually ignored in previous studies

    Economic analysis and optimization of a renewable energy based power supply system with different energy storages for a remote island

    This study investigates and compares the various combinations of renewable energies (solar, wind) and storage technologies (battery, pumped hydro storage, hybrid storage) for an off-grid power supply system. Four configurations (i.e., single RE source system, double RE source system, single storage, and double storage system) based on two scenarios (self-discharge equal to 0% and 1%) are considered, and their operational performance is compared and analyzed. The energy management strategy created for the hybrid pumped battery storage (HPBS) considers that batteries cover low energy surplus/shortages while pumped hydro storage (PHS) is the primary energy storage device for serving high-energy generations/deficits. The developed mathematical model is optimized using Particle Swarm Optimization and the performance and results of the optimizer are discussed in particular detail. The results evidence that self-discharge has a significant impact on the cost of energy (13%–50%) for all configurations due to the substantial increase in renewable energy (RE) generators size compared to the energy storage capacity. Even though solar-wind-PHS is the cost-optimal arrangement, it exhibits lower reliability when compared to solar-wind-HPBS. The study reveals the significance of HPBS in the off-grid RE environment, allowing more flexible energy management, enabling to guarantee a 100% power supply with minimum cost and reducing energy curtailment. Additionally, this study presents and discuss the results of a sensitivity analysis conducted by varying load demand and energy balance of all considered configurations is performed, which reveals the effectiveness of the supplementary functionality of both storages in hybrid mode. Overall, the role of energy storage in hybrid mode improved, and the total energy covered by hybrid storage increased (48%), which reduced the direct dependency on variable RE generation

    Homozygosity Mapping and Genetic Analysis of Autosomal Recessive Retinal Dystrophies in 144 Consanguineous Pakistani Families.

    PurposeThe Pakistan Punjab population has been a rich source for identifying genes causing or contributing to autosomal recessive retinal degenerations (arRD). This study was carried out to delineate the genetic architecture of arRD in the Pakistani population.MethodsThe genetic origin of arRD in a total of 144 families selected only for having consanguineous marriages and multiple members affected with arRD was examined. Of these, causative mutations had been identified in 62 families while only the locus had been identified for an additional 15. The remaining 67 families were subjected to homozygosity exclusion mapping by screening of closely flanking microsatellite markers at 180 known candidate genes/loci followed by sequencing of the candidate gene for pathogenic changes.ResultsOf these 67 families subjected to homozygosity mapping, 38 showed homozygosity for at least one of the 180 regions, and sequencing of the corresponding genes showed homozygous cosegregating mutations in 27 families. Overall, mutations were detected in approximately 61.8 % (89/144) of arRD families tested, with another 10.4% (15/144) being mapped to a locus but without a gene identified.ConclusionsThese results suggest the involvement of unmapped novel genes in the remaining 27.8% (40/144) of families. In addition, this study demonstrates that homozygosity mapping remains a powerful tool for identifying the genetic defect underlying genetically heterogeneous arRD disorders in consanguineous marriages for both research and clinical applications

    WLAN aware cognitive medium access control protocol for IoT applications

    © 2020 by the authors. Internet of Things (IoT)-based devices consist of wireless sensor nodes that are battery-powered; thus, energy efficiency is a major issue. IEEE 802.15.4-compliant IoT devices operate in the unlicensed Industrial, Scientific, and Medical (ISM) band of 2.4 GHz and are subject to interference caused by high-powered IEEE 802.11-compliant Wireless Local Area Network (WLAN) users. This interference causes frequent packet drop and energy loss for IoT users. In this work, we propose a WLAN Aware Cognitive Medium Access Control (WAC-MAC) protocol for IoT users that uses techniques, such as energy detection based sensing, adaptive wake-up scheduling, and adaptive backoff, to reduce interference with the WSN and improve network lifetime of the IoT users. Results show that the proposedWAC-MAC achieves a higher packet reception rate and reduces the energy consumption of IoT nodes
