FGF2 overrides key pro-fibrotic features of bone marrow stromal cells isolated from Modic type 1 change patients
Extensive extracellular matrix production and increased cell-matrix adhesion by bone marrow stromal cells (BMSCs) are hallmarks of fibrotic alterations in the vertebral bone marrow known as Modic type 1 changes (MC1). MC1 are associated with non-specific chronic low-back pain. To identify treatment targets for MC1, in vitro studies using patient BMSCs are important to reveal pathological mechanisms. For the culture of BMSCs, fibroblast growth factor 2 (FGF2) is widely used. However, FGF2 has been shown to suppress matrix synthesis in various stromal cell populations. The aim of the present study was to investigate whether FGF2 affected the in vitro study of the fibrotic pathomechanisms of MC1-derived BMSCs. Transcriptomic changes and changes in cell-matrix adhesion of MC1-derived BMSCs were compared to intra-patient control BMSCs in response to FGF2. RNA sequencing and quantitative real-time polymerase chain reaction revealed that pro-fibrotic genes and pathways were not detectable in MC1-derived BMSCs when cultured in the presence of FGF2. In addition, significantly increased cell-matrix adhesion of MC1-derived BMSCs was abolished in the presence of FGF2. In conclusion, the data demonstrated that FGF2 overrides key pro-fibrotic features of MC1 BMSCs in vitro. Usage of FGF2-supplemented media in studies of fibrotic mechanisms should be critically evaluated as it could override normally dominant biological and biophysical cues
Sustained suppression of neointimal proliferation by sirolimus-eluting stents: one-year angiographic and intravascular ultrasound follow-up
BACKGROUND: We have previously reported a virtual absence of neointimal hyperplasia 4 months after implantation of sirolimus-eluting stents. The aim of the present investigation was to determine whether these results are sustained over a period of 1 year. METHODS AND RESULTS: Forty-five patients with de novo coronary disease were successfully treated with the implantation of a single sirolimus-eluting Bx VELOCITY stent in Sao Paulo, Brazil (n=30, 15 fast release [group I, GI] and 15 slow release [GII]) and Rotterdam, The Netherlands (15 slow release, GIII). Angiographic and volumetric intravascular ultrasound (IVUS) follow-up was obtained at 4 and 12 months (GI and GII) and 6 months (GIII). In-stent minimal lumen diameter and percent diameter stenosis remained essentially unchanged in all groups (at 12 months, GI and GII; at 6 months, GIII). Follow-up in-lesion minimal lumen diameter was 2.28 mm (GIII), 2.32 mm (GI), and 2.48 mm (GII). No patient approached the >/=50% diameter stenosis at 1 year by angiography or IVUS assessment, and no edge restenosis was observed. Neointimal hyperplasia, as detected by IVUS, was virtually absent at 6 months (2+/-5% obstruction volume, GIII) and at 12 months (GI=2+/-5% and GII=2+/-3%). CONCLUSIONS: This study demonstrates a sustained suppression of neointimal proliferation by sirolimus-eluting Bx VELOCITY stents 1 year after implantation

Search for CP violation in decays
A model-independent search for direct CP violation in the Cabibbo suppressed
decay in a sample of approximately 370,000 decays is
carried out. The data were collected by the LHCb experiment in 2010 and
correspond to an integrated luminosity of 35 pb. The normalized Dalitz
plot distributions for and are compared using four different
binning schemes that are sensitive to different manifestations of CP violation.
No evidence for CP asymmetry is found.Comment: 13 pages, 8 figures, submitted to Phys. Rev.
Measurement of the ratio of branching fractions BR(B0 -> K*0 gamma)/BR(Bs0 -> phi gamma)
The ratio of branching fractions of the radiative B decays B0 -> K*0 gamma
and Bs0 -> phi gamma has been measured using 0.37 fb-1 of pp collisions at a
centre of mass energy of sqrt(s) = 7 TeV, collected by the LHCb experiment. The
value obtained is BR(B0 -> K*0 gamma)/BR(Bs0 -> phi gamma) = 1.12 +/- 0.08
^{+0.06}_{-0.04} ^{+0.09}_{-0.08}, where the first uncertainty is statistical,
the second systematic and the third is associated to the ratio of fragmentation
fractions fs/fd. Using the world average for BR(B0 -> K*0 gamma) = (4.33 +/-
0.15) x 10^{-5}, the branching fraction BR(Bs0 -> phi gamma) is measured to be
(3.9 +/- 0.5) x 10^{-5}, which is the most precise measurement to date.Comment: 15 pages, 1 figure, 2 table
Measurements of the branching fractions of the decays B°s â Dâs K± and B°s â DÂŻsÏ+
The decay mode B°s â Dâs K± allows for one of the theoretically cleanest measurements of the CKM angle Îł through the study of time-dependent CP violation. This paper reports a measurement of its branching fraction relative to the Cabibbo-favoured mode B°s â DÂŻsÏ+ based on a data sample corresponding to 0.37 fbÂŻÂč of proton-proton collisions at âs = 7TeV collected in 2011 with the LHCb detector. In addition, the ratio of B meson production fractions fs/fd, determined from semileptonic decays, together with the known branching fraction of the control channel B°s â DÂŻsÏ+ is used to perform an absolute measurement of the branching fractions: B(B°s â DÂŻsÏ+) = (2.95 ± 0.05 ± 0.17 -0.22 +0.18) Ă 10ÂŻÂł ; B(B°s â Dâs K±) = (1.90 ± 0.12 ± 0.13 -0.14 +0.12) Ă 10ÂŻ4 ; where the first uncertainty is statistical, the second the experimental systematic uncertainty, and the third the uncertainty due to f s/f

CP violation in the decays of mesons to pairs,
, with a significance of 3.3. Furthermore, we
report the first observation of CP violation in decays at a hadron
collider, , with a significance exceeding 6.Comment: 8 pages, 2 figures, 2 tables; v2 with minor changes after journal

Measurement of the CP-violating phase phi_s in the decay Bs->J/psi phi
We present a measurement of the time-dependent CP-violating asymmetry in B_s
-> J/psi phi decays, using data collected with the LHCb detector at the LHC.
The decay time distribution of B_s -> J/psi phi is characterized by the decay
widths Gamma_H and Gamma_L of the heavy and light mass eigenstates of the
B_s-B_s-bar system and by a CP-violating phase phi_s. In a sample of about 8500
B_s -> J/psi phi events isolated from 0.37 fb^-1 of pp collisions at sqrt(s)=7
TeV we measure phi_s = 0.15 +/- 0.18 (stat) +/- 0.06 (syst) rad. We also find
an average B_s decay width Gamma_s == (Gamma_L + Gamma_H)/2 = 0.657 +/- 0.009
(stat) +/- 0.008 (syst) ps^-1 and a decay width difference Delta Gamma_s ==
Gamma_L - Gamma_H} = 0.123 +/- 0.029 (stat) +/- 0.011 (syst) ps^-1. Our
measurement is insensitive to the transformation (phi_s,DeltaGamma_s --> pi -
phi_s, - Delta Gamma_s.Comment: 9 pages, 3 figure
Strong constraints on the rare decays Bs -> mu+ mu- and B0 -> mu+ mu-
A search for Bs -> mu+ mu- and B0 -> mu+ mu- decays is performed using 1.0
fb^-1 of pp collision data collected at \sqrt{s}=7 TeV with the LHCb experiment
at the Large Hadron Collider. For both decays the number of observed events is
consistent with expectation from background and Standard Model signal
predictions. Upper limits on the branching fractions are determined to be BR(Bs
-> mu+ mu-) mu+ mu-) < 1.0 (0.81) x 10^-9 at
95% (90%) confidence level.Comment: 2+6 pages; 4 figures; Accepted for publication in Physical Review
