    Baseline Trauma Symptoms: Residential and Non-Residential Survivors of Intimate Partner Violence

    Intimate Partner Violence (IPV) is a preventable public health problem that’s literature has documented the clinical presentations of those who have experienced IPV. These presentations include what is generally consistent with post-traumatic stress disorder, as well as a wide range of other symptoms including different medical comorbidities, defensiveness, difficulties in self-regulation, externalizing behavior, difficulties in relationships, withdrawal, and somatic preoccupations. These presentations are typically assumed to be symptoms of IPV but some argue that some of these, such as insecure attachment or trauma exposure, may be precursors to experiencing IPV. This has been discussed with great caution to avoid victim blaming, while still attempting to identify if certain characteristics could increase one’s likelihood of experiencing IPV. Treatment approaches have attempted to respond to several of these differing symptoms with evidenced-based practices such as prolonged-exposure therapy, cognitive-processing therapy, stress inoculation training, eye-movement desensitization and reprocessing, and medication therapy. This project reviewed how victims of IPV present to IPV service providers as those needing residential reprieve from IPV or those who are suffering from IPV but are not at immediate risk. Results from this study concluded that those who require these residential services experience higher levels of attachment difficulties, specifically rejection sensitivity, displayed level of mental distress, and traumatic symptomology than those who are seeking non-residential IPV services.     Elder Abuse in the European Union

    The rapid rise in persons over age 60 has created a platform for the rise of elder abuse all over the world. Increasing strains on caregivers and the realization that elder abuse is a serious issue have only recently produced research on the topic. In light of this, this presentation evaluates the state of the aging population in the European Union where more research has been done on elder abuse than in other areas of the world. This presentation focuses specifically on elder abuse in Germany and in Greece, as these countries represent the cultural and geographical extremes in Europe today. First, the presentation evaluates the risk factors and forms that elder abuse is taking today in the European Union. The presentation then describes the state of elder abuse in both Germany and Greece, taking into consideration demographics, culture, and current responses. Finally, this presentation evaluates possible steps to implement for effective change in the area of elder abuse

    Mapping heterogeneous buried archaeological features using multisensor data from unmanned aerial vehicles

    There is a long history of the use of aerial imagery for archaeological research, but the application of multisensor image data has only recently been facilitated by the development of unmanned aerial vehicles (UAVs). Two archaeological sites in the East Midlands U.K. that differ in age and topography were selected for survey using multisensor imaging from a fixed-wing UAV. The aim of this study was to determine optimum methodology for the use of UAVs in examining archaeological sites that have no obvious surface features and examine issues of ground control target design, thermal effects, image processing and advanced filtration. The information derived from the range of sensors used in this study enabled interpretation of buried archaeology at both sites. For any archaeological survey using UAVs, the acquisition of visible colour (RGB), multispectral, and thermal imagery as a minimum are advised, as no single technique is sufficient to attempt to reveal the maximum amount of potential information

    Discovery of an orally active benzoxaborole prodrug effective in the treatment of Chagas disease in non-human primates

    Trypanosoma cruzi, the agent of Chagas disease, probably infects tens of millions of people, primarily in Latin America, causing morbidity and mortality. The options for treatment and prevention of Chagas disease are limited and underutilized. Here we describe the discovery of a series of benzoxaborole compounds with nanomolar activity against extra- and intracellular stages of T. cruzi. Leveraging both ongoing drug discovery efforts in related kinetoplastids, and the exceptional models for rapid drug screening and optimization in T. cruzi, we have identified the prodrug AN15368 that is activated by parasite carboxypeptidases to yield a compound that targets the messenger RNA processing pathway in T. cruzi. AN15368 was found to be active in vitro and in vivo against a range of genetically distinct T. cruzi lineages and was uniformly curative in non-human primates (NHPs) with long-term naturally acquired infections. Treatment in NHPs also revealed no detectable acute toxicity or long-term health or reproductive impact. Thus, AN15368 is an extensively validated and apparently safe, clinically ready candidate with promising potential for prevention and treatment of Chagas disease

    The Astropy Problem

    The Astropy Project (http://astropy.org) is, in its own words, "a community effort to develop a single core package for Astronomy in Python and foster interoperability between Python astronomy packages." For five years this project has been managed, written, and operated as a grassroots, self-organized, almost entirely volunteer effort while the software is used by the majority of the astronomical community. Despite this, the project has always been and remains to this day effectively unfunded. Further, contributors receive little or no formal recognition for creating and supporting what is now critical software. This paper explores the problem in detail, outlines possible solutions to correct this, and presents a few suggestions on how to address the sustainability of general purpose astronomical software

    Birth weight in a large series of triplets

    Background: Triplets are often born premature and with a low birth weight. Because the incidence of triplet births is rare, there are relatively few studies describing triplet birth weight characteristics. Earlier studies are often characterized by small sample sizes and lack information on important background variables such as zygosity. The objective of this study is to examine factors associated with birth weight in a large, population-based sample of triplets registered with the Netherlands Twin Register (NTR).Methods: In a sample of 1230 triplets from 410 families, the effects of assisted reproductive techniques, zygosity, birth order, gestational age, sex, maternal smoking and alcohol consumption during pregnancy on birth weight were assessed. The resemblance among triplets for birth weight was estimated as a function of zygosity. Birth weight discordance within families was studied by the pair-wise difference between triplets, expressed as a percentage of the birth weight of the heaviest child. We compare data from triplets registered with the NTR with data from population records, which include live births, stillbirths and children that have deceased within days after birth.Results: There was no effect of assisted reproductive techniques on triplet birth weight. At gestational age 24 to 40 weeks triplets gained on average 130 grams per week; boys weighed 110 grams more than girls and triplets of smoking mothers weighted 104 grams less than children of non-smoking mothers. Monozygotic triplets had lower birth weights than di- and trizygotic triplets and birth weight discordance was smaller in monozygotic triplets than in di- and trizygotic triplets. The correlation in birth weight among monozygotic and dizygotic triplets was 0.42 and 0.32, respectively. In nearly two-thirds of the families, the heaviest and the lightest triplet had a birth weight discordance over 15%. The NTR sample is representative for the Dutch triplet population that is still alive 28 days after birth.Conclusion: Birth weight is an important determinant of childhood development. Triplet status, gestational age, sex, zygosity and maternal smoking affect birth weight. The combined effects amount to a difference of 364 grams between monozygotic girl triplets of smoking mothers compared to dizygotic boy triplets of non-smoking mothers of the same gestational age. Birth weight in triplets is also influenced by genetic factors, as indicated by a larger correlation in monozygotic than in di- and trizygotic triplets

    ADAMS project: a genetic Association study in individuals from Diverse Ancestral backgrounds with Multiple Sclerosis based in the UK

    PURPOSE: Genetic studies of multiple sclerosis (MS) susceptibility and severity have focused on populations of European ancestry. Studying MS genetics in other ancestral groups is necessary to determine the generalisability of these findings. The genetic Association study in individuals from Diverse Ancestral backgrounds with Multiple Sclerosis (ADAMS) project aims to gather genetic and phenotypic data on a large cohort of ancestrally-diverse individuals with MS living in the UK. PARTICIPANTS: Adults with self-reported MS from diverse ancestral backgrounds. Recruitment is via clinical sites, online (https://app.mantal.co.uk/adams) or the UK MS Register. We are collecting demographic and phenotypic data using a baseline questionnaire and subsequent healthcare record linkage. We are collecting DNA from participants using saliva kits (Oragene-600) and genotyping using the Illumina Global Screening Array V.3. FINDINGS TO DATE: As of 3 January 2023, we have recruited 682 participants (n=446 online, n=55 via sites, n=181 via the UK MS Register). Of this initial cohort, 71.2% of participants are female, with a median age of 44.9 years at recruitment. Over 60% of the cohort are non-white British, with 23.5% identifying as Asian or Asian British, 16.2% as Black, African, Caribbean or Black British and 20.9% identifying as having mixed or other backgrounds. The median age at first symptom is 28 years, and median age at diagnosis is 32 years. 76.8% have relapsing-remitting MS, and 13.5% have secondary progressive MS. FUTURE PLANS: Recruitment will continue over the next 10 years. Genotyping and genetic data quality control are ongoing. Within the next 3 years, we aim to perform initial genetic analyses of susceptibility and severity with a view to replicating the findings from European-ancestry studies. In the long term, genetic data will be combined with other datasets to further cross-ancestry genetic discoveries

    Structural/mechanistic insights into the efficacy of non-classical β-lactamase inhibitors against extensively drug resistant Stenotrophomonas maltophilia clinical isolates

    Clavulanic acid and avibactam are clinically deployed serine β-lactamase inhibitors, important as a defence against antibacterial resistance. Bicyclic boronates are recently discovered inhibitors of serine and some metallo β-lactamases. Here we show that avibactam and a bicyclic boronate inhibit L2 (serine β-lactamase) but not L1 (metallo β-lactamase) from the extensively drug resistant human pathogen Stenotrophomonas maltophilia. X-ray crystallography revealed that both inhibitors bind L2 by covalent attachment to the nucleophilic serine. Both inhibitors reverse ceftazidime resistance in S. maltophilia because, unlike clavulanic acid, they do not induce L1 production. Ceftazidime/inhibitor resistant mutants hyper-produce L1, but retain aztreonam/inhibitor susceptibility because aztreonam is not an L1 substrate. Importantly, avibactam, but not the bicyclic boronate is deactivated by L1 at a low rate; the utility of avibactam might be compromised by mutations that increase this deactivation rate. These data rationalize the observed clinical efficacy of ceftazidime/avibactam plus aztreonam as combination therapy for S. maltophilia infections and confirm that aztreonam-like β-lactams plus non-classical β-lactamase inhibitors, particularly avibactam-like and bicyclic boronate compounds, have potential for treating infections caused by this most intractable of drug resistant pathogens
