The aim of this thesis is to point out the mistakes made by the authorities in public tenders. The contracting authority often makes mistakes in processing the specifications not by making errors due to unclear wording of the law, but in incorrectly defined parameters of the public tenders. The thesis consists of three chapters. The first chapter describes the basic terms of contracts, the second contains mistakes the authorities make in preparation of tender specifications, which are divided into separate phases. The practical part of this work involves supervising the observance of law and the possible consequences of violations. The work of the Office for Protection of Competition, the actions taken by the candidates against improper practices of the contracting entity and the consequences of non-compliance are also mentioned.117 - Katedra účetnictvívelmi dobř

Variation in Egg Yolk Colour in Different Systems of Rearing Laying Hens

A total of 72 laying hens (ISA BROWN hybrid combination) were divided into two groups. Thirty-six layers in the first group were reared on litter while the second group of layers was reared using the cage rearing system. Samples of eggs were collected from both groups for a period of seven months. Yolk colour was determined using a portable spectrophotometer using a newly developed method, which complied with the CIELAB system. Yolk colour indicator L* decreased for the rearing system on litter, i.e. egg yolk colour became darker in the course of the whole experiment (α = 0.05). Indicator a* showed a distinct increase in the case of the rearing system on litter, with the differences being significant (α = 0.05) over the last three months of the experiment. Differences for the whole experiment were highly conclusive (α = 0.01), as seen from the increasing red colour intensity. Similarly, indicator b* also increased, i.e. egg yolk was of more distinct yellow colour. In the case of the cage-rearing system, egg yolk colour indicators correlated well with egg weight indicators, as compared to the rearing system on litter. The rearing system on litter resulted in significantly increased intensity of orange colour compared to the cage rearing system

Effect of Oregano and Marjoram Essential Oils on the Physical and Antimicrobial Properties of Chitosan Based Systems

    Get PDF
The effect of two essential oils (EOs), from Origanum vulgare and Origanum majorana, their structural properties, and concentration on physical and antimicrobial characteristics of chitosan based solutions as well as films was investigated. Results showed that significantly different behaviour was induced by variation in the compositions of given essential oils. Film-forming solutions (FFS) containing oregano oil comprised considerably greater particles in comparison with the marjoram samples. Similarly, structural changes were confirmed by SEM analysis of chitosan films modified with the EOs; the smaller particles of the marjoram oil demonstrated better compatibility with chitosan matrix. However, chitosan films enriched with the oregano oil showed significantly superior antimicrobial activity compared to the marjoram. The dissimilar effects of the two EOs were also observed by water vapour pressure (WVP) measurement; increasing the amount of oregano oil triggered a drop in the WVP of the prepared films, whereas the marjoram oil had a negligible impact in this respect. These results suggest that the structural features of active substances in the EOs play a crucial role in determining the final properties of FFS and biofilm systems. © 2017 Jana Sedlaříková et al

Genetic characterization of nodular worm infections in Asian Apes

    Get PDF
Parasitic nematodes of Oesophagostomum spp., commonly known, as 'nodular worms' are emerging as the most widely distributed and prevalent zoonotic nematodes. Oesophagostomum infections are well documented in African non-human primates; however, the taxonomy, distribution and transmission of Oesophagostomum in Asian non-human primates are not adequately studied. To better understand which Oesophagostomum species infect Asian non-human primates and determine their phylogeny we analysed 55 faecal samples from 50 orangutan and 5 gibbon individuals from Borneo and Sumatra. Both microscopy and molecular results revealed that semi-wild animals had higher Oesophagostomum infection prevalence than free ranging animals. Based on sequence genotyping analysis targeting the Internal transcribed spacer 2 of rDNA, we report for the first time the presence of O. aculeatum in Sumatran apes. Population genetic analysis shows that there is significant genetic differentiation between Bornean and Sumatran O. aculeatum populations. Our results clearly reveal that O. aculeatum in free-ranging animals have a higher genetic variation than those in semi-wild animals, demonstrating that O. aculeatum is circulating naturally in wildlife and zoonotic transmission is possible. Further studies should be conducted to better understand the epidemiology and dynamics of Oesophagostomum transmission between humans, non-human primates and other wild species and livestock in Southeast Asia

How many species of whipworms do we share? Whipworms from man and other primates form two phylogenetic lineages

    Get PDF
The whipworms, i.e. parasitic nematodes of the genus Trichuris Roederer, 1761, infect a variety of mammals. Apparently low diversity of primate-infecting species of Trichuris strongly contrasts with the high number of species described in other mammalian hosts. The present study addresses the diversity of whipworms in captive and free-ranging primates and humans by analysing nuclear (18S rRNA, ITS2) and mitochondrial (cox1) DNA. Phylogenetic analyses revealed that primate whipworms form two independent lineages: (i) the Trichuris trichiura (Linnaeus, 1771) clade comprised of genetically almost identical whipworms from human and other primates, which suggests the ability of T. trichiura to infect a broader range of primates; (ii) a clade containing primarily Trichuris suis Schrank, 1788, where isolates from human and various primates formed a sister group to isolates from pigs; the former isolates thus may represent of more species of Trichuris in primates including humans. The analysis of cox1 has shown the polyphyly of the genera Trichuris and Capillaria, Zeder, 1800. High sequence similarity of the T. trichiura isolates from humans and other primates suggests their zoonotic potential, although the extent of transmission between human and other non‐human primates remains questionable and requires further stud

Muckle-Wells Syndrome Across Four Generations in One Czech Family: Natural Course of the Disease

Background: Muckle-Wells syndrome (MWS) represents a moderate phenotype of cryopyrinopathies. Sensorineural hearing loss and AA amyloidosis belong to the most severe manifestations of uncontrolled disease. Simultaneous discovery of MWS in four generations of one large kindred has enabled us to document natural evolution of untreated disease and their response to targeted therapy. Methods: A retrospective case study, clinical assessment at the time of diagnosis and 2-year prospective follow-up using standardized disease assessments were combined. Results: Collaborative effort of primary care physicians and pediatric and adult specialists led to identification of 11 individuals with MWS within one family. Presence of p.Ala441Val mutation was confirmed. The mildest phenotype of young children suffering with recurrent rash surprised by normal blood tests and absence of fevers. Young adults all presented with fevers, rash, conjunctivitis, and arthralgia/arthritis with raised inflammatory markers. Two patients aged over 50 years suffered with hearing loss and AA amyloidosis. IL-1 blockade induced disease remission in all individuals while hearing mildly improved or remained stable in affected patients as did renal function in one surviving individual with amyloidosis. Conclusions: We have shown that severity of MWS symptoms gradually increased with age toward distinct generation-specific phenotypes. A uniform trajectory of disease evolution has encouraged us to postpone institution of IL-1 blockade in affected oligosymptomatic children. This report illustrates importance of close interdisciplinary collaboration
