    Where Does Blood Flow Restriction Fit in the Toolbox of Athletic Development? A Narrative Review of the Proposed Mechanisms and Potential Applications

    Blood flow-restricted exercise is currently used as a low-intensity time-efficient approach to reap many of the benefits of typical high-intensity training. Evidence continues to lend support to the notion that even highly trained individuals, such as athletes, still benefit from this mode of training. Both resistance and endurance exercise may be combined with blood flow restriction to provide a spectrum of adaptations in skeletal muscle, spanning from myofibrillar to mitochondrial adjustments. Such diverse adaptations would benefit both muscular strength and endurance qualities concurrently, which are demanded in athletic performance, most notably in team sports. Moreover, recent work indicates that when traditional high-load resistance training is supplemented with low-load, blood flow-restricted exercise, either in the same session or as a separate training block in a periodised programme, a synergistic and complementary effect on training adaptations may occur. Transient reductions in mechanical loading of tissues afforded by low-load, blood flow-restricted exercise may also serve a purpose during de-loading, tapering or rehabilitation of musculoskeletal injury.     Ibuprofen ingestion does not affect markers of post-exercise muscle inflammation

    PURPOSE: We investigated if oral ingestion of ibuprofen influenced leucocyte recruitment and infiltration following an acute bout of traditional resistance exercise Methods: Sixteen male subjects were divided into two groups that received the maximum over-the-counter dose of ibuprofen (1200mg d(-1)) or a similarly administered placebo following lower body resistance exercise. Muscle biopsies were taken from m.vastus lateralis and blood serum samples were obtained before and immediately after exercise, and at 3 and 24 h after exercise. Muscle cross-sections were stained with antibodies against neutrophils (CD66b and MPO) and macrophages (CD68). Muscle damage was assessed via creatine kinase and myoglobin in blood serum samples, and muscle soreness was rated on a ten-point pain scale. RESULTS: The resistance exercise protocol stimulated a significant increase in the number of CD66b(+) and MPO(+) cells when measured 3 h post exercise. Serum creatine kinase, myoglobin and subjective muscle soreness all increased post-exercise. Muscle leucocyte infiltration, creatine kinase, myoglobin and subjective muscle soreness were unaffected by ibuprofen treatment when compared to placebo. There was also no association between increases in inflammatory leucocytes and any other marker of cellular muscle damage. CONCLUSION: Ibuprofen administration had no effect on the accumulation of neutrophils, markers of muscle damage or muscle soreness during the first 24 h of post-exercise muscle recovery

    Superior Physiological Adaptations After a Microcycle of Short Intervals Versus Long Intervals in Cyclists

    Purpose: To compare the effects of a 1-week high-intensity aerobic-training shock microcycle composed of either 5 short-interval sessions (SI; n = 9, 5 series with 12 × 30-s work intervals interspersed with 15-s recovery and 3-min recovery between series) or 5 long-interval sessions (LI; n = 8, 6 series of 5-min work intervals with 2.5-min recovery between series) on indicators of endurance performance in well-trained cyclists. Methods: Before and following 6 days with standardized training loads after the 1-week high-intensity aerobic-training shock microcycle, both groups were tested in physiological determinants of endurance performance. Results: From pretraining to posttraining, SI achieved a larger improvement than LI in maximal oxygen uptake (5.7%; 95% confidence interval, 1.3–10.3; P = .015) and power output at a blood lactate concentration of 4 mmol·L−1 (3.8%; 95% confidence interval, 0.2–7.4; P = .038). There were no group differences in changes of fractional use of maximal oxygen uptake at a workload corresponding to a blood lactate concentration of 4 mmol·L−1, gross efficiency, or the 1-minute peak power output from the maximal-oxygen-uptake test. Conclusion: The SI protocol may induce superior changes in indicators of endurance performance compared with the LI protocol, indicating that SI can be a good strategy during a 1-week high-intensity aerobic-training shock microcycle in well-trained cyclists

    Signal Propagation in Feedforward Neuronal Networks with Unreliable Synapses

    In this paper, we systematically investigate both the synfire propagation and firing rate propagation in feedforward neuronal network coupled in an all-to-all fashion. In contrast to most earlier work, where only reliable synaptic connections are considered, we mainly examine the effects of unreliable synapses on both types of neural activity propagation in this work. We first study networks composed of purely excitatory neurons. Our results show that both the successful transmission probability and excitatory synaptic strength largely influence the propagation of these two types of neural activities, and better tuning of these synaptic parameters makes the considered network support stable signal propagation. It is also found that noise has significant but different impacts on these two types of propagation. The additive Gaussian white noise has the tendency to reduce the precision of the synfire activity, whereas noise with appropriate intensity can enhance the performance of firing rate propagation. Further simulations indicate that the propagation dynamics of the considered neuronal network is not simply determined by the average amount of received neurotransmitter for each neuron in a time instant, but also largely influenced by the stochastic effect of neurotransmitter release. Second, we compare our results with those obtained in corresponding feedforward neuronal networks connected with reliable synapses but in a random coupling fashion. We confirm that some differences can be observed in these two different feedforward neuronal network models. Finally, we study the signal propagation in feedforward neuronal networks consisting of both excitatory and inhibitory neurons, and demonstrate that inhibition also plays an important role in signal propagation in the considered networks.Comment: 33pages, 16 figures; Journal of Computational Neuroscience (published

    Penelitian ini dilakukan untuk melihat pengaruh model cooperative learning metode TSTS dan NHT terhadap pemahaman konsep akuntansi ditinjau dari kemampuan awal siswa dalam mata pelajaran akuntansi kelas X Akuntansi SMKN 1 Kota Tasikmalaya. Metode yang digunakan adalah quasi eksperimen dengan pengambilan sampel cluster random sampling yaitu kelas X-Ak 1 dan X-Ak 3. Kemampuan awal setiap kelas dibagi menjadi dua kategori, yaitu kemampuan awal tinggi dan kemampuan awal rendah. Data penelitian dikumpulkan melalui tes tulis yaitu tes kemampuan awal, pre test, dan pos test. Uji Hipotesis menggunakan uji anova between subject design faktorial 2x2. Hasil penelitian menunjukan ada pengaruh metode pembelajaran NHT dan TSTS terhadap pemahaman konsep akuntansi. Ada pengaruh kemampuan awal terhadap pemahaman konsep akuntansi. Ada pengaruh dan interaksi penggunaan metode TSTS dan NHT terhadap peningkatan pemahaman konsep akuntansi ditinjau dari kemampuan awal. Pemahaman konsep siswa yang memiliki kemampuan awal tinggi dengan metode NHT lebih tinggi dari penggunaan metode TSTS dengan kemampuan awal yang sama. Peningkatan pemahaman konsep siswa dengan kemampuan awal rendah metode TSTS lebih baik dari metode NHT. This research was conducted to see the effect of cooperative learning model of Two Stay Two Stray method and Numbered Head Together method toward the understanding of accounting concepts seen from the student’s prior knowledge.The research method was is quasi experimental with cluster random sampling in class X-Ak 1 and X-Ak 3. Prior Knowledge of each class is divided into two categories, namely high prior knowledge and low prior knowledge. The instrument used in this research was tests, was prerequisite test, pre test and post test. Hypothesis test using ANOVA test between subject 2x2 factorial design.The findings of this research are: there is the influence of the use of methods TSTS and NHT toward the increase of the understanding of accounting concepts. There is also the influence of the prior knowledge toward the increase of the understanding of accounting concepts. Moreover an effect of interaction between TSTS method and NHT method with the student’s prior knowledge toward the increase of the understanding of accounting concepts. The use of NHT method and TSTS method increased the understanding of the concept of students who have a high prior knowledge. Meanwhile, the use of TSTS method is better than NHT method in improving student comprehension with low prior knowledge

    MicroRNAs in muscle: Characterizing the powerlifter phenotype

    Powerlifters are the epitome of muscular adaptation and are able to generate extreme forces. The molecular mechanisms underpinning the significant capacity for force generation and hypertrophy are not fully elucidated. MicroRNAs (miRs) are short non-coding RNA sequences that control gene expression via promotion of transcript breakdown and/or translational inhibition. Differences in basal miR expression may partially account for phenotypic differences in muscle mass and function between powerlifters and untrained age-matched controls. Muscle biopsies were obtained from m. vastus lateralis of 15 national level powerlifters (25.1 ± 5.8 years) and 13 untrained controls (24.1 ± 2.0 years). The powerlifters were stronger than the controls (isokinetic knee extension at 60°/s: 307.8 ± 51.6 Nm vs. 211.9 ± 41.9 Nm, respectively P < 0.001), and also had larger muscle fibers (type I CSA 9,122 ± 1,238 vs. 4,511 ± 798 μm2 p < 0.001 and type II CSA 11,100 ± 1,656 vs. 5,468 ± 1,477 μm2 p < 0.001). Of the 17 miRs species analyzed, 12 were differently expressed (p < 0.05) between groups with 7 being more abundant in powerlifters and five having lower expression. Established transcriptionally regulated miR downstream gene targets involved in muscle mass regulation, including myostatin and MyoD, were also differentially expressed between groups. Correlation analysis demonstrates the abundance of eight miRs was correlated to phenotype including peak strength, fiber size, satellite cell abundance, and fiber type regardless of grouping. The unique miR expression profiles between groups allow for categorization of individuals as either powerlifter or healthy controls based on a five miR signature (miR-126, -23b, -16, -23a, -15a) with considerable accuracy (100%).     The effect of session order on the physiological, neuromuscular, and endocrine responses to maximal speed and weight training sessions over a 24-h period

    Objectives: Athletes are often required to undertake multiple training sessions on the same day with these sessions needing to be sequenced correctly to allow the athlete to maximize the responses of each session. We examined the acute effect of strength and speed training sequence on neuromuscular, endocrine, and physiological responses over 24 hours. Design: 15 academy rugby union players completed this randomized crossover study. Method: Players performed a weight traiing session followed 2 hours later by a speed training session (WS) and on a separate day reversed the order (SW). Countermovement jumps (CMJ), perceived muscle soreness (MS), and blood samples were collected immediately prior, immediately post, and 24 hours post sessions one and two respectively. Jumps were analyzed for power, jump height, rate of force development, and velocity. Blood was analyzed for testosterone (T), cortisol (C), lactate and creatine kinase (CK). Results: There were no differences between CMJ variables at any of the post training time points (p > 0.05). Likewise, CK, T, C , and MS were unaffected by session order (p > 0.05). However, 10 meter sprint time was significantly faster (Mean ± SD; SW 1.80s ± 0.11 vs. WS 1.76 ± 0.08s; p > 0.05) when speed was sequenced second. Lactate levels were significantly higher immediately post speed sessions versus weight training sessions at both time points (p < 0.05). Conclusions: The sequencing of strength and speed training does not affect the neuromuscular, endocrine, and physiological recovery over 24 hours. However, speed may be enhanced when performed as the second session

    Patient perspectives on infection prevention and control in cancer care : a survey of knowledge and attitudes among persons with cancer and their next of kin

    Background: With the growing prevalence of antimicrobial resistance, there may be a need to strengthen infection prevention and control (IPC) measures in cancer care. When developing clinical guidelines, it is important to incorporate patient perspectives. Aim: To determine the knowledge of, and attitudes towards, IPC among persons with cancer and their next of kin in Norway. Methods: Through discussions in expert panels and a pilot study, a survey was developed consisting of 13 knowledge statements to be judged true/false and 40 attitude items to be judged using a Likert scale and was sent to a panel of people with cancer experience on August 22nd, 2023. The mean correct responses and attitude scores were reported. Findings: Of 551 respondents, the mean correct response to IPC-related knowledge questions was 79% (95% confidence interval: 78e80). Respondents were most knowledgeable about hand hygiene (99%, 546/551), but least knowledgeable about its role in preventing antibiotic resistance (41%, 225/551). Strong support was noted for IPC, especially within the patient responsibilities theme, with a mean score of 4.83. However, there was a notable reluctance towards some selected intrusive IPC measures, such as reducing contact with close relations. Conclusion: This survey revealed a high level of knowledge and attitudes that support the importance of IPC among persons with cancer in Norway and their next of kin. We recommend including patient perspectives in future development of IPC guidelines.peer-reviewe

    Human skeletal muscle plasmalemma alters its structure to change its Ca2+-handling following heavy-load resistance exercise

    High-force eccentric exercise results in sustained increases in cytoplasmic Ca2+ levels ([Ca2+]cyto), which can cause damage to the muscle. Here we report that a heavy-load strength training bout greatly alters the structure of the membrane network inside the fibres, the tubular (t-) system, causing the loss of its predominantly transverse organization and an increase in vacuolation of its longitudinal tubules across adjacent sarcomeres. The transverse tubules and vacuoles displayed distinct Ca2+-handling properties. Both t-system components could take up Ca2+ from the cytoplasm but only transverse tubules supported store-operated Ca2+ entry. The retention of significant amounts of Ca2+ within vacuoles provides an effective mechanism to reduce the total content of Ca2+ within the fibre cytoplasm. We propose this ability can reduce or limit resistance exercise-induced, Ca2+-dependent damage to the fibre by the reduction of [Ca2+]cyto to help maintain fibre viability during the period associated with delayed onset muscle soreness

    Functional Organization of Locomotor Interneurons in the Ventral Lumbar Spinal Cord of the Newborn Rat

    Although the mammalian locomotor CPG has been localized to the lumbar spinal cord, the functional-anatomical organization of flexor and extensor interneurons has not been characterized. Here, we tested the hypothesis that flexor and extensor interneuronal networks for walking are physically segregated in the lumbar spinal cord. For this purpose, we performed optical recordings and lesion experiments from a horizontally sectioned lumbar spinal cord isolated from neonate rats. This ventral hemi spinal cord preparation produces well-organized fictive locomotion when superfused with 5-HT/NMDA. The dorsal surface of the preparation was visualized using the Ca2+ indicator fluo-4 AM, while simultaneously monitoring motor output at ventral roots L2 and L5. Using calcium imaging, we provided a general mapping view of the interneurons that maintained a stable phase relationship with motor output. We showed that the dorsal surface of L1 segment contains a higher density of locomotor rhythmic cells than the other segments. Moreover, L1 segment lesioning induced the most important changes in the locomotor activity in comparison with lesions at the T13 or L2 segments. However, no lesions led to selective disruption of either flexor or extensor output. In addition, this study found no evidence of functional parcellation of locomotor interneurons into flexor and extensor pools at the dorsal-ventral midline of the lumbar spinal cord of the rat
