    Nanoparticles from biodegradable polymers are considered one of the most promising systems for biomedical application as drug delivery systems. Therefore, the synthesis and characterization of a new aliphatic biodegradable copolyester named PBS/PBDL (poly(butylene succinate-co- butylene dilinoleate)) intended to the application as drug delivery system is reported in the thesis. Surfactant-free biodegradable and narrowly distributed, nanosized spherical particles (RH < 60 nm) have been produced from the biodegradable material by applying a single-step nanoprecipitation protocol. The size of the generated polymer nanoparticles (PNPs) could be controlled by adjusting the polymer concentration, the choice of organic solvent, mixing different organic solvents or by changing temperature and ionic strength. By optimizing such parameters sub-100 nm uniform PNPs can be produced through this methodology including the advantage and ability to scale-up production. The nanoparticles structure was characterized in detail by employing a variety of scattering techniques and transmission electron microscopy (TEM). Combined static light scattering (SLS) and dynamic light scattering (DLS) measurements suggested that the nanoparticles comprise a porous core conferring them a non-compact characteristic. Tato disertační práce je proto zaměřena na syntézu a charakterizaci nových biodegradovatelných alifatických kopolyesterů PBS/PBDL (poly(butylen sukcinát-co-butylen dilinoleát) pro cílenou dopravu léčiv. Tyto polymery byly použity pro přípravu sférických nanočástic (poloměr < 60 nm) s úzkou distribucí rozměrů pomocí jednokrokové metody nanoprecipitace bez přítomnosti povrchově aktivních látek. Rozměr vytvořených polymerních nanočástic bylo možno řídit v požadovaném rozsahu volbou koncentrace polymeru, výběrem organického rozpouštědla, použitím směsi organických rozpouštědel nebo nastavením teploty a iontové síly. Optimalizací těchto parametrů bylo možno připravit nanočástice s rozměry menšími než 100 nm, výhodou tohoto postupu je možnost přípravy ve velkých množstvích. Struktura nanočástic byla podrobně charakterizována rozptylovými technikami a transmisní elektronovou mikroskopií. Kombinace statického a dynamického rozptylu světla ukázala, že nanočástice jsou porézní a tedy jejich interní struktura není kompaktní.     Paclitaxel-loaded biodegradable ROS-sensitive nanoparticles for cancer therapy

Background: Reactive oxygen species (ROS), such as hydrogen peroxide and superoxide, trigger biodegradation of polymer-based nanoparticles (NPs) bearing pinacol-type boronic ester groups. These NPs may selectively release their cargo, in this case paclitaxel (PTX), at the high levels of ROS present in the intracellular environment of inflamed tissues and most tumors. Purpose: The main objective was to determine anti-tumor efficacy of PTX-loaded ROS-sensitive NPs and to examine whether macrophage infiltration had any impact on treatment efficacy. Methods: NPs were synthesized and their characteristics in the presence of H2O2 were demonstrated. Both confocal microscopy as well as flow cytometry approaches were used to determine degradation of ROS-sensitive NPs. HeLa cells were cultured in vitro and used to establish tumor xenografts in nude mice. In vivo experiments were performed to understand toxicity, biodistribution and anti-tumor efficacy of the NPs. Moreover, we performed immunohistochemistry on tumor sections to study infiltration of M1 and M2 subsets of macrophages. Results: We demonstrated that PTX delivered in NPs containing a ROS-sensitive polymer exhibits a better anti-tumor efficacy than PTX in NPs containing ROS-non-sensitive polymer, free PTX or Abraxane® (nab-PTX). The biodistribution revealed that ROS-sensitive NPs exhibit retention in liver, spleen and lungs, suggesting a potential to target cancer metastasizing to these organs. Finally, we demonstrated a correlation between infiltrated macrophage subsets and treatment efficacy, possibly contributing to the efficient anti-tumor effects. Conclusion: Treatment with ROS-sensitive NPs containing PTX gave an improved therapeutic effect in HeLa xenografts than their counterpart, free PTX or nab-PTX. Our data revealed a correlation between macrophage infiltration and efficiency of the different antitumor treatments, as the most effective NPs resulted in the highest infiltration of the anti-tumorigenic M1 macrophages

    A complete parton level analysis of lv + four jets production at the LHC is presented, including all processes at order O(alpha^6), O(alpha^4*alpha_s^2) and O(alpha^2*alpha_s^4). The infinite Higgs mass scenario, which is considered as a benchmark for strong scattering theories and is the limiting case for composite Higgs models, is confronted with the Standard Model light Higgs predictions in order to determine whether a composite Higgs signal can be detected as an excess of events in boson--boson scattering.Comment: More detailed discussion of the effects of the reconstruction of the longitudinal neutrino momentum.     A complete parton level analysis of ll + four jets l = e,mu and 3lv + two jets production at the LHC is presented, including all processes at order \ordEW, \ordQCD and \ordQCDsq when appropriate. The infinite Higgs mass scenario, which is considered as a benchmark for strong scattering theories and is the limiting case for composite Higgs models, and one example of a model incorporating a Strongly Interacting Light Higgs are confronted with the Standard Model light Higgs predictions.     PHANTOM: a Monte Carlo event generator for six parton final states at high energy colliders

    PHANTOM is a tree level Monte Carlo for six parton final states at proton--proton, proton--antiproton and electron--positron collider at O(alpha_ew^6) and O(alpha_ew^4*alpha_s^2) including possible interferences between the two sets of diagrams. This comprehends all purely electroweak contribution as well as all contributions with one virtual or two external gluons. It can generate unweighted events for any set of processes and it is interfaced to parton shower and hadronization packages via the last Les Houches Accord protocol.     In-vivo multilaboratory investigation of the optical properties of the human head

    The in-vivo optical properties of the human head are investigated in the 600–1100 nm range on different subjects using continuous wave and time domain diffuse optical spectroscopy. The work was performed in collaboration with different research groups and the different techniques were applied to the same subject. Data analysis was carried out using homogeneous and layered models and final results were also confirmed by Monte Carlo simulations. The depth sensitivity of each technique was investigated and related to the probed region of the cerebral tissue. This work, based on different validated instruments, is a contribution to fill the existing gap between the present knowledge and the actual in-vivo values of the head optical properties

    A novel cell-based immunofluorescence assay for the detection of autoantibodies to myelin-associated glycoprotein

    Peripheral neuropathy with antibodies to myelin-associated glycoprotein (MAG) is an autoimmune demyelinating disorder of the peripheral nervous system caused by pathogenic IgM recognizing the human natural killer-1 glycoepitope expressed on MAG. This study aimed to analyze the performance of a new indirect immunofluorescence cell-based assay (CBA, EUROIMMUN) for the detection of anti-MAG IgM. Antibody reactivity was determined in sera from 95 patients with clinical and neurophysiological evidence of anti-MAG-associated neuropathy and in control samples from 55 patients with other forms of peripheral neuropathy. Compared to the results of the gold standard method (ELISA, Bühlmann) and using samples at a dilution of 1:100, the CBA had a sensitivity of 98.9% and a specificity of 100% (PPV 100%, NPV 98.2%). In conclusion, the CBA allows the detection of antibodies to MAG using an easy and standardized technique, and it presents a sensitive and specific alternative to the more time-consuming ELISA. Larger studies are needed to address anti-MAG titer monitoring in parallel with clinical activity

    Treatment characteristics and outcomes of pure Acinar cell carcinoma of the pancreas - A multicentric European study on radically resected patients

    Background: Acinar cell carcinomas (ACC) belong to the exocrine pancreatic malignancies. Due to their rarity, there is no consensus regarding treatment strategies for resectable ACC. Methods: This is a retrospective multicentric study of radically resected pure pancreatic ACC. Primary endpoints were overall survival (OS) and disease-free survival (DFS). Further endpoints were oncologic outcomes related to tumor stage and therapeutic protocols. Results: 59 patients (44 men) with a median age of 64 years were included. The median tumor size was 45.0 mm. 61.0% were pT3 (n = 36), nodal positivity rate was 37.3% (n = 22), and synchronous distant metastases were present in 10.1% of the patients (n = 6). 5-Years OS was 60.9% and median DFS 30 months. 24 out of 31 recurred systemically (n = 18 only systemic, n = 6 local and systemic). Regarding TNM-staging, only the N2-stage negatively influenced OS and DFS (p = 0.004, p = 0.001). Adjuvant treatment protocols (performed in 62.7%) did neither improve OS (p = 0.542) nor DFS (p = 0.159). In 9 cases, radical resection was achieved following neoadjuvant therapy. Discussion: Radical surgery is currently the mainstay for resectable ACC, even for limited metastatic disease. Novel (neo)adjuvant treatment strategies are needed, since current systemic therapies do not result in a clear survival benefit in the perioperative setting

    Cashew gum (Anacardium occidentale) as a potential source for the production of tocopherol-loaded nanoparticles: formulation, release profile and cytotoxicity

    Every year, more than thirty thousand tons of Cashew gum (Anacardium occidentale, family: Anacardiaceae) are produced in Brazil; however, only a small amount is used for different applications in foodstuff and in pharmaceutical industries. As a raw material for the production of drug delivery systems, cashew gum is still regarded as an innovative compound worth to be exploited. In this work, cashew gum was extracted from the crude exudate of cashew tree employing four methodologies resulting in a light brown powder in different yields (40.61% to 58.40%). The total ashes (0.34% to 1.05%) and moisture (12.90% to 14.81%) were also dependent on the purification approach. FTIR spectra showed the typical bands of purified cashew gum samples, confirming their suitability for the development of a pharmaceutical product. Cashew gum nanoparticles were produced by nanoprecipitation resulting in particles of low polydispersity (<0.2) and an average size depending on the percentage of the oil. The zeta potential of nanoparticles was found to be below 20 mV, which promotes electrostatic stability. Encapsulation efficiencies were above 99.9%, while loading capacity increased with the increase of the percentage of the oil content of particles. The release of the oil from the nanoparticles followed the KorsmeyerPeppas kinetics model, while particles did not show any signs of toxicity when tested in three distinct cell lines (LLC-MK2, HepG2, and THP-1).
