Selecció clonal i sanitària de la varietat picapoll blanc a la denominació d'origen Pla de Bages
El picapoll blanc, varietat cultivada a la denominació d'origen (DO) Pla de Bages, és considerada autòctona catalana. La selecció clonal i sanitària s'ha realitzat per permetre la certificació i multiplicació dels millors clons, amb una elevada adaptació a les condicions ambientals i amb una producció per a l'obtenció de vins de qualitat. També, aquest programa de selecció permet la conservació de la diversitat intravarietal d'una de les varietats que va ser més difosa al segle passat a Catalunya i que resta oblidada a gairebé totes les zones vitícoles excepte a aquesta DO, i procura mantenir així el nivell de variabilitat que presenta encara la població actual.El picapoll blanco, variedad cultivada en la denominación de origen (DO) Pla de Bages, es considerada autóctona catalana. Component-Resolved in Vitro Diagnosis in Peach-Allergic Patients
BACKGROUND: The in vitro diagnosis of pollen-related food allergy presents low specifi city and reproducibility with many conventional extracts. This can be improved using natural purifi ed allergens, recombinant purifi ed allergens, or both. OBJECTIVE: We compared specifi c immunoglobulin (Ig) E determination (sIgE), the basophil activation test (BAT), the histamine release test (HRT), and the cellular allergen stimulation test (CAST) using natural and recombinant allergens in the diagnosis of peach allergy. METHODS: Thirty-two peach allergic patients were studied. Skin prick tests were performed with commercial peach and extract with Mal d 1, nPru p 3, and profi lin (nPho d 2). sIgE, BAT, CAST, and HRT were determined using rPru p 3, rMal d 3, rBet v 1, rMal d 1, and rMal d 4. RESULTS: Agreement between the techniques was good with all the allergens, except HRT with rMal d 1 and rMal d 4. With rPru p 3, sIgE, CAST, BAT, and HRT showed sensitivity values of 88%, 81%, 72%, and 69% and specifi city values of 100%, 93%, 97%, and 83%, respectively. In patients with systemic symptoms or contact urticaria, the values were 100%, 85%, 81%, and 81%. In patients with oral allergy syndrome, sensitivity to profi lins or homologues of Bet v 1 was detected in 100% of the cases by all the techniques, except by HRT with rMal d 1, which detected 66% of the cases. CONCLUSIONS: The use of single allergens in the in vitro diagnosis of peach allergy by specifi c IgE determination, BAT, and CAST offers high specifi city and sensitivity, with better results than the HRT
The Great American Biotic Interchange revisited: a new perspective from the stable isotope record of Argentine Pampas fossil mammals
This study aims at assessing resource and habitat use, niche occupation and trophic interactions from a stable isotope perspective on fossil mammals from the Argentine Pampas during the Great American Biotic Interchange (GABI). We present stable isotope data of more than 400 samples belonging to 10 mammalian orders and spanning a temporal range from ~9.5 Ma to ~12 ky. Rodents, notoungulates and pilosians record an increase in the consumption of C4 plants, whereas litopterns and cingulates show δ13C values that remain mostly within a C3-dominated diet. Our stable isotope data indicates that the expansion of C4 vegetation opened up new niche opportunities, probably alleviating resource competition among endemic taxa. Gomphothere, equid and camelid δ13C records show a broad variability pointing to consumption of C3 and mixed C3-C4 vegetation. This flexible dietary behavior may have facilitated the successful settlement of immigrant groups in South America. In the case of carnivorous taxa, Late Miocene pre-GABI endemic sparassodonts consumed prey from C3 environments, whereas immigrant carnivorans preferred prey from mixed C3-C4 areas. Gregariousness in the giant sloth Lestodon (Xenarthra): multi-proxy approach of a bonebed from the Last Maximum Glacial of Argentine Pampas
Megamammals constituted an important component in the Pleistocene faunal communities of South America. Paleobiological and paleoecological studies involving different megamammal taxa have increased significantly in the last years, but there are still several poorly-known issues of its life history. In this work, we analyze an assemblage composed of 13 individuals of different ontogenetic stages, and possibly different sex, belonging to the giant ground sloth Lestodon armatus (Xenarthra, Folivora), recovered from Playa del Barco site (Pampean Region, Argentina). A dating of 19,849 years Cal BP allows assigning this assemblage to a period of the MIS (Marine Isotope Stage) 2 related to the end of the Last Glacial Maximum. Based on multiple lines of research (e.g. taphonomy, paleopathology, osteohistology, isotopy), we interpret the origin of the assemblage and diverse paleobiological and paleoecological aspects (e.g. social behavior, ontogenetic changes, sexual dimorphism, diseases, resource and habitat use, trophic relationships) of L. armatus. Evidence suggests that the assemblage was formed by a local single event of catastrophic mortality, which affected different members of a social group. This record represents the first accurate evidence of gregariousness for this ground sloth, providing new data on a poorly-known behavior among extinct Folivora
SERCA inhibition improves lifespan and healthspan in a chemical model of Parkinson disease in Caenorhabditis elegans
Introduction: The high prevalence of neurodegenerative diseases in our population and the lack of effective treatments encourage the search for new therapeutic targets for these pathologies. We have recently described that submaximal inhibition of the Sarco-Endoplasmic Reticulum Ca2+ ATPase (SERCA), the main responsible for ER calcium storage, is able to increase lifespan in Caenorhabditis elegans worms by mechanisms involving mitochondrial metabolism and nutrient-sensitive pathways.Methods: We have studied here the effects of submaximal SERCA inhibition in a chemical model of Parkinson’s disease (PD) induced in C. elegans worms by treatment with the mitochondrial complex I inhibitor rotenone. For specific SERCA inhibition, we treated worms with RNAi against sca-1, the sole orthologue of SERCA in C. elegans.Results and Discussion: Our results show that rotenone produces alterations in worms that include decreased lifespan, smaller size, reduced fertility, decreased motility, defecation and pumping rate, increased mitochondrial ROS production, reduced mitochondrial membrane potential and oxygen consumption rate, altered mitochondrial structure, and altered ethanol preference in behavioral studies. Most of these alterations were either fully or partially reversed in worms treated with sca-1 RNAi, suggesting that SERCA inhibition could be a novel pharmacological target in the prevention or treatment of neurodegeneration
Post-transcriptional control of a stemness signature by RNA-binding protein MEX3A regulates murine adult neurogenesis
Neural stem cells (NSCs) in the adult murine subependymal zone balance their self-renewal capacity and glial identity with the potential to generate neurons during the lifetime. Adult NSCs exhibit lineage priming via pro-neurogenic fate determinants. However, the protein levels of the neural fate determinants are not sufficient to drive direct differentiation of adult NSCs, which raises the question of how cells along the neurogenic lineage avoid different conflicting fate choices, such as self-renewal and differentiation. Here, we identify RNA-binding protein MEX3A as a post-transcriptional regulator of a set of stemness associated transcripts at critical transitions in the subependymal neurogenic lineage. MEX3A regulates a quiescence-related RNA signature in activated NSCs that is needed for their return to quiescence, playing a role in the long-term maintenance of the NSC pool. Furthermore, it is required for the repression of the same program at the onset of neuronal differentiation. Our data indicate that MEX3A is a pivotal regulator of adult murine neurogenesis acting as a translational remodeller.© 2023. The Author(s)
The Sunrise Mission
The first science flight of the balloon-borne \Sunrise telescope took place
in June 2009 from ESRANGE (near Kiruna/Sweden) to Somerset Island in northern
Canada. We describe the scientific aims and mission concept of the project and
give an overview and a description of the various hardware components: the 1-m
main telescope with its postfocus science instruments (the UV filter imager
SuFI and the imaging vector magnetograph IMaX) and support instruments (image
stabilizing and light distribution system ISLiD and correlating wavefront
sensor CWS), the optomechanical support structure and the instrument mounting
concept, the gondola structure and the power, pointing, and telemetry systems,
and the general electronics architecture. We also explain the optimization of
the structural and thermal design of the complete payload. The preparations for
the science flight are described, including AIV and ground calibration of the
instruments. The course of events during the science flight is outlined, up to
the recovery activities. Finally, the in-flight performance of the
instrumentation is briefly summarized.Comment: 35 pages, 17 figure
The biota of the Upper Cretaceous site of Lo Hueco (Cuenca, Spain)
The Late Cretaceous (Campanian-Maastrichtian) fossil site of Lo Hueco was recently discovered close to the village of Fuentes (Cuenca,
Spain) during the cutting of a little hill for installation of the railway of the Madrid-Levante high-speed train. To date, it has yielded a rich
collection of well-preserved Cretaceous macrofossils, including plants, invertebrates, and vertebrates. The recovered fossil assemblage
is mainly composed of plants, molluscs (bivalves and gastropods), actinopterygians and teleosteans fishes, amphibians, panpleurodiran
(bothremydids) and pancryptodiran turtles, squamate lizards, eusuchian crocodyliforms, rhabdodontid ornithopods, theropods (mainly
dromaeosaurids), and titanosaur sauropods. This assemblage was deposited in a near-coast continental muddy floodplain crossed by distributary
sandy channels, exposed intermittently to brackish or marine and freshwater flooding as well as to partial or total desiccation events.The Konzentrat-Lagerstatt of Lo Hueco constitutes a singular accumulation of fossils representing individuals of some particular lineages
of continental tetrapods, especially titanosaurs, eusuchians and bothremydid turtles. In the case of the titanosaurs, the site has yielded
multiple partial skeletons in anatomical connection or with a low dispersion of their skeletal elements. A combination of new taxa, new
records of taxa previously known in the Iberian Peninsula, and relatively common taxa in the European record compose the Lo Hueco biota.
The particular conditions of the fossil site of Lo Hueco and the preliminary results indicate that the analysis of the geological context, the
floral and faunal content, and the taphonomical features of the site provide elements that will be especially useful for reassess the evolutionary
history of some lineages of European Late Cretaceous reptiles.Peer reviewe
