Functional Haplotypes of the hTERT Gene, Leukocyte Telomere Length Shortening, and the Risk of Peripheral Arterial Disease
Background: The development of peripheral arterial disease (PAD) is heterogeneous even in the presence of similar risk factors. Our aim was to determine whether inter-individual differences in leukocyte telomere length contribute to the susceptibility of PAD. Methods A total of 485 patients with PAD (defined by the ankle-brachial index) and 970 age- and gender-matched controls were recruited from seven rural communities in Henan Province in China. The relative leukocyte telomere length was determined by a quantitative PCR-based method. Two common promoter variants of the hTERT gene were genotyped to assess their effects on telomere length and the risk of PAD. In vivo luciferase assay was performed to study the transcriptional activity. Results: After adjustment for vascular risk factors and genetic variants in the hTERT gene, individuals in the lowest and middle tertiles of telomere length had a significantly higher risk of PAD than did those in the highest tertile (odds ratio [OR] 1.73, 95% confidence interval [CI] 1.29–2.49 in the middle tertile; 3.15, 95%CI 2.31–4.29 in the lowest tertile). Haplotype analysis using the 2 variants (rs2735940 and rs2853669) showed that subjects with the at-risk C-C haplotype had shorter telomere length than those individuals with the T-T haplotype and consistently had 1.30-fold (OR 1.30, 95%CI 1.06–1.58; P = 0.005) increased risk for PAD. The C-C haplotype had 43% lowered transcription activity of hTERT promoter (P<0.001). Conclusion: The associations between the functional haplotype of hTERT gene and telomere length and the risk of atherosclerotic PAD suggested that mean leukocyte telomere length may independently serve as a potential predictor of PAD
The sky brightness and transparency in i-band at Dome A, Antarctica
The i-band observing conditions at Dome A on the Antarctic plateau have been
investigated using data acquired during 2008 with the Chinese Small Telescope
ARray. The sky brightness, variations in atmospheric transparency, cloud cover,
and the presence of aurorae are obtained from these images. The median sky
brightness of moonless clear nights is 20.5 mag arcsec^{-2} in the SDSS
band at the South Celestial Pole (which includes a contribution of about 0.06
mag from diffuse Galactic light). The median over all Moon phases in the
Antarctic winter is about 19.8 mag arcsec^{-2}. There were no thick clouds in
2008. We model contributions of the Sun and the Moon to the sky background to
obtain the relationship between the sky brightness and transparency. Aurorae
are identified by comparing the observed sky brightness to the sky brightness
expected from this model. About 2% of the images are affected by relatively

Transcriptome and network analyses reveal key pathways and genes involved in response to carotenoid deposition in scallop muscle
Carotenoids are essential nutrients for humans and animals, and carotenoid content has become an important trait to evaluate the nutritional value of many cultured animals. Marine animals provide humans with diverse carotenoids, and developing carotenoid-enriched varieties has been the focus of marine animal breeding. Understanding the molecular mechanism of carotenoid deposition could benefit marine animal breeding for carotenoid content improvement. In the present study, transcriptomic analysis of adductor muscle was performed between Yesso scallop (Patinopecten yessoensis) with white muscle (WM) and carotenoid-enriched orange muscle (OM). A total of 683 differentially expressed genes (DEGs) were identified, with 302 and 381 genes being up- and down-regulated in OM scallop. Gene co-expression network analysis identified four carotenoid accumulation−related modules, including three up-regulated modules and one down-regulated module. The genes in up-regulated modules mainly participate in the pathways of translation and transcription (MEgreen), immune system (MElightyellow), and lipid metabolism (MEpink), while the down-regulated module is mainly enriched with genes involved in various metabolic pathways (MEturquoise). As the causal gene responsible for muscle coloration in scallop, PyBCO-like 1 is the hub gene of MEturquoise and showed strong connectivity with NR2F1A, a transcriptional factor involved in the regulation of retinoic acid. In addition, the up-regulated DEGs, including WDR3, RPP29, TBL3, RIOK2, and NOB1 from “ribosome biogenesis”, HSP70s and HSP702Bs from “antigen processing and presentation”, and ACOX1 from “PPAR signaling pathway” were identified as hub genes, indicating the potential regulatory role of these genes and pathways in response to carotenoid accumulation. Our data contribute to a deeper understanding of the regulatory and response mechanisms of carotenoid accumulation in marine animals
The First Release of the CSTAR Point Source Catalog from Dome A, Antarctica
In 2008 January the 24th Chinese expedition team successfully deployed the
Chinese Small Telescope ARray (CSTAR) to DomeA, the highest point on the
Antarctic plateau. CSTAR consists of four 14.5cm optical telescopes, each with
a different filter (g, r, i and open) and has a 4.5degree x 4.5degree field of
view (FOV). It operates robotically as part of the Plateau Observatory, PLATO,
with each telescope taking an image every 30 seconds throughout the year
whenever it is dark. During 2008, CSTAR #1 performed almost flawlessly,
acquiring more than 0.3 million i-band images for a total integration time of
1728 hours during 158 days of observations. For each image taken under good sky
conditions, more than 10,000 sources down to 16 mag could be detected. We
performed aperture photometry on all the sources in the field to create the
catalog described herein. Since CSTAR has a fixed pointing centered on the
South Celestial Pole (Dec =-90 degree), all the sources within the FOV of CSTAR
were monitored continuously for several months. The photometric catalog can be
used for studying any variability in these sources, and for the discovery of

A High Intrinsic Peculiarity Rate Among Type Ia Supernovae
We have compiled a sample of 45 Type Ia supernovae (SNe Ia) discovered by the
Lick Observatory Supernova Search (LOSS) and the Beijing Astronomical
Observatory Supernova Survey (BAOSS), and determined the rate of
spectroscopically peculiar SNe Ia (i.e., SN 1986G-like, SN 1991bg-like, and SN
1991T-like objects) and the luminosity function of SNe Ia. Because of the
nature of the two surveys (distance-limited with small baselines and deep
limiting magnitudes), nearly all SNe Ia have been discovered in the sample
galaxies of LOSS and BAOSS; thus, the observed peculiarity rate and luminosity
function of SNe Ia are intrinsic. We find that 369% of nearby SNe Ia are
peculiar; specifically, the luminosity function of SNe Ia consists of 20% SN
1991T-like, 64% normal, and 16% SN 1991bg-like objects. We have compared our
results to those found by earlier studies, and to those found at high redshift.
The apparent dearth of SN 1991T-like objects at high redshift may be due to
extinction, and especially to the difficulty of recognizing them from spectra
obtained past maximum brightness or from spectra with low signal-to-noise
ratios. Implications of the high peculiarity rate for the progenitor systems of

Overexpression of the FBA and TPI genes promotes high production of HDMF in Zygosaccharomyces rouxii
4-Hydroxy-2,5-dimethyl-3 (2H)-furanone (HDMF) is widely used in the food industry as a spice and flavoring agent with high market demand. In this study, fructose-1,6-bisphosphate aldolase (FBA) and triose phosphate isomerase (TPI) were overexpressed in Zygosaccharomyces rouxii in the form of single and double genes, respectively, via electroporation. High-yield HDMF-engineered yeast strains were constructed by combining the analysis of gene expression levels obtained by real-time fluorescence quantitative PCR technology and HDMF production measured by HPLC. The results showed that there was a significant positive correlation between the production of HDMF and the expression levels of the FBA and TPI genes in yeast; the expression levels of the FBA and TPI genes were also positively correlated (p < 0.05). Compared with the wild type (WT), the engineered strains F10-D, T17-D, and TF15-A showed marked increases in HDMF production and FBA and TPI gene expression (p < 0.05) and exhibited great genetic stability with no obvious differences in biomass or colony morphology. In addition, the exogenous addition of d-fructose promoted the growth of Z. rouxii. Among the engineered strains, when fermented in YPD media supplemented with d-fructose for 5 days, TF15-A (overexpressing the FBA and TPI genes) generated the highest HDMF production of 13.39 mg/L, which is 1.91 times greater than that of the wild-type strain. The results above indicated that FBA and TPI, which are key enzymes involved in the process of HDMF biosynthesis by Z. rouxii, positively regulate the synthesis of HDMF at the transcriptional level. d-fructose can be used as a precursor for the biosynthesis of HDMF by engineered yeast in industrial production
Association of sleep duration and sleep quality with the risk of metabolic syndrome in adults: a systematic review and meta-analysis
Introduction: The association between sleep duration and metabolic syndrome (MetS) remains controversial, and few have considered the effects of sleep quality. We performed a meta-analysis to clarify the relationship of sleep duration and sleep quality with the risk of MetS.
Material and methods: We conducted a systematic and comprehensive literature search of electronic databases from inception to 17 February 2022. The effect sizes of covariates from each study were pooled using a random or fixed model, and a restricted cubic spline random-effects meta-analysis was performed to examine the dose-response relationship between sleep duration and MetS.
Results: A total of 62 studies were included in this meta-analysis. Compared to normal sleep duration, short sleep duration [odds ratio (OR) = 1.14, 95% confidence interval (CI): 1.10–1.19] and long sleep duration (OR = 1.15, 95% CI: 1.09–1.23) were associated with an increased risk of MetS. The restricted cubic spline analysis indicated that sleep durations of 8.5 h (OR = 0.95, 95% CI: 0.92–0.97) and 11 h (OR = 1.58, 95% CI: 1.31–1.91) were significantly associated with the risk of MetS. The pooled results showed that poor sleep quality (OR = 1.46, 95% CI: 1.03–2.06) and sleep complaints had significant positive associations with MetS.
Conclusion: Our results demonstrated that short sleep duration increased the risk of developing MetS. Long sleep duration was also associated with MetS, especially for 11 h. 8.5 h can be considered the recommended sleep duration for MetS. Poor sleep quality and sleep complaints were also associated with MetS
Identification of Major QTLs Associated With First Pod Height and Candidate Gene Mining in Soybean
First pod height (FPH) is a quantitative trait in soybean [Glycine max (L.) Merr.] that affects mechanized harvesting. A compatible combination of the FPH and the mechanized harvester is required to ensure that the soybean is efficiently harvested. In this study, 147 recombinant inbred lines, which were derived from a cross between ‘Dongnong594’ and ‘Charleston’ over 8 years, were used to identify the major quantitative trait loci (QTLs) associated with FPH. Using a composite interval mapping method with WinQTLCart (version 2.5), 11 major QTLs were identified. They were distributed on five soybean chromosomes, and 90 pairs of QTLs showed significant epistatic associates with FPH. Of these, 3 were main QTL × main QTL interactions, and 12 were main QTL × non-main QTL interactions. A KEGG gene annotation of the 11 major QTL intervals revealed 8 candidate genes related to plant growth, appearing in the pathways K14486 (auxin response factor 9), K14498 (serine/threonine-protein kinase), and K13946 (transmembrane amino acid transporter family protein), and 7 candidate genes had high expression levels in the soybean stems. These results will aid in building a foundation for the fine mapping of the QTLs related to FPH and marker-assisted selection for breeding in soybean
Moving from contractor to owner operator: Impact on safety culture; a case study
Purpose – The purpose of this paper is to investigate whether a change in staffing contractual arrangements, specific training in hazard identification, mentoring of supervisors and the introduction of a robust safety system could improve an organisation\u27s safety culture. How safety conditions change under contracted out labour compared to direct labour and the influence that contracting out has on organisational safety culture is explored. Design/methodology/approach – The study used a case study methodology to detail how the change occurred over a six month period in 2011. As part of the analysis a model of the change process and push-pull factors is offered. Findings – As a result of the change, all areas saw some improvement. Work-related injury statistics dropped significantly, supervisors were clear of their roles, actively monitoring their crews to ensure they worked in a safer manner than before, and staff were actively addressing work-place hazards. With the safety system in place the organisation should be deemed compliant and diligent by the state auditing authorities. This study has also shown that using contractor workers together with in-house workers that are managed under different safety regimes is problematic. The problems don’t occur due to the contractor\u27s safety systems being less robust than the parent company\u27s or that contract workers are themselves less safe; it is the added complexity of managing multiple safety regimes and the lack of trust of the robustness of each system that create conflict. Research limitations/implications – The paper reports on the change process of one mining organisation in Western Australia as a case study from a managerial sample and is thereby limited. Practical implications – This study demonstrates the difficulties in changing safety culture in an underground mining organisation. The paper argues the need for specialised training in identifying hazards by the staff, the mentoring of supervisory staff and the adoption of a robust safety system to support improved safety culture. Originality/value – There is little research conducted in the resources sector researching changes in human resource supply and OHS management, in particular moving from contracted labour to hiring in-house. This case provides an insight into how a change in staffing hiring arrangements, together with specific safety initiatives, has a positive impact on safety performance
