    Cooperative approach in GPS training

    As Global Positioning System (GPS) technology becomes more commonly used in many aspects of natural resource management, the need for education and training in this area has also increased. However, the high cost of the equipment and the high level of technical knowledge required has been a barrier to including GPS in forestry and other natural resources curricula. This fall the forest technology program at Penn State-Mont Alto and the Bartlett Tree Experts Company collaborated on a two-day training session in GPS using Trimble receivers and data collectors. Ten students and faculty from Mont Alto and ten Bartlett personnel participated in the program. The first day of field procedures and data processing was taught by a Trimble-certified trainer. The second day consisted of training by Bartlett Tree Research Laboratory staff in Bartlett’s tree inventory system and management plan writing. As a practical project over 400 trees in the campus’s arboretum were inventoried with the Bartlett tree inventory and appraisal system. The workshop was mutually beneficial to both groups. Bartlett was able to train its personnel in a well-equipped computer lab and typical landscaped environment on campus. The university students used the latest equipment and were able to get career and practical insights from arborists employing the technology in the field. The combined efforts of all the participants in the tree inventory facilitated a long-standing need in the arboretum’s management. Sharing resources in joint training exercises such as this one provides a realistic teaching opportunity in a time of budget restraint

    Formation and capture of thiiranium ion intermediates using Lewis acids

    The aprotic formation of thiiranium ions and subsequent nucleophilic capture was investigated. It was discovered that β-methoxy and β-acetoxy sulfides could form thiiranium ions in the presences of BF3•OEt2, TMSOTf and various organoaluminum reagents. Treatment of β-methoxy and β-acetoxy sulfides with BF3•OEt2 or TMSOTf allowed for the capture of thiiranium ions by silyl enol ethers, serving as external nucleophiles. However, treatment with Lewis acid caused a drop in the enantiopurity of the thiiranium ion. Varying the amount of Lewis acid, amount and type of nucleophile, temperature and concentration failed to provide conditions where the reaction could proceed in an enantiospecific manner. While investigating new Lewis acids that would allow for enantiospecific formation and capture of thiiranium ions, it was serendipitously discovered that trialkylaluminum reagents could form thiiranium ions and transfer an alkyl group. Further investigation showed that dimethyl(phenylethynyl)aluminum could selectively transfer an alkynyl group. A library of β-acetoxy (2,6-diisopropylphenyl) sulfides was synthesized and underwent both methylation and alkynylation with high yields. It was later determined that the installation of a 2,6-diisopropylphenyl group was unnecessary to achieve high yields and 100% enantiospecificity. A library of various β-acetoxy (phenyl) sulfides underwent methylation and alkynylation with very high yields and 100% enantiospecificity in almost all cases. Arylation and alkenylation via organoaluminum reagents were briefly investigated; however, further studies are needed to successfully prepare the organoaluminum reagent and treat it with β-acetoxy (phenyl) sulfides

    Falsification Of The Atmospheric CO2 Greenhouse Effects Within The Frame Of Physics

    The atmospheric greenhouse effect, an idea that many authors trace back to the traditional works of Fourier (1824), Tyndall (1861), and Arrhenius (1896), and which is still supported in global climatology, essentially describes a fictitious mechanism, in which a planetary atmosphere acts as a heat pump driven by an environment that is radiatively interacting with but radiatively equilibrated to the atmospheric system. According to the second law of thermodynamics such a planetary machine can never exist. Nevertheless, in almost all texts of global climatology and in a widespread secondary literature it is taken for granted that such mechanism is real and stands on a firm scientific foundation. In this paper the popular conjecture is analyzed and the underlying physical principles are clarified. By showing that (a) there are no common physical laws between the warming phenomenon in glass houses and the fictitious atmospheric greenhouse effects, (b) there are no calculations to determine an average surface temperature of a planet, (c) the frequently mentioned difference of 33 degrees Celsius is a meaningless number calculated wrongly, (d) the formulas of cavity radiation are used inappropriately, (e) the assumption of a radiative balance is unphysical, (f) thermal conductivity and friction must not be set to zero, the atmospheric greenhouse conjecture is falsified.Comment: 115 pages, 32 figures, 13 tables (some typos corrected

    A prospective cohort study assessing clinical referral management & workforce allocation within a UK regional medical genetics service

    Abstract Ensuring patient access to genomic information in the face of increasing demand requires clinicians to develop innovative ways of working. This paper presents the first empirical prospective observational cohort study of UK multi-disciplinary genetic service delivery. It describes and explores collaborative working practices including the utilisation and role of clinical geneticists and non-medical genetic counsellors. Six hundred and fifty new patients referred to a regional genetics service were tracked through 850 clinical contacts until discharge. Referral decisions regarding allocation of lead health professional assigned to the case were monitored, including the use of initial clinical contact guidelines. Significant differences were found in the cases led by genetic counsellors and those led by clinical geneticists. Around a sixth, 16.8% (109/650) of referrals were dealt with by a letter back to the referrer or re-directed to another service provider and 14.8% (80/541) of the remaining patients chose not to schedule an appointment. Of the remaining 461 patients, genetic counsellors were allocated as lead health professional for 46.2% (213/461). A further 61 patients did not attend. Of those who did, 86% (345/400) were discharged after one or two appointments. Genetic counsellors contributed to 95% (784/825) of total patient contacts. They provided 93.7% (395/432) of initial contacts and 26.8% (106/395) of patients were discharged at that point. The information from this study informed a planned service re-design. More research is needed to assess the effectiveness and efficiency of different models of collaborative multi-disciplinary working within genetics services.     A systems approach to evaluate One Health initiatives

    Challenges calling for integrated approaches to health, such as the One Health (OH) approach, typically arise from the intertwined spheres of humans, animals, and ecosystems constituting their environment. Initiatives addressing such wicked problems commonly consist of complex structures and dynamics. As a result of the EU COST Action (TD 1404) “Network for Evaluation of One Health” (NEOH), we propose an evaluation framework anchored in systems theory to address the intrinsic complexity of OH initiatives and regard them as subsystems of the context within which they operate. Typically, they intend to influence a system with a view to improve human, animal, and environmental health. The NEOH evaluation framework consists of four overarching elements, namely: (1) the definition of the initiative and its context, (2) the description of the theory of change with an assessment of expected and unexpected outcomes, (3) the process evaluation of operational and supporting infrastructures (the “OH-ness”), and (4) an assessment of the association(s) between the process evaluation and the outcomes produced. It relies on a mixed methods approach by combining a descriptive and qualitative assessment with a semi-quantitative scoring for the evaluation of the degree and structural balance of “OH-ness” (summarised in an OH-index and OH-ratio, respectively) and conventional metrics for different outcomes in a multi-criteria-decision-analysis. Here, we focus on the methodology for Elements (1) and (3) including ready-to-use Microsoft Excel spreadsheets for the assessment of the “OH-ness”. We also provide an overview of Element (2), and refer to the NEOH handbook for further details, also regarding Element (4) (http://neoh.onehealthglobal.net). The presented approach helps researchers, practitioners, and evaluators to conceptualise and conduct evaluations of integrated approaches to health and facilitates comparison and learning across different OH activities thereby facilitating decisions on resource allocation. The application of the framework has been described in eight case studies in the same Frontiers research topic and provides first data on OH-index and OH-ratio, which is an important step towards their validation and the creation of a dataset for future benchmarking, and to demonstrate under which circumstances OH initiatives provide added value compared to disciplinary or conventional health initiatives

    Increased lipophilicity and subsequent cell partitioning decrease passive transcellular diffusion of novel, highly lipophilic antioxidants

    ABSTRACT Oxidative stress is considered a cause or propagator of acute and chronic disorders of the central nervous system. Novel 2,4-diamino-pyrrolo [2,3-d]pyrimidines are potent inhibitors of iron-dependent lipid peroxidation, are cytoprotective in cell culture models of oxidative injury, and are neuroprotective in brain injury and ischemia models. The selection of lead candidates from this series required that they reach target cells deep within brain tissue in efficacious amounts after oral dosing. A homologous series of 26 highly lipophilic pyrrolopyrimidines was examined using cultured cell monolayers to understand the structure-permeability relationship and to use this information to predict brain penetration and residence time. Pyrrolopyrimidines were shown to be a more permeable structural class of membrane-interactive antioxidants where transepithelial permeability was inversely related to lipophilicity or to cell partitioning. Pyrrole substitutions influence cell partitioning where bulky hydrophobic groups increased partitioning and decreased permeability and smaller hydrophobic groups and more hydrophilic groups, especially those capable of weak hydrogen bonding, decreased partitioning, and increased permeability. Transmonolayer diffusion for these membrane-interactive antioxidants was limited mostly by desorption from the receiver-side membrane into the buffer. Thus, in this case, these in vitro cell monolayer models do not adequately mimic the in vivo situation by underestimating in vivo bioavailability of highly lipophilic compounds unless acceptors, such as serum proteins, are added to the receiving buffer. A series of novel 2,4-diamino-pyrrolo[2,3-d]pyrimidines were described as potent inhibitors of iron-dependent lipid peroxidation, and proved to be cytoprotective in cell culture models of oxidative injury and neuroprotective in brain injury and ischemia models Structural determinants of permeability and partitioning are discussed for a series of structurally similar homologs. In addition, detailed studies were conducted concurrently with two radiolabled compounds from the pyrrolopyrimidine series representing different physicochemical, permeability, and cell partitioning attributes to discern the roles of protein binding and cell partitioning on permeation and to complement ongoing pharmacological and pharmacokinetic studies. The data proved useful in predicting which compounds were most likely to leave the blood and penetrate underlying tissue.     Comparison of observed and model-computed low frequency circulation and hydrography on the New England Shelf

The FVCOM-GoM system incorporates realistic time-dependent surface forcing derived from a high-resolution mesoscale meteorological model (MM5) and assimilation of observed quantities including sea surface temperature and salinity and temperature fields on the open boundary. An evaluation of FVCOM-GoM model skill on the New England shelf is made by comparison of computed fields and data collected during the Coastal Mixing and Optics (CMO) Program (August 1996–June 1997). Model mean currents for the full CMO period compare well in both magnitude and direction in fall and winter but overpredict the westward flow in spring. The direction and ellipticity of the subtidal variability correspond but computed magnitudes are around 20% below observed, partially due to underprediction of the variability by MM5. Response of subtidal currents to wind-forcing shows the model captures the directional dependence, as well as seasonal variability of the lag. Hydrographic results show that FVCOM-GoM resolves the spatial and temporal evolution of the temperature and salinity fields. The model-computed surface salinity field compares well, except in May when there is no indication of the fresh surface layer from the Connecticut River discharge noted in the observations. Analysis of model-computed results indicates that the plume was unable to extend to the mooring location due to the presence of a westward mean model-computed flow during that time that was stronger than observed. Overall FVCOM-GoM captures well the dynamics of the mean and subtidal flow on the New England shelf.G.     An expert-driven framework for applying eDNA tools to improve biosecurity in the Antarctic

    Signatories to the Antarctic Treaty System’s Environmental Protocol are committed to preventing incursions of non-native species into Antarctica, but systematic surveillance is rare. Environmental DNA (eDNA) methods provide new opportunities for enhancing detection of non-native species and biosecurity monitoring. To be effective for Antarctic biosecurity, eDNA tests must have appropriate sensitivity and specificity to distinguish non-native from native Antarctic species, and be fit-for-purpose. This requires knowledge of the priority risk species or taxonomic groups for which eDNA surveillance will be informative, validated eDNA assays for those species or groups, and reference DNA sequences for both target non-native and related native Antarctic species. Here, we used an expert elicitation process and decision-by-consensus approach to identify and assess priority biosecurity risks for the Australian Antarctic Program (AAP) in East Antarctica, including identifying high priority non-native species and their potential transport pathways. We determined that the priority targets for biosecurity monitoring were not individual species, but rather broader taxonomic groups such as mussels (Mytilus species), tunicates (Ascidiacea), springtails (Collembola), and grasses (Poaceae). These groups each include multiple species with high risks of introduction to and/or establishment in Antarctica. The most appropriate eDNA methods for the AAP must be capable of detecting a range of species within these high-risk groups (e.g., eDNA metabarcoding). We conclude that the most beneficial Antarctic eDNA biosecurity applications include surveillance of marine species in nearshore environments, terrestrial invertebrates, and biofouling species on vessels visiting Antarctica. An urgent need exists to identify suitable genetic markers for detecting priority species groups, establish baseline terrestrial and marine biodiversity for Antarctic stations, and develop eDNA sampling methods for detecting biofouling organisms
