    Advanced control strategies for bioprocess chromatography: Challenges and opportunities for intensified processes and next generation products

    Recent advances in process analytical technologies and modelling techniques present opportunities to improve industrial chromatography control strategies to enhance process robustness, increase productivity and move towards real-time release testing. This paper provides a critical overview of batch and continuous industrial chromatography control systems for therapeutic protein purification. Firstly, the limitations of conventional industrial fractionation control strategies using in-line UV spectroscopy and on-line HPLC are outlined. Following this, an evaluation of monitoring and control techniques showing promise within research, process development and manufacturing is provided. These novel control strategies combine rapid in-line data capture (e.g. NIR, MALS and variable pathlength UV) with enhanced process understanding obtained from mechanistic and empirical modelling techniques. Finally, a summary of the future states of industrial chromatography control systems is proposed, including strategies to control buffer formulation, product fractionation, column switching and column fouling. The implementation of these control systems improves process capabilities to fulfil product quality criteria as processes are scaled, transferred and operated, thus fast tracking the delivery of new medicines to market

    “Only your first yes will count”: The impact of pre-lineup instructions on sequential lineup decisions

    When administering sequential lineups, researchers often inform their participants that only their first yes response will count. This instruction differs from the original sequential lineup protocol and from how sequential lineups are conducted in practice. Participants (N = 896) viewed a videotaped mock crime and viewed a simultaneous lineup, a sequential lineup with a first-yes-counts instruction, or a sequential control lineup (with no first-yes-counts instruction); the lineup was either target-present or target-absent. Participants in the first-yes-counts condition were less likely to identify the suspect and more likely to reject the lineup than participants in the simultaneous and sequential control conditions, suggesting a conservative criterion shift. The diagnostic value of suspect identifications, as measured by partial Area Under the Curve, was lower in the first-yes-counts lineup than in the simultaneous lineup. Results were qualitatively similar for other metrics of diagnosticity, though the differences were not statistically significant. Differences between the simultaneous and sequential control lineups were negligible on all outcomes. The first-yes-counts instruction undermines sequential lineup performance and produces an artefactual simultaneous lineup advantage. Researchers should adhere to sequential lineup protocols that maximize diagnosticity and that would feasibly be implemented in practice, allowing them to draw more generalizable conclusions from their data.This research was supported in part by a grant to the third-author from the Centre for Applies Social Sciences at Queen Margaret University.https://doi.org/10.1037/xap000033727pubpub

    Alcohol-induced retrograde facilitation renders witnesses of crime less suggestible to misinformation

    RATIONALE: Research has shown that alcohol can have both detrimental and facilitating effects on memory: intoxication can lead to poor memory for information encoded after alcohol consumption (anterograde amnesia) and may improve memory for information encoded before consumption (retrograde facilitation). This study examined whether alcohol consumed after witnessing a crime can render individuals less vulnerable to misleading post-event information (misinformation). METHOD: Participants watched a simulated crime video. Thereafter, one third of participants expected and received alcohol (alcohol group), one third did not expect but received alcohol (reverse placebo), and one third did not expect nor receive alcohol (control). After alcohol consumption, participants were exposed to misinformation embedded in a written narrative about the crime. The following day, participants completed a cued-recall questionnaire about the event. RESULTS: Control participants were more likely to report misinformation compared to the alcohol and reverse placebo group. CONCLUSION: The findings suggest that we may oversimplify the effect alcohol has on suggestibility and that sometimes alcohol can have beneficial effects on eyewitness memory by protecting against misleading post-event information

    Importance of individual events in temporal networks

    Records of time-stamped social interactions between pairs of individuals (e.g., face-to-face conversations, e-mail exchanges, and phone calls) constitute a so-called temporal network. A remarkable difference between temporal networks and conventional static networks is that time-stamped events rather than links are the unit elements generating the collective behavior of nodes. We propose an importance measure for single interaction events. By generalizing the concept of the advance of event proposed by [Kossinets G, Kleinberg J, and Watts D J (2008) Proceeding of the 14th ACM SIGKDD International conference on knowledge discovery and data mining, p 435], we propose that an event is central when it carries new information about others to the two nodes involved in the event. We find that the proposed measure properly quantifies the importance of events in connecting nodes along time-ordered paths. Because of strong heterogeneity in the importance of events present in real data, a small fraction of highly important events is necessary and sufficient to sustain the connectivity of temporal networks. Nevertheless, in contrast to the behavior of scale-free networks against link removal, this property mainly results from bursty activity patterns and not heterogeneous degree distributions.Comment: 36 pages, 13 figures, 2 table

    Foundation characteristics of edible Musa triploids revealed from allelic distribution of SSR markers

    Background and Aims The production of triploid banana and plantain (Musa spp.) cultivars with improved characteristics (e.g. greater disease resistance or higher yield), while still preserving the main features of current popular cultivars (e.g. taste and cooking quality), remains a major challenge for Musa breeders. In this regard, breeders require a sound knowledge of the lineage of the current sterile triploid cultivars, to select diploid parents that are able to transmit desirable traits, together with a breeding strategy ensuring final triploidization and sterility. Highly polymorphic single sequence repeats (SSRs) are valuable markers for investigating phylogenetic relationships. Methods Here, the allelic distribution of each of 22 SSR loci across 561 Musa accessions is analysed. Key Results and ConclusionsWe determine the closest diploid progenitors of the triploid 'Cavendish' and 'Gros Michel' subgroups, valuable information for breeding programmes. Nevertheless, in establishing the likely monoclonal origin of the main edible triploid banana subgroups (i.e. 'Cavendish', 'Plantain' and 'Mutika- Lujugira'), we postulated that the huge phenotypic diversity observed within these subgroups did not result from gamete recombination, but rather from epigenetic regulations. This emphasizes the need to investigate the regulatory mechanisms of genome expression on a unique model in the plant kingdom.     A Phenomenological Inquiry of Building and Living in European Earthship Homes

    Earthships remain a relatively underexplored type of sustainable/alternative home. This is the first study to investigate the personal experiences of constructing and living in European Earthship homes. The aim of this study is to reveal insights into the ‘hands-on’ practical experience of persons who have constructed their own Earthship homes and to gather insights into the collective experience of these dwellers on the realities of living full time in an Earthship home. A phenomenological methodology, using an inductive research approach, was utilised through a qualitative research strategy to solicit insights into the personal experiences of these unique persons. The main themes and sub-themes that emerge from the analysis are that anyone from any background can make the shift to building/living in an Earthship home. However, a mixed skill set and knowledge of building trades, plus physical and emotional prowess, are needed for the long-haul build process, which, together with access to financial means and/or available materials, are critical factors that influence the outcome of the Earthship building. Meanwhile, once constructed, off-grid living in an Earthship home gives dwellers a greater connectivity with the natural world, raises awareness of consumerism and leads to enhanced appreciation of human impacts

    Original multidisciplinary research hereby clarifies the complex geodomestication pathways that generated the vast range of banana cultivars (cvs). Genetic analyses identify the wild ancestors of modern-day cvs and elucidate several key stages of domestication for different cv groups. Archaeology and linguistics shed light on the historical roles of people in the movement and cultivation of bananas from New Guinea to West Africa during the Holocene.
