    Dietary selenium required to achieve body homeostasis and attenuate pro-inflammatory responses in Atlantic salmon post-smolt exceeds the present EU legal limit

    Selenium (Se) supplementation either as inorganic or organic form was evaluated in Atlantic salmon post-smolt in vivo and in vitro. The basal diet was formulated to be low in fish meal and contain 0.24 mg Se kg−1; six other diets with Se inclusion of 0.15, 0.4, 0.7 or 1.1 mg kg−1 as sodium selenite (SS) and 0.15 or 0.4 mg kg−1 as L-selenomethionine (SM) were formulated from the basal diet. The diets were fed to Atlantic salmon post-smolt (mean initial weight, 216 ± 27 g) in triplicate groups (35 fish tank−1) and reared in flow-through seawater (33 ppt) at 10–12 °C for 9 weeks. At the end of the feeding trial, whole fish and tissues were sampled for in vivo assessment; whereas, liver cells and head kidney leukocytes (HKL) were isolated and their primary cultures used for in vitro assessment following exposure to hydrogen peroxide (H2O2), lipopolysaccharide (LPS) or poly I:C (PIC). Growth, feed intake, feed conversion ratio, specific growth rate, hepato-somatic index, proximate composition and mineral concentration of the whole fish (except for Se) were unaffected by dietary Se (p > .05). Hematocrit was significantly higher in fish fed the 0.4 mg Se supplemented feeds, irrespective of the Se source (p = .02). The Se concentration in whole body, liver, muscle, plasma, kidney and liver/kidney Se ratio increased with increasing dietary Se concentration (p < .0001). Level of oxidised glutathione (GSSG) in liver and head kidney followed a quadratic function (p < .05) indicating that the concentrations were lower at intermediate SS supplementation of 0.4 and 0.65 mg Se kg−1 (total Se, 0.65 and 0.87 mg Se kg−1). Impact of Se sources on glutathione redox status was similar. Slope-ratio analysis revealed SM to be more efficient than SS in improving apparent availability, whole body or tissues Se status, Se retention and reducing Se loss to the environment. In vitro, the mRNA expression of p38mapk and aif, in liver cells were affected by the impact of dietary Se, but not by the treatment of H2O2 (p < .05). In the HKL, the LPS and PIC induced pro-inflamatory action of il-1β, cox2, nfkβ and viperin were attenuated by SM supplementation, but not by SS (p < .001). Dietary Se supplementation required to the basal diet containing 0.24 mg Se kg−1 was 0.41 mg Se as SS (total 0.65 mg kg−1) or 0.17 mg Se as SM (total 0.41 mg kg−1) based on body Se homeostasis or tissue Se status. SM inclusion at 0.4 mg kg−1 diet (total, 0.65 mg kg−1) attenuated LPS or PIC induced pro-inflammatory responses in vitro. Overall, Se requirement of Atlantic salmon post smolt was 0.27 mg kg−1 diet, on availale basis. Dietary Se level required to maintain body Se homeostasis and improved health status of Atlantic salmon fed plant-based diets (0.65 mg kg−1 diet) exceed the existing EU maximum limit of 0.5 mg Se kg−1 diet. SM as the Se source in salmon feeds has the potential to improve salmon health and reduce Se emissions from Norwegan salmon farming by 60 to 70%.publishedVersio

    A novel mutation causing mild, atypical fumarylacetoacetase deficiency (Tyrosinemia type I): a case report

    A male patient, born to unrelated Belgian parents, presented at 4 months with epistaxis, haematemesis and haematochezia. On physical examination he presented petechiae and haematomas, and a slightly enlarged liver. Serum transaminases were elevated to 5-10 times upper limit of normal, alkaline phosphatases were 1685 U/L (<720), total bilirubin was 2.53 mg/dl (<1.0), ammonaemia 69 μM (<32), prothrombin time less than 10%, thromboplastin time >180 s (<60) and alpha-fetoprotein 29723 μg/L (<186). Plasma tyrosine (651 μM) and methionine (1032 μM) were strongly increased. In urine, tyrosine metabolites and 4-oxo-6-hydroxyheptanoic acid were increased, but succinylacetone and succinylacetoacetate - pathognomonic for tyrosinemia type I - were repeatedly undetectable. Delta-aminolevulinic acid was normal, which is consistent with the absence of succinylacetone. Abdominal ultrasound and brain CT were normal

    Use of Swedish smokeless tobacco during pregnancy: a systematic review of pregnancy and early life health risk

Tobacco use during pregnancy is concerning due to the risk of adverse pregnancy outcomes and effects on child health. Nicotine may mediate several of these effects. This systematic review measured health outcomes from Swedish smokeless tobacco (snus) use during pregnancy. Method: Literature search was conducted by an information specialist in May 2022. We included human studies of snus use during pregnancy compared with no tobacco use, assessed risk of bias, conducted a meta-analysis and assessed confidence in effectestimates using Grading of Recommendations, Assessment, Development and Evaluations (GRADE). Results: We included 18 cohort studies (42 to 1 006 398 participants). Snus use during pregnancy probably (moderate confidence in risk estimates) increase the risk of neonatal apnea, adjusted odds ratio 95% confidence interval [aOR (95% CI)] 1.96 (1.30 to 2.96). Snus use during pregnancy possibly (low confidence in risk estimates) increase the risk of stillbirths aOR 1.43 (1.02 to 1.99), extremely premature births aOR 1.69 (1.17 to 2.45), moderately premature birth aOR 1.26 (1.15 to 1.38), SGA aOR 1.26 (1.09 to 1.46), reduced birth weight mean difference of 72.47 g (110.58 g to 34.35 g reduction) and oral cleft malformations aOR 1.48 (1.00 to 2.21). It is uncertain (low confidence in risk estimates, CI crossing 1) whether snus use during pregnancy affects risk of preeclampsia aOR 1.11 (0.97 to 1.28), antenatal bleeding aOR 1.15 (0.92 to 1.44) and very premature birth aOR 1.26 (0.95 to 1.66). Risk of early neonatal mortality and altered heart rate variability is uncertain, very low confidence. Snus using mothers had increased prevalence of caesarean sections, low confidence.     The Role of Vaccine Coverage within Social Networks in Cholera Vaccine Efficacy

    Traditional vaccine trial methods have an underlying assumption that the effect of a vaccine is the same throughout the trial area. There are, however, many spatial and behavioral factors that alter the rates of contact among infectious and susceptible individuals and result in different efficacies across a population. We reanalyzed data from a field trial in Bangladesh to ascertain whether there is evidence of indirect protection from cholera vaccines when vaccination rates are high in an individual's social network.We analyzed the first year of surveillance data from a placebo-controlled trial of B subunit-killed whole-cell and killed whole-cell-only oral cholera vaccines in children and adult women in Bangladesh. We calculated whether there was an inverse trend for the relation between the level of vaccine coverage in an individual's social network and the incidence of cholera in individual vaccine recipients or placebo recipients after controlling for potential confounding variables.Using bari-level social network ties, we found incidence rates of cholera among placebo recipients were inversely related to levels of vaccine coverage (5.28 cases per 1000 in the lowest quintile vs 3.27 cases per 1000 in the highest quintile; p = 0.037 for trend). Receipt of vaccine by an individual and the level of vaccine coverage of the individual's social network were independently related to a reduced risk of cholera.Findings indicate that progressively higher levels of vaccine coverage in bari-level social networks can lead to increasing levels of indirect protection of non-vaccinated individuals and could also lead to progressively higher levels of total protection of vaccine recipients

    Versailles project on advanced materials and standards (VAMAS) interlaboratory study on measuring the number concentration of colloidal gold nanoparticles

    We describe the outcome of a large international interlaboratory study of the measurement of particle number concentration of colloidal nanoparticles, project 10 of the technical working area 34, "Nanoparticle Populations" of the Versailles Project on Advanced Materials and Standards (VAMAS). A total of 50 laboratories delivered results for the number concentration of 30 nm gold colloidal nanoparticles measured using particle tracking analysis (PTA), single particle inductively coupled plasma mass spectrometry (spICP-MS), ultraviolet-visible (UV-Vis) light spectroscopy, centrifugal liquid sedimentation (CLS) and small angle X-ray scattering (SAXS). The study provides quantitative data to evaluate the repeatability of these methods and their reproducibility in the measurement of number concentration of model nanoparticle systems following a common measurement protocol. We find that the population-averaging methods of SAXS, CLS and UV-Vis have high measurement repeatability and reproducibility, with between-labs variability of 2.6%, 11% and 1.4% respectively. However, results may be significantly biased for reasons including inaccurate material properties whose values are used to compute the number concentration. Particle-counting method results are less reproducibile than population-averaging methods, with measured between-labs variability of 68% and 46% for PTA and spICP-MS respectively. This study provides the stakeholder community with important comparative data to underpin measurement reproducibility and method validation for number concentration of nanoparticles

    Potentials and challenges of ecotourism for conservation and community development : a case study of Mafia Island Marine Park

    Ecotourism is one of the fastest growing segment within tourism, and are often used as a strategy in protected areas to generate revenues for conservation and management of the protected areas. As coral reefs and marine life are facing increasing pressure from destructive harvesting methods and global warming, marine parks have been established to protect and conserve sensitive ecosystems. However, such areas are often a home for local communities where they are depended on the natural resources. The establishment of protected areas where local communities resides are often complicated and requires broad involvement of all local residents. This study examines the relationship between the Mafia Island Marine Park (MIMP), tourism and the local community, by utilizing the framework of Ross and Wall (1999a) to conceptualize and evaluate ecotourism, and determine the current level of ecotourism within the marine park. MIMP was created in 1995 after much protesting from community leaders about increased destructive fishing methods by residents outside of Mafia. The park was the first marine protected area in Tanzania, and had the intention to be a premier example of community-based conservation and development project. However, the objectives of community involvement were eventually not fulfilled in the further development of the park. This study focused on the core area of tourism in MIMP, more specifically, Utende and Utende Village. The local authorities, the tourism industry and the local community of Utende were identified, and in-depth semi-structured interviews were conducted to investigate their previous experiences and future prospects of tourism and conservation in MIMP. The relationship between local community and MIMP were assessed by looking at the resource use natural resources, local attitudes towards conservation and relations between park staff and residents. The relationship between local community and tourism were exploring the local attitudes towards tourism, economic benefits from tourism, negative impacts from tourism, economic leakage, benefits from tourism, and intercultural exchanges opportunities. Assessing the relationship between MIMP and tourism were including factors such as revenues generated for conservation, environmental education opportunities, Environmental Impact Assessment (EIA), and management and policies. This study reveals that MIMP have created limited involvement of the local community in decision-making concerning both tourism development and conservation. However, the local community have positive attitude towards tourism development with the hope of more job opportunities and community development, and recognizes the importance of conservation of the park as tourism is dependent on the natural areas.M-IE
