    This open access book discusses British literature as part of a network of global entangled modernities and shared aesthetic concerns, departing from the retrospective model of a postcolonial “writing back” to the centre. Accordingly, the narrative strategies in the texts of early Black Atlantic authors, like Equiano, Sancho, Wedderburn, and Seacole, and British canonical novelists, such as Defoe, Sterne, Austen, and Dickens, are framed as entangled tonalities. Via their engagement with discourses on slavery, abolition, and imperialism, these texts shaped an understanding of national belonging as a form of familial feeling. This study thus complicates the “rise of the novel” framework and British middle-class identity formation from a transnational perspective combining approaches in narrative studies with postcolonial and queer theory

    PrĂ€disponierende Faktoren von aktinischen Keratosen bei einer westfĂ€lisch – niedersĂ€chsischen Bevölkerung

    Die wachsende Inzidenzrate von Hautkrebserkrankungen bei Kaukasiern weist auf ein weltweit relevantes gesundheitliches Problem hin. Die aktinische Keratose (AK) stellt das Carcinoma in Situ des Plattenepithelkarzinom der Haut dar. Das Erstellen eines Risikoprofils fĂŒr die Entstehung von AKs ist fĂŒr zukĂŒnftige PrĂ€ventionsprogramme von großer Bedeutung. Die Datenerhebung dieser multizentrischen Fall–Kontroll–Studie erfolgte im Zeitraum von Oktober 2005 bis Oktober 2006. FĂŒr das Auftreten von AKs wurde auf Basis logistischer Regressionsanalysen (stepwise-forward-Modell) ein klinisches Risikoprofil ermittelt: MĂ€nnlich, Hauttumoren in der Eigenanamnese , heller Hauttyp und berufliche Sonnenexposition. Die vorliegende Untersuchung prĂ€sentiert erstmalig prĂ€disponierende Faktoren fĂŒr die Entstehung von aktinischen Keratosen in einer zentraleuropĂ€ischen Bevölkerungsgruppe. Sowohl Genetische bzw. konstitutionelle Faktoren als auch durch die Lebensweise bedingte Faktoren haben einen Einfluss auf die Entstehung der Erkrankung

    Insulin-like growth factor binding protein-2 (IGFBP2) is a key molecule in the MACC1-mediated platelet communication and metastasis of colorectal cancer cells

    Tumor cell crosstalk with platelets and, subsequently, their activation are key steps in hematogenous tumor metastasis. MACC1 is an oncogene involved in molecular pathogenesis of colorectal cancer (CRC) and other solid tumor entities, mediating motility and metastasis, making MACC1 an accepted prognostic biomarker. However, the impact of MACC1 on platelet activation has not yet been addressed. Here, we investigated the activation of platelets by human CRC cells upon MACC1 modulation, indicated by platelet aggregation and granule release. These approaches led to the identification of insulin-like growth factor binding protein-2 (IGFBP2) as a functional downstream molecule of MACC1, affecting communication with platelets. This was confirmed by an shRNA-mediated IGFBP2 knockdown, while maintaining MACC1 activity. Although IGFBP2 displayed an attenuated platelet activation potential, obviously by scavenging IGF-I as a platelet costimulatory mediator, the MACC1/IGFBP2 axis did not affect the thrombin formation potential of the cells. Furthermore, the IGFBP2/MACC1-driven cell migration and invasiveness was further accelerated by platelets. The key role of IGFBP2 for the metastatic spread in vivo was confirmed in a xenograft mouse model. Data provide evidence for IGFBP2 as a downstream functional component of MACC1-driven metastasis, linking these two accepted oncogenic biomarkers for the first time in a platelet context

    First two unrelated cases of isolated sedoheptulokinase deficiency: A benign disorder?

    We present the first two reported unrelated patients with an isolated sedoheptulokinase (SHPK) deficiency. The first patient presented with neonatal cholestasis, hypoglycemia, and anemia, while the second patient presented with congenital arthrogryposis multiplex, multiple contractures, and dysmorphisms. Both patients had elevated excretion of erythritol and sedoheptulose, and each had a homozygous nonsense mutation in SHPK. SHPK is an enzyme that phosphorylates sedoheptulose to sedoheptulose-7-phosphate, which is an important intermediate of the pentose phosphate pathway. It is questionable whether SHPK deficiency is a causal factor for the clinical phenotypes of our patients. This study illustrates the necessity of extensive functional and clinical workup for interpreting a novel variant, including nonsense variants

    A short-term plastic adherence incubation of the stromal vascular fraction leads to a predictable GMP-compliant cell-product

    Introduction: Mesenchymal stromal/stem cells (MSCs) derived from fat tissue are an encouraging tool for regenerative medicine. They share properties similar to the bone marrow-derived MSCs, but the amount of MSCs per gram of fat tissue is 500x higher. The fat tissue can easily be digested by collagenase, releasing a heterogeneous cell fraction called stromal vascular fraction (SVF) which contains a variable amount of stromal/stem cells. In Europe, cell products like the SVF derived from fat tissue are considered advanced therapy medicinal product (ATMPs). As a consequence, the manufacturing process has to be approved via GMP-compliant process validation. The problem of the process validation for SVF is the heterogeneity of this fraction. Methods: Here, we modified existing purification strategies by adding an additional plastic adherence incubation of maximal 20 hours after SVF isolation. The resulting cell fraction was characterized and compared to SVF as well as cultivated adipose-derived stromal/stem cells (ASCs) with respect to viability and cell yield, the expression of surface markers, differentiation potential and cytokine expression. Results: Short-term incubation significantly reduced the heterogeneity of the resulting cell fraction compared to SVF. The cells were able to differentiate into adipocytes, chondrocytes, and osteoblasts. More importantly, they expressed trophic proteins which have been previously associated with the beneficial effects of MSCs. Furthermore, GMP compliance of the production process described herein was acknowledged by the national regulatory agencies (DE_BB_01_GMP_2017_1018). Conclusion: Addition of a short purification-step after the SVF isolation is a cheap and fast strategy to isolate a homogeneous uncultivated GMP-compliant cell fraction of ASCs
