India is a country with an ancient history. The crafts of this country are diverse and are a reflection of the country’s cultural heritage. According to Dasra (2013), the craft industry provides millions of artisans with employment and is one of the largest small scale or cottage industries. For centuries, these craft communities have specialized in traditions that are specific to the regions and respective skills, which have been handed down from generations of artisans. Yet in recent years, as Wood (2011) notes, this sector faces a major threat as a result of growing preference for mass produced designs that are cheap and easy to afford. This has contributed to the steady decline of craft based products. This, alongside a lack of knowledge of new innovative ways to design, and decreasing demand for their products have left the craftspeople very vulnerable and has posed a big threat to their sustainable livelihoods. Integrating fashion design with artisan textiles can help regenerate the community and village culture while enhancing the value of products. This practice led research project offers a framework to meet the emerging challenges associated with ethical and environmental principles linked with fashion by incorporating sustainable processes used in traditional craft production. Importantly, it can also offer one strategy to support economic sustainability issues linked to the livelihood of craftspeople at the village level. The project develops an example of a sustainable fashion design process by drawing from the rich heritage of Indian crafts and focuses on developing a fashion collection that integrates artisan craft textiles into the design process. This reaches out to a market that is sensitive towards sustainability issues linked to artisan livelihood and fashion and values the handmade and organic as an alternative approach. This exploration of issues related to sustainability both for fashion design and craft is an attempt to uncover why developing a relationship between the two is significantly more relevant today than it ever has been

AI Quantification of Language Puzzle to Language Learning Generalization

Online language learning applications provide users multiple ways/games to learn a new language. Some of the ways include rearranging words in the foreign language sentences, filling in the blanks, providing flashcards, and many more. Primarily this research focused on quantifying the effectiveness of these games in learning a new language. Secondarily my goal for this project was to measure the effectiveness of exercises for transfer learning in machine translation. Currently, very little research has been done in this field except for the research conducted by the online platforms to provide assurance to their users [12]. Machine learning has been used in this research to achieve the goals mentioned earlier. Specifically, deep learning models with Recurrent Neural Network (RNN) were employed to process the data. Models were designed on popular exercises from these platforms using sequence-to-sequence learning. Our research discovered that most of the models had cross-validation accuracy in the range of 70%- 80%. This result shows that knowledge learned from one model is transferrable to the other

The rapid spread of antimicrobial resistance (AMR) jeopardizes the effectiveness of modern medicine in treating infections. This thesis describes the use of nanomedicine in addressing the clinical challenges posed by multi-drug resistant Staphylococcus aureus bacteria. Nanoparticles possess unique properties that can be engineered for various biomedical applications, including the potential bypass of antibiotic resistance mechanisms. Two central aims of the presented research are to gain a better understanding of S. aureus interactions with human host cells, and to investigate a nanomedicine-based solution for targeting intracellular S. aureus infections. The thesis begins by introducing S. aureus and the epidemiology of S. aureus infections, detailing this bacterium’s ability to develop AMR, to invade different tissues and to form biofilms. Several intracellular infection models are presented, along with potential methods to identify infected cells, thereby providing a better understanding of how S. aureus interacts with and persists inside host cells. The results support the identification of potential nanomedicinal strategies to treat intracellular S. aureus infections. The second part of the thesis describes the feasibility of using gold nanoparticles as carriers for antibiotic delivery to target intracellular S. aureus bacteria, as well as planktonic or biofilm-resident bacteria. Altogether, the results presented in this thesis show that a multi-pronged approach is needed to combat AMR. It is concluded that knowledge gained from further fundamental research into the biology of microbes and their interactions with host cells will be instrumental for developing new strategies to combat AMR and to ensure the continued effectiveness of antimicrobial treatments

Pyruvate kinase M2 (PKM2), a glycolytic enzyme, is required to maintain vascular barrier function

RATIONALE - Metabolic enzymes, like pyruvate kinase M2 (PKM2), play an essential role in altering endothelial cell (EC) phenotypes and behavior. Extensive research has elucidated the function of PKM2, a rate-limiting glycolytic enzyme, in the context of cancer cells and in activated immune cells, but its role in EC biology is only newly emerging. Recent findings show PKM2 acts as a key regulator of angiogenesis. Where exogenous circulating PKM2 induces EC cell proliferation leading to increased tumor angiogenesis and growth. Also, PKM2 deficient ECs exhibit decreased proliferation and migration. The relevance of PKM2 in modulating vascular barrier function is yet to be defined. OBJECTIVE -This study attempts to elucidate the role of PKM2 in regulating vascular barrier function. METHODS AND RESULTS - In vivo, EC specific deletion of PKM2 promotes increased vascular permeability in pulmonary capillary vessels and increased VEGF-induced acute vessel permeability in mouse dermal vessels. Similarly, in vitro, PKM2 deficient ECs exhibit decreased electrical resistance, disrupted VE-cadherin junctions and gap formations (illustrated via florescent VE-cadherin staining and phosphorylation of VE- cadherin protein at tyrosine residue Y658). Mechanistically, the deletion of PKM2 in ECs leads to increased angiopoietin-2 (Ang-2) expression, a well-known modulator of vascular permeability. Also, deletion of Ang-2 was sufficient to attenuate vascular leakage in PKM2 deficient endothelium, indicating that vascular leaky phenotype observed in PKM2 deficient endothelium is mediated by increased Ang-2 expression. CONCLUSIONS - PKM2, by modulating Ang-2 expression, plays a vital role in maintaining vascular barrier function.2019-07-11T00:00:00
