    Using Fuzzy Linear Regression to Estimate Relationship between Forest Fires and Meteorological Conditions

    Each year, millions of hectares of forest land are destroyed by fires causing great financial loss and ecological damage. In this paper, our aim is to study the effect of the variation of meteorological conditions on the total burned area in hectares, by using fuzzy linear regression analysis based on Tanaka’s approaches. The total burned area is considered a dependent variable. Air temperature (in ºC), relative humidity (in %), wind speed (in km/h) and rainfall (in mm/m2 ) are considered to be independent variables. The relationship between input and output data is estimated using data provided in data mining literature. In our study, we apply fuzzy regression, using crisp/fuzzy input data and fuzzy output data expressed in linguistic terms

    Contribution of heme oxygenase 2 to blood pressure regulation in response to swimming exercise and detraining in spontaneously hypertensive rats

    Background: We aimed to determine the effects of exercise followed by detraining on systolic blood pressure (SBP), heme oxygenase 2 (HO-2) expression, and carboxyhemoglobin (COHb) concentration in spontaneously hypertensive rats (SHR) to explain the role of carbon monoxide (CO) in this process. Material/Methods: Animals were randomized into exercised and detrained groups. Corresponding sedentary rats were grouped as Time 1–2. Swimming of 60 min/5 days/week for 10 weeks was applied. Detraining rats discontinued training for an additional 5 weeks. Gene and protein expressions were determined by real-time PCR and immunohistochemistry. Results: Aorta HO-2 histological scores (HSCORE) of hypertensive rats were lower, while SBP was higher. Swimming caused enhancement of HO-2 immunostaining in aorta endothelium and adventitia of SHR. Exercise induced elevation of blood COHb index in SHR. Synchronous BP lowering effect of exercise was observed. HO-2 mRNA expression, HSCORE, and blood COHb index were unaltered during detraining, while SBP was still low in SHR. Conclusions: CO synthesized by HO-2 at least partly plays a role in SBP regulation in the SHR-and BP-lowering effect of exercise. Regular exercise with short-term pauses may be advised to both hypertensives and individuals who are at risk. © Med Sci Monit

    A Forward Genetic Approach in Chlamydomonas reinhardtii as a Strategy for Exploring Starch Catabolism

    International audienceA screen was recently developed to study the mobilization of starch in the unicellular green alga Chlamydomonas reinhardtii. This screen relies on starch synthesis accumulation during nitrogen starvation followed by the supply of nitrogen and the switch to darkness. Hence multiple regulatory networks including those of nutrient starvation, cell cycle control and light to dark transitions are likely to impact the recovery of mutant candidates. In this paper we monitor the specificity of this mutant screen by characterizing the nature of the genes disrupted in the selected mutants. We show that one third of the mutants consisted of strains mutated in genes previously reported to be of paramount importance in starch catabolism such as those encoding β-amylases, the maltose export protein, and branching enzyme I. The other mutants were defective for previously uncharacterized functions some of which are likely to define novel proteins affecting starch mobilization in green algae

    Genome-wide modeling of transcription kinetics reveals patterns of RNA production delays

    Genes with similar transcriptional activation kinetics can display very different temporal mRNA profiles because of differences in transcription time, degradation rate, and RNA-processing kinetics. Recent studies have shown that a splicing-associated RNA production delay can be significant. To investigate this issue more generally, it is useful to develop methods applicable to genome-wide datasets. We introduce a joint model of transcriptional activation and mRNA accumulation that can be used for inference of transcription rate, RNA production delay, and degradation rate given data from high-throughput sequencing time course experiments. We combine a mechanistic differential equation model with a nonparametric statistical modeling approach allowing us to capture a broad range of activation kinetics, and we use Bayesian parameter estimation to quantify the uncertainty in estimates of the kinetic parameters. We apply the model to data from estrogen receptor alpha activation in the MCF-7 breast cancer cell line. We use RNA polymerase II ChIP-Seq time course data to characterize transcriptional activation and mRNA-Seq time course data to quantify mature transcripts. We find that 11% of genes with a good signal in the data display a delay of more than 20 min between completing transcription and mature mRNA production. The genes displaying these long delays are significantly more likely to be short. We also find a statistical association between high delay and late intron retention in pre-mRNA data, indicating significant splicing-associated production delays in many genes.Peer reviewe

    Plantar pressure distribution profile of type 2 diabetes mellitus with diabetic foot syndrome: A hospital-based observational study

    INTRODUCTION: The prevalence of diabetic foot syndrome is increasing in the Indian population. It is a triad of neurological, vascular, and biomechanical changes due to long term diabetes mellitus (DM). Altered plantar pressure distribution is a risk factor for developing diabetic foot ulcers. The purpose of this study is to evaluate the altered plantar pressure distribution in diabetic peripheral neuropathy individuals with peripheral vascular disease in comparison with diabetic peripheral neuropathy and non-diabetic neuropathy. Therefore, the objective of the study is to evaluate the plantar pressure distribution and parameters in type 2 DM (T2DM) with diabetic foot syndrome.METHODS: In this study, 60 participants with T2DM were recruited in either one of the three groups: 20 Diabetic non-neuropathy (DNN), 20 Diabetic peripheral neuropathy (DPN), and 20 Diabetic peripheral neuropathy with the peripheral arterial disease (DNPAD). Then we compared the plantar pressure parameters like maximum plantar pressure, pressure-time integral, fore foot-hind foot ratio, and total contact area were measured using a WinTrack plantar pressure system.RESULTS: The diabetic peripheral neuropathy with peripheral vascular disease group shown a significant difference in all the  plantar pressure parameters measured in comparison with the diabetic peripheral neuropathy group. There was increased Maximum plantar pressure, pressure-time integral and forefoot hindfoot ratio, and reduced total contact area of the foot (p < 0.05).CONCLUSION: For patients, with combined peripheral neuropathy and peripheral vascular disease have increased plantar pressure distribution, are at higher risks of developing neuro-ischemic foot, which further leads to diabetic foot ulcers

    Effect of balance strategies on fall risk in type 2 diabetes mellitus with peripheral neuropathy: a systematic review and meta-analysis

    Individuals with type 2 diabetes mellitus with peripheral neuropathy are at a high risk of developing unsteadiness and falls. This systematic review and meta-analysis evaluated the effect of balance strategies on fall risk in these conditions. Records from six electronic databases were reviewed. Only randomized control trials focusing on balance strategies and outcomes linked to fall risk were included. Preferred reporting items for systematic reviews and meta-analyses guidelines were followed. Of 2,373 studies, 8 were in-cluded. Risk-of-bias analysis showed that 6 were of good quality, 1 was fair, and 1 was of poor. Meta-analysis of 4 outcome measures was conducted. A meta-analysis of the Berg balance scale (MD 1.08, 95% CI 0.42, 3.19), the functional reach test in centimeters (MD 3.82, 95% CI 0.82, 6.83), the timed up and go test in seconds (MD −1.41, 95% CI −2.14, −0.69), and the one-leg stance test in seconds (right eye open: MD 7.86, 95% CI 1.97, 13.94; left eye open: MD 6.14, 95% CI 2.64, 9.64; right eye closed: MD 2.45, 95% CI 0.61, 4.28; and left eye closed: MD 1.80, 95% CI 0.86, 2.75) It suggested that balance strategies improve fall risk in diabetic neuropathy. The meta-analysis also showed low to very low evidence of a large effect on all the outcomes. The findings of this review show that balance strategies positively influence balancing abilities and reduce fall risk in type 2 diabetes mellitus with peripheral neuropathy

    Mutations in LAMB1 Cause Cobblestone Brain Malformation without Muscular or Ocular Abnormalities

    Cobblestone brain malformation (COB) is a neuronal migration disorder characterized by protrusions of neurons beyond the first cortical layer at the pial surface of the brain. It is usually seen in association with dystroglycanopathy types of congenital muscular dystrophies (CMDs) and ocular abnormalities termed muscle-eye-brain disease. Here we report homozygous deleterious mutations in LAMB1, encoding laminin subunit beta-1, in two families with autosomal-recessive COB. Affected individuals displayed a constellation of brain malformations including cortical gyral and white-matter signal abnormalities, severe cerebellar dysplasia, brainstem hypoplasia, and occipital encephalocele, but they had less apparent ocular or muscular abnormalities than are typically observed in COB. LAMB1 is localized to the pial basement membrane, suggesting that defective connection between radial glial cells and the pial surface mediated by LAMB1 leads to this malformation

    DNA topoisomerases participate in fragility of the oncogene RET

    Fragile site breakage was previously shown to result in rearrangement of the RET oncogene, resembling the rearrangements found in thyroid cancer. Common fragile sites are specific regions of the genome with a high susceptibility to DNA breakage under conditions that partially inhibit DNA replication, and often coincide with genes deleted, amplified, or rearranged in cancer. While a substantial amount of work has been performed investigating DNA repair and cell cycle checkpoint proteins vital for maintaining stability at fragile sites, little is known about the initial events leading to DNA breakage at these sites. The purpose of this study was to investigate these initial events through the detection of aphidicolin (APH)-induced DNA breakage within the RET oncogene, in which 144 APHinduced DNA breakpoints were mapped on the nucleotide level in human thyroid cells within intron 11 of RET, the breakpoint cluster region found in patients. These breakpoints were located at or near DNA topoisomerase I and/or II predicted cleavage sites, as well as at DNA secondary structural features recognized and preferentially cleaved by DNA topoisomerases I and II. Co-treatment of thyroid cells with APH and the topoisomerase catalytic inhibitors, betulinic acid and merbarone, significantly decreased APH-induced fragile site breakage within RET intron 11 and within the common fragile site FRA3B. These data demonstrate that DNA topoisomerases I and II are involved in initiating APH-induced common fragile site breakage at RET, and may engage the recognition of DNA secondary structures formed during perturbed DNA replication

    New insight into the effects of lead modulation on antioxidant defense mechanism and trace element concentration in rat bone

    Risks of heavy metals-induced severe bone disorders generate interest to their toxicity. The present study was undertaken to monitor the biochemical and antioxidant status of bone of 30 and 80 days old male Wistar rats exposed to 5 week lead treatment. At the end of study, the rats were sacrificed, their long bone i.e. femur were excised, cleaned of soft tissue, minced and homogenized. Nucleic acid content, alkaline phosphatase, lipid peroxidation, catalase, glutathione S-transferase and superoxide dismutase were determined in bone. In both groups of treated animals lead treatment increased the production of malondialdehyde, while reducing activities of catalase, glutathione S-transferase and superoxide dismutase, indicating that it causes oxidative stress. Parallely with these effects lead significantly reduced the nucleic acid content and the activity of alkaline phosphatase, considered as biomarkers of osteoblast's function, conditions and development of bones. Moreover the concentrations of copper, zinc, iron and sodium were reduced in the excised bones. The present study indicates that the lead induced bone toxicity and its deteriorated development is the consequence of a primary oxidative stress. Our results may be helpful in understanding the modulation of biochemical parameters under lead toxicity

    Downlink user selection and resource allocation for semi-elastic flows in an OFDM cell

    We are concerned with user selection and resource allocation in wireless networks for semi-elastic applications such as video conferencing. While many packet scheduling algorithms have been proposed for elastic applications, and many user selection algorithms have been proposed for inelastic applications, little is known about optimal user selection and resource allocation for semi-elastic applications in wireless networks. We consider user selection and allocation of downlink transmission power and subcarriers in an orthogonal frequency division multiplexing cellular system. We pose a utility maximization problem, but find that direct solution is computationally intractable. We first propose a method that makes joint decisions about user selection and resource allocation by transforming the utility function into a concave function so that convex optimization techniques can be used, resulting in a complexity polynomial in the number of users with a bounded duality gap. This method can be implemented if the network communicates a shadow price for power to power allocation modules, which in turn communicate shadow prices for rate to individual users. We then propose a method that makes separate decisions about user selection and resource allocation, resulting in a complexity linear in the number of users
