Parasite and host kinases as targets for antimalarials
Introduction:
The deployment of Artemisinin-based combination therapies and transmission control measures led to a decrease in the global malaria burden over the recent decades. Unfortunately, this trend is now reversing, in part due to resistance against available treatments, calling for the development of new drugs against untapped targets to prevent cross-resistance.
Areas covered:
In view of their demonstrated druggability in noninfectious diseases, protein kinases represent attractive targets. Kinase-focussed antimalarial drug discovery is facilitated by the availability of kinase-targeting scaffolds and large libraries of inhibitors, as well as high-throughput phenotypic and biochemical assays. We present an overview of validated Plasmodium kinase targets and their inhibitors, and briefly discuss the potential of host cell kinases as targets for host-directed therapy.
Expert opinion:
We propose priority research areas, including (i) diversification of Plasmodium kinase targets (at present most efforts focus on a very small number of targets); (ii) polypharmacology as an avenue to limit resistance (kinase inhibitors are highly suitable in this respect); and (iii) preemptive limitation of resistance through host-directed therapy (targeting host cell kinases that are required for parasite survival) and transmission-blocking through targeting sexual stage-specific kinases as a strategy to protect curative drugs from the spread of resistance
VLBI imaging of a flare in the Crab Nebula: More than just a spot
We report on very long baseline interferometry (VLBI) observations of the
radio emission from the inner region of the Crab Nebula, made at 1.6 GHz and 5
GHz after a recent high-energy flare in this object. The 5 GHz data have
provided only upper limits of 0.4 milli-Jansky (mJy) on the flux density of the
pulsar and 0.4 mJy/beam on the brightness of the putative flaring region. The
1.6 GHz data have enabled imaging the inner regions of the nebula on scales of
up to ~40". The emission from the inner "wisps" is detected for the first time
with VLBI observations. A likely radio counterpart (designated "C1") of the
putative flaring region observed with Chandra and HST is detected in the radio
image, with an estimated flux density of \,mJy and a size of
0.2-0.6". Another compact feature ("C2") is also detected in the VLBI image
closer to the pulsar, with an estimated flux density of 0.4 +- 0.2 mJy and a
size smaller than 0{\farcs}2. Combined with the broad-band SED of the flare,
the radio properties of C1 yield a lower limit of ~0.5 mG for the magnetic
field and a total minimum energy of 1.2*10^41 ergs vested in the flare
(corresponding to using about 0.2% of the pulsar spin-down power). The 1.6 GHz
observations provide upper limits for the brightness (0.2 mJy/beam) and total
flux density (0.4 mJy) of the optical Knot 1 located at 0.6" from the pulsar.
The absolute position of the Crab pulsar is determined, and an estimate of the

PharmGED: Pharmacogenetic Effect Database
Prediction and elucidation of pharmacogenetic effects is important for facilitating the development of personalized medicines. Knowledge of polymorphism-induced and other types of drug-response variations is needed for facilitating such studies. Although databases of pharmacogenetic knowledge, polymorphism and toxicogenomic information have appeared, some of the relevant data are provided in separate web-pages and in terms of relatively long descriptions quoted from literatures. To facilitate easy and quick assessment of the relevant information, it is helpful to develop databases that provide all of the information related to a pharmacogenetic effect in the same web-page and in brief descriptions. We developed a database, Pharmacogenetic Effect Database (PharmGED), for providing sequence, function, polymorphism, affected drugs and pharmacogenetic effects. PharmGED can be accessed at free of charge for academic use. It currently contains 1825 entries covering 108 disease conditions, 266 distinct proteins, 693 polymorphisms, 414 drugs/ligands cited from 856 references
Effects of metal-contaminated soils on the accumulation of heavy metals in gotu kola (Centella asiatica) and the potential health risks: a study in Peninsular Malaysia
Centella asiatica is a commonly used medicinal plant in Malaysia. As heavy metal accumulation in medicinal plants which are highly consumed by human is a serious issue, thus the assessment of heavy metals in C. asiatica is important for the safety of consumers. In this study, the heavy metal accumulation in C. asiatica and the potential health risks were investigated. Samples of C. asiatica and surface soils were collected from nine different sites around Peninsular Malaysia. The concentration of six heavy metals namely Cd, Cu, Ni, Fe, Pb and Zn were determined by air-acetylene flame atomic absorption spectrophotometer (AAS). The degree of anthropogenic influence was assessed by calculating the enrichment factor (EF) and index of geoaccumulation (Igeo). The heavy metal uptake into the plant was estimated through the calculation of translocation factor (TF), bioconcentration factor (BCF) and correlation study. Estimated daily intakes (EDI) and target hazard quotients (THQ) were used to determine the potential health risk of consuming C. asiatica. The results showed that the overall surface soil was polluted by Cd, Cu and Pb, while the uptake of Zn and Ni by the plants was high. The value of EDI and THQ showed that the potential of Pb toxicity in C. asiatica was high as well. As heavy metal accumulation was confirmed in C. asiatica, daily consumption of the plant derived from polluted sites in Malaysia was not recommended
Gauged Flavor Group with Left-Right Symmetry
We construct an anomaly-free extension of the left-right symmetric model,
where the maximal flavor group is gauged and anomaly cancellation is guaranteed
by adding new vectorlike fermion states. We address the question of the lowest
allowed flavor symmetry scale consistent with data. Because of the mechanism
recently pointed out by Grinstein et al. tree-level flavor changing neutral
currents turn out to play a very weak constraining role. The same occurs, in
our model, for electroweak precision observables. The main constraint turns out
to come from WR-mediated flavor changing neutral current box diagrams,
primarily K - Kbar mixing. In the case where discrete parity symmetry is
present at the TeV scale, this constraint implies lower bounds on the mass of
vectorlike fermions and flavor bosons of 5 and 10 TeV respectively. However,
these limits are weakened under the condition that only SU(2)_R x U(1)_{B-L} is
restored at the TeV scale, but not parity. For example, assuming the SU(2)
gauge couplings in the ratio gR/gL approx 0.7 allows the above limits to go
down by half for both vectorlike fermions and flavor bosons. Our model provides
a framework for accommodating neutrino masses and, in the parity symmetric
case, provides a solution to the strong CP problem. The bound on the lepton
flavor gauging scale is somewhat stronger, because of Big Bang Nucleosynthesis
constraints. We argue, however, that the applicability of these constraints

Mid-infrared plasmons in scaled graphene nanostructures
Plasmonics takes advantage of the collective response of electrons to
electromagnetic waves, enabling dramatic scaling of optical devices beyond the
diffraction limit. Here, we demonstrate the mid-infrared (4 to 15 microns)
plasmons in deeply scaled graphene nanostructures down to 50 nm, more than 100
times smaller than the on-resonance light wavelength in free space. We reveal,
for the first time, the crucial damping channels of graphene plasmons via its
intrinsic optical phonons and scattering from the edges. A plasmon lifetime of
20 femto-seconds and smaller is observed, when damping through the emission of
an optical phonon is allowed. Furthermore, the surface polar phonons in SiO2
substrate underneath the graphene nanostructures lead to a significantly
modified plasmon dispersion and damping, in contrast to a non-polar
diamond-like-carbon (DLC) substrate. Much reduced damping is realized when the
plasmon resonance frequencies are close to the polar phonon frequencies. Our
study paves the way for applications of graphene in plasmonic waveguides,
modulators and detectors in an unprecedentedly broad wavelength range from
sub-terahertz to mid-infrared.Comment: submitte
'Prechronous' metastasis in clear cell renal cell carcinoma: a case report
<p>Abstract</p> <p>Introduction</p> <p>Although metastatic carcinoma in the presence of an occult primary tumor is well recognized, underlying reasons for the failure of the primary tumor to manifest are uncertain. Explanations for this phenomenon have ranged from spontaneous regression of the primary tumor to early metastasis of the primary tumor before manifestation of a less aggressive primary tumor. We report a case of 'prechronous' metastasis arising from clear cell renal cell carcinoma, where metastatic disease initially manifested in the absence of a primary renal tumor, followed by aggressive growth of the primary renal lesion.</p> <p>Case presentation</p> <p>A 43-year-old Malay man initially presented to our facility with fever and cough. He subsequently underwent surgical resection of a 9 cm right-sided lung mass found on radiological examination. Histology showed a high-grade clear cell tumor with sarcomatoid differentiation, suggestive of a metastasis from clear cell renal cell carcinoma. However, no concurrent renal lesions were noted on computed tomographic evaluation at that time. Then, four months after lung resection, he presented with a subcutaneous mass in the left loin, as well as right loin discomfort. Computed tomography scanning revealed a 10 cm right renal mass, with renal vein and inferior vena cava invasion, as well as recurrent disease in the right thorax. Histological examination of the excised subcutaneous mass revealed a high-grade carcinoma consistent with clear cell renal cell carcinoma.</p> <p>Conclusions</p> <p>This is the first reported case of prechronous metastasis of renal cell carcinoma, with metastatic disease manifesting prior to the development of the primary lesion. The underlying mechanism is uncertain, but our patient's case provides anecdotal support for the early dissemination model of metastasis.</p

Characteristics of stable chronic obstructive pulmonary disease patients in the pulmonology clinics of seven Asian cities
Background and objectives Chronic obstructive pulmonary disease (COPD) is responsible for significant morbidity and mortality worldwide. We evaluated the characteristics of stable COPD patients in the pulmonology clinics of seven Asian cities and also evaluated whether the exposure to biomass fuels and dusty jobs were related to respiratory symptoms, airflow limitation, and quality of life in the COPD patients. Methods: This cross-sectional observational study recruited 922 COPD patients from seven cities of Asia. The patients underwent spirometry and were administered questionnaires about their exposure to cigarette smoking, biomass fuels, and dusty jobs in addition to respiratory symptoms and health related quality of life. Results: Of the patients, there appeared to be variations from city to city in the history of exposure to biomass fuels and dusty jobs and also in respiratory symptoms of cough, phlegm, wheeze, and dyspnea. These symptoms were more frequent in those COPD patients with a history of exposure to biomass fuels than without and those with a history of exposure to dusty jobs than without (P < 0.01 for all comparisons). Airflow limitation was more severe in those COPD patients with a history of exposure to biomass fuels than without (52.2% predicted versus 55.9% of post-bronchodilator forced expiratory volume in 1 second [FEV1], P = 0.009); quality of life was poorer in those with exposure to biomass fuels than without (40.4 versus 36.2 of the St George’s Respiratory Questionnaire [SGRQ] total score, P = 0.001). Airflow limitation was more severe in those COPD patients with a history of exposure to dusty jobs than without (51.2% predicted versus 57.3% of post-bronchodilator FEV1, P < 0.001); quality of life was poorer in those with dusty jobs than without (41.0 versus 34.6 of SGRQ score, P = 0.006). Conclusion: In Asian cities, the characteristics of COPD patients vary and the history of exposure to biomass fuels or dusty jobs was related to frequency of symptoms, severe airflow limitation, and poor quality of life
