    Divergent responses of Atlantic cod to ocean acidification and food limitation

    In order to understand the effect of global change on marine fishes, it is imperative to quantify the effects on fundamental parameters such as survival and growth. Larval survival and recruitment of the Atlantic cod (Gadus morhua) were found to be heavily impaired by end-of-century levels of ocean acidification. Here, we analysed larval growth among 35–36 days old surviving larvae, along with organ development and ossification of the skeleton. We combined CO2treatments (ambient: 503 µatm, elevated: 1,179 µatm) with food availability in order to evaluate the effect of energy limitation in addition to the ocean acidification stressor. As expected, larval size (as a proxy for growth) and skeletogenesis were positively affected by high food availability. We found significant interactions between acidification and food availability. Larvae fed ad libitum showed little difference in growth and skeletogenesis due to the CO2 treatment. Larvae under energy limitation were significantly larger and had further developed skeletal structures in the elevated CO2 treatment compared to the ambient CO2 treatment. However, the elevated CO2 group revealed impairments in critically important organs, such as the liver, and had comparatively smaller functional gills indicating a mismatch between size and function. It is therefore likely that individual larvae that had survived acidification treatments will suffer from impairments later during ontogeny. Our study highlights important allocation trade-off between growth and organ development, which is critically important to interpret acidification effects on early life stages of fish

    Renal Function but Not Asymmetric Dimethylarginine Is Independently Associated with Retinopathy in Type 2 Diabetes

    Background. Asymmetric dimethylarginine (ADMA) is associated with macrovascular disease and possibly with microangiopathy in type 2 diabetes (T2DM). We tested the hypothesis that ADMA is related to diabetic retinopathy (DR) independently of macrovascular disease. Methods. This cross-sectional study included 127 T2DM patients selected to achieve equal distributions of patients with and without macrovascular disease in the groups with and without DR. Results. Patients with DR had increased ADMA, longer diabetes duration, and reduced glomerular filtration rate (GFR). ADMA correlated with GFR (ρ = -0.35; P < .001), diabetes duration (ρ = 0.19; P = .048), and age (ρ = 0.19; P = .033). Logistic regression analysis revealed an association of ADMA with DR. After adjustment for macrovascular disease, this association remained significant (OR 1.48; 95% CI: 1.02–2.15; P = .039). Inclusion of GFR and T2DM duration into the model abolished this significant relationship. GFR remained the only independent predictor for DR. A 10 mL/min/1.73 m2 GFR decrease was associated with DR in a multivariate model (OR 1.30; 95% CI: 1.08–1.56; P = .006). Conclusions. These findings indicate an association between ADMA and DR in T2DM independent of macrovascular disease. This relationship is modified by GFR, the only parameter significantly related to DR in multivariate analysis

    Intravenous iron increases labile serum iron but does not impair forearm blood flow reactivity in dialysis patients.BackgroundThere are concerns about adverse vascular effects of intravenous iron by inducing oxidative stress. We therefore examined the effect of a single high dose of intravenous iron on endothelial function and biochemical markers of iron homeostasis.MethodsIn a randomized, placebo-controlled, double-blind, parallel-group study, forearm blood flow (FBF) was assessed by strain-gauge plethysmography in 38 peritoneal dialysis patients before and after a single intravenous infusion of 300mg iron sucrose.ResultsIron infusion increased total (Δ 601 μg/100mL, CI 507, 696) and non-transferrin-bound iron (Δ 237.2 μmol/L, CI 173.6, 300.8) approximately 10-fold, as well as redox-active iron nearly five-fold (Δ 0.76 μmol/L, CI 0.54, 0.98). After iron infusion basal FBF was 59% higher than after placebo. FBF response to acetylcholine before and after iron infusion was 263 ± 32% and 310 ± 33%, corresponding to 304 ± 43% and 373 ± 29% in the placebo group, respectively. Before and after iron or placebo infusion, glyceryl-trinitrate increased resting FBF to 232 ± 22% and 258 ± 21% in the iron group, and to 234 ± 18% and 270 ± 30% in the placebo group. L-N-monomethyl-arginine decreased FBF to 70 ± 4% and 72 ± 3% before and after iron, and to 74 ± 4% and 73 ± 4% before and after placebo infusions, respectively. Despite higher basal FBF after iron infusion, absolute and relative FBF changes in response to vasoactive substances were not significantly different between iron and placebo groups.ConclusionOur data suggest that 300mg intravenous iron sucrose has a vasodilatory effect, but does not impair vascular reactivity in dialysis patients, despite a significant increase in non-transferrin-bound and redox-active iron

    Transcriptome profiling reveals exposure to predicted end-of-century ocean acidification as a stealth stressor for Atlantic cod larvae

    Ocean acidification (OA), a direct consequence of increasing atmospheric CO2 concentration dissolving in ocean waters, is impacting many fish species. Little is known about the molecular mechanisms underlying the observed physiological impacts in fish. We used RNAseq to characterize the transcriptome of 3 different larval stages of Atlantic cod (Gadus morhua) exposed to simulated OA at levels (1179 µatm CO2) representing end-of-century predictions compared to controls (503 µatm CO2), which were shown to induce tissue damage and elevated mortality in G. morhua. Only few genes were differentially expressed in 6 and 13 days-post-hatching (dph) (3 and 16 genes, respectively), during a period when maximal mortality as a response to elevated pCO2 occurred. At 36 dph, 1413 genes were differentially expressed, most likely caused by developmental asynchrony between the treatment groups, with individuals under OA growing faster. A target gene analysis revealed only few genes of the universal and well-defined cellular stress response to be differentially expressed. We thus suggest that predicted ocean acidification levels constitute a “stealth stress” for early Atlantic cod larvae, with a rapid breakdown of cellular homeostasis leading to organismal death that was missed even with an 8-fold replication implemented in this study

    Asteroseismology with the WIRE satellite. I. Combining Ground- and Space-based Photometry of the Delta Scuti Star Epsilon Cephei

    We have analysed ground-based multi-colour Stromgren photometry and single-filter photometry from the star tracker on the WIRE satellite of the delta scuti star Epsilon Cephei. The ground-based data set consists of 16 nights of data collected over 164 days, while the satellite data are nearly continuous coverage of the star during 14 days. The spectral window and noise level of the satellite data are superior to the ground-based data and this data set is used to locate the frequencies. However, we can use the ground-based data to improve the accuracy of the frequencies due to the much longer time baseline. We detect 26 oscillation frequencies in the WIRE data set, but only some of these can be seen clearly in the ground-based data. We have used the multi-colour ground-based photometry to determine amplitude and phase differences in the Stromgren b-y colour and the y filter in an attempt to identify the radial degree of the oscillation frequencies. We conclude that the accuracies of the amplitudes and phases are not sufficient to constrain theoretical models of Epsilon Cephei. We find no evidence for rotational splitting or the large separation among the frequencies detected in the WIRE data set. To be able to identify oscillation frequencies in delta scuti stars with the method we have applied, it is crucial to obtain more complete coverage from multi-site campaigns with a long time baseline and in multiple filters. This is important when planning photometric and spectroscopic ground-based support for future satellite missions like COROT and KEPLER.Comment: 13 pages, 12 figures, 4 tables.
