    Faster monitoring of the invasive alien species (IAS) Dreissena polymorpha in river basins through isothermal amplification

    Zebra mussel (Dreissena polymorpha) is considered as one of the 100 most harmful IAS in the world. Traditional detection methods have limitations, and PCR based environmental DNA detection has provided interesting results for early warning. However, in the last years, the development of isothermal amplification methods has received increasing attention. Among them, loop-mediated isothermal amplification (LAMP) has several advantages, including its higher tolerance to the presence of inhibitors and the possibility of naked-eye detection, which enables and simplifies its potential use in decentralized settings. In the current study, a real-time LAMP (qLAMP) method for the detection of Dreissena polymorpha was developed and tested with samples from the Guadalquivir River basin, together with two real-time PCR (qPCR) methods using different detection chemistries, targeting a specific region of the mitochondrial gene cytochrome C oxidase subunit I. All three developed approaches were evaluated regarding specificity, sensitivity and time required for detection. Regarding sensitivity, both qPCR approaches were more sensitive than qLAMP by one order of magnitude, however the qLAMP method proved to be as specific and much faster being performed in just 9 min versus 23 and 29 min for the qPCR methods based on hydrolysis probe and intercalating dye respectivelyThis work was supported by a partnership agreement project between the Confederación Hidrográfica del Guadalquivir and INL for the development of a system for early detection of zebra mussels through analysis of environmental DNA, and by project Nanotechnology Based Functional Solutions (NORTE-01-0145-FEDER-000019), supported by Norte Portugal Regional Operational Programme (NORTE2020), under the PORTUGAL 2020 Partnership Agreement, through the European Regional Development Fund.     Tumor Associated Stromal Cells Play a Critical Role on the Outcome of the Oncolytic Efficacy of Conditionally Replicative Adenoviruses

    The clinical efficacy of conditionally replicative oncolytic adenoviruses (CRAd) is still limited by the inefficient infection of the tumor mass. Since tumor growth is essentially the result of a continuous cross-talk between malignant and tumor-associated stromal cells, targeting both cell compartments may profoundly influence viral efficacy. Therefore, we developed SPARC promoter-based CRAds since the SPARC gene is expressed both in malignant cells and in tumor-associated stromal cells. These CRAds, expressing or not the Herpes Simplex thymidine kinase gene (Ad-F512 and Ad(I)-F512-TK, respectively) exerted a lytic effect on a panel of human melanoma cells expressing SPARC; but they were completely attenuated in normal cells of different origins, including fresh melanocytes, regardless of whether cells expressed or not SPARC. Interestingly, both CRAds displayed cytotoxic activity on SPARC positive-transformed human microendothelial HMEC-1 cells and WI-38 fetal fibroblasts. Both CRAds were therapeutically effective on SPARC positive-human melanoma tumors growing in nude mice but exhibited restricted efficacy in the presence of co-administered HMEC-1 or WI-38 cells. Conversely, co-administration of HMEC-1 cells enhanced the oncolytic efficacy of Ad(I)-F512-TK on SPARC-negative MIA PaCa-2 pancreatic cancer cells in vivo. Moreover, conditioned media produced by stromal cells pre-infected with the CRAds enhanced the in vitro viral oncolytic activity on pancreatic cancer cells, but not on melanoma cells. The whole data indicate that stromal cells might play an important role on the outcome of the oncolytic efficacy of conditionally replicative adenoviruses

    碩士[[abstract]]本研究旨在探討高中體育班學生在使用數位遊戲式APP學習基礎物理-力學單元的學習成效及其學習態度與動機。本研究以徐新逸及周雲虎(2012)研發之兩種數位遊戲式學習力學APP-牛頓兔與超級平衡為教材,將數位遊戲式APP應用於課堂學習,使用兩種遊戲軟體作為學習工具,探討其是否可提高學習態度與動機,增進其學習成效。 本研究採「準實驗設計」,採隨機分派分組,研究對象為89位新北市兩所高中的高一、高二體育班學生,分為實驗組與控制組進行實驗,實施「物理學習成效試卷」、「學習態度問卷(包含動機)」之前後測,並進行描述性統計、獨立樣本t檢定、課程觀察及訪談紀錄來檢視實驗結果。 研究結果顯示,數位遊戲式APP在高中體育班學習物理比對傳統教學法時學生的學習成效有顯著差異(p<.05),而學生的學習態度無顯著差異,但學習動機有顯著差異(p<.05)。結果建議,體育班高中生在經過數位遊戲式APP實驗後,其物理成績有顯著提升,且對其學習物理時也有助於改善其學習動機。[[abstract]]The purpose of this research was to investigate learning achievement, learning attitude, and learning motivation on athletic-class students in high school who thus received the digital game-based learning application to learn basic physics. This study utilized Newton Rabbit and Super Balance (Game-based APP) developed by Shyu & Chou (2012) as learning tools to explore whether it could improve learning attitude, motivation and enhance learning achievement in physics. This study adopted quasi-experimental research method. There were 89 high-school students from New Taipei City who were divided into an experimental group and a control group. The results showed that high school students learning physics with digital game-based learning application had significant differences on learning achievement and learning motivation (p <.05) but not learning attitude. Argumento que, agindo assim, elas mobilizam e vão além da retórica desenvolvimentista do Estado e do discurso de uma etnia negra 'gendrada' e da tradição das organizações políticas negras da região.<br>In this paper I explore how Afro-Colombian women's organizations and networks shape and are shaped by state initiatives to develop and modernize the Pacific region. I argue that in doing so these mobilize and go beyond the developmentalist rhetoric of the state and the discourse of gendered black ethnicity and tradition of black political organizations in the region
