    Initial Stage of Fluid-Structure Interaction of a Celestial Icosahedron Shaped Vacuum Lighter Than Air Vehicle

    The analysis of a celestial icosahedron geometry is considered as a potential design for a Vacuum Lighter than Air Vehicle (VLTAV). The goal of the analysis is ultimately to understand the initial fluid-structure interaction of the VLTAV and the surrounding airflow. Up to this point, previous research analyzed the celestial icosahedron VLTAV in relation to withstanding a symmetric sea-level pressure applied to the membrane of the structure. This scenario simulates an internal vacuum being applied in the worst-case atmospheric environmental condition. The next step in analysis is to determine the aerodynamic effects of the geometry. The experimental setup for obtaining aerodynamic effects is validated prior to analyzing the deformed celestial icosahedron geometry by analyzing a perfect sphere utilizing the same manufacturing process and setup expected to be adopted for the celestial icosahedron experiments. The data received from Computational Fluid Dynamics (CFD) analysis of the deformed structure in collaboration with Wright State University is used to determine the significance of aerodynamic effects on the structure. The pressure profiles experienced in the wind tunnel experiments and CFD analysis are comparatively similar. Therefore, the CFD data is used to conduct a structural analysis in which aerodynamic effects are incorporated. The research concluded that the aerodynamic pressures do not significantly affect the stress on the structure. As a result, it recommended that a full nonlinear fluid-structure interaction analysis is not necessary for this structure

    The activity of TRAF RING homo- and heterodimers is regulated by zinc finger 1

    Ubiquitin chains linked through lysine63 (K63) play a critical role in inflammatory signalling. Following ligand engagement of immune receptors, the RING E3 ligase TRAF6 builds K63-linked chains together with the heterodimeric E2 enzyme Ubc13-Uev1A. Dimerisation of the TRAF6 RING domain is essential for the assembly of K63-linked ubiquitin chains. Here, we show that TRAF6 RING dimers form a catalytic complex where one RING interacts with a Ubc13~Ubiquitin conjugate, while the zinc finger 1 (ZF1) domain and linker-helix of the opposing monomer contact ubiquitin. The RING dimer interface is conserved across TRAFs and we also show that TRAF5–TRAF6 heterodimers form. Importantly, TRAF5 can provide ZF1, enabling ubiquitin transfer from a TRAF6-bound Ubc13 conjugate. Our study explains the dependence of activity on TRAF RING dimers, and suggests that both homo- and heterodimers mediated by TRAF RING domains have the capacity to synthesise ubiquitin chains

    Neurofibrillary tangles are a neuronal change observed in various conditions, linked with dementia when affecting the cerebral cortex as in Alzheimer's disease. They may be found locally close to fibrous or vascular tumors, or affect extensive regions of the neocortex while the cerebellum and the medulla are not affected. Recent immunological and biochemical studies demonstrate that the MT-associated protein tau is the main component of the tangles, in an abnormally phosphorylated state. A consequence of the formation of tangles is a decreased assembly of MT in axons and dendrites, with disturbances of neuroplasmic flow. The relations between tangles and amyloid, as seen in Alzheimer's and Down's diseases are topographical, tangles accumulating in particular in neurites close to the amyloid in the senile plaques (but also at distance in cell bodies and neurites). Genetically and biochemically A4 or beta-amyloid and tau differ.     Dense sampling of bird diversity increases power of comparative genomics

    Whole-genome sequencing projects are increasingly populating the tree of life and characterizing biodiversity(1-4). Sparse taxon sampling has previously been proposed to confound phylogenetic inference(5), and captures only a fraction of the genomic diversity. Here we report a substantial step towards the dense representation of avian phylogenetic and molecular diversity, by analysing 363 genomes from 92.4% of bird families-including 267 newly sequenced genomes produced for phase II of the Bird 10,000 Genomes (B10K) Project. We use this comparative genome dataset in combination with a pipeline that leverages a reference-free whole-genome alignment to identify orthologous regions in greater numbers than has previously been possible and to recognize genomic novelties in particular bird lineages. The densely sampled alignment provides a single-base-pair map of selection, has more than doubled the fraction of bases that are confidently predicted to be under conservation and reveals extensive patterns of weak selection in predominantly non-coding DNA. Our results demonstrate that increasing the diversity of genomes used in comparative studies can reveal more shared and lineage-specific variation, and improve the investigation of genomic characteristics. We anticipate that this genomic resource will offer new perspectives on evolutionary processes in cross-species comparative analyses and assist in efforts to conserve species. A dataset of the genomes of 363 species from the Bird 10,000 Genomes Project shows increased power to detect shared and lineage-specific variation, demonstrating the importance of phylogenetically diverse taxon sampling in whole-genome sequencing.Peer reviewe
