    Hot primary melts and mantle source for the Paraná-Etendeka flood basalt province: New constraints from Al-in-olivine thermometry

    Continental flood basalts (CFB) are amongst the most voluminous volcanic eruptions in Earth's history. They are rapidly emplaced, and in rare cases the thick lava piles are associated with primitive magmas that have high MgO contents. The compositions of these primitive melts are consistent with a deep-sourced, high-temperature mantle plume origin. Whilst the association of CFBs with impacting mantle plumes is widely accepted, the magnitude of the thermal anomaly is not yet resolved. The development of Al-in-olivine thermometry, however, allows the crystallisation temperature of (near-)liquidus olivine to be determined without using the composition of the co-existing melt. This provides both a robust minimum estimate of mantle temperature and a value from which potential temperature (TP) can be back-calculated. This technique has previously confirmed that crystallisation temperatures in CFB settings can be a few hundred degrees greater than those estimated for MORB, and the results hint at a diversity in crystallisation temperatures between different CFB settings

    The Composition of Melts from a Heterogeneous Mantle and the Origin of Ferropicrite: Application of a Thermodynamic Model

    Evidence for chemical and lithological heterogeneity in the Earth’s convecting mantle is widely acknowledged, yet the major element signature imparted on mantle melts by this heterogeneity is still poorly resolved. In this study, a recent thermodynamic melting model is tested on a range of compositions that correspond to potential mantle lithologies (harzburgitic to pyroxenitic), to demonstrate its applicability over this compositional range, in particular for pyroxenite melting. Our results show that, despite the model’s calibration in peridotitic systems, it effectively reproduces experimental partial melt compositions for both Si-deficient and Si-excess pyroxenites. Importantly, the model accurately predicts the presence of a free silica phase at high pressures in Si-excess pyroxenites, indicating the activation of the pyroxene–garnet thermal divide. This thermal divide has a dominant control on solidus temperature, melt productivity and partial melt composition. The model is used to make new inferences on the link between mantle composition and melting behaviour. In silica-deficient and low-pressure (olivine-bearing) lithologies, melt composition is not very sensitive to source composition. Linearly varying the source composition between peridotite and basaltic pyroxenite, we find that the concentration of oxides in the melt tends to be buffered by the increased stability of more fusible phases, causing partial melts of even highly fertile lithologies to be similar to those of peridotite. An exception to this behaviour is FeO, which is elevated in partial melts of silica-deficient pyroxenite even if the bulk composition does not have a high FeO content relative to peridotite. Melt Al2_2O3_3 and MgO vary predominantly as a function of melting depth rather than bulk composition. We have applied the thermodynamic model to test the hypothesis that Fe-rich mantle melts such as ferropicrites are derived by partial melting of Si-deficient pyroxenite at elevated mantle potential temperatures. We show that the conspicuously high FeO in ferropicrites at a given MgO content does not require a high-Fe mantle source and is indeed best matched by model results involving around 0–20% melting of silica-deficient pyroxenite. A pyroxenite source lithology also accounts for the low CaO content of ferropicrites, whereas their characteristic low Al2_2O3_3 is a function of their high pressure of formation. Phanerozoic ferropicrites are exclusively located in continental flood basalt (CFB) provinces and this model of formation confirms that lithological heterogeneity (perhaps recycled oceanic crust) is present in CFB mantle sources.Natural Environment Research Council (NE/J500070/1, NE/J021539/1

    SIRT3 and SIRT5 regulate the enzyme activity and cardiolipin binding of very long-chain Acyl-CoA dehydrogenase

    SIRT3 and SIRT5 have been shown to regulate mitochondrial fatty acid oxidation but the molecular mechanisms behind the regulation are lacking. Here, we demonstrate that SIRT3 and SIRT5 both target human very long-chain acyl-CoA dehydrogenase (VLCAD), a key fatty acid oxidation enzyme. SIRT3 deacetylates and SIRT5 desuccinylates K299 which serves to stabilize the essential FAD cofactor in the active site. Further, we show that VLCAD binds strongly to cardiolipin and isolated mitochondrial membranes via a domain near the C-terminus containing lysines K482, K492, and K507. Acetylation or succinylation of these residues eliminates binding of VLCAD to cardiolipin. SIRT3 deacetylates K507 while SIRT5 desuccinylates K482, K492, and K507. Sirtuin deacylation of recombinant VLCAD rescues membrane binding. Endogenous VLCAD from SIRT3 and SIRT5 knockout mouse liver shows reduced binding to cardiolipin. Thus, SIRT3 and SIRT5 promote fatty acid oxidation by converging upon VLCAD to promote its activity and membrane localization. Regulation of cardiolipin binding by reversible lysine acylation is a novel mechanism that is predicted to extrapolate to other metabolic proteins that localize to the inner mitochondrial membrane

    Trichotillometry: the reliability and practicality of hair pluckability as a method of nutritional assessment

    <p>Abstract</p> <p>Background</p> <p>A nutritional assessment method that is quick and easy to conduct would be extremely useful in a complex emergency, where currently there is no agreed practical and acceptable method. Hair pluckability has been suggested to be a useful method of assessing protein nutritional status. The aim was to investigate the reliability of the trichotillometer and to explore the effects of patient characteristics on hair epilation force.</p> <p>Methods</p> <p>Three observers plucked hair from twelve participants to investigate the within- and between-observer reliability. To investigate the effect of patient characteristics on hair pluckability, 12 black African and 12 white volunteers were recruited. Participants completed a short questionnaire to provide basic information on their characteristics and hair.</p> <p>Results</p> <p>Mean hair pluckability measurements for the 12 participants obtained by the three observers (39.5 g, 41.2 g and 32.7 g) were significantly different (p < 0.001). Significant variation between patients was also found (p < 0.001). None of the patient characteristics significantly affected hair pluckability, with the exception of age, although this relationship was not consistent.</p> <p>Conclusion</p> <p>Due to significant variation in measurements, hair pluckability does not appear to be a reliable method for assessing adult nutritional status. Hair pluckability could be a useful method of nutritional assessment in complex humanitarian emergencies but only if the reliability was improved.</p

    Cognitive appraisal of environmental stimuli induces emotion-like states in fish

    The occurrence of emotions in non-human animals has been the focus of debate over the years. Recently, an interest in expanding this debate to non-tetrapod vertebrates and to invertebrates has emerged. Within vertebrates, the study of emotion in teleosts is particularly interesting since they represent a divergent evolutionary radiation from that of tetrapods, and thus they provide an insight into the evolution of the biological mechanisms of emotion. We report that Sea Bream exposed to stimuli that vary according to valence (positive, negative) and salience (predictable, unpredictable) exhibit different behavioural, physiological and neuromolecular states. Since according to the dimensional theory of emotion valence and salience define a two-dimensional affective space, our data can be interpreted as evidence for the occurrence of distinctive affective states in fish corresponding to each the four quadrants of the core affective space. Moreover, the fact that the same stimuli presented in a predictable vs. unpredictable way elicited different behavioural, physiological and neuromolecular states, suggests that stimulus appraisal by the individual, rather than an intrinsic characteristic of the stimulus, has triggered the observed responses.     Circulating leukocyte telomere length is highly heritable among families of Arab descent

    Background Telomere length, an indicator of ageing and longevity, has been correlated with several biomarkers of cardiometabolic disease in both Arab children and adults. It is not known, however, whether or not telomere length is a highly conserved inheritable trait in this homogeneous cohort, where age-related diseases are highly prevalent. As such, the aim of this study was to address the inheritability of telomere length in Saudi families and the impact of cardiometabolic disease biomarkers on telomere length. Methods A total of 119 randomly selected Saudi families (123 adults and 131 children) were included in this cross-sectional study. Anthropometrics were obtained and fasting blood samples were taken for routine analyses of fasting glucose and lipid profile. Leukocyte telomere length was determined using quantitative real time PCR. Results Telomere length was highly heritable as assessed by a parent-offspring regression [h2 = 0.64 (p = 0.0006)]. Telomere length was modestly associated with BMI (R2 0.07; p-value 0.0087), total cholesterol (R2 0.08; p-value 0.0033), and LDL-cholesterol (R2 0.15; p-value 3 x 10-5) after adjustments for gender, age and age within generation. Conclusion The high heritability of telomere length in Arab families, and the associations of telomere length with various cardiometabolic parameters suggest heritable genetic fetal and/or epigenetic influences on the early predisposition of Arab children to age-related diseases and accelerated ageing

    Seeing is believing: the nocturnal malarial mosquito Anopheles coluzzii responds to visual host-cues when odour indicates a host is nearby

    Background: The immediate aim of our study was to analyse the behaviour of the malarial mosquito Anopheles coluzzii (An. gambiae species complex) near a human host with the ultimate aim of contributing to our fundamental understanding of mosquito host-seeking behaviour and the overall aim of identifying behaviours that could be exploited to enhance sampling and control strategies. Results: Based on 3D video recordings of individual host-seeking females in a laboratory wind-tunnel, we found that despite being a nocturnal species, An. coluzzii is highly responsive to a visually conspicuous object, but only in the presence of host-odour. Female mosquitoes approached and abruptly veered away from a dark object, which suggests attraction to visual cues plays a role in bringing mosquitoes to the source of host odour. It is worth noting that the majority of our recorded flight tracks consisted of highly stereotyped ‘dipping’ sequences near the ground, which have been mentioned in the literature, but never before quantified. Conclusions: Our quantitative analysis of female mosquito flight patterns within ~1.5 m of a host has revealed highly relevant information about responsiveness to visual objects and flight height that could revolutionise the efficacy of sampling traps; the capturing device of a trap should be visually conspicuous and positioned near the ground where the density of host-seeking mosquitoes would be greatest. These characteristics are not universally present in current traps for malarial mosquitoes. The characterisation of a new type of flight pattern that is prevalent in mosquitoes suggests that there is still much that is not fully understood about mosquito flight behaviour
