    Non-Markoffian effects of a simple nonlinear bath

    We analyze a model of a nonlinear bath consisting of a single two-level system coupled to a linear bath (a classical noise force in the limit considered here). This allows us to study the effects of a nonlinear, non-Markoffian bath in a particularly simple situation. We analyze the effects of this bath onto the dynamics of a spin by calculating the decay of the equilibrium correlator of the spin's z-component. The exact results are compared with those obtained using three commonly used approximations: a Markoffian master equation for the spin dynamics, a weak-coupling approximation, and the substitution of a linear bath for the original nonlinear bath.Comment: 7 pages, 6 figure

    Probing Ionization Energies for Trace Gas Identification: The Micro Photo Electron Ionization Detector (PEID)

    Micro gas sensors detect the presence of substances, but can hardly identify them. We developed a novel approach of probing referenceable ionization energies. It extends the photoionization principle towards tunable energies via replacement of photons by accelerated photo electrons. The device comprises UV-LED illumination, an atmospherically stable photoelectron emission layer with a nano-vacuum electronics accelerator realized in thin film technology and charged particle measurement. A voltage variation at the accelerator provides electrons of tunable energies. We were able to prove that variable electron energies can be used for substance detection. The resulting system reaches ambient conditions operability. The actual limitations and challenges are discussed

    The nuclear immune receptor RPS4 is required for RRS1SLH1-dependent constitutive defense activation in Arabidopsis thaliana

    Plant nucleotide-binding leucine-rich repeat (NB-LRR) disease resistance (R) proteins recognize specific ‘‘avirulent’’ pathogen effectors and activate immune responses. NB-LRR proteins structurally and functionally resemble mammalian Nod-like receptors (NLRs). How NB-LRR and NLR proteins activate defense is poorly understood. The divergently transcribed Arabidopsis R genes, RPS4 (resistance to Pseudomonas syringae 4) and RRS1 (resistance to Ralstonia solanacearum 1), function together to confer recognition of Pseudomonas AvrRps4 and Ralstonia PopP2. RRS1 is the only known recessive NBLRR R gene and encodes a WRKY DNA binding domain, prompting suggestions that it acts downstream of RPS4 for transcriptional activation of defense genes. We define here the early RRS1-dependent transcriptional changes upon delivery of PopP2 via Pseudomonas type III secretion. The Arabidopsis slh1 (sensitive to low humidity 1) mutant encodes an RRS1 allele (RRS1SLH1) with a single amino acid (leucine) insertion in the WRKY DNA-binding domain. Its poor growth due to constitutive defense activation is rescued at higher temperature. Transcription profiling data indicate that RRS1SLH1-mediated defense activation overlaps substantially with AvrRps4- and PopP2-regulated responses. To better understand the genetic basis of RPS4/RRS1-dependent immunity, we performed a genetic screen to identify suppressor of slh1 immunity (sushi) mutants. We show that many sushi mutants carry mutations in RPS4, suggesting that RPS4 acts downstream or in a complex with RRS1. Interestingly, several mutations were identified in a domain C-terminal to the RPS4 LRR domain. Using an Agrobacterium-mediated transient assay system, we demonstrate that the P-loop motif of RPS4 but not of RRS1SLH1 is required for RRS1SLH1 function. We also recapitulate the dominant suppression of RRS1SLH1 defense activation by wild type RRS1 and show this suppression requires an intact RRS1 P-loop. These analyses of RRS1SLH1 shed new light on mechanisms by which NB-LRR protein pairs activate defense signaling, or are held inactive in the absence of a pathogen effector

    Modelado del Tratamiento de Efluentes Lácteos por Electrofloculación mediante Aplicación del Método de Taguchi Residuales

    Los efluentes industriales en la actualidad poseen una matriz constitutiva de gran complejidad y el tratamiento de los mismos plantea un problema medioambiental y económico para las industrias lácteas. La electrofloculación es una alternativa atractiva debido a los numerosos beneficios que presenta. En este trabajo se estudia el empleo de esta tecnología en el tratamiento de un efluente lácteo sintético, con el fin de disminuir la demanda química de oxígeno. Se aplica un diseño experimental de Taguchi para evaluar la influencia de parámetros del proceso, como voltaje, tiempo, conductividad y composición del efluente. Se analizan los efectos de estos factores y se desarrolla un modelo predictivo para el proceso.De los factores considerados, se determinó que la composición del efluente es la más significativa seguida por el voltaje aplicado, mientras que los demás factores tienen efectos similares. Se plantea la ampliación del espacio del diseño para futuros ensayos

    Cellular and developmental control of O2 homeostasis by hypoxia-inducible factor 1 alpha.

    Hypoxia is an essential developmental and physiological stimulus that plays a key role in the pathophysiology of cancer, heart attack, stroke, and other major causes of mortality. Hypoxia-inducible factor 1 (HIF-1) is the only known mammalian transcription factor expressed uniquely in response to physiologically relevant levels of hypoxia. We now report that in Hif1a-/- embryonic stem cells that did not express the O2-regulated HIF-1alpha subunit, levels of mRNAs encoding glucose transporters and glycolytic enzymes were reduced, and cellular proliferation was impaired. Vascular endothelial growth factor mRNA expression was also markedly decreased in hypoxic Hif1a-/- embryonic stem cells and cystic embryoid bodies. Complete deficiency of HIF-1alpha resulted in developmental arrest and lethality by E11 of Hif1a-/- embryos that manifested neural tube defects, cardiovascular malformations, and marked cell death within the cephalic mesenchyme. In Hif1a+/+ embryos, HIF-1alpha expression increased between E8.5 and E9.5, coincident with the onset of developmental defects and cell death in Hif1a-/- embryos. These results demonstrate that HIF-1alpha is a master regulator of cellular and developmental O2 homeostasis

    Competition between uptake of ammonium and potassium in barley and Arabidopsis roots: molecular mechanisms and physiological consequences

    Plants can use ammonium (NH4+) as the sole nitrogen source, but at high NH4+ concentrations in the root medium, particularly in combination with a low availability of K+, plants suffer from NH4+ toxicity. To understand the role of K+ transporters and non-selective cation channels in K+/NH4+ interactions better, growth, NH4+ and K+ accumulation and the specific fluxes of NH4+, K+, and H+ were examined in roots of barley (Hordeum vulgare L.) and Arabidopsis seedlings. Net fluxes of K+ and NH4+ were negatively correlated, as were their tissue concentrations, suggesting that there is direct competition during uptake. Pharmacological treatments with the K+ transport inhibitors tetraethyl ammonium (TEA+) and gadolinium (Gd3+) reduced NH4+ influx, and the addition of TEA+ alleviated the NH4+-induced depression of root growth in germinating Arabidopsis plants. Screening of a barley root cDNA library in a yeast mutant lacking all NH4+ and K+ uptake proteins through the deletion of MEP1–3 and TRK1 and TRK2 resulted in the cloning of the barley K+ transporter HvHKT2;1. Further analysis in yeast suggested that HvHKT2;1, AtAKT1, and AtHAK5 transported NH4+, and that K+ supplied at increasing concentrations competed with this NH4+ transport. On the other hand, uptake of K+ by AtHAK5, and to a lesser extent via HvHKT2;1 and AtAKT1, was inhibited by increasing concentrations of NH4+. Together, the results of this study show that plant K+ transporters and channels are able to transport NH4+. Unregulated NH4+ uptake via these transporters may contribute to NH4+ toxicity at low K+ levels, and may explain the alleviation of NH4+ toxicity by K+

    TRIP13 and APC15 drive mitotic exit by turnover of interphase- and unattached kinetochore-produced MCC

    The mitotic checkpoint ensures accurate chromosome segregation through assembly of the mitotic checkpoint complex (MCC), a soluble inhibitor of the anaphase-promoting complex/cyclosome (APC/C) produced by unattached kinetochores. MCC is also assembled during interphase by Mad1/Mad2 bound at nuclear pores, thereby preventing premature mitotic exit prior to kinetochore maturation and checkpoint activation. Using degron tagging to rapidly deplete the AAA+ ATPase TRIP13, we show that its catalytic activity is required to maintain a pool of open-state Mad2 for MCC assembly, thereby supporting mitotic checkpoint activation, but is also required for timely mitotic exit through catalytic disassembly of MCC. Strikingly, combining TRIP13 depletion with elimination of APC15-dependent Cdc20 ubiquitination/degradation results in a complete inability to exit mitosis, even when MCC assembly at unattached kinetochores is prevented. Thus, mitotic exit requires MCC produced either in interphase or mitosis to be disassembled by TRIP13-catalyzed removal of Mad2 or APC15-driven ubiquitination/degradation of its Cdc20 subunit

    [EN] The development of hardfacing coatings has become technologically significant in many industries A common approach is the production of metal matrix composites (MMC) layers. In this work NiCr-WC MMC hardfacing layers are deposited on C25 steel by means of laser cladding. Spheroidal fused tungsten carbides is used as reinforcement phase. Three different NiCr alloys with different Cr content were tested. Optimum conditions to obtain dense, uniform carbide distribution and hardness close to nominal values were defined.
