    Piles of scats for piles of DNA: deriving DNA of lizards from their faeces

Although there are problems, such as contamination from prey DNA, in deriving donor DNA in this way, non-invasive genetic sampling using scats has a well established role in conservation biology. Using scats from captive and wild Egernia stokesii (Squamata, Scincidae) we evaluated two storage and four DNA extraction methods and assessed the reliability of assessing subsequent genotypes and sequences. Reliable genotypes and sequences were obtained from frozen and dried captive lizard scat DNA extracted using a QIAamp ® DNA Stool Mini Kit and a modified Gentra ®Puregene ® method; yet success rates deteriorated for wild lizard scats. Wild E. stokesii eat more plants than their captive counterparts; DNA extraction may be impeded by plant inhibitors present in scats of wild lizards . Notably, reliable genotypes and sequences were obtained from wild E. stokesii scat DNA extracted using a Qiagen DNeasy ® Plant Mini Kit, a method designed to remove plant inhibitors. Results highlight the opportunity for using scat derived DNA in lizard studies, particularly for species that deposit scats in piles

    ‘I’d rather you didn’t come’: The impact of stigma on exercising with epilepsy

    Epilepsy is a common but hidden disorder, leading to stigma in everyday life. Despite stigma being widely researched, little is known about the impact of stigma for people with epilepsy within a sports and exercise setting. Using constructionist grounded theory, we explored the barriers and adaptations to exercise for people with epilepsy. Three focus groups (2-3 participants per group) and three semi-structured interviews were conducted (11 participants total). Stigma negatively impacted joining team sports, running groups, and disclosure to others. The effect of stigma was reduced by educating others about epilepsy, thus creating more awareness and understanding

    "There's not enough knowledge out there": examining older adults' perceptions of digital technology use and digital inclusion classes

    Older adults' definitions of digital technology, and experiences of digital inclusion sessions, were examined using qualitative approaches. Seventeen older adults (aged between 54 and 85) participated in two focus groups that each lasted approximately 90 minutes to explore how older adults understood technology within their lived experience. Interpretative Phenomenological Analysis yielded two main themes: Thirst for knowledge and a wish list for digital technology sessions. A separate content analysis was performed to identify what technology older adults identified as digital technology. This analysis revealed that the older adults most frequently defined digital technology as computers and telephones. The findings support the conclusions that this group of older adults, some of whom were 'successful users', have a wide knowledge of digital technology, are interested in gaining more skills, and desire knowledge acquisition through personalised one-to-one learning sessions

    Labour market experiences of young UK Bangladeshi men: Identity, inclusion and exclusion in inner-city London

    Detailed qualitative data are used to explore the processes perpetuatinglabour market disadvantage among young UK-Bangladeshi men living in central London. Strong forces of inclusion within the Bangladeshi community are found to interact with forces of exclusion from ‘mainstream’ society to constrain aspirations and limit opportunities. Though diverse forms of young Bangladeshi masculinity are found, a common pattern is heavy dependency on intra-ethnic networks. Negative experiences of and isolation from ‘mainstream’ society further reinforce reliance on ‘our own people’. However, acute ambivalence towards belonging to a dense Bangladeshi community exists, exemplified in the widespread denigration of the restaurant trade. Many respondents express the desire to ‘break out’ and access new experiences. The findings support current policy emphasis on ‘connecting people to work’ but highlight the more fundamental need to connect people across ethnic boundaries. The paper urges researchers to ‘unpack’ ethnicity to consider carefully what ethnic identity implies in terms of access to resources and opportunities for different individuals in different contexts in order better to understand the diversity of labour market outcomes and the persistence of disadvantage

    Iterative development of Vegethon: a theory-based mobile app intervention to increase vegetable consumption.

    BACKGROUND: Mobile technology may serve as a cost-effective and scalable tool for delivering behavioral nutrition interventions. This research sought to iteratively develop a theory-driven mobile app, Vegethon, to increase vegetable consumption. METHODS: Development of Vegethon followed phases outlined by the IDEAS framework: 1) empathize with users (qualitative interviews, n = 18); 2) specify target behavior; 3) ground in behavioral theory; 4) ideate implementation strategies; 5) prototype potential products; 6) gather user feedback (qualitative interviews, n = 14; questionnaire, n = 41); 7) build minimum viable product; and 8) pilot potential efficacy and usability (pilot RCT, n = 17). Findings from each phase informed subsequent phases. The target population that informed intervention development was 18-50 years of age, had BMIs of 28-40 kg/m(2), and lived in the geographical area surrounding Stanford University. A full description of the final version of Vegethon is included in the paper. RESULTS: Qualitative findings that shaped initial intervention conception were: participants' interests in accountability without judgment; their desire for simple and efficient dietary self-monitoring; and the importance of planning meals in advance. Qualitative findings identified during intervention refinement were the need for a focus on vegetable self-monitoring; inclusion of vegetable challenges; simplification of features; advice and inspiration for eating vegetables; reminder notifications; and peer comparison. Pilot RCT findings suggested the initial efficacy, acceptance, and feasibility of the intervention. The final version of Vegethon enabled easy self-monitoring of vegetable consumption and included a range of features designed to engage the user (e.g., surprise challenges; leaderboard; weekly reports). Vegethon was coded for its inclusion of 18 behavior change techniques (BCTs) (e.g., goal setting; feedback; social comparison; prompts/cues; framing/reframing; identity). CONCLUSIONS: Vegethon is a theory-based, user-informed mobile intervention that was systematically developed using the IDEAS framework. Vegethon targets increased vegetable consumption among overweight adults and is currently being evaluated in a randomized controlled efficacy trial.     Development and delivery of an exercise programme for falls prevention: the Prevention of Falls Injury Trial (PreFIT)

    This paper describes the development and implementation of an exercise intervention to prevent falls within The Prevention of Fall Injury Trial (PreFIT), which is a large multi-centred randomised controlled trial based in the UK National Health Service (NHS).Using the template for intervention description and replication (TIDieR) checklist, to describe the rationale and processes for treatment selection and delivery of the PreFIT exercise intervention.Based on the results of a validated falls and balance survey, participants were eligible for the exercise intervention if they were at moderate or high risk of falling.Intervention development was informed using the current evidence base, published guidelines, and pre-existing surveys of clinical practice, a pilot study and consensus work with therapists and practitioners. The exercise programme targets lower limb strength and balance, which are known, modifiable risk factors for falling. Treatment was individually tailored and progressive, with seven recommended contacts over a six-month period. Clinical Trials Registry (ISCTRN 71002650)

    Translating a health behavior change intervention for delivery to 2-year college students: the importance of formative research

    Young adults are at risk for weight gain in the transition to independent adulthood; 2-year college students are at greater risk and understudied relative to 4-year students. This project conducted formative research for a randomized controlled weight gain prevention trial among 2-year college students, to ensure appropriateness of content and delivery of a curriculum originally developed for 4-year college students. Data were collected from community college students, faculty, and staff from October 2009 to August 2011. Work included focus groups and key informant interviews, curriculum pilot testing, and social network and support website beta testing. Based on focus groups and interviews, program content, course delivery modes, and communication channels were adjusted to meet population interests and preferences. The course was delivered successfully in pilot testing, and the website was received well by beta testers. Formative work successfully guided program adaptations to address population needs

    Adolescents' experiences of victimization: the role of attribution style and generalized trust

    Positive attribution style, negative attribution style, and generalised peer trust beliefs were examined as mediators in the relationship between adolescents’ peer victimisation experiences and psychosocial and school adjustment. Two hundred and eighty (150 female and 130 males, Mage = 13 years 4 months, SDage = 1 year 1 month) adolescents completed measures of peer victimisation, global self-worth, depressive symptoms, social confidence, school liking, loneliness, attribution styles, and generalised trust beliefs. Multigroup path analysis revealed that: (a) negative attribution style mediated the relationship between cyber victimisation and school liking and depressive symptoms for males and females; (b) positive attribution style mediated the relationship between cyber victimisation, school liking, global self-worth, and depressive symptoms for females; and (c) generalised peer trust beliefs mediated the relationship between social victimisation, depressive symptoms, social confidence, and loneliness for females. Consequently, attribution style and generalised trust beliefs differentially influence the relationship between peer victimisation and adjustment

    Gametocytes: insights gained during a decade of molecular monitoring

    In vertebrate hosts, malaria parasites produce specialized male and female sexual stages (gametocytes). Soon after being taken up by a mosquito, gametocytes rapidly produce gametes and, once mated, they infect their vector and can be transmitted to new hosts. Despite being the parasite stages that were first identified (over a century ago), gametocytes have remained elusive, and basic questions remain concerning their biology. However, the postgenomic era has substantiated information on the specialized molecular machinery of gametocytogenesis and expedited the development of molecular tools to detect and quantify gametocytes. The application of such highly sensitive and specific tools has opened up novel approaches and provided new insights into gametocyte biology. Here, we review the discoveries made during the past decade, highlight unanswered questions and suggest new directions
