    Metodología para la elaboración de guías de intervención basadas en la evidencia en psicología y salud mental: procedimientos del NICE

    El presente material explica el proceso de desarrollo de guías de práctica clínica que utiliza el NICE (National Institute for Health and Care Excellence), una de las instituciones de referencia en la elaboración de guías de intervenciones basadas en la evidencia. La primera parte está dirigida a la descripción de las característica generales de esta institución, que incluye la explicación de su organización y la estructuración en grupos de trabajo con perfiles y funciones bien definidas. Posteriormente se exponen los elementos claves de la metodología que utiliza el NICE para la elaboración de sus recomendaciones. Si bien en principio, estos procedimientos son generales y están fuertemente vinculados a las intervenciones en el ámbito sanitario, en los últimos años se han extendido al ámbito de las intervenciones sociales, educativas y de salud mental. Por lo cual, la metodología que aquí se expone, puede aplicarse para la generación de guías de intervención psicológica o psicoeducativas aplicables en distintos contextos

    RanBP2-Mediated SUMOylation Promotes Human DNA Polymerase Lambda Nuclear Localization and DNA Repair

    Cellular DNA is under constant attack by a wide variety of agents, both endogenous and exogenous. To counteract DNA damage, human cells have a large collection of DNA repair factors. Among them, DNA polymerase lambda (Polλ) stands out for its versatility, as it participates in different DNA repair and damage tolerance pathways in which gap-filling DNA synthesis is required. In this work we show that human Polλ is conjugated with Small Ubiquitin-like MOdifier (SUMO) proteins both in vitro and in vivo, with Lys27 being the main target of this covalent modification. Polλ SUMOylation takes place in the nuclear pore complex and is mediated by the E3 ligase RanBP2. This post-translational modification promotes Polλ entry into the nucleus, which is required for its recruitment to DNA lesions and stimulated by DNA damage induction. Our work represents an advance in the knowledge of molecular pathways that regulate cellular localization of human Polλ, which are essential to be able to perform its functions during repair of nuclear DNA, and that might constitute an important point for the modulation of its activity in human cells

    Multivariate optimization of the KharascheSosnovsky allylic oxidation of olefins

    The multivariate optimization method known as simplex is applied to the KharascheSosnovsky allylic oxidation of double bonds. By applying this method, the amounts of three variables (copper source, oxidant, and additive) are optimized at the same time. Under the conditions thus obtained the reaction takes place in a considerable shorter time, being the alkene the limiting reagent. These conditions are applied to some monoterpenes and sesquiterpenes leading regioselectively to the corresponding benzoate esters, opening a route to the employment of this reaction in the synthesis of more complex molecules

    Early and intermediate age-related macular degeneration: update and clinical review

    Age-related macular degeneration (AMD) is the leading cause of irreversible central vision loss in developed countries. With the aging of population, AMD will become globally an increasingly important and prevalent disease worldwide. It is a complex disease whose etiology is associated with both genetic and environmental risk factors. An extensive decline in the quality of life and progressive need of daily living assistance resulting from AMD among those most severely affected highlights the essential role of preventive strategies, particularly advising patients to quit smoking. In addition, maintaining a healthy diet, controlling other risk factors (such as hypertension, obesity, and atherosclerosis), and the use of nutritional supplements (antioxidants) are recommendable. Genetic testing may be especially important in patients with a family history of AMD. Recently, unifying criteria for the clinical classification of AMD, defining no apparent aging changes; normal aging changes; and early, intermediate, and late AMD stages, are of value in predicting AMD risk of progression and in establishing recommendations for the diagnosis, therapeutic approach, and follow-up of patients. The present review is focused on early and intermediate AMD and presents a description of the clinical characteristics and ophthalmological findings for these stages, together with algorithms for the diagnosis and management of patients, which are easily applicable in daily clinical practice

    Dentre as tipologias de revestimentos cerâmicos atualmente produzidos o porcelanato tem se destacado pela grande expansão em sua produção, observada nos últimos anos. O sucesso do produto se deve pelas propriedades técnicas e estéticas oferecidas. Dentro deste contexto a técnica de coloração a seco surge como alternativa a ser adotada no processamento da massa do material, pois confere ao produto características estéticas que podem ser obtidas através de uma simples mistura de pigmentos e grânulos atomizados. Entretanto, pouco se conhece sobre as variáveis envolvidas neste processo de mistura e a real interferência de cada uma delas nas propriedades apresentadas pelo produto queimado. PIE 0013/2016 and UESC/PROPP 0010299-61.A primary goal of the upcoming Deep Underground Neutrino Experiment (DUNE) is to measure the Oo10 thorn MeV neutrinos produced by a Galactic core-collapse supernova if one should occur during the lifetime of the experiment. The liquid-argon-based detectors planned for DUNE are expected to be uniquely sensitive to the & nu;e component of the supernova flux, enabling a wide variety of physics and astrophysics measurements. A key requirement for a correct interpretation of these measurements is a good understanding of the energy-dependent total cross section & sigma;oE & nu; thorn for charged-current & nu;e absorption on argon. In the context of a simulated extraction of supernova & nu;e spectral parameters from a toy analysis, we investigate the impact of & sigma;oE & nu; thorn modeling uncertainties on DUNE's supernova neutrino physics sensitivity for the first time. We find that the currently large theoretical uncertainties on & sigma;oE & nu; thorn must be substantially reduced before the & nu;e flux parameters can be extracted reliably; in the absence of external constraints, a measurement of the integrated neutrino luminosity with less than 10% bias with DUNE requires & sigma;oE & nu; thorn to be known to about 5%. The neutrino spectral shape parameters can be known to better than 10% for a 20% uncertainty on the cross-section scale, although they will be sensitive to uncertainties on the shape of & sigma;oE & nu; thorn .     Characterization and immunolocalization of RNA polymerase I transcription factor UBF with anti-NOR serum in protozoa, higher plant and vertebrate cells

    We have used anti-NOR serum from a patient with rheumatoid arthritis, to study its reactivity on different phylogenetically separated species such as protozoa, higher plants, birds and mammals. The biochemical characteristics of the antigens detected after applying mono- and two-dimensional electrophoresis and electrophoretic transfers confirm that they correspond to the rRNA polymerase I transcription factor UBF. We have demonstrated the different molecular sizes, depending on the cell complexity, but the same neutral isoelectric points in whole cell extracts of the different species. We have also demonstrated an immunolocalization of this transcription factor to the fibrillar component in all the species studied. These results suggest a high conservation of UBF throughout evolution and the possibility of using this anti-NOR serum as a tool for the study of the structure, nucleolar organization and functional roles of the different nucleolar components

    Synthesis of Porous Clay Heterostructures Modified with SiO2–ZrO2 Nanoparticles for the Valorization of Furfural in One-Pot Process

    The layered structure of two smectites (montmorillonite and saponite) are modified by the insertion of SiO2–ZrO2 nanoparticles, with a Si/Zr molar ratio of 5, to form porous clay heterostructures (PCHs). These PCHs exhibit a clear improvement of the textural and acid properties in comparison to the starting clays, due to the formation of a pillared structure and the presence of Zr species, which provide an increase in the amount of Lewis acid sites. These materials are studied in one-pot catalytic processes to transform furfural into valuable products, such as furfuryl alcohol, alkyl furfuryl ethers, alkyl levulinates or γ-valerolactone, depending on the experimental conditions. Thus, the use of milder reaction temperature (110 °C) favors the formation of furfuryl alcohol and alkyl furfuryl ethers, while higher temperatures (170 °C) promote the formation of alkyl levulinate and γ-valerolactone, as well as an increase in nondetected products associated to the formation of humins due to the polymerization of furfural and/or furfuryl alcohol.The authors are grateful for financial support from the Spanish Ministry of Science, Innovation and Universities (RTI2018-94918-B-C44 project), FEDER (European Union) funds (UMA18-FEDERJA-171), and the University of Malaga.     The MRC1/CD68 ratio is positively associated with adipose tissue lipogenesis and with muscle mitochondrial gene expression in humans

    This is an open-access article distributed under the terms of the Creative Commons Attribution License.[Background]: Alternative macrophages (M2) express the cluster differentiation (CD) 206 (MCR1) at high levels. Decreased M2 in adipose tissue is known to be associated with obesity and inflammation-related metabolic disturbances. Here we aimed to investigate MCR1 relative to CD68 (total macrophages) gene expression in association with adipogenic and mitochondrial genes, which were measured in human visceral [VWAT, n = 147] and subcutaneous adipose tissue [SWAT, n = 76] and in rectus abdominis muscle (n = 23). The effects of surgery-induced weight loss were also longitudinally evaluated (n = ).[Results]: MCR1 and CD68 gene expression levels were similar in VWAT and SWAT. A higher proportion of CD206 relative to total CD68 was present in subjects with less body fat and lower fasting glucose concentrations. The ratio MCR1/CD68was positively associated with IRS1gene expression and with the expression of lipogenic genes such as ACACA, FASN and THRSP, even after adjusting for BMI. The ratio MCR1/CD68 in SWAT increased significantly after the surgery-induced weight loss (+44.7%; p = 0.005) in parallel to the expression of adipogenic genes. In addition, SWAT MCR1/CD68ratio was significantly associated with muscle mitochondrial gene expression (PPARGC1A, TFAM and MT-CO3). AT CD206 was confirmed by immunohistochemistry to be specific of macrophages, especially abundant in crown-like structures.
