    Bacteria-Carried Iron Oxide Nanoparticles for Treatment of Anemia

    The efficiency of maghemite nanoparticles for the treatment of anemia was sensibly higher when nanoparticles were incorporated onto the probiotic bacterium Lactobacillus fermentum (MNP-bacteria) than when administrated as uncoated nanoparticles (MNP). Plasma iron and hemoglobin, intestine expression of divalent metal transporter 1 (DMT1) and duodenal Cytochrome b (DcytB), as well as hepatic expression of the hormone hepcidin were fully restored to healthy levels after administration of MNP-bacteria but not of MNP. A magnetic study on biodistribution and biodegradation showed accumulation of maghemite nanoparticles in intestine lumen when MNP-bacteria were administrated. In contrast, MNP barely reached intestine. In vivo MRI studies suggested the internalization of MNP-bacteria into enterocytes, which did not occur with MNP. Transmission electronic microscopy confirmed this internalization. The collective analysis of results point out that L. fermentum is an excellent carrier to overcome the stomach medium and drive maghemite nanoparticles to intestine, where iron absorption occurs. Due the probiotic ability to adhere to the gut wall, MNP-bacteria internalize into the enterocyte, where maghemite nanoparticles are delivered, providing an adequate iron level into enterocyte.     Role of drug transporters in the sensitivity of acute myeloid leukemia to sorafenib

    [EN]Chemoresistance often limits the success of the pharmacological treatment in acute myeloid leukemia (AML) patients. Although positive results have been obtained with tyrosine kinase inhibitors (TKIs), such as sorafenib, especially in patients with Fms-like tyrosine kinase 3 (FLT3)-positive AML, the success of chemotherapy is very heterogeneous.The sensitivity to sorafenib-induced cell death (MTT test and anexin V/7-AAD method) was evaluated in five different cell lines: MOLM-13, OCI-AML2, HL-60, HEL and K-562. The transportome was characterized by measuring mRNA using RT-qPCR. Drug uptake/efflux was determined by flow cytometry using specific substrates and inhibitors. The cytostatic response to sorafenib was: MOLM-13»OCI-AML2>HL- 60>HEL≈K-562. Regarding efflux pumps, MDR1 was highly expressed in HEL>K- 562≈MOLM-13, but not in OCI-AML2 and HL-60. BCRP and MPR3 expression was low in all cell lines, whereas MRP4 and MRP5 expression was from moderate to high. Flow cytometry studies demonstrated that MRP4, but not MRP5, was functional

    Structural origin for the dipole x-ray resonant scattering in the low temperature phase of Nd0.5Sr0.5MnO3 manganite

    We have investigated the low temperature phase of a Nd0.5Sr0.5MnO3 single crystal by x-ray resonant scattering at the Mn K-edge of the (3 0 0), (0 3 0) and (0 5/2 0) reflections. Strong resonances were observed for the s-s' channel in the (3 0 0) and (0 3 0) reflections and for the s-p' channel in the (0 5/2 0) reflection. These resonances show a p-periodicity on the azimuthal angle, having the intensity at the minimum position almost zero. The intensity dependence on the photon energy, azimuthal angle and polarisation dependencies has been analysed using a semi-empirical structural model. Contrary to previous claims of charge (Mn3+-Mn4+) and orbital ordering in this compound, our results show that the dipole resonant superlattice reflections can be explained by the presence of two types of Mn sites with different local geometric structure. One of the Mn sites is surrounded by a tetragonal-distorted oxygen octahedron whereas the other site has a symmetric octahedral environment. This model also establishes that no real space charge ordering is needed to explain the experimental data.     La pluralidad de membranas utilizadas en la fabricación de dializadores, contribuye a que se presente en el mercado gran número de ellos, más de 400 en el ámbito de la EDTA, con características muy diferentes, tanto en lo relativo a sus prestaciones funcionales como a su "biocompatibilidad" y su precio. La trascendencia clínica y económica del uso de uno u otro tipo de dializador ha sido objeto de multitud de publicaciones y considerable debate que no ha llegado a materializarse en un cuerpo de doctrina universalmente aceptado, que sitúe con precisión la contribución relativa de cada tipo de dializador al resultado clínico del tratamiento en diálisis de la insuficiencia renal; y, consecuentemente, la efectividad y eficiencia de este elemento, fundamental en dicho tratamiento.     Dating the setting of a late prehistoric statue-menhir at Cruz de Cepos, NE Portugal

    The emergence of ‘standing stone’ monuments within the European Late Prehistoric landscape is considered to be associated with a pivotal human cultural transition from hunting and gathering to agriculture and permanent settlement, being the earliest monuments currently dated by radiocarbon to the 5th millennium BCE. However, many standing stones were first erected, subsequently collapsed, and then re-erected during the following three millennia. The excavation of the site of an apparently in situ statue-menhir at Cruz de Cepos in NE Portugal provided the rare opportunity in Iberian prehistory to apply radiocarbon and luminescence techniques to establish the date of construction. On the basis of the iconography, the standing stone was assigned to a sculptural tradition of north-western and western Iberia, loosely dated to the Early/Middle Bronze Age (ca. 2000/1900–1250 BCE). The optically stimulated luminescence (OSL) and dosimetry characteristics of quartz extracted from sediment samples taken from locations associated with the socket pit and surrounding backfilling deposits were examined, producing OSL single grain ages at eight locations. Comparison of the OSL and calibrated radiocarbon ages shows very good agreement, with the mid-3rd millennium BCE dates confirming original erection during the Copper Age and not a much later transformation of the monument. These encouraging results indicate that OSL has the potential to provide reliable dating of depositional processes related to the construction process and is suitable for wider application to megalithic monuments of this type

    Allogeneic Hematopoietic Stem Cell Transplantation in Transformed Follicular Lymphoma (tFL): Results of a Retrospective Multicenter Study from GELTAMO/GETH-TC Spanish Groups

    Simple Summary Follicular lymphoma (FL) is the most prevalent subtype of indolent lymphoma, accounting for 70% of all cases. The estimated risk of histological transformation (tFL), such as diffuse large B cell lymphoma (DLBCL), varies from 2-3% per year to 7-8% at 10 years in different series. Treatment after transformation is not clearly established. Allogeneic hematopoietic stem cell transplantation (alloSCT) could be an option for these patients, but it has not been widely explored. We analyze the efficacy and toxicity of alloSCT in 43 patients from 14 Spanish centers. We observed long-term survival in around one third of the patients, especially those who developed chronic graft versus host disease, indicating that alloSCT continues to be a potentially curative option for patients with tFL, mainly due to the graft versus lymphoma effect. Background: Transformation of follicular lymphoma into an aggressive lymphoma (tFL) worsens the prognosis and the standard treatment is not completely defined. Allogeneic hematopoietic stem cell transplantation (alloSCT) could be a potentially curative option for these patients, but it has not been widely explored. Methods: We designed a retrospective multicenter study to analyze the efficacy and toxicity of alloSCT in tFL patients and potential prognostic factors of survival. Results: A total of 43 patients diagnosed with tFL who underwent alloSCT in 14 Spanish centers between January 2000 and January 2019 were included. Median age was 44 (31-67) years. After a median follow-up of 58 months, estimated 5-year overall survival (OS) and progression-free survival (PFS) were both 35%. Estimated 100-day and 1-year non-relapse mortality (NRM) were 20% and 34%, respectively. The type of conditioning regimen (3-year OS of 52% vs. 20%, respectively, for reduced-intensity vs. myeloablative conditioning) and development of chronic graft versus host disease (cGVHD) (3-year OS of 75% vs. 40%) were the only factors significantly associated with OS. The only variable with an independent association with OS was cGVHD (HR, 3.4; 95% CI, 1.2-9.6). Conclusions: Our results indicate that alloSCT continues to be a potentially curative option for patients with tFL

    Restauración de las olmedas ibéricas (Ulmus minor y U. laevis) en zonas riparias de la Comunidad de Madrid

    El proyecto LIFE+ “Olmos Vivos” (LIFE13 BIO/ES/000556) tiene como objetivo la restauración de unas formaciones vegetales, las olmedas, que por diversos motivos prácticamente han desaparecido del paisaje forestal español, a pesar de haber poseído un alto valor ecológico y cultural. La grafiosis ha constituido una de las enfermedades forestales más devastadoras, y supuso la práctica desaparición de las olmedas ibéricas de Ulmus minor . En el caso de Ulmus laevis , la alteración de su hábitat ligado a zonas temporalmente encharcadas y riberas ha provocado que las poblaciones españolas se encuentren en riesgo de desaparición. Gracias a la obtención de siete clones de U. minor resistentes a la grafiosis como fruto del Programa del Olmo (UPM-MAGRAMA), el presente proyecto permitirá reintroducir la especie en diversas localizaciones de ribera de los ríos Jarama y Tajo. Asimismo, se plantarán brinzales obtenidos de agrupaciones relícticas de U. laevis , con el fin de aumentar sus poblaciones y conservar sus recursos genéticos, en terrenos de naturaleza silícea de las riberas del río Jarama y Arroyo Viñuelas. El desarrollo del proyecto, de cinco años y tres meses de duración 2014-2019), prevé la plantación de unas 16 700 plantas. Al mismo tiempo se recuperará en nuestras ciudades y pueblos la presencia del olmo, mediante plantaciones con fines divulgativos en entornos urbanos y paseos históricos, para devolver así el protagonismo cultural a un árbol que siempre estuvo entre nosotros, presidiendo las reuniones en las plazas y acompañando con su sombra en muchos caminos de nuestra geografía

    The Relationship between Depressive Symptoms, Quality of Life and miRNAs 8 Years after Bariatric Surgery

We aim to determine whether BS outcomes are associated with QoL/depressive symptomatology in studied patients at the 8-year follow-up after BS, as well as their relationship with different serum proteins and miRNAs. Methods: A total of 53 patients with class III obesity who underwent BS, and then classified into "good responders" and "non-responders" depending on the percentage of excess weight lost (%EWL) 8 years after BS (%EWL ≥ 50% and %EWL < 50%, respectively), were included. Basal serum miRNAs and different proteins were analysed, and patients completed tests to evaluate QoL/depressive symptomatology at 8 years after BS. Results: The good responders group showed higher scores on SF-36 scales of physical functioning, role functioning-physical, role functioning-emotional, body pain and global general health compared with the non-responders. The expression of hsa-miR-101-3p, hsa-miR-15a-5p, hsa-miR-29c-3p, hsa-miR-144-3p and hsa-miR-19b-3p were lower in non-responders. Hsa-miR-19b-3p was the variable associated with the response to BS in a logistic regression model. Conclusions: The mental health of patients after BS is limited by the success of the intervention. In addition, the expression of basal serum miRNAs related to depression/anxiety could predict the success of BS
