STUDIES ON THE FEASIBILITY OF PREDICTING FEEDLOT PERFORMANCE FROM CERTAIN LABORATORY GRAIN ANALYSES
Data from 14 cattle feeding trials were utilized to study the relationship between several laboratory analyses and animal feed intake (INTAKE), gain (ADG) and feed efficiency (F/G). Laboratory analyses considered were 6, 12 and 24 hr in vitro dry matter disappearance (IV6, IV12, IV24, respectively); in vitro gas production in 1 hr and 6 hr (GP1 and GP6, respectively); and degree of gelatinization (GEL). A multiple regression equation with variables for treatment and trial classification, initial weight and the quadratic effect of initial weight was fit to the data. The effect of initial weight was significant for all three performance variables, and the quadratic effect was significant for ADG and F/G. A second model was fit excluding the treatment classification, and the maximum R2 procedure was utilized to examine how well laboratory analyses accounted for variation among residuals from this second model. More variation was accounted for in the dependent variables F/G (34.96%) and INTAKE (17.81%) than ADG (5.16%) when a combination of all laboratory analyses except GEL was included in the model. Moreover, correlations between residuals of the second model and the laboratory analyses were higher for INTAKE and F/G than ADG and were all negative for INTAKE and F/G, suggesting a negative response in intake and an improved F/G ratio as starch alteration increases. Correlations between the laboratory analyses were generally quite high. This study suggests that no single laboratory analysis considered would be useful for the development of accurate, reliable equations for the prediction of feedlot performance, and combinations appear to have value only in the case of FIG and INTAKE
Stable Aqueous Nanoparticle Film Assemblies with Covalent and Charged Polymer Linking Networks
The construction of highly stable and efficiently assembled multilayer films of purely water soluble gold nanoparticles is reported. Citrate-stabilized nanoparticles (CS-NPs) of average core diameter of 10 nm are used as templates for stabilization-based exchange reactions with thioctic acid to form more robust aqueous NPs that can be assembled into multilayer films. The thioctic acid stabilized nanoparticles (TAS-NPs) are networked via covalent and electrostatic linking systems, employing dithiols and the cationic polymer poly(l-lysine), respectively. Multilayer films of up to 150 nm in thickness are successfully grown at biological pH with no observable degradation of the NPs within the film. The characteristic surface plasmon band, an optical feature of certain NP film assemblies that can be used to report the local environment and core spacing within the film, is preserved. Growth dynamics and film stability in solution and in the air are examined, with poly(l-lysine) linked films showing no evidence of aggregation for at least 50 days. We believe these films represent a pivotal step toward exploring the potential of aqueous NP film assemblies as a sensing apparatus
SAPHIRE 8 Volume 2 - Technical Reference
The Systems Analysis Programs for Hands-on Integrated Reliability Evaluations (SAPHIRE) refers to a set of computer programs that were developed to create and analyze probabilistic risk assessment (PRAs). Herein information is provided on the principles used in the construction and operation of Version 8.0 of the SAPHIRE system. This report summarizes the fundamental mathematical concepts of sets and logic, fault trees, and probability. This volume then describes the algorithms used to construct a fault tree and to obtain the minimal cut sets. It gives the formulas used to obtain the probability of the top event from the minimal cut sets, and the formulas for probabilities that apply for various assumptions concerning reparability and mission time. It defines the measures of basic event importance that SAPHIRE can calculate. This volume gives an overview of uncertainty analysis using simple Monte Carlo sampling or Latin Hypercube sampling, and states the algorithms used by this program to generate random basic event probabilities from various distributions. Also covered are enhance capabilities such as seismic analysis, Workspace algorithms, cut set "recovery," end state manipulation, and use of "compound events.

SPAR-H Step-by-Step Guidance
Step-by-step guidance was developed recently at Idaho National Laboratory for the US Nuclear Regulatory Commission on the use of the Standardized Plant Analysis Risk-Human Reliability Analysis (SPAR-H) method for quantifying Human Failure Events (HFEs). This work was done to address SPAR-H user needs, specifically requests for additional guidance on the proper application of various aspects of the methodology. This paper overviews the steps of the SPAR-H analysis process and highlights some of the most important insights gained during the development of the step-by-step directions. This supplemental guidance for analysts is applicable when plant-specific information is available, and goes beyond the general guidance provided in existing SPAR-H documentation. The steps highlighted in this paper are: Step-1, Categorizing the HFE as Diagnosis and/or Action; Step-2, Rate the Performance Shaping Factors; Step-3, Calculate PSF-Modified HEP; Step-4, Accounting for Dependence, and; Step-5, Minimum Value Cutoff

Systems analysis programs for Hands-on integrated reliability evaluations (SAPHIRE) Version 5.0: Verification and validation (V&V) manual. Volume 9
A verification and validation (V&V) process has been performed for the System Analysis Programs for Hands-on Integrated Reliability Evaluation (SAPHIRE) Version 5.0. SAPHIRE is a set of four computer programs that NRC developed for performing probabilistic risk assessments. They allow an analyst to perform many of the functions necessary to create, quantify, and evaluate the risk associated with a facility or process being analyzed. The programs are Integrated Reliability and Risk Analysis System (IRRAS) System Analysis and Risk Assessment (SARA), Models And Results Database (MAR-D), and Fault tree, Event tree, and Piping and instrumentation diagram (FEP) graphical editor. Intent of this program is to perform a V&V of successive versions of SAPHIRE. Previous efforts have been the V&V of SAPHIRE Version 4.0. The SAPHIRE 5.0 V&V plan is based on the SAPHIRE 4.0 V&V plan with revisions to incorporate lessons learned from the previous effort. Also, the SAPHIRE 5.0 vital and nonvital test procedures are based on the test procedures from SAPHIRE 4.0 with revisions to include the new SAPHIRE 5.0 features as well as to incorporate lessons learned from the previous effort. Most results from the testing were acceptable; however, some discrepancies between expected code operation and actual code operation were identified. Modifications made to SAPHIRE are identified
A mixed treatment comparison meta-analysis of metaphylaxis treatments for bovine respiratory disease in beef cattle

Journal of Animal Science, 95(2), 626-635. doi:10.2527/jas2016.1062The objective of this project was to evaluate the effects of antimicrobials approved for parenteral metaphylactic use in feeder and stocker calves on morbidity and mortality for bovine respiratory disease with the use of a mixed treatment comparison meta-analysis. An initial literature review was conducted in April 2016 through Pubmed, Agricola, and CAB (Commonwealth Agricultural Bureau) for randomized controlled trials for metaphylaxis antimicrobial administered parentally to incoming feedlot or stocker calves within 48 h of arrival. The final list of publications included 29 studies, with a total of 37 trials. There were 8 different metaphylactic antimicrobials. Final event outcomes were categorized into bovine respiratory disease (BRD) morbidity cumulative incidence d 1 to <= 60 of the feeding period, BRD morbidity cumulative incidence d 1 to closeout of the feeding period, BRD mortality cumulative incidence d 1 to closeout of the feeding period, and BRD retreatment cumulative incidence morbidity d 1 to closeout of the feeding period. Network meta-analysis combined direct and indirect evidence for all the event outcomes to determine mean odds ratio (OR) with 95% credibility intervals (CrIs) for all metaphylactic antimicrobial comparisons. The "upper tier" treatment arms for morbidity d 1 to <= 60 included tulathromycin, gamithromycin, and tilmicosin. For BRD mortality cumulative incidence d 1 to closeout and BRD retreatment morbidity d 1 to closeout, classifying the treatment arms into tiers was not possible due to overlapping 95% CrIs. The results of this project accurately identified differences between metaphylactic antimicrobials, and metaphylactic antimicrobial options appear to offer different outcomes on BRD morbidity and mortality odds in feedlot cattle
Subacute ruminal acidosis reduces sperm quality in beef bulls
Breeding bulls are commonly fed high-energy diets, which may induce subacute ruminal acidosis (SARA). In this experiment, 8 Santa Gertrudis bulls (age 20 ± 6 mo) were used to evaluate the extent and duration of effects of SARA on semen quality and the associated changes in circulating hormones and metabolites. The bulls were relocated and fed in yards with unrestricted access to hay and daily individual concentrate feeding for 125 d before SARA challenge. Semen was collected and assessed at 14-d intervals before the challenge to ensure acclimatization and the attainment of a stable spermiogram. The challenge treatments consisted of either a single oral dose of oligofructose (OFF; 6.5 g/kg BW) or an equivalent sham dose of water (Control). Locomotion, behavior, respiratory rate, and cardiovascular and gastrointestinal function were intensively monitored during the 24-h challenge period. Rumen fluid samples were retained for VFA, ammonia, and lactate analysis. After the challenge, semen was then collected every third day for a period of 7 wk and then once weekly until 12 wk, with associated blood collection for FSH, testosterone, inhibin, and cortisol assay. Percent normal sperm decreased in bulls dosed with OFF after the challenge period (P < 0.05) and continued to remain lower on completion of the study at 88 d after challenge. There was a corresponding increase in sperm defects commencing from 16 d after challenge. These included proximal cytoplasmic droplets (P < 0.001), distal reflex midpieces (P = 0.01), and vacuole and teratoid heads (P < 0.001). Changes in semen quality after challenge were associated with lower serum testosterone (P < 0.001) and FSH (P < 0.05). Serum cortisol in OFF bulls tended to be greater (P = 0.07) at 7 d after challenge. This study shows that SARA challenge causes a reduction in sperm quality sufficient to preclude bulls from sale as single sire breeding animals 3 mo after the event occurred
Pharmacological Characterization of FE 202158, a Novel, Potent, Selective, and Short-Acting Peptidic Vasopressin V 1a Receptor Full Agonist for the Treatment of Vasodilatory Hypotension
ABSTRACT R), and oxytocin receptor, respectively] contrasting with AVP's lack of selectivity, especially versus the V 2 R (selectivity ratio of 1:18:0.2:92; human V 1a R EC 50 ϭ 0.24 nM). This activity and selectivity profile was confirmed in radioligand binding assays. FE 202158 was a potent vasoconstrictor in the isolated rat common iliac artery ex vivo (EC 50 ϭ 3.6 nM versus 0.8 nM for AVP) and reduced rat ear skin blood flow after intravenous infusion in vivo (ED 50 ϭ 4.0 versus 3.4 pmol/kg/min for AVP). The duration of its vasopressor effect by intravenous bolus in rats was as short as AVP at submaximally effective doses. FE 202158 had no V 2 R-mediated antidiuretic activity in rats by intravenous infusion at its ED 50 for reduction of ear skin blood flow, in contrast with the pronounced antidiuretic effect of AVP. Thus, FE 202158 seems suitable for treatment of conditions where V 1a R activity is desirable but V 2 R activity is potentially deleterious, such as vasodilatory hypotension in septic shock. In addition to the desirable selectivity profile, its shortacting nature should allow dose titration with rapid onset and offset of action to optimize vasoconstriction efficacy and safety
