    Near-Infrared Variability Study of the Central 2.3 arcmin x 2.3 arcmin of the Galactic Centre I. Catalog of Variable Sources

    We used four-year baseline HST/WFC3 IR observations of the Galactic Centre in the F153M band (1.53 micron) to identify variable stars in the central ~2.3'x2.3' field. We classified 3845 long-term (periods from months to years) and 76 short-term (periods of a few days or less) variables among a total sample of 33070 stars. For 36 of the latter ones, we also derived their periods (<3 days). Our catalog not only confirms bright long period variables and massive eclipsing binaries identified in previous works, but also contains many newly recognized dim variable stars. For example, we found \delta Scuti and RR Lyrae stars towards the Galactic Centre for the first time, as well as one BL Her star (period < 1.3 d). We cross-correlated our catalog with previous spectroscopic studies and found that 319 variables have well-defined stellar types, such as Wolf-Rayet, OB main sequence, supergiants and asymptotic giant branch stars. We used colours and magnitudes to infer the probable variable types for those stars without accurately measured periods or spectroscopic information. We conclude that the majority of unclassified variables could potentially be eclipsing/ellipsoidal binaries and Type II Cepheids.     Because of strong and spatially highly variable interstellar extinction and extreme source crowding, the faint (K>15) stellar population in the Milky Way's nuclear cluster is still poorly studied. RR Lyrae stars provide us with a tool to estimate the mass of the oldest, relative dim stellar population. Recently, we analyzed HST/WFC3/IR observations of the central 2.3'x2.3' of the Milky Way and found 21 variable stars with periods between 0.2 and 1d. Here, we present a further comprehensive analysis of these stars. The period-luminosity relationship of RR Lyrae is used to derive their extinctions and distances. Using multiple approaches, we classify our sample as four RRc, four RRab and three candidates, ten binaries. Especially, the four RRabs show sawtooth light curves and fall exactly onto the Oosterhoff I division in the Bailey diagram. Compared to the RRabs reported by Minniti et al, 2016, our new RRabs have higher extinction (A_K>1.8) and should be closer to the Galactic Centre. The extinction and distance of one RRab match those for the nuclear star cluster given in previous works. We perform simulations and find that after correcting for incompleteness, there could be no more than 40 RRabs within the nuclear star cluster and in our field-of-view. Through comparing with the known globular clusters of the Milky Way, we estimate that if there exists an old, metal-poor (-1.5<[Fe/H]<-1) stellar population in the Milky Way nuclear star cluster on a scale of 5x5pc, then it contributes at most 4.7x10^5 solar mass, i.e., ~18% of the stellar mass.Comment: 33 pages, 10 figures.     Modelling with a CFD code the near-range dispersion of particles unexpectedly released from a nuclear power plant

    An event in November 2007 in Ascó-1 nuclear power plant (Spain) originated the release of a significant amount of hot metallic particles through the discharge stack. Particles were dispersed and deposited in roofs and neighbouring areas within the NPP controlled area. However, the event was not detected until March 2008. More than 1,300 hot points with radioactive particles were found, 94% located inside the double fenced controlled area and 6% within the exclusion area; 5 particles were out of the exclusion area, across the river. To provide additional insights on the potential consequences of the release, a computational fluid dynamics (CFD) code, Ansys-CFX-11, has been used to simulate the near-range atmospheric dispersion and deposition of the particles. The purpose of the analysis was to assess the distance travelled by particles of different sizes. A very detailed model of the site was built, taking into account the buildings and the terrain features including the river valley and the surrounding hills. The modelled domain was 3.2 x 5.2 km, with the atmospheric layer up to 4 km height. The atmospheric conditions recorded during different periods of time were classified into 37 representative categories. In general, the distribution of the particles found was adequately reproduced. Particles larger than 100 microns could not travel beyond the double fence. Particles between 50 and 100 microns could have been deposited mainly within the exclusion area, with a small probability of travelling farther. Smaller particles could have travelled beyond, but also should have been deposited in the nearby area, while the majority of particles found are larger, thus indicating that the size of the released particles should be above 50 microns.     Pd-Catalyzed α-Arylation of Sulfones in a Three-Component Synthesis of 3-[2-(Phenyl/methylsulfonyl)ethyl]indoles

    A novel four-step domino process for the synthesis of 3-[2-(aryl/alkylsulfonyl)ethyl]indoles starting from readily available 2-iodoanilines is reported. The domino reaction is based on the intramolecular palladium-catalyzed α-arylation of sulfones, which was combined with both intermolecular aza-Michael and Michael addition reactions using vinyl sulfones as the electrophile. The domino process produced good yields and tolerated the presence of substituents with different electronic properties on the aniline ring. In addition, density functional theory (DFT) calculations were carried out to gain more insight into the formation of the observed indole derivatives.     Identification of new sources of resistance to RHBV- rice hoja blanca virus

    With the aim to find new sources of resistance to rice hoja blanca (white leaf) disease, transmitted by the insect Tagosodes orizicolus, 660 genotypes were evaluated under greenhouse and field conditions. Seven resistant genotypes were identified, and genomic studies were performed to demonstrate that the resistance in these sources is genetically different from that of Fedearroz 2000, which is currently the variety with the most resistance to hoja blanca. These new resistance sources constitute a resource that can be used to sustainably extend hoja blanca disease management throughout all of the rice-growing regions of tropical America. This is the first report of hoja blanca resistance in indica rice and different from that of Fedearroz 2000

    FASN-dependent de novo lipogenesis is required for brain development

    Fate and behavior of neural progenitor cells are tightly regulated during mammalian brain development. Metabolic pathways, such as glycolysis and oxidative phosphorylation, that are required for supplying energy and providing molecular building blocks to generate cells govern progenitor function. However, the role of de novo lipogenesis, which is the conversion of glucose into fatty acids through the multienzyme protein fatty acid synthase (FASN), for brain development remains unknown. Using Emx1Cre-mediated, tissue-specific deletion of Fasn in the mouse embryonic telencephalon, we show that loss of FASN causes severe microcephaly, largely due to altered polarity of apical, radial glia progenitors and reduced progenitor proliferation. Furthermore, genetic deletion and pharmacological inhibition of FASN in human embryonic stem cell-derived forebrain organoids identifies a conserved role of FASN-dependent lipogenesis for radial glia cell polarity in human brain organoids. Thus, our data establish a role of de novo lipogenesis for mouse and human brain development and identify a link between progenitor-cell polarity and lipid metabolism

    Fibroblast Growth Factor 21 and the Adaptive Response to Nutritional Challenges.

    The Fibroblast Growth Factor 21 (FGF21) is considered an attractive therapeutic target for obesity and obesity-related disorders due to its beneficial effects in lipid and carbohydrate metabolism. FGF21 response is essential under stressful conditions and its metabolic effects depend on the inducer factor or stress condition. FGF21 seems to be the key signal which communicates and coordinates the metabolic response to reverse different nutritional stresses and restores the metabolic homeostasis. This review is focused on describing individually the FGF21-dependent metabolic response activated by some of the most common nutritional challenges, the signal pathways triggering this response, and the impact of this response on global homeostasis. We consider that this is essential knowledge to identify the potential role of FGF21 in the onset and progression of some of the most prevalent metabolic pathologies and to understand the potential of FGF21 as a target for these diseases. After this review, we conclude that more research is needed to understand the mechanisms underlying the role of FGF21 in macronutrient preference and food intake behavior, but also in β-klotho regulation and the activity of the fibroblast activation protein (FAP) to uncover its therapeutic potential as a way to increase the FGF21 signaling

    The death receptor antagonist FAIM promotes neurite outgrowth by a mechanism that depends on ERK and NF-κB signaling

    Fas apoptosis inhibitory molecule (FAIM) is a protein identified as an antagonist of Fas-induced cell death. We show that FAIM overexpression fails to rescue neurons from trophic factor deprivation, but exerts a marked neurite growth–promoting action in different neuronal systems. Whereas FAIM overexpression greatly enhanced neurite outgrowth from PC12 cells and sympathetic neurons grown with nerve growth factor (NGF), reduction of endogenous FAIM levels by RNAi decreased neurite outgrowth in these cells. FAIM overexpression promoted NF-κB activation, and blocking this activation by using a super-repressor IκBα or by carrying out experiments using cortical neurons from mice that lack the p65 NF-κB subunit prevented FAIM-induced neurite outgrowth. The effect of FAIM on neurite outgrowth was also blocked by inhibition of the Ras–ERK pathway. Finally, we show that FAIM interacts with both Trk and p75 neurotrophin receptor NGF receptors in a ligand-dependent manner. These results reveal a new function of FAIM in promoting neurite outgrowth by a mechanism involving activation of the Ras–ERK pathway and NF-κB

    Isolation of human bone marrow mesenchymal stem cells and evaluation of their osteogenic potential

    Las células madre mesenquimatosas de médula ósea humana (abreviadas hBMSCs) constituyen una fuente de células auto-renovables con alto potencial de diferenciación, comúnmente aisladas a partir de los aspirados medulares en huesos largos. Su diferenciación hacia el linaje osteogénico, por ejemplo, ha sido ampliamente utilizada para la evaluación biológica de biomateriales o matrices con aplicaciones en la ingeniería de tejidos óseos. El objetivo de este trabajo consistió en aislar hBMSCs a partir de la cabeza femoral de pacientes sometidos a artroplastia total de cadera, así como evaluar su potencial osteogénico. Brevemente, se extrajo el hueso esponjoso y se disgregó mecánicamente; las células desprendidas se cultivaron y las células no adherentes se eliminaron luego de 4 días. El potencial osteogénico se evaluó en la quinta generación de cultivo, mediante ensayos de diferenciación a 14 y 20 días donde se compararon cultivos con y sin suplementos osteogénicos. La evaluación se realizó mediante tinción con Alizarina Roja y la cuantificación de los niveles de expresión génica de los marcadores osteogénicos colágeno tipo I, osteonectinca y sialoprotiena ósea mediante RT-PCR en tiempo real. Las hBMSCs obtenidas presentaron un fenotipo no-diferenciado estable, así como la capacidad de mineralizar la matriz extracelular y expresar un fenotipo similar al osteoblasto durante la inducción osteogénica. Los tres marcadores evaluados se sobre-expresaron en los cultivos en condiciones osteogénicas, y se encontró que cambios hasta de 2X en sus niveles de expresión son relevantes para el desarrollo del proceso de diferenciación. El modelo de hBMSCS presentado podría ser utilizado para la evaluación in vitro de la osteoinductividad de diferentes biomateriales, moléculas bioactivas o matrices para ingeniería de tejidos.Human bone marrow mesenchymal stem cells (hBMSCs) comprise a cell population capable of self-renewal and multilineage differentiation commonly isolated from bone marrow aspirates of large bones. Their osteogenic potential has been extensively exploited for the biological evaluation of scaffolds or biomaterials with applications in bone tissue engineering. This work aimed to isolate hBMSCs from femoral heads of patients undergoing total hip arthroplasty and to evaluate their osteogenic potential. Briefly, the trabecular bone was extracted and mechanically disaggregated; the released cells were cultured and non-adherent cells were removed after 4 days. The osteogenic potential was evaluated at the fifth passage after 14 and 20 days of induction, comparing cultures with and without osteogenic supplements, via Alizarin red staining and the quantification of the gene expression levels of the osteogenic markers collagen type I, osteonectin and bone sialoprotein through real-time RT-PCR. The obtained hBMSCs presented a stable undifferentiated phenotype after prolonged cell culture, matrix mineralization capabilities and expression of osteoblast phenotype upon osteogenic induction. The three markers were up-regulated in cultures under osteogenic conditions and 2 fold differences in their expression levels were found to be significant for the onset of the differentiation process. The obtained hBMSCs may have applications on the in vitro evaluation of the osteoinductivity of different biomaterials, bioactive molecules or tissue engineering scaffolds
