Effects of ageing on the vulnerability of the substantia nigra pars compacta dopaminergic neurons: implications in Parkinson’s disease

The degeneration of dopaminergic neurons (DAn) in the midbrain substantia nigra pars compacta (SNpc) is a definitive feature of Parkinson's disease (PD). Although age has been established as one of the main risk factors, the role that ageing has in the development of the disease is not yet fully understood. The aim of this Thesis, therefore, was to enhance our understanding of how ageing may increase the vulnerability of SNpc DAn. To achieve this aim, isobaric tags for relative and absolute quantitation (iTRAQ) labelling combined with liquid chromatography tandem mass spectrometry (LC-MS/MS) was used to quantitatively compare the SNpc proteome of rats during ageing. Western blot and immunofluorescence analyses were subsequently conducted to verify some of the differentially expressed proteins from the analysis (e.g., GFAP), while immunohistochemistry analyses were performed to quantitatively and morphologically characterize the DAn of the rat SNpc themselves during ageing. From a total of 1,953 proteins that were identified and quantified in the proteomic study, the expression levels of 66 proteins were altered throughout ageing. Bioinformatic analysis revealed that proteins related to glial cells (e.g., GFAP) and the extracellular matrix (ECM) were differentially expressed in the old rat SNpc. Importantly, an unusual form of the GFAP protein (i.e., GFAPδ) was showed for the first time to be differentially expressed during ageing. In addition to this, the level of tyrosine hydroxylase (TH) expression in the SNpc throughout ageing was maintained. This was somewhat surprising as it appears that, independent of the proteomic changes, there was a general decrease in the density of rat SNpc DAn together with an increase of their soma size with ageing, which might indicate that the remaining DAn are able to maintain the level of tyrosine hydroxylase (TH) expression in the SNpc throughout ageing. These results were followed by an in vitro investigation of the role astrocytes play in the vulnerability of DAn. To do this, primary cultures of embryonic DAn were challenged with the toxin 6-hydroxidopamine (6-OHDA) after reducing the number of astrocytes in the cultures in a unique way (i.e., by using the anti-mitotic drug paclitaxel). Though the anti-mitotic drug was successful in reducing astrocytes in the cultures, it was difficult to test the effect this had on combating the effects of 6-OHDA on DAn because the toxin also affected the viability of the remaining astrocytes in culture. Lastly, multiple multi-study proteomic comparisons of published studies on the ageing nervous system and PD demonstrated that metabolism, oxidation-reduction mechanisms, mitochondrial function and immune system were biological processes and pathways enriched in both ageing and PD. Because some of these biological processes were the same as those found differentially expressed in the proteomics study of ageing conducted here, this support the idea that they may be key toward understanding how ageing is involved in the development of PD. In conclusion, this Thesis showed that ageing alters the metabolic support associated with mitochondrial and oxidation-reduction functions (as it happens in PD) and suggests that this might have considerable repercussions on highly reactive oxygen species (ROS) sensitive neurons such as SNpc DAn. Adding to this, the alteration of proteins related to glial cells (e.g., astrocytes) might affect their protective function in the SNpc during a time when they are become even more essential to the survival of DAn

Scutoids unveil the three-dimensional packing in curved epithelia

As animals develop, the initial simple planar epithelia of the early embryos must acquire complex three-dimensional architectures to form the final functional tissues of the organism. Epithelial bending is, therefore, a general principle of all developing systems. Scholarly publications depict epithelial cells as prisms where their basal and apical faces resemble polygons with the same number of sides. The accepted view is that, when a tissue bend, the cells of the epithelia modify their shape from columnar to what has been traditionally called “bottle shape”. However, the morphology and packing of curved epithelia remain largely unknown. Here, through mathematical and computational modelling, we show that cells in bent epithelia necessarily undergo intercalations along the apico-basal axis. This event forces cells to exchange their neighbours between their basal and apical surfaces. Therefore, the traditional view of epithelial cells as simple prisms is incompatible with this phenomenon. Consequently, epithelial cells are compelled to adopt a novel geometrical shape that we have named “scutoid”. The in-depth analysis of diverse epithelial tissues and organs confirm the generation of apico-basal transitions among cell during morphogenesis. Using biophysics arguments, we determine that scutoids support the energetic minimization on the tissue and conclude that the transitions along the apico-basal axis stabilize the threedimensional packing of the tissue. Altogether, we argue that scutoids are nature’s solution to bend efficiently epithelia, and the missing piece for developing a unifying and realistic model of epithelial architecture

The Arthurian World of J.R.R. Tolkien's The Lord of the Rings: a Reassessment

This PhD thesis intends to explore the possible influences and rewritings of Arthurian legends in J.R.R. Tolkien's The Lord of the Rings. Tolkien's work seems to be strongly influenced by Arthurian literature. The themes of Quest for the Ring and the Fellowship of the Ring echo the Grail Quest and the Order of the Round Table. The Ring as the centre of the plot in The Lord of the Rings is a clear allusion to the Grail legend while the Arthurian myth of the Wasteland is represented in Rohan, whose ruler resembles the Fisher King, and in Gondor, which echoes the Arthurian idea of a ruler and his land as one. The tragic love between Aragorn and Arwen has some parallelisms with Tristan and Isolde's fatal love while Frodo and Bilbo's uncle-nephew relationship reminds us of King Arthur and Gawain. Also, Galadriel, Arwen or Éowyn share many resemblances with some Arthurian heroines such has Morgan Le Fay, the Lady of the Lake or Guenevere, among others. Similarly, landscapes and objects from The Lord of the Rings have Arthurian resonances. Indeed, the term 'Middle-earth' appears in medieval literature, including Arthurian texts, while Aragorn's sword clearly resembles King Arthur's legendary blade, Excalibur

Desde este proyecto se pretende realizar un análisis de la violencia de género, de dónde viene y cómo influye la sociedad en ello. Se ha hecho visible esta nueva realidad que se ha ido incrementando desde edades más tempranas, fomentadas en algunos casos por las redes sociales y nuevas tecnologías. Tras un recorrido por los diversos factores que pueden afectar a sus relaciones y cómo influye el entorno que les rodea a nivel de escuela, familia, amigos y sociedad en general, se pretende diseñar un proyecto a través del análisis, reflexión y pensamiento crítico que dote de herramientas a las chicas y chicos antes de introducirse en este tipo de relaciones, educándolos y educándolas en la igualdad social, igualdad en las relaciones afectivas y en la detección de relaciones tóxicas que puedan derivar en violencia de género.Grado en Educación Socia

Microstructural development of Powder Metallurgy Cobalt-based superalloys processed by Field Assisted Sintering Techniques (FAST)

This study reports the microstructural evolution, physical and mechanical properties of cobalt-based superalloys processed from mechanical alloyed powders and consolidated by field-assisted sintering techniques (FAST). After an initial thermodynamic simulation of the ternary diagram by Thermocalc (R) to determine the composition, a sequential milling process was carried out at room temperature up to 40 h milling of two different alloying systems: CO-12Al-10W (at%) and Co-12Al-10W-2Ti-2Ta (at%). Characterization of the powders was performed by using X-Ray diffraction, scanning electron microscope with energy-dispersive X-ray spectroscopy (EDS) and particle-size analyzer. Consolidated samples were also characterized in terms of density, microhardness and hardness. In order to promote the dual gamma/gamma microstructure, both alloys were aged after solution annealing heat treatment, improving a new route of consolidation with a new level of performance.Project funded by the Madrid Region under the program S2013/MIT-277

Propuesta de Infraestructura de la Escuela de Arquitectura – Universidad Señor de Sipán para optimizar su nivel de enseñanza – aprendizaje

Las escuelas de arquitectura cumplen un rol importante en la ciudad, ya que el proyecto del arquitecto, a diferencia de otras profesiones, es el que más cambio puede producir en la sociedad o en una comunidad determinada. Los proyectos de arquitectura a menudo están relacionados con el progreso, la cultura, la evolución y el paso del tiempo. En la actualidad la escuela de arquitectura de la Universidad Señor de Sipán no cuenta con los espacios necesarios, debido a que el edificio en el que funciona no fue pensado para este fin sino que fue realizado para que se desarrollen las actividades integradoras de la universidad. Es por este motivo que con el presente trabajo de investigación se pretende cubrir las necesidades de la escuela a través de un edificio que cuente con las comodidades espaciales y funcionales que la escuela requiere, con el fin de brindar un mejor servicio a sus usuarios en general y generar a la vez un aporte a la ciudad.Tesi

Las Delicias. Centro Residencial Geriátrico

El proyecto propuesto para el Centro Geriátrico en el actual aparcamiento de las Delicias se ha realizado teniendo como prioridad el lugar y los edificios existentes, poniéndolos en valor y haciendo uso de lo existente. La parcela durante más de cuatro siglos ha acogido numerosos usos públicos y lúdicos, teniendo un marcado carácter social en el barrio. Por ello, se propone recuperar esta tradición social y cultural (patrimonio inmaterial) perdida con la aparición del uso de aparcamiento, para crear un proyecto cercano a los vecinos del barrio, donde tengan cabida todas las generaciones existentes. La geometría irregular de la parcela y la suma de las diferentes arquitecturas patrimoniales encontradas, dificultan y enriquecen la estrategia del proyecto. Mediante una limpieza de los elementos superfluos y no protegidos, se propone poner en valor el alzado lateral de la sala Mª Cristina para que vuelva a ser telón de fondo de usos públicos y lúdicos del barrio (memoria colectiva), a la vez que se conserva la nave abovedada de hormigón. Por otra parte, el proyecto también conserva la portada neoclásica de entrada como actual imagen reconocible de la parcela hacia la ciudad. Para explicar la propuesta hablaremos de dos partes diferenciadas: la intervención principal y las intervenciones secundarias. - La intervención principal es la realizada en el edificio de viviendas existente. Tras el análisis en profundidad de dicho edificio, se propone realizar un edificio que complete la manzana. Como si de un organismo vivo se tratase, la nueva edificación se adhiere a lo existente. Comienza haciéndose sitio por las viviendas abandonadas, rehabilitando los núcleos verticales para así hacer del edificio un espacio accesible, y se formaliza al exterior como una infraestructura sencilla dispuesta para ser habitada, para ser colonizada. Este organismo se irá modificando a medida que pase el tiempo, siempre respondiendo a las necesidades de los vecinos y del barrio. Una estructura que se construye como un espejo de la edificación existente. Respetando las dimensiones de la estructura muraria antigua, pero con un lenguaje contemporáneo, y ampliando los patios existentes, que configuran el edificio. La fachada se compone en cierta simetría con la existente, aumentando de tamaño el elemento balcón que aparece en el edificio, se configura junto a escaleras que conectan estos espacios exteriores en altura. El edificio se cierra con un elemento tradicional llevado a otro papel. Una celosía compuesta por perfiles pequeños a modo de reja, que nos transporta a las casas de los pueblos. Se podría decir que la fachada es casi inexistente dada su permeabilidad, permitiendo ver la vida dentro del edificio y haciendo partícipes a todos de ella. A nivel programático, se disponen los usos públicos en la planta baja aprovechando así la plaza generada. En las plantas primera y segunda se desarrolla la vida de los mayores, con las viviendas y servicios para mayores. Los usos se modifican según las necesidades pues, los espacios libres son capaces de albergar cualquier tipo de uso: terapia ocupacional, talleres, espacios de descanso,… además de ser beneficioso para los mayores permanecer en el exterior para su salud. Por último, la cubierta transitable acoge cualquier tipo de actividad al aire libre: huertos, espacios de juego, guardería… Con la posibilidad de colocar pabellones o pequeñas instalaciones. E incluso, llegado el momento en el que la estructura esté completa, colonizar también la cubierta. - Las intervenciones secundarias son las realizadas en torno al barrio, pequeñas propuestas para poner en valor lo existente: La plaza: se libera la parte norte de la nave abriendo así la plaza creada con la plaza de Los cristos. De esta forma se pretende fomentar el flujo entre ambas, generando un espacio público amable y habitable. La nave abovedada: se considera la nave como elemento único dentro del conjunto. Por ello, se alberga en ella la parte del programa menos común: hidroterapia. Se elimina el forjado intermedio, construido a posteriori en la nave, para así tener un espacio único, amplio y aprovechar ese espacio abovedado. Además, de esta forma, se libera el patio junto a la sala María Cristina para poner el valor esa fachada. Esta pieza está dirigida tanto al público de los mayores del barrio como a cualquier habitante del barrio y la ciudad. El pasadizo: el espacio comprendido entre la nave, la sala María Cristina y el convento se reajusta, abriendo puertas en el convento y conectando éste con el resto del conjunto. Consiguiendo así un espacio interesante junto a la sala María Cristina, donde disfrutar de la fachada histórica. La nave industrial: una de las naves industriales existentes se mantiene como espacio polivalente para todo el conjunto. Un espacio diáfano reciclado, para eventos y cualquier necesidad. En conclusión, el proyecto ha sido un ejercicio de síntesis en todos los aspectos. Teniendo como primera prioridad la puesta en valor de lo existente, buscando la esencia de cada elemento y poniéndola por delante. Intentado siempre actuar sin hacer ruido, leyendo y dejándose llevar por el lugar

Sistema para la gestión informatizada de stock de productos

El objetivo principal de este proyecto es conseguir crear un sistema que simule la gestión de un almacén, tanto la parte de almacenamiento de material y entrada de productos nuevos, como la simulación de realizar pedidos donde se solicitará material que es almacenado en el almacén y donde deberemos controlar el stock. El proyecto se centrará en dos grandes partes: • El almacenamiento de los datos del material que se encuentra en el almacén, la información que van a contener las bases de datos sobre estos, y el control del stock donde se informará en todo momento del stock disponible y reservado para pedidos. • La simulación de realizar pedidos donde reservaremos material disponible para los pedidos, asegurándonos que no se podrá reservar más de la cantidad disponible para que todos los pedidos puedan completarse con éxito.The main objective of this project is to create a system that simulates the management of a warehouse, both the material storage part and the entry of new products, as well as the simulation of placing orders where the material that is stored in the warehouse will be requested and where we will have to control the stock. The project will focus on two main parts: • The storage of the data of the material that is in the warehouse, the information which the databases will contain about these and the control of the stock. ¿Estamos preparados para la primera asistencia en víctimas de accidentes deportivos? Revisión bibliográfica y propuesta de intervención acerca del running y la muerte súbita

En este trabajo se ha realizado una revisión bibliográfica sobre la muerte súbita en la población que practica running. Se han estudiado las causas, circunstancias, consecuencias, así como la asistencia que reciben estas personas, después del trágico suceso. Concluyendo que, con un correcto examen médico realizado al deportista, la mayoría de estos casos podría haberse evitado
