Tonsillar transcriptional profiles in atopic and non‐atopic subjects
Background: Emerging research suggests that local lymphatic tissue such as tonsils have important role in regulating the immune responses. However, allergen sensitization-induced alterations in transcriptome of tonsils are not known.
Objectives: To examine the key differences in tonsillar gene expression between atopic and non-atopic subjects and further by type of sensitization.
Methods: RNA-sequencing was performed on 52 tonsillar samples from atopic and non-atopic tonsillectomy patients. Sensitization to common food- and aero-allergen was defined by allergen specific IgE. Following groups were studied: (1) aero- and food-allergen sensitized (AS+FS) versus non-sensitized (NS), (2) aeroallergen-sensitized (AS) versus food-allergen sensitized (FS), (3) AS versus NS, (4) FS versus NS. Bioinformatics analysis was done using DESeq2(v3.10.2), WGCNA and GATK pipeline in R software (v3.3.1). Protein-protein interaction network was made from String database.
Results: We studied 13 aeroallergen-sensitized, 6 food-allergen sensitized, 4 both food-and aero-allergen-sensitized and 29 non-sensitized tonsillectomy patients. Overall, 697 unique differentially expressed genes (DEGs) were detected in all sensitized subgroups including chemokines (CXCL2, CXCL8, CXCL10, CXCL11), IL-20RA, MUC1 and MUC20. When comparing different groups, the gene expression profiles overlapped except the AS versus FS group comparison, suggesting significantly different gene expression between the two sensitization subgroups. Furthermore, aeroallergen-sensitized subjects had more prominent immune responses compared with non-sensitized and food-allergen sensitized subjects including gene expression for IL-17 pathway and Toll-like receptor signalling pathway.
Conclusion: Allergic sensitization is associated with extensive tonsillar transcriptomic alterations and changes in immune related genes and pathways. Distinct differences were found between aero-allergen and food-allergen sensitization

Performance of new gellan gum hydrogels combined with human articular chondrocytes for cartilage regeneration when subcutaneously implanted in nude mice
Gellan gum is a polysaccharide that has been recently proposed by our group for cartilage tissueengineering
applications. It is commonly used in the food and pharmaceutical industry and has
the ability to form stable gels without the use of harsh reagents. Gellan gum can function as a
minimally invasive injectable system, gelling inside the body in situ under physiological conditions
and efficiently adapting to the defect site. In this work, gellan gum hydrogels were combined with
human articular chondrocytes (hACs) and were subcutaneously implanted in nude mice for 4 weeks.
The implants were collected for histological (haematoxylin and eosin and Alcian blue staining),
biochemical [dimethylmethylene blue (GAG) assay], molecular (real-time PCR analyses for collagen
types I, II and X, aggrecan) and immunological analyses (immunolocalization of collagen types I and
II). The results showed a homogeneous cell distribution and the typical round-shaped morphology
of the chondrocytes within the matrix upon implantation. Proteoglycans synthesis was detected by
Alcian blue staining and a statistically significant increase of proteoglycans content was measured
with the GAG assay quantified from 1 to 4 weeks of implantation. Real-time PCR analyses showed a
statistically significant upregulation of collagen type II and aggrecan levels in the same periods. The
immunological assays suggest deposition of collagen type II along with some collagen type I. The
overall data shows that gellan gum hydrogels adequately support the growth and ECM deposition
of human articular chondrocytes when implanted subcutaneously in nude mice.J. T. Stress transfer among en echelon and opposing thrusts and tear faults : triggering caused by the 2003 Mw = 6.9 Zemmouri, Algeria, earthquake

The definitive version was published in Journal of Geophysical Research 116 (2011): B03305, doi:10.1029/2010JB007654.The essential features of stress interaction among earthquakes on en echelon thrusts and tear faults were investigated, first through idealized examples and then by study of thrust faulting in Algeria. We calculated coseismic stress changes caused by the 2003 Mw = 6.9 Zemmouri earthquake, finding that a large majority of the Zemmouri afterslip sites were brought several bars closer to Coulomb failure by the coseismic stresses, while the majority of aftershock nodal planes were brought closer to failure by an average of ∼2 bars. Further, we calculated that the shallow portions of the adjacent Thenia tear fault, which sustained ∼0.25 m slip, were brought >2 bars closer to failure. We calculated that the Coulomb stress increased by 1.5 bars on the deeper portions of the adjacent Boumerdes thrust, which lies just 10–20 km from the city of Algiers; both the Boumerdes and Thenia faults were illuminated by aftershocks. Over the next 6 years, the entire south dipping thrust system extending 80 km to the southwest experienced an increased rate of seismicity. The stress also increased by 0.4 bar on the east Sahel thrust fault west of the Zemmouri rupture. Algiers suffered large damaging earthquakes in A.D. 1365 and 1716 and is today home to 3 million people. If these shocks occurred on the east Sahel fault and if it has a ∼2 mm/yr tectonic loading rate, then enough loading has accumulated to produce a Mw = 6.6–6.9 shock today. Variability of Female Responses to Conspecific vs. Heterospecific Male Mating Calls in Polygynous Deer: An Open Door to Hybridization?
Males of all polygynous deer species (Cervinae) give conspicuous calls during the reproductive season. The extreme interspecific diversity that characterizes these vocalizations suggests that they play a strong role in species discrimination. However, interbreeding between several species of Cervinae indicates permeable interspecific reproductive barriers. This study examines the contribution of vocal behavior to female species discrimination and mating preferences in two closely related polygynous deer species known to hybridize in the wild after introductions. Specifically, we investigate the reaction of estrous female red deer (Cervus elaphus) to playbacks of red deer vs. sika deer (Cervus nippon) male mating calls, with the prediction that females will prefer conspecific calls. While on average female red deer preferred male red deer roars, two out of twenty females spent more time in close proximity to the speaker broadcasting male sika deer moans. We suggest that this absence of strict vocal preference for species-specific mating calls may contribute to the permeability of pre-zygotic reproductive barriers observed between these species. Our results also highlight the importance of examining inter-individual variation when studying the role of female preferences in species discrimination and intraspecific mate selection

This review focuses on behavioral genetic studies of sweet, umami, bitter and salt taste responses in mammals. Studies involving mouse inbred strain comparisons and genetic analyses, and their impact on elucidation of taste receptors and transduction mechanisms are discussed. Finally, the effect of genetic variation in taste responsiveness on complex traits such as drug intake is considered. Recent advances in development of genomic resources make behavioral genetics a powerful approach for understanding mechanisms of taste
The population of merging compact binaries inferred using gravitational waves through GWTC-3
We report on the population properties of 76 compact binary mergers detected with gravitational waves below a false alarm rate of 1 per year through GWTC-3. The catalog contains three classes of binary mergers: BBH, BNS, and NSBH mergers. We infer the BNS merger rate to be between 10 and 1700 and the NSBH merger rate to be between 7.8 and 140 , assuming a constant rate density versus comoving volume and taking the union of 90% credible intervals for methods used in this work. Accounting for the BBH merger rate to evolve with redshift, we find the BBH merger rate to be between 17.9 and 44 at a fiducial redshift (z=0.2). We obtain a broad neutron star mass distribution extending from to . We can confidently identify a rapid decrease in merger rate versus component mass between neutron star-like masses and black-hole-like masses, but there is no evidence that the merger rate increases again before 10 . We also find the BBH mass distribution has localized over- and under-densities relative to a power law distribution. While we continue to find the mass distribution of a binary's more massive component strongly decreases as a function of primary mass, we observe no evidence of a strongly suppressed merger rate above . The rate of BBH mergers is observed to increase with redshift at a rate proportional to with for . Observed black hole spins are small, with half of spin magnitudes below . We observe evidence of negative aligned spins in the population, and an increase in spin magnitude for systems with more unequal mass ratio
