    Mamíferos em remanescentes florestais de um ecótono Mata Atlântica-Cerrado no sudeste do Brasil

    Forest areas in southeastern Brazil were intensively fragmented by historical processes of land use, and even today this region is under severe anthropic pressure due to agricultural, mining, tourism, and high urban expansion. However, contemporaneous studies on its biodiversity, particularly those focusing on remaining fauna in higly fragmented landscapes are necessary. In the present study, we present a species list of mammals in forest remnants of an agricultural landscape from southeastern Brazil (RPPN Fazenda Lagoa), located in an ecotone of Atlantic Forest and Cerrado, with additional information on habitat use, vulnerability to roadkill and considerations for conservation purposes. We recorded 63 species of Chiroptera, Carnivora, Rodentia, Didelphimorphia, Primates, Cingulata, Pilosa, Cetartiodactyla and Lagomorpha. Approximately 15% of the species are under some risk of extinction, including the endemic buffy-tufted-ear marmoset (Callithrix aurita). We also recorded invasive species and domestic dogs and cats living in feral condition. Ten species were reported as roadkills in the roads that give access to the RPPN. We concluded that these remnants play an important role in the maintenance and conservation of the mammals, serving as one of the last refuges for wildlife in a severely deforested region. No entanto, estudos contemporâneos sobresua biodiversidade, particularmente sobre a biodiversidade remanescente em paisagensprofundamente fragmentadas são necessários. No presente estudo, apresentamos uma listade espécies de mamíferos em remanescentes florestais de uma paisagem agrícola do sudestedo Brasil (RPPN Fazenda Lagoa), localizada em ecótono de Mata Atlântica e Cerrado,com informações adicionais sobre uso de habitats, vulnerabilidade a atropelamentos e consideraçõespara a conservação. Registramos 63 espécies de Chiroptera, Carnivora, Rodentia,Didelphimorphia, Primates, Cingulata, Pilosa, Cetartiodactyla e Lagomorpha. Cerca de 15%das espécies estão em algum risco de extinção, incluindo o endêmico sagui-da-serra-escuro, Callithrix aurita. Também registramos espécies silvestres invasoras, cães e gatos domésticosvivendo em condição feral. Dez espécies foram registradas como vítimas de atropelamentonas estradas de acesso a RPPN.     Climatic and local stressor interactions threaten tropical forests and coral reefs

    Tropical forests and coral reefs host a disproportionately large share of global biodiversity and provide ecosystem functions and services used by millions of people. Yet, ongoing climate change is leading to an increase in frequency and magnitude of extreme climatic events in the tropics, which, in combination with other local human disturbances, is leading to unprecedented negative ecological consequences for tropical forests and coral reefs. Here, we provide an overview of how and where climate extremes are affecting the most biodiverse ecosystems on Earth and summarize how interactions between global, regional and local stressors are affecting tropical forest and coral reef systems through impacts on biodiversity and ecosystem resilience. We also discuss some key challenges and opportunities to promote mitigation and adaptation to a changing climate at local and global scales.     Full genome sequence and sfRNA interferon antagonist activity of Zika virus from Recife, Brazil

    Background: The outbreak of Zika virus (ZIKV) in the Americas has transformed a previously obscure mosquito-transmitted arbovirus of the Flaviviridae family into a major public health concern. Little is currently known about the evolution and biology of ZIKV and the factors that contribute to the associated pathogenesis. Determining genomic sequences of clinical viral isolates and characterization of elements within these are an important prerequisite to advance our understanding of viral replicative processes and virus-host interactions. Methodology/Principal findings: We obtained a ZIKV isolate from a patient who presented with classical ZIKV-associated symptoms, and used high throughput sequencing and other molecular biology approaches to determine its full genome sequence, including non-coding regions. Genome regions were characterized and compared to the sequences of other isolates where available. Furthermore, we identified a subgenomic flavivirus RNA (sfRNA) in ZIKV-infected cells that has antagonist activity against RIG-I induced type I interferon induction, with a lesser effect on MDA-5 mediated action. Conclusions/Significance: The full-length genome sequence including non-coding regions of a South American ZIKV isolate from a patient with classical symptoms will support efforts to develop genetic tools for this virus. Detection of sfRNA that counteracts interferon responses is likely to be important for further understanding of pathogenesis and virus-host interactions

    Quantifying and addressing the prevalence and bias of study designs in the environmental and social sciences

    Building trust in science and evidence-based decision-making depends heavily on the credibility of studies and their findings. Researchers employ many different study designs that vary in their risk of bias to evaluate the true effect of interventions or impacts. Here, we empirically quantify, on a large scale, the prevalence of different study designs and the magnitude of bias in their estimates. Randomised designs and controlled observational designs with pre-intervention sampling were used by just 23% of intervention studies in biodiversity conservation, and 36% of intervention studies in social science. We demonstrate, through pairwise within-study comparisons across 49 environmental datasets, that these types of designs usually give less biased estimates than simpler observational designs. We propose a model-based approach to combine study estimates that may suffer from different levels of study design bias, discuss the implications for evidence synthesis, and how to facilitate the use of more credible study designs.Fil: Christie, Alec P..     Connected Conservation: Rethinking conservation for a telecoupled world

    The convergence of the biodiversity and climate crises, widening of wealth inequality, and most recently the COVID-19 pandemic underscore the urgent need to mobilize change to secure sustainable futures. Centres of tropical biodiversity are a major focus of conservation efforts, delivered in predominantly site-level interventions often incorporating alternative-livelihood provision or poverty-alleviation components. Yet, a focus on site-level intervention is ill-equipped to address the disproportionate role of (often distant) wealth in biodiversity collapse. Further these approaches often attempt to ‘resolve’ local economic poverty in order to safeguard biodiversity in a seemingly virtuous act, potentially overlooking local communities as the living locus of solutions to the biodiversity crisis. We offer Connected Conservation: a dual-branched conservation model that commands novel actions to tackle distant wealth-related drivers of biodiversity decline, while enhancing site-level conservation to empower biodiversity stewards. We synthesize diverse literatures to outline the need for this shift in conservation practice. We identify three dominant negative flows arising in centres of wealth that disproportionately undermine biodiversity, and highlight the three key positive, though marginalized, flows that enhance biodiversity and exist within biocultural centres. Connected Conservation works to amplify the positive flows, and diminish the negative flows, and thereby orientates towards desired states with justice at the centre. We identify connected conservation actions that can be applied and replicated to address the telecoupled, wealth-related reality of biodiversity collapse while empowering contemporary biodiversity stewards. The approach calls for conservation to extend its collaborations across sectors in order to deliver to transformative change

    Dormancy within Staphylococcus epidermidis biofilms : a transcriptomic analysis by RNA-seq

    The proportion of dormant bacteria within Staphylococcus epidermidis biofilms may determine its inflammatory profile. Previously, we have shown that S. epidermidis biofilms with higher proportions of dormant bacteria have reduced activation of murine macrophages. RNA-sequencing was used to identify the major transcriptomic differences between S. epidermidis biofilms with different proportions of dormant bacteria. To accomplish this goal, we used an in vitro model where magnesium allowed modulation of the proportion of dormant bacteria within S. epidermidis biofilms. Significant differences were found in the expression of 147 genes. A detailed analysis of the results was performed based on direct and functional gene interactions. Biological processes among the differentially expressed genes were mainly related to oxidation-reduction processes and acetyl-CoA metabolic processes. Gene set enrichment revealed that the translation process is related to the proportion of dormant bacteria. Transcription of mRNAs involved in oxidation-reduction processes was associated with higher proportions of dormant bacteria within S. epidermidis biofilm. Moreover, the pH of the culture medium did not change after the addition of magnesium, and genes related to magnesium transport did not seem to impact entrance of bacterial cells into dormancy.The authors thank Stephen Lorry at Harvard Medical School for providing CLC Genomics software.     First report on dung beetles in intra-Amazonian savannahs in Roraima, Brazil

    This is the first study to address the dung beetle (Coleoptera: Scarabaeidae: Scarabaeinae) diversity in intra-Amazonian savannahs in the state of Roraima, Brazil. Our aim was to survey the dung beetle fauna associated with these savannahs (regionally called 'lavrado'), since little is known about the dung beetles from this environment. We conducted three field samples using pitfall traps baited with human dung in savannah areas near the city of Boa Vista during the rainy seasons of 1996, 1997, and 2008. We collected 383 individuals from ten species, wherein six have no previous record in intra-Amazonian savannahs. The most abundant species were Ontherus appendiculatus (Mannerheim, 1829), Canthidium aff. humerale (Germar, 1813), Dichotomius nisus (Olivier, 1789), and Pseudocanthon aff. xanthurus (Blanchard, 1846). We believe that knowing the dung beetles diversity associated with the intra-Amazonian savannahs is ideal for understanding the occurrence and distribution of these organisms in a highly threatened environment, it thus being the first step towards conservation strategy development
