    Intracellular \u3cem\u3eListeria monocytogenes\u3c/em\u3e Comprises a Minimal but Vital Fraction of the Intestinal Burden Following Foodborne Infection

    Listeria monocytogenes is a highly adaptive bacterium that replicates as a free-living saprophyte in the environment as well as a facultative intracellular pathogen that causes invasive foodborne infections. The intracellular life cycle of L. monocytogenes is considered to be its primary virulence determinant during mammalian infection; however, the proportion of L. monocytogenes that is intracellular in vivo has not been studied extensively. In this report, we demonstrate that the majority of wild-type (strain EGDe) and mouse-adapted (InlAm-expressing) L. monocytogenes recovered from the mesenteric lymph nodes (MLN) was extracellular within the first few days after foodborne infection. In addition, significantly lower burdens of L. monocytogenes were recovered from the colon, spleen, and liver of gentamicin-treated mice than of control mice. This led us to investigate whether intracellular replication of L. monocytogenes was essential during the intestinal phase of infection. We found that lipoate protein ligase-deficient L. monocytogenes (ΔlplA1) mutants, which display impaired intracellular growth, were able to colonize the colon but did not persist efficiently and had a significant defect in spreading to the MLN, spleen, and liver. Together, these data indicate that the majority of the L. monocytogenes burden in the gastrointestinal tract is extracellular, but the small proportion of intracellular L. monocytogenes is essential for dissemination to the MLN and systemic organs

    Leavers and Remainers After Brexit: More United Than Divided After All?

    Since the British “Brexit referendum” in 2016, tensions between ‘leave’ and ‘remain’ voters have been growing. Using a novel analytical approach based on the full distribution of responses rather than their arithmetic means, Study 1 (N=1,506) showed on average 90% of overlap among Leavers and Remainers across a range of important variables. Even on the variables that are commonly used to illustrate how Leavers and Remainers differ (e.g., prejudice against immigrants), the amount of overlap was larger than 50%. In Study 2 (N=206), we demonstrate the importance of focusing on similarities between groups: Presenting the full distribution of responses from Leavers and Remainers rather than their mean differences caused more accurate perceptions of the actual differences and similarities between both groups and higher perceived intergroup harmony. We conclude that similarities between Leavers and Remainers are substantial and that our proposed approach may help to de-escalate tensions between these and other groups

    COVID‐19, nationalism, and the politics of crisis: A scholarly exchange

    In this article, several scholars of nationalism discuss the potential for the COVID‐19 pandemic to impact the development of nationalism and world politics. To structure the discussion, the contributors respond to three questions: (1) how should we understand the relationship between nationalism and COVID‐19; (2) will COVID‐19 fuel ethnic and nationalist conflict; and (3) will COVID‐19 reinforce or erode the nation‐state in the long run? The contributors formulated their responses to these questions near to the outset of the pandemic, amid intense uncertainty. This made it acutely difficult, if not impossible, to make predictions. Nevertheless, it was felt that a historically and theoretically informed discussion would shed light on the types of political processes that could be triggered by the COVID‐19 pandemic. In doing so, the aim is to help orient researchers and policy‐makers as they grapple with what has rapidly become the most urgent issue of our times

    Locating mathematics within post-16 vocational education in England

    The political importance of mathematics in post-16 education is clear. Far less clear is how mathematics does and should relate to vocational education. Successive mathematics curricula (e.g. core skills, key skills) have been developed in England with vocational learners in mind. Meanwhile, general mathematics qualifications remain largely disconnected from vocational learning. Following a brief historical survey of mathematics within vocational education, the paper presents findings from a nested case study of student groups in three large Further Education colleges in England. The primary unit of analysis herein is student groups learning Functional Mathematics in two vocational areas: construction and hairdressing. We show how approaches to organising teaching, developing connected curricula and classroom pedagogy tend to isolate or integrate mathematics from/with the vocational experience. Integrated approaches are shown to impact positively on student engagement and attitudes to learning mathematics. The paper concludes by discussing the potential impact of academic qualifications displacing vocationally relevant mathematics

    ‘Growing your own’: a multi-level modelling approach to understanding personal food growing trends and motivations in Europe

    Growing food for personal and family consumption is a significant global activity, but one that has received insufficient academic attention, particularly in developed countries. This paper uses data from the European Quality of Life Survey (EQLS) to address three areas of particular concern: the prevalence of growing your own food and how this has changed over time; the individual and household context in which growing takes place; and whether those who grow their own food are happier than those who do not. Results showed that there was a marked increase in growing your own food in Europe, in the period 2003–2007. This increase is largely associated with poorer households and thus, possibly, economic hardship. In the UK however the increase in growing your own food is predominantly associated with older middle class households. Across Europe, whether causal or not, those who grew their own were happier than those who did not. The paper therefore concludes that claims about the gentrification of growing your own may be premature. Despite contrary evidence from the UK, the dominant motive across Europe appears to be primarily economic — to reduce household expenditure whilst ensuring a supply of fresh food

    Social cognition differentiates phenocopy syndrome of behavioural variant frontotemporal dementia from behavioural variant frontotemporal dementia

    Background: Patients displaying clinical features of behavioural variant of frontotemporal dementia (bvFTD) but lacking both neuroimaging abnormalities and clinical progression are considered to represent the phenocopy syndrome of bvFTD (phFTD). Extensive clinical overlap between early phase bvFTD and phFTD hampers diagnostic distinction. We aimed to assess the diagnostic value of clinician-rated, self-reported and caregiver-reported symptoms for clinical distinction between phFTD and bvFTD. Methods: There were 33 phFTD and 95 probable bvFTD patients included in the study (total N = 128). Clinician-rated, self-reported tests and caregiver-reported symptoms were compared between phFTD and bvFTD on social cognition, behaviour, mood and activities of daily living (ADL). Scores were compared between groups, followed by multiple logistic regression analysis, adjusted for age and sex. Receiver operating characteristic curves were plotted to assess diagnostic value. Results: Using clinician-rated and self-reported tests, phFTD patients performed better on facial emotion recognition and reported more depressive symptoms. Caregiver-reported behavioural symptoms indicated higher behavioural and ADL impairment in phFTD compared to bvFTD. Facial emotion recognition provided highest diagnostic accuracy for distinction of phFTD from bvFTD (area under the curve (AUC) 0.813 95% CI 0.735–0.892, P < 0.001, sensitivity 81%, specificity 74%) followed by depressive symptoms (AUC 0.769 95% 0.674–0.864, P < 0.001 sensitivity 81%, specificity of 63%). Conclusion: Social cognition tests are most suitable for distinction of phFTD from bvFTD. Caregiver-reported questionnaires and phFTD diagnosis seemed inversely correlated, showing more symptoms in phFTD. Further research is needed on phFTD aetiology and in caregivers taking into account disease burden to assess what explains this discrepancy between clinician-rated and caregiver-based tools

    The pursuit for markers of disease progression in behavioral variant frontotemporal dementia: a scoping review to optimize outcome measures for clinical trials

    Behavioral variant frontotemporal dementia (bvFTD) is a neurodegenerative disorder characterized by diverse and prominent changes in behavior and personality. One of the greatest challenges in bvFTD is to capture, measure and predict its disease progression, due to clinical, pathological and genetic heterogeneity. Availability of reliable outcome measures is pivotal for future clinical trials and disease monitoring. Detection of change should be objective, clinically meaningful and easily assessed, preferably associated with a biological process. The purpose of this scoping review is to examine the status of longitudinal studies in bvFTD, evaluate current assessment tools and propose potential progression markers. A systematic literature search (in PubMed and Embase.com) was performed. Literature on disease trajectories and longitudinal validity of frequently-used measures was organized in five domains: global functioning, behavior, (social) cognition, neuroimaging and fluid biomarkers. Evaluating current longitudinal data, we propose an adaptive battery, combining a set of sensitive clinical, neuroimaging and fluid markers, adjusted for genetic and sporadic variants, for adequate detection of disease progression in bvFTD

    Q methodology and rural research

    Traditionally, rural scholarship has been limited in its methodological approach. This has begun to change in recent years as rural researchers have embraced a range of different methodological tools. The aim of this article is to contribute to greater methodological pluralism in rural sociology by introducing readers to a method of research that is rarely engaged in the field, that is, Q methodology. The article describes the defining features of the approach as well as providing examples of its application to argue that it is a method that offers particular opportunities and synergies for rural social science research

    British trade unions and the academics: the case of Unionlearn

    Unionlearn and union learning representatives were developed by the British TUC to match workers with education and training opportunities, strengthen the economy, foster market inclusion and facilitate social mobility. Their contribution to union revitalisation was emphasised. This article questions whether, with unions confronting global crisis, this is a necessary initiative. It stemmed from TUC failure to achieve policy goals, institutional needs, consequent acceptance of a lesser role, and the availability of state finance. Claims by academics that it provides influence over state policy and contributes to revitalisation remain inadequately evidenced. Union resurgence is not immanent. The way forward is through adversarial grassroots organising and socialist education, not through retooling capital, improving members’ marketability and partnership with a hostile state
